In order to screen new allogeneic mixed lymphocyte culture (allo-MLR) inhibitor which is expected to be immunomodulating drug lead, we have investigated allo-MLR inhibitory activity on the marine-derived symbiotic microorganisms (1,895 strains) from the marine algae. The potent inhibitory activities (over 45% inhibition of proliferation at 10 and 2 ${\mu}g/ml$) without cytotoxicity were observed in the extracts of 46 strains. While, the significant stimulating activities (over 100% proliferation at 10 and 2 ${\mu}g/ml$) without cytotoxicity were observed in the extracts of 5 strains. In the second assay using 46 bioactive strains, 14 strains exhibited again significant allo-MLR inhibitory activity. Finally, 11 strains among the 14 strains inhibited proliferation and IFN-${\gamma}$ production of CD4+ T cells during the stimulation with specific antigen in the third assay. On the basis of above results, the marine algae is nice source for isolation of immunomodulating microorganism, and the marine algae-associated microorganism is also nice target for development of the new immunomodulating drug lead.

Marine pathogenic bacteria, such as Streptococcus parauberis, Edwardsiella tarda and Vibrio harveyi, can cause lethal infections in farmed fish, ozone and antibiotics, are employed to sterilize waters used for rearing fish to mitigate this threat. The most widely used method is treatment with sodium hypochlorite solution. However, the maintenance of a constant concentration of chlorine in rearing waters can be difficult. We investigated the potential of a mixed oxidant (MO) solution generated by electrolysis of sea water to improve water quality. We measured the survival rates of fish pathogenic bacteria exposed to different concentrations of MO (0.5, 1.0, 1.5 and 2.0 MO) and sodium hypochlorite (0.5, 1.0, 1.5 and 2.0 ppm) for various lengths of time (0, 5, 10, 15, 20, 25 and 30 min). We found a time-dependent decrease in the survival rates of the tested pathogenic microorganisms. The sterilization effect of the MO solution on pathogenic organisms was greater than that of sodium hypochlorite for gram-negative and gram-positive bacteria. We conclude that MO solution produced by electrolysis could be used to maintain a constant chlorine concentration in aquaculture systems.
Objective: This experiment was conducted to compare the structure and composition of ruminal microorganisms in goats with high and low neutral detergent fibre (NDF) digestibility. Methods: Nineteen crossbred goats were used as experimental animals and fed the same total mixed rations during the 30-day pre-treatment and 6-day digestion trialperiods. All faeces were collected during the digestion period for measuring the NDF digestibility. Then, high and the low NDF digestibility individuals were chosen for the high NDF digestibility group (HFD) and low NDF digestibility group (LFD), respectively. Rumen contents were collected for total microbial DNA extraction. The V4 region of the bacterial 16S rRNA gene was amplified using universal primers of bacteria and sequenced using high-throughput sequencer. The sequences were mainly analysed by QIIME 1.8.0. Results: A total of 18,694 operational taxonomic units were obtained, within 81.98% belonged to bacteria, 6.64% belonged to archaea and 11.38% was unassigned microorganisms. Bacteroidetes, Firmicutes, and Proteobacteria were the predominant microbial phyla in both groups. At the genus level, the relative abundance of fifteen microorganisms were significantly higher (p<0.05) and six microorganisms were extremely significantly higher (p<0.01) in LFD than HFD. Overall, 176 core shared genera were identified in the two groups. The relative abundance of 2 phyla, 5 classes, 10 orders, 13 families and 15 genera had a negative correlation with NDF digestibility, but only the relative abundance of Pyramidobacter had a positive correlation with NDF digestibility. Conclusion: There were substantial differences in NDF digestibility among the individual goats, and the NDF digestibility had significant correlation with the relative abundance of some ruminal microorganisms.

