Treatment of central diabetes insipidus with anemia in a dog

  • A 10-year-old, spayed female miniature schnauzer was referred to the Veterinary Medical Teaching Hospital of Chungnam National University due to evaluation of sudden polyuria (PU) and, polydipsia (PD) (540 mL/kg/day) with severe anemia and weight loss. Blood examination results were normal except for severe anemia (hematocrit, [HCT]: 11.8%). Urinalysis revealed a urine specific gravity (USG) of 1.003, whereas urine sediment was not specific. Urine osmolality was 90 mOsm (reference range: 800~2500 mOsm), and plasma osmolality was 303 mOsm. No specific lesions were found using diagnostic imaging including radiography, ultrasonography and magnetic resonance imaging (MRI). The serum cortisol level was normal in cosyntropin stimulation test. Plasma arginine vasopressin (AVP) concentration was <0.4 pg/mL (reference range: 3.49~5.45 pg/mL). Blood transfusion was initiated in addition to an oral prescription of desmopressin acetate (DDAVP, 0.1 mg/head) thrice a day for one week. The patient was rechecked for clinical signs, urine osmolality, and USG; the clinical signs of PU/PD were resolved, urine osmolality increased to 1106 mOsm, and, USG increased to 1.021. Considering the improved clinical signs, and increased urine osmolality, and USG after DDAVP treatment, the dog was diagnosed with central diabetes insipidus. USG and urine osmolality increased to >1.030 and 2200 mOsm, respectively. Anemia also gradually improved and HCT increased to >37%. DDAVP was tapered to 0.1 mg/head twice a day and all clinical signs in the patient have completely resolved.

Evaluation of porcine intestinal organoids as an in vitro model for mammalian orthoreovirus 3 infection

  • Background: Mammalian orthoreovirus type 3 (MRV3), which is responsible for gastroenteritis in many mammalian species including pigs, has been isolated from piglets with severe diarrhea. However, the use of pig-derived cells as an infection model for swine-MRV3 has rarely been studied. Objectives: This study aims to establish porcine intestinal organoids (PIOs) and examine their susceptibility as an in vitro model for intestinal MRV3 infection. Methods: PIOs were isolated and established from the jejunum of a miniature pig. Established PIOs were characterized using polymerase chain reaction (PCR) and immunofluorescence assays (IFAs) to confirm the expression of small intestine-specific genes and proteins, such as Lgr5, LYZI, Mucin-2, ChgA, and Villin. The monolayered PIOs and three-dimensional (3D) PIOs, obtained through their distribution to expose the apical surface, were infected with MRV3 for 2 h, washed with Dulbecco's phosphate-buffered saline, and observed. Viral infection was confirmed using PCR and IFA. We performed quantitative real-time reverse transcription-PCR to assess changes in viral copy numbers and gene expressions linked to intestinal epithelial genes and antiviral activity. Results: The established PIOs have molecular characteristics of intestinal organoids. Infected PIOs showed delayed proliferation with disruption of structures. In addition, infection with MRV3 altered the gene expression linked to intestinal epithelial cells and antiviral activity, and these effects were observed in both 2D and 3D models. Furthermore, viral copy numbers in the supernatant of both models increased in a time-dependent manner. Conclusions: We suggest that PIOs can be an in vitro model to study the infection mechanism of MRV3 in detail, facilitating pharmaceutical development.

A retrospective study of age-specific disease incidence in major popular breed dogs in Republic of Korea

  • Dogs exhibit patterns of health issues that vary by life stage. An understanding of disease incidence with respect to breed and age/life stage could be an important component of canine health management and welfare. This study aimed to describe the age-specific disease incidence of 3 small dog breeds that attended veterinary clinics in the Republic of Korea, based on data from electronic veterinary medical records (EVMRs). A total of 40,785 EVMRs from Maltese (n = 21,355), Miniature Poodle (n = 11,658) and Shih Tzu dogs (n = 7,772) were analyzed. Common health problems in 3 small dog breeds were 'diseases of the skin' and 'diseases of the ear,' respectively. Among dogs aged ≤ 3 years, 'preventive medicine' was the most common cause cited for veterinary clinic visits. Among dogs aged 4 to 8 years, the most frequent health problems were 'diseases of the skin.' Among dogs aged 9 to 13 years, 'heart diseases,' 'kidney diseases,' 'mammary gland tumor,' and 'neoplasia (unspecified)' were considerably more frequent, compared to the rates in dogs ≤3 years. Among dogs aged ≥ 14 years, 'heart diseases' and 'sneezing/cough' were the main health problems. In all breeds, the frequencies of 'diseases of circulatory system,' 'diseases of respiratory system,' 'diseases of the nervous system,' 'endocrine' and 'neoplasia' increased rapidly with aging. This surveillance could inform strategies for disease screening tests and management based on life stage in these dog breeds and enable more effective health management.

