Reproduction and molt are costly processes in avian life histories. These two fitness-related traits are expected to be under one of physiological trade-offs. Age-related molt is known to be higher in young birds than that in adults presumably due to the cost of reproduction in adults. The present study partially replicated a previous study using a non-invasive method of seasonal wing feather loss instead of capture-inspection for molting progress in oriental storks (Ciconia boyciana). We first examined characteristics of the known six wing feather types (i.e., primaries [P], primary coverts [PC], secondaries [S], secondary coverts [SC], and tertials [T]) from two specimens with four wings. Results were utilized as references for further investigation. We then collected a total of 3,807 wing feathers shedded by 61 captive storks for one year and classified them into six wing feather types based on the reference with structures of vane (i.e., how asymmetrical) and calamus (i.e., how rigidly attached to skin) of wing feathers. Our results indicated that annual losses of all six-type wing feathers decreased with increasing ages, ranging from 29% to 58% for PC, alula, SC, P, S, and T in order. Our results were also comparable to those of a former study, suggesting that the pattern of age-specific molt might be associated with the cost of reproduction in adults. However, juveniles might shed more wing feathers with low quality formed during the previous development stage than older birds.
This study aimed at evaluating the effects of a molting diet method in molt induction and post-molt performance of laying hens. Eighty-one ISA Brown hens at 62 wk of age were randomly divided into three groups. After a 4-wk preliminary period, a control group was fed a corn-soybean-based layer diet, and for the other groups, molting was induced by starvation (MS) or feeding a molting diet (MD). For the MS group, feed was withdrawn for 2 wk; this was followed by feeding a layer diet every other day for 1 wk and then the control diet. The MD group was fed a molting diet containing low-protein and low-energy diet based for 4 wk; this was followed by feeding a layer diet. They had a free access to their diet and water. Egg production, egg quality, feed intake, and ovary and oviduct weights were measured throughout the experimental period. During molting, the feed intake in the MD group was lower than that of the control. Body weight of the molted groups was significantly reduced. The MS group feeding totally ceased egg production within 4d; after the initiation of feeding and decreased; in the MD group, egg production to 9.3% by d 10. On d 14, the ovaries and oviducts of the molted groups were distinctly lighter than those of the control. Throughout the post-molt period, egg production and egg shell thickness of the molted group improved; but there were no significant differences. Eggs from the MD-fed or control group were heavier than those of the MS-fed or control group. Finally, feeding of a low-protein and low-energy diet effectively induces molting and increase post-molt production, but further research will be conducted to determine the effects of the molt diet with other ingredients and to reduce the energy level of the molt diet for maximizing molt induction and post-molt egg quality.
The larvae of Palaemon gravieri were reared in the laboratory at three different temperature regimes ($15^{\circ}C,\;20^{\circ}C,\;and\;25^{\circ}C$) with the salinity ranges (28-32 psu) to understand how temperature and body size influence survival rate, and growth components (molt increment and intermolt period). The optimum temperature for the highest survival rate was $25^{\circ}C$. The intermolt periods consistently increased with an increase in size and instar number; however, the molt increments at successive instars generally decreased with an increase in size and instar number. The shortest intermolt period and the highest larval growth rate both occurred at $25^{\circ}C$. Thus, the optimum temperature for larval survival and growth rate was found to be $25^{\circ}C$ which was the temperature at which the larvae actually appear in nature.
