Yield characteristics and medium composition of winter mushroom in cultivation farms in Korea (국내 팽이버섯 재배농가의 배지조성 및 수량 특성)

  • This study was conducted to provide basic data for setting a standard medium for winter mushroom cultivation. Investigation of medium composition in winter mushroom farms in Korea revealed that the types of medium used for each farm were slightly different and that the mixing ratio of the medium also varied. All farmers used corncob, rice bran, and beet pulp as the main media, and calcium carbonate and ground oyster shells were used at 1.0~3.8% to adjust the pH of the medium. Analysis of the physicochemical properties of the mixed media showed nitrogen content of 1.28~1.52%, carbon content of 45.7~48.5%, and C/N ratio of 30.7~37.9. The content of inorganic components was significantly different in each farm, depending on the type and amount of minerals in the mixed medium. Mycelial growth was the fastest at Farm 2, reaching 6.5 cm within 34 days of culture, followed by Farm 4 at 5.7 cm, whereas Farm 1, 3, and 5 showed similar growth. Mycelial density was high without any significant difference between farms. Survey of yield according to medium composition in each farm showed the highest yield in Farm 2 at 173.4 g/bottle, followed by Farm 1 at 168.4 g/bottle, whereas Farm 3 and 4 showed similar yield. However, Farm 4 had the lowest yield at 145.4 g/bottle. Therefore, mushroom yield was different depending on the medium composition in the farm, and thus selection of a medium that can be used universally in multiple mushroom farms was considered possible.

  • This report is the result of devising a method for utilizing the device of the load cell to maintain a constant water content of the medium every day to prepare a cultural substrates with the mixer for growing mushrooms bottle cultivation. A load cell was device under the medium mixer. It is developed when the device reaches the weight calculated as amount of substrate bottled and number of the bottle, it is automatically terminated by water injection. In addition, measuring the water content of each medium and the total weight of the medium reaches the target moisture content were calculated by using the program Cheong et al. (2015). Enter the total weight of the medium on the display unit of the load cell, when starting the water supply to reach the weight-based mixing media, the water supply is stopped. This method can improve the convenience by reducing the user's trouble in repeated work medium prepared by automating water supply. The suitable moisture content of the mixed medium for some kind of mushroom can be improved by the composition accuracy. And mycelial culture period, primordial period, mushroom growing period is maintained even of the medium can be produced stably. Therefore, it is possible to achieve a stable management of the mushroom farm according to mushroom quality and quantity stable throughout the year.

Characteristics of Crude Polysaccharide Separated from the Herbal Medium of Trichloloma Matsutake Mycelium and its Anti-diabetic Effect

  • As part of studies to develop new materials to lower blood glucose levels using crude polysaccharide, this study was attempted to analyze the characteristics of crude polysaccharide obtained from the extracts of a mixed herbal medium(OCM) where Trichloloma matsutake mycelium and Cordyceps militaris mycelium were cultured together and to look into the influence of administering these by concentration upon the blood glucose and serum lipid levels of rats with diabetes which was induced by STZ(Streptozotosin). Experimental group was divided into 6 groups: first, it was divided into normal control group(NC group) and diabetes-induced group, and diabetes-induced group was subdivided into diabetic control group(DC group), acarbose-treated group(PC group), 100 mg/kg/body weight-treated by crude polysaccharide of OCM(UE) group(UE100 group), 200 mg/kg/body weight-treated group(UE200 group), and 300 mg/kg/body weight-treated group(UE300 group). In diabetic-induced groups, after streptozotocin was melted in 0.01M citrate buffer at 50 mg/kg/body weight, when the non-fasting blood glucose level not on an empty stomach was 300 mg/dl or more in blood collected from the tail vein, it was regarded as diabetic induction and then such diabetic-induced experimental animals were used in this experiment. The yield of crude polysaccharide obtained from OCM was found to be 0.31% and the ${\beta}$-glucan content 39.40%. As a result of analyzing NO on immune function, which is known as major physiological activity of crude polysaccharide, high NO viability was shown; when 1 mg/ml LPS was treated at 1 ug/ml, it was found to be 50.77 uM, and when LPS was treated at 10 ug/m, it was found to be 53.78 uM. Also, regarding cancer cells, cell count was decreased by about 26% in proportion to sample concentration, while for normal cells, it was a little decreased in proportion to concentration, however, cell count was maintained in the range of $81.92{\sim}98.16%$ at all concentrations. In case of blood glucose level, it was decreased in all extract-treated groups compared to DC group and in the cases of ALT and AST, they were found to be lower in extract-treated groups compared to PC group and for serum lipid, it was found to be lower in UE100 group compared to PC group. Thus this study tried to utilize these results as fundamental data for development of preventive and therapeutic agents against diabetes as well as functional foods using the crude polysaccharide of mushrooms.