Bacterio-mineral water (BMW) produced from manure has been known to exert a number of positive effects on animal production and odor control. An experiment was conducted to examine the effects of BMW produced from bio-reacted swine manure on in vitro gas production, cellulose degradation, microbial growth and fibrolytic enzyme activities of mixed rumen microorganisms. The five levels of 0, 0.001, 0.005, 0.01 and 1.0% BMW were supplemented into serum vials containing mixed rumen microorganisms. Incubations were carried out anaerobically at $39^{\circ}C$ without shaking for 0, 12, 24, 48, 72 and 96 h. There were no significant (p>0.05) differences among the treatments for the initial rate of gas production. At 72 h incubation, the gas production tended (p<0.1) to be increased by the 0.01 and 1.0% BMW treatments compared with control and the 0.001% BMW treatment. At the end of incubation (96 h), the sample supplemented with 0.01% BMW was higher (p<0.05) than control (0% BMW) in the gas production. The microbial growth rate was increased by all the BMW treatments, while 0.01% BMW was most effective in stimulating the growth rate. Although the addition of BMW on the filter paper DM degradation was not significantly influenced throughout the incubation period except the 48 h incubation, DM degradation tended to be increased by all BMW treatments compared with control. The addition of both 0.005 and 0.01% BMW highly increased (p<0.05) CMCase activity compared with control after 24 h and 48 h incubation, while at the 72 h incubation the 0.01% BMW addition only significantly increased (p<0.05). After 72 h incubation, the xylanase activity was significantly (p<0.05) increased with the addition of 1.0% BMW compared with the addition of 0.001 and 0.005% BMW, while at the other incubation times, the xylanase activity was not different among the treatments. In conclusion, the 0.01% BMW of supplementation level would be the suitable addition level to stimulate rumen fermentation increasing microbial growth and cellulose degradation.
High substrate costs decrease the profitability of polyhydroxyalkanoates (PHAs) production, and thus low-cost carbon substrates coming from agricultural and industrial residuals are tested for the production of these biopolymers. Among them, crude glycerol, formed as a by-product during biodiesel production, seems to be the most promising source of carbon. The object of this study was to characterize the mixed population responsible for the conversion of crude glycerol into PHAs by cultivation-dependent and -independent methods. Enrichment of the microbial community was monitored by applying the Ribosomal Intergenic Spacer Analysis (RISA), and the identification of community members was based on 16S rRNA gene sequencing of cultivable species. Molecular analysis revealed that mixed populations consisted of microorganisms affiliated with four bacterial lineages: ${\alpha}$, ${\gamma}$-Proteobacteria, Actinobacteria, and Bacteroides. Among these, three Pseudomonas strains and Rhodobacter sp. possessed genes coding for polyhydroxyalkanoates synthase. Comparative analysis revealed that most of the microorganisms detected by direct molecular analysis were obtained by the traditional culturing method.
To explore the effective treatment methods for waste food generating malodor compounds, mixed microorganisms (MM) capable of deodorizing of domestic animal waste were applied using household composting box. The mixture of 5 kg whole chips as a bulk agent, 2 kg MM and 1 kg of waste food were input into the compost reactor and agitated. Waste food was supplemented every 24 hours. As the results, the composting volume was stable at 13∼14 L for 10 days. In the initial compost process with MM, the pH and temperature were increased more quickly than that of without MM. Also, the conductivity recognized as a barometer of compost was increased from 0.2 to 2.4 mS/cm that was higher than 1.3 mS/cm of without MM, for 10 days. The malodor compounds generated from waste food treatment such as sulfur compounds and volatile fatty acids were effectively reduced about 90∼l00%, and 70∼80% for 8 days, respectively. The microorganisms growing under the condition of alkaline phase and higher temperature were dominated during the compost Moreover, it was demonstrated that inoculated Bacillus cereus HY15 dominated during the compost results in responsible to the effective treatment of waste food.
Several microorganisms were isolated and characterized for the development of fermentation accelerator of animal manure. Firstly, 61 species were isolated from rice bran extract. Secondly, five strains of microorganisms were screened by the analysis of hydrolysis activities for organic compounds including protease, cellulase, amylase, and lipase. From a deodorization test for ammonia gas using the isolated strains, finally three bacterial strains were selected (NA 2, 12, 15). The selected strains, NA 2 and 15 were identified as Bacillus acidocaldarius and Planococcus sp. respectively. The media composition of key nutrients and pH for the mixed culture of the three selected strains were optimized using an experimental design method (response surface method) as follows : beef extract (4.59g/L), peptone (8.72g/L) and pH 6.3. Consequently, the isolated microorganisms seem to have potential applicability in the animal manure treatment.

