Roles of the Hsp90-Calcineurin Pathway in the Antifungal Activity of Honokiol

  • Honokiol, a bioactive compound isolated from the cone and bark of Magnolia officinalis, has been shown to have various activities including inhibition of the growth of Candida albicans. We investigated the roles of the Hsp90-calcineurin pathway in the antifungal activity of honokiol. The pharmacologic tool was employed to evaluate the effects of Hsp90 and calcineurin in the antifungal activity of honokiol. We also evaluated the protective effects of the calcineurin inhibitor cyclosporin A (CsA) on honokiol-induced mitochondrial dysfunction by the fluorescence staining method. The Hsp90 inhibitor potentiated the antifungal activity of honokiol. A C. albicans strain with the calcineurin gene deleted displayed enhanced sensitivity to honokiol. However, co-treatment with calcineurin inhibitor CsA attenuated the cytotoxic activity of honokiol due to the protective effect on mitochondria. Our results provide insight into the action mechanism of honokiol.

Effect of Prostaglandin $F_2\alpha$ Administration on the Luteal Cell of Korean Native Cattle (Prostaglandin $F_2\alpha$의 투여가 한우황체의 조직상에 미치는 영향)

  • This experiment was carried out to observe early morphological changes of luteal cells in Korean native cattle treated with prostaglandin F2$\alpha$. Twenty five gram of prostaglandin F2$\alpha$ was administrated per cow at 10 days after ovulation and luteal cells were removed 30, 60, 120 and 180 minites after administration. Morphological changes of each luteal cell was observed by electron microscope. the results obtained were summarized as followings: 1. Many electron-dense granules were observed in luteal cells obtained from control cow but those granules were decreased rapidly after 30 minutes of administration and no granules were obresved after 180 minutes of administration. 2. In control, the shape of mitochondria were begining to collapse from the time of 60 minutes after administration. After 180 minutes of administration, mitochondria were swelled extreamly. 3. Lipid droplets in luteal cell were increased in its size and number with the duration of time after administration. 4. Shape of smooth endoplasmic recticulum was vesicular and its dimension and number were increased according to time course after administration.

Mitochondrial Protein Nfu1 Influences Homeostasis of Essential Metals in the Human Fungal Pathogen Cryptococcus neoformans

  • Mitochondrial protein Nfu1 plays an important role in the assembly of mitochondrial Fe-S clusters and intracellular iron homeostasis in the model yeast Saccharomyces cerevisiae. In this study, we identified the Nfu1 ortholog in the human fungal pathogen Cryptococcus neoformans. Our data showed that C. neoformans Nfu1 localized in the mitochondria and influenced homeostasis of essential metals such as iron, copper and manganese. Marked growth defects were observed in the mutant lacking NFU1, which suggests a critical role of Nfu1 in Fe-S cluster biosynthesis and intracellular metal homeostasis in C. neoformans.

Overexpressed Mitochondrial Thioredoxin Protects PC12 Cells from Hydrogen Peroxide and Serum-deprivation

  • Oxidative damage to mitochondria is a critical mechanism in necrotic or apoptotic cell death induced by many kinds of toxic chemicals. Thioredoxin (Trx) family proteins are known to play protective roles in organisms under oxidative stress through redox reaction by using reducing equivalents of cysteines at a conserved active site, Cys-X-X-Cys. Whereas biological and physiological properties of Trx1 are well characterized, significance of mitochondrial thioredoxin (Trx2) is not well known. Therefore, we addressed physiological role of Trx2 in PC12 cells under oxidative stress. In PC12 cells, transiently overexpressed Trx2 significantly reduced cell death induced by hydrogen peroxide, whereas mutant Trx2, having serine residues instead of two cysteine residues at the active site did not. In addition, stably expressed Trx2 protected PC12 cells from serum deprivation. These results suggest that Trx2 may play defensive roles in PC12 cells by reducing oxidative stress to mitochondria.