Comparative study on the external micro-morphology of 3 kinds of minute pollen and spore Herbs (Pini Pollen, Typhae Pollen, Lygodii Spora) utilizing scanning electron microscope (주사전자현미경을 이용한 3종 화분포자류(花粉胞子類) 한약재(송화분(松花粉), 포황(蒲黃), 해금사(海金沙))의 미세형태 비교연구)

  • Objectives : We tried to establish standards for genuine by discriminating 3 kinds of pollen and spore herbs that are highly to be mixed with each other. Using an scanning electron microscope, we prepare a standard for discriminating external morphological characters of minute herbs, thereby contributing to the stable supply of genuine herbal medicines. Methods : Standard samples were confirmed by literature review on external morphological characteristics of original plants and herbal medicines, and collection and identification of original plants. The herbal medicines on the market were purchased and classified with using naked eye observation and magnifier. Finally, micromorphological identifications were conducted using an scanning electron microscope. Results : 1. Pini Pollen was clearly distinguished by its relatively medium size and a pollen grain with two swollen reticulate sacci at both ends. The verrucate ornamentation on the exine surface of the corpus and a sunken leptoma germ pore may be used as a discrimination criteria. 2. Typhae Pollen was distinguished by its relatively small size and a saccus on the end of a pollen grain. Reticulate ornamentation of exine surface of the corpus, and a slightly clear ulcerate germ pore can be used as a discrimination criteria. 3. Lygodii Spora was distinguished by its relatively large triangular-ovate shape and trichotomous fissure. Verrucate-tuberculate ornamentation of exine surface and trilete aperture could be used as a discrimination criteria. Conclusion : These results indicate that the use of electron microscopy is very effective for discriminating the external morphology of minute herbal medicines.

Growth Responses of Potted Gerbera 'Sunny Lemon' under Non-Nutrient Solution Recycling System by Media and Nutrient Contents (비순환식 분화 양액재배시 배지와 양액함량에 따른 거베라 'Sunny Lemon'의 생육반응)

  • To investigate the characteristics of plant growth and flower quality of gerbera 'Sunny Lemon' by amount of nutrient solution, young seedling plants, 'Sunny Lemon' were transplanted to rock-wool and medium of peat moss and perlite mixed with 1 to 2 and they were acclimatized in greenhouse during about 1 month. Nutrient solution supplied to the plants is sonneveld solution of 1/2 concentration and treatments launched June 24, 2010 when average plant height was $20{\pm}1cm$. Nutrient contents as a standard for starting point of irrigation by time domain reflectometry (TDR) were determined with 60-65%, 70-75%, and 80-85%. Results of growth during vegetative growth, plant height, leaf width and leaf number increased by 10% in rockwool, but they were not significantly different. As for plant growth depending on nutrient content, 80-85% treatment showed the highest values. Leaf number increased by 60%, and leaf width and plant height had a about 40% increase than initial growth. Effectiveness for flower quality, yield and days to flowering were superior when nutrient content of media was higher than in the others. Especially, average days to flowering in 80-85% content was advanced by 7-10 days compared to the day in 60-65% treatment. The total amount of nutrient supply per plant was higher in mixed medium than in rockwool, but change patterns of EC and pH were enhanced in rockwool. Based on our results, we recommended that growth, cut flower, and yield of gerbera 'Sunny Lemon' were more effective when nutrient content of mixed medium was maintained at 80-85%.

Metabolizing analysis according to the sawdust media of the known anticancer trees by Pleurotus ostreatuss (느타리버섯의 항암수목자원 배지속 함유성분의 분해능 평가)

  • The transitivity of Chemical constituents by Pleurotus ostreatus cultivated in different raw sawdusts, which are Juglans mandchurica, Cudrania tricuspidata and Lindera glauca, was investigated. The HPLC chromatography patterns on the chemical constituents of P. ostreatus showed the similar chromatography patterns in all different raw sawdusts and control sawdust. The unknown chemical constituents of P. ostreatus cultivated in the 10%, 20% mixed medium added 10 %, 20% different raw sawdusts, respectively, were increased. But the significance results in the mixed medium added 50% different raw sawdusts were not showed. The chromatography patterns of mycelia grown in media added the 80% MeOH extracts of three tree species showed the similar patterns in comparison with control mycelia. In the results, the secondary metabolites of functional media were not degrade and changed to other derivatives compounds by P. ostreatus.