Background: Hepatitis C virus(HCV), a family of Flaviviridae, has a host cell-derived envelope containing a positive-stranded RNA genome, and has been known as the maj or etiological agent for chronic hepatitis, hepatic cirrhosis, and hepatocellular carcinoma. There remains a need to dissect a molecular mechanism of pathogenesis for the development of therapeutic and effective preventive measure for HCV. Identification of cellular receptor is of central importance not only to understand the viral pathogenesis, but also to exploit strategies for prevention of HCV. This study was aimed at identifying peptide mimotopes inhibiting the binding of E2 protein of HCV to MOLT-4 cell. Methods: In this study, phage peptide library displaying a random peptides consisting of 7 or 12 random peptides was employed in order to pan against E2 protein. Free HCV particles were separated from the immune complex forms by immunoprecipitation using anti-human IgG antibody, and used for HCV-capture ELISA. To identify the peptides inhibiting E2-binding to MOLT-4 cells, E2 protein was subj ect to bind to MOLT-4 cells under the competition with phage peptides. Results: Several phage peptides were selected for their specific binding to E2 protein, which showed the conserved sequence of SHFWRAP from 3 different peptide sequences. They were also able to recognize the HCV particles in the sera of HCV patients captured by monoclonal antibody against E2 protein. Two of them, showing peptide sequence of HLGPWMSHWFQR and WAPPLERSSLFY respectively, were revealed to inhibit the binding of E2 protein to MOLT-4 cell efficiently in dose dependent mode. However, few membrane-associated receptor candidates were seen using Fasta3 programe for homology search with these peptides. Conclusion: Phage peptides containing HLGPWMSHWFQR and WAPPLERSSLFY respectively, showed the inhibition of E2-binding to MOLT-4 cells. However, they did not reveal any homologues to cellular receptors from GenBank database. In further study, cellular receptor could be identified through the screening of cDNA library from MOLT-4 or hepatocytes using antibodies against these peptide mimotopes.
This study has been conducted to investigate the molting behavior and the effects of rearing temperature and day length on molting in the behavior and the effects of rearing temperature and day length on molting in the giant freshwater prawn, Macrobrachium rosenbergii reared in the laboratory. The results obtained were summarized as follow: 1. After pre-spawning molting, the protopodites of 1st, 2nd, 3rd, and 4th except 5th pleopod bore new breeding setae which conserve eggs in the brooding chamber and the basis of 3rd, 4th, and 5th pereiopods bore new breeding dresses which transport the ovulated eggs into the brooding chamber. 2. Adult females reared in 27.5-$29.4^{\circ}C$ molted 10-12 times per year at interval of 27-35 days, of which four or six moltings were common molting for growth and another four or five moltings were pre-spawning molting for spwaning and brooding. In winter season, pre-spawning molting did not happen to most of adult females in spite of the same temperature. 3. Duration of intermolt cycle was 31-38 days and 26-30 days at 25.3- $26.5^{\circ}C$ and 28.7- $30.4^{\circ}C$ of rearing temperature, respectively.
Homogeneous human deoxycytidine kinase was purified in one step from a variety of spontaneous human leukemic cells (T-ALL, B-ALL, B-CLL, AML, CML), and from cultured T-lymphoblast cells (MOLT-4) using the newly developed affinity medium, $dCp_4$-Sepharose. Starting with an ammonium sulfate fraction, purification was achieved in one step with the kinase being eluted from a column by the end product inhibitor, dCTP. The purified deoxycytidine kinase from T-ALL cells phosphorylated deoxyadenosine and deoxyguanosine, as well as deoxycytidine. The enzyme purified from T-ALL and B-CLL cells yielded one major band with a molecular weight of 52 kDa determined by SDS-polyacrylamide gel electrophoresis. AML and CML cells yielded one 52 kDa band and an extra band of 30 kDa molecular weight. On the other hand, B-ALL and MOLT-4 cells showed a low molecular weight band of 30 kDa only. However, the electrophoretic mobilities of enzymatic activity in 12% non-denaturing gels were identical for the dCyd kinase from all different kinds of leukemic cell lines, except that the B-ALL, B-CLL, and MOLT-4 cell preparations had an extra minor peak, all at the same position. dAdo and dCyd phosphorylating activities comigrated indicating that these activities are all associated with the same protein. Two new methods, a disk implantation method and a nitrocellulose powder method were used with a small amount of enzyme protein to raise polyclonal antibodies against dCyd kinase purified from T-ALL cells.