To test the potential effects of extracellular electron shuttles (EES) on the rate and extent of heavy metal release from contaminated soils during microbial iron reduction, we created anaerobic batch systems with anthraquinone-2,6-disulfonate (AQDS) as a surrogate of EES, and with contaminated soils as mixed iron (hydr)oxides and microbial sources. Two types of soils were tested: Zn-contaminated soil A and As/Pb-contaminated soil B. In soil A, the rate of iron reduction was fastest in the presence of AQDS and > 3500 mg/L of total Fe(II) was produced within 2 d. This suggests that indigenous microorganisms can utilize AQDS as EES to stimulate iron reduction. In the incubations with soil B, the rate and extent of iron reduction did not increase in the presence of AQDS likely because of the low pH (< 5.5). In addition, less than 2000 mg/L of total Fe(II) was produced in soil B within 52 d suggesting that iron reduction by subsurface microorganisms in soil B was not as effective as that in soil A. Relatively high amount of As (~500 mg/L) was released to the aqueous phase during microbial iron reduction in soil B. The release of As might be due to the reduction of As-associated iron (hydr)oxides and/or direct enzymatic reduction of As(V) to As(III) by As-reducing microorganisms. However, given that Pb in liquid phase was < 0.3 mg/L for the entire experiment, the microbial reduction As(V) to As(III) by As-reducing microorganisms has most likely occurred in this system. This study suggests that heavy metal release from contaminated soils can be strongly controlled by subsurface microorganisms, soil pH, presence of EES, and/or nature of heavy metals.
Laboratory-scale investigation into initial anaerobic biofilm development was carried out by circulating mixed liquor from a steady-state anaerobic reactor through silicone tubing and then rerurning the mixed liquor to the reactor. The wall of the silicone tubing was the surface upon which anaerobic biofilm accumulation or development was monitored. Methanogenic bacteria accumulation was monitored by F$_{420}$ fluorescence (picomoles F$_{420}$/cm$^{2}$) of the extracted biofilm material. Biofilm accumulation was measured by the increase in COD of the extracted material ($\mu $g COD/cm$^{2}$). Experiments were conducted for 25 days, and biofilm analyses were performed at 5 days intervals. The results indicated that the initial rates of methangen and anaerobic biofilm accumulation increased with increasing organic loading rate and higher initial rates were observed for 15 days than 15 day liquid HRT or SRT. When the initial rates were plotted against the corresponding mixed liquor volatile suspended solids the difference between the results at the two HRT's became much less significant. Thus, the concentration of mixed liquor volatile suspended solids was found to be a very important parameter affecting initial anaerobic biofilm development. The ratio of methanogens to anaerobic biofilm was also investigated. The results showed that the ratio remained constant through the 25 days of each experiment and for high organic loading rates. Based on the results of this research, a reduction, a reduction of start-up period of anaerobic fixed film reactors might be achieved by maintaining a high organic loading and a large concentration of anaerobic microorganisms in the mixed liquor during the start-up period.
Purpose: To evaluate correlation between the clinical results and causative bacteria in diabetic foot patients with lower extremity amputation. Materials and Methods: One hundred twenty nine patients(131 feet) of diabetic foot amputations were followed for more than one year. Wound cultures were done by deep tissue or bone debris at first visit to our clinics. Retrospective analysis was performed using chart review and interview with the patients. Depending on the culture result, level of amputation, reinfection, duration of treatment, death rate, patient satisfaction and admission dates were evaluated. Results: Microorganisms were confirmed in 114 cases. In the other 17 cases, there were no cultured microorganisms. In bacterial growth group, Methicillin-sensitive Staphylococcus aureus was the most common pathogen and accounted for 34 cases. As other common pathogens, there were Methicillin-resistant Staphylococcus aureus(24 cases) and mixed infection(14 cases). Mortality is no difference in each infected group. Mixed bacterial infected patients have higher reinfection, longer hospital day and duration of treatment, but there is no difference in patients satisfaction and pain at last follow up. Conclusion: The most common pathogen in diabetic foot patients with lower extremity amputation was Methicillin-sensitive Staphylococcus aureus, and mixed bacterial infected patients have higher reinfection rate, longer admission date and duration of treatment than other bacterial infected patients.