Identification of Isoleucine-Accepting tRNA in Maize Mitochondria

  • Maize mitochondrial tRNAs for isoleucine have been isolated using a putative $tRNA^{Ile}$ gene probe which has been previously isolated and characterized. It contains the 5'-CAT anticodon which would normally recognize the AUG methionine codon. The nucleotide sequence of one of these tRNAs has been partially determined, and contains a modified nucleotide at the first position of the anticodon. This type of posttranscriptional modification event could change the specificity of amino acid acceptance of a tRNA, unlike that deduced from the corresponding gene. An aminoacylation experiment also demonstrated that these purified tRNAs have isoleucine acceptance activity but no methionine-accepting activity.

Electron Microscopic Study of Hop Shoot Apex by Gibberellin Treatment (Gibberellin 처리에 의한 Hop Shoot Apex의 전자현미경적인 연구)

  • This experiment was undertaken with the purpose of investigating effect of gibberellin on the growth and electron microscopic ultrastructure of hop(Humulus lupulus var. Hallertau) shoot apex. The results are as follows. 1) The growth of stem in the plots treated with GA(10, 25, and 50ppm) is more predominant than control. The leaves and cones of treated plots turn to pale green. 10ppm plot of GA treatment is the most effective in growth and yield than the others. 2) Under the GA-treated condition, it is believed that the nucleoplasmic index (NP) are higher than the control, and so nucleus, nucleolus, mitochondria, primary cell wall, middle lamella, and etc. tends to be larger than the control. 3) Mitochondria contained in the cell of shoot apex treated with concentration of 10ppm GA was seemed to increases in number and be larger in size than the control.

  • This paper aims to probe the effect of SQ in the rat liver which pretreated with CP was examined by transmission electron microscope. In the A group, the difference between the normal and the treated groups were not detected at 24 hours, but the few mitochondria were expanded at the 72 hours. In the B group, the cisternae of rough-surfaced endoplasmic reticulum were partially destructed and attached ribosomes were remarkably decreased at 24 hours. A number of the mitochondria were dilated and increased in number, the filamentous materials also detected at 72 hours. These results suggest that SQ is not only concerned with construction of the membrane of the cell organelles but also decreased the cellular toxicity in the hepatic cells.

  • The spermatogenetic process in the edible giant snail is similar to those in the other snails, except for the axoneme formation process. In this study, the axoneme formation process in the giant snail was mainly examined by means of electron microscopy. The tail portion of a spermatozoon is about $160{\mu}m$ long, and extends straight to the rear, surrounded by two large and long mitochondria in spiral forms. A number of glycogen particles $(40\sim70nm)$ are found in the swollen matrix of the mitochodria. The axoneme which composes the tail of a spermatozoon is surrounded by $7\sim10$ lamella-form fibrous sheaths of about $0.2{\mu}m$ in thickness. Most of the mature spermatozoa are found to be clustered into a group of $5\sim7$ ea in syncytial bridges formed by cytoplasmic processes. Sertoli cells contain glycogen particles, endoplasmic reticulum, a lot of mitochondria, and lipids in their cytoplasm. They protrude their filiform pseudopodia and phagocytize abnormal spermatids or spermaozoa.

  • The glandular secretory system of the capitate gandular trichomes in leaf of Thymus quinquecostatus Celakovsky was examined by transmission electron microscope. The glandular trichome was consisted of three cell layers; an basal cell layer, a stalk cell with single-celled intermediate layer and a discoid secretory layer with thickened cuticle. The secretory cell was dense, rich in mitochondria, rER, plastds, Golgi complex and had many vesicular structure. Typical plastids with reticulate body and plastoglobule were present in glandular trichome. The tytoplasm of secretory cell was filled with osmiophilic secretory materials. The secretory vesicles, originated from Golgi complex, appeared as membrane bounded vesicles and secreted to the outer wall surface. The presences of well developed rER, mitochondria, Golgi complex, and membrane-bounded vesicles fused with plasmalemma in the secreting cells indicate that the granulocrine mechanism of secretion was occurring in T. quinquecostatus. Subcuticular cavity was developed between the cuticular layer and the secretory cell wall, and it formed above the secretory cell upon separation of cuticle-wall.