In the present investigation, a series of some novel 1,3-diaryl/heteroarylprop-2-en-1-one derivatives (3a-j) have been synthesized and evaluated for their in vitro cytotoxicity against three cancer cell lines, two hepatocarcinoma cell lines HUH-7, Hep-3b and one leukemia cancer cell line MOLT-4. Based on these results, structure-activity relationship (SAR) was studied on modification of $R^1$ and $R^2$ to identify the compound with maximum potency. Amongst the compounds, 3b and 3d strongly inhibited the growth of Hep-3b and MOLT-4 cells with $IC_{50}$ value of 3.39 and $3.63{\mu}M$ respectively. The results obtained from the inhibitory study had further been supported by the reactive oxygen species (ROS) measurement using flow cytometry in MOLT-4 cells. These observations collectively reveal that compounds comprising 1,3-diarylprop-2-en-1-one framework with pyrazole ring at position-3 and heteroaryl/aryl substituents at position-1 can be used as promising anticancer agents.
Larvae of Palaemon serrifer were reared in the laboratory under three different temperature regimes ($15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$) to study the effects of rearing temperature on larval survival and growth, as well as other traits such as embryo volume, number of embryos (fecundity), incubation period, development. Mode and development period. Growth pattern was analyzed by measuring the molt increment and intermolt period. The intermolt period consistently increased with size and instar number and was shortest at $25^{\circ}C$. However, molt increments generally decreased with instar number. Number of embryos varied from 552 to 1355. The relationship between the number of embryos and carapace length was expressed by the equation (fecundity) y=2.7744x+0.208 ($R^2$=0.7961). Egg volume was a primary factor affecting other life-history traits. Egg volume was $0.078\;m^3$, which is relatively small thus embryos exhibited a relatively short incubation period and a comparatively short development period, and the nutritional mode was planktotrophic. Brood production was followed by a fast parturitional pattern. Most ovigerous females had mature ovaries when the parturial molt occurred soon after eclosion.

Younggaechulgam-tang has been used for treating skin diseases. In this study, I investigated the immunosuppressive effect of Younggaechul-tang in the human T cell line MOLT-4 cells. MOLT-4 cells were stimulated with the phytohemagglutinin (PHA) and phorbol 12-myristate 13-acetate (PMA) + A23187. The secretion appeared to be greater when cells were stimulated with PHA than with PMA + A23187. Younggaechulgam-tang had no affect proliferation stimulated by PHA. I showed that IL-2 secretion and expression by PHA stimulated MOLT-4 cells were inhibited by Younggaechugam-tang treatment. Maximal inhibition rate of IL-2, TNF-${\alpha}$ secretion was 80$\%$ and 30$\%$, respectively. Younggaechulgam-tang also inhibited nuclear translocation of p65 subunit of nuclear factor-kB and nuclear factor of activated T cells (NFAT). In conclusion, these results suggest that Younggaechulgam-tang may contribute to the immunosuppressive oriental drug clinically.
Larvae of the freshwater shrimp, Macrobrachium nipponense(De Haan) were reared in the laboratory under constant conditions ($25^{\circ}C,\;7\%0$ S), and their feeding rate, oxygen consumption, ammonia nitrogen excretion, and growth were measured at regular intervals during development from hatching to post larval stage. Growth was measured as dry weight, carbon, nitrogen, hydrogen, protein and lipid. All these physiological and biochemical traits revealed significant changes from instar to instar. Average feeding rate was high in intermolt stage of the molt cycle and it showed a bell-shaped pattern. Respiration(R) increased from hatching to post larval stage. Excretion(U) increased in intermolt phase of larvae and it showed a bell-shaped variation pattern, in all larval instars with a maximum near the middle of the molt cycle. Regression equations describing rates of feeding, growth, respiration and ammonia excretion as functions of time during individual larval molt cycles were inserted in a simulation model, in order to analyse time-dependent patterns of variation as well as in bioenergetic efficiencies. Carbon was initially increased and nitrogen showed a tendency to increase in premolt phase during individual molt cycles. Protein remained clearly the predominant biochemical constituent in larval biomass.