Growth characteristics of oyster mushroom upon addition of barley flour as a substitute for cotton seed meal (면실박 대체 보리가루 첨가에 따른 느타리버섯의 생육 특성)

  • This study was conducted to investigate the optimum additive ratio of barley flour when used instead of cotton seed meal for the cultivation of oyster mushrooms. Mycelial growth was the fastest to 11.4 cm in medium containing 5% barley flour and to 9.0 cm in medium containing 30% barley flour. The mycelial density was slightly lower at 5% and 10% of barley powder addition, but there was no significant difference between the other treatments. The yield of the fruiting bodies was 158 g/850 mL, which was highest on treatment with 10% barley flour as a substitute for cotton seed meal. However, this was lower on treatments with only barley flour than in the control (156 g/850 mL). The diameter of the pileus and the hardness of stipes were highest with 10% barley flour and 10% in cotton seed meal, respectively. The L value was highest with 10% added barley flour, but the a-value and the b-value were not significantly different between treatments. The pH of the mixed media before inoculation with Pleurotus ostreatus was 4.7-5.0, and the total nitrogen content was 1.7-2.2, which was slightly lower than that of the control. The total carbon content was 44.6-45.0, which was not significantly different from that of the control. However, the content of trace elements such as Cu and Fe was lower than those of the control.

Efficiency of virus elimination in apple calli (cv. Hongro) derived from meristem culture of dormant buds (사과 품종 홍로의 휴면아 분열조직 배양을 통해 형성된 캘러스에서의 바이러스 제거효율)

  • Various sizes (0.2 ~ 1.2 mm) and developmental stages (referred to as Stage 1 ~ 3) of apical and lateral meristems were excised, together or separately, directly from dormant buds of apple 'Hongro'. They were mixed infected by Apple scar skin viroid (ASSVd), Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV) and Apple stem grooving virus (ASGV), which are major viruses attacking apples. A total of 31 callus lines (> 10 mm in diameter) were obtained by culturing the explants on Murashige and Skoog (MS) medium supplemented with 3% sucrose, 3.0 mg/L benzyladenine (BA) and 0.1 mg/L indole-3-butyric acid (IBA), and they were subjected to RT-PCR analysis for virus detection. A high rate of virus elimination (expressed as the percentage of calli that did not amplify during RT-PCR, i.e., RT-PCR negative calli per total number of calli obtained) was achieved for ACLSV (100%), ASSVd (93.7%), and ASPV (93.7%), whereas it was only 25.8% for ASGV. ASPV was detected in the presence of 2 ~ 3 bracts. Simultaneous virus elimination of ASSVd, ASPV, ACLSV, and ASGV occurred during the meristem culture, in which the early stages of the dormant buds (Stage 1) were used, because ASGV was mostly eliminated during that stage. The results of the present study will be valuable for the production of virus-free apple trees.

Determination of Salt Type, Salt Concentration, and Salt Application Method and Timing for Suppression of Stem Elongation in Grafted Cucumber Seedlings (오이 접목묘의 도장억제를 위한 염 스트레스 처리 효과)

  • This study aimed to examine a suppression effect of salinity on extension rate of stem elongation of cucumber seedlings (Cucumis sativus L.) grafted with roots of figleaf gourd plant. The effects of application methods, timings, and concentrations of two salt types, sodium chloride (NaCl) and calcium chloride ($CaCl_2$), were compared to untreated control plants. In result, an obvious suppression effect on the excessive elongation of stem was obtained by both sub-irrigated and medium-mixed NaCl salt. An improvement in quality of transplants was also obtained by the sub-irrigated NaCl salt. Foliar-applied NaCl caused visible leaf injury when the concentration was higher than 40 mM; but, with no effect on suppressing the stem elongation. When the NaCl was applied at 7 days after grafting, a higher concentration of NaCl was demanded for suppressing the stem elongation compared to an application at the day of grafting. No effect of the NaCl salt on the fresh weights of 36-day grown plants was observed; but, there was a negative effect on the number of female flowers at a high temperature season. Overall, the NaCl salt was more effective on slowing down the stem elongation and had the lower incidence of leaf injury than the $CaCl_2$ salt.