External Attachment of Pop-up Satellite Archival Tag (PSAT) and Water Temperature Affect Oxygen Consumption Rate of the Olive Flounder Paralichthys olivaceus (넙치(Paralichthys olivaceus) 산소 소비율에 미치는 Pop-up Satellite Archival Tag (PSAT) 체외 부착과 수온의 영향)

  • This study aimed to examine the effect of external pop-up satellite archival tags (PSATs) attachment and water temperature on the oxygen consumption rate (OCR) of the olive flounder (mean body weight 2281.7 g). The OCRs of fish were measured under conditions of three different water temperature conditions (15, 20, and 25℃) and two different tagging methods [non-tagging, control; bio-logger external attachment with a miniature PSAT (dummy mrPAT), BEA] using a closed flow-through respirometer. The OCRs of fish linearly increased with the increase in water temperature in both the control and BEA (P<0.001); however, the OCRs of BEA were approximately 1.8-1.9 times lower than those of the control at each water temperature (P<0.001). The Q10 values of the control and BEA were the highest in the water temperature range of 15 to 20℃, but sensitivity to water temperature changes was higher in BEA than in the control. The metabolic energy loss rate (MEL) of fish increased with increasing water temperature regardless of external tagging, but the MEL of the control was higher than that of BEA (P<0.001). These results demonstrate that OCR, thermal sensitivity, and energy expenditure are all affected in adult olive flounder with external PSAT attachment.

Computed Tomography for Diagnosing Chylothorax Associated with Cranial Vena Cava Thrombosis in a Dog

  • A 13-year-old male neutered Miniature Pinscher presented with coughing and dyspnea. The dog had been coughing for the past 4 weeks. The patient had mild dehydration on physical examination, and muffled heart sounds were detected. Thoracic radiographs revealed pleural effusion, which was consistent with chylous effusion based on cytological and biochemical evaluations. Computed tomography (CT) lymphangiography, which was performed via intrametatarsal pad injection, revealed no evidence of thoracic duct rupture or obvious leakage. On CT angiography (CTA), an intraluminal filling defect was identified in the cranial vena cava (CrVC). CrVC thrombosis with secondary chylothorax was diagnosed based on CT lymphangiography and CTA. Clopidogrel, rivaroxaban, and recombinant tissue-plasminogen activator were prescribed. The follow-up CTA, 4 months after diagnosis, revealed a decrease in the thrombus, and no pleural effusion was identified. Although CrVC thrombosis is an uncommon presentation in veterinary patients, thrombus in the CrVC should be considered as a differential diagnosis of chylothorax in dogs. CT lymphangiography and CTA could be helpful in identifying and differentiating the underlying etiologies of chylothorax.

Direct Action of Relaxin on the Pig Mammary Glands to Promote Development of the Alveoli and Remodeling of the Extracellular Matrix (Relaxin Hormone의 직접젇인 작용에 의한 돼지 유선상피세포의 발달과 세포외간 기질의 분해개조 촉진)

  • Our previous studies demonstrated that relaxin in concert with estrogen promotes development of the mammary parenchyma during the last third of gestation in gilts, and the specific relaxin-binding sites were present in the mammary gland. This study was conducted to determine if relaxin-binding sites in the mammary gland were functional relaxin receptors. Three cycling cross-bred gilts were bilaterally ovariectomized on day 0 of the experiment. Beginning on day 15 and continuing through day 29 post-surgery, the gilt received an im. injection of estradiol benzoate at 12-hr intervals. Beginning on day 22 post-surgery, highly purified porcine relaxin was administered(lug/hr) into the left fourth mammary gland from the anterior end via miniature osmotic pump. Physiological saline was administered to the right fourth mammary gland. The gilt was sacrificed on day 29 post-surgery and histological characteristics of the mammary parenchyma were examined. The mammary glands treated locally with saline showed little, if any, lobulo-alveolar development, whereas the mammary glands treated with relaxin showed not only marked lobulo-alveolar development but also prominent secretions in the alveoli. The saline-treated glands were characterized by relatively dense and highly organized collagen fiber bundles. Whereas, in the relaxin-treated mammary glands, collagen fiber bundles were dispersed and loosely organized. In conclusion, relaxin-binding sites in the mammary gland are functional relaxin receptors and relaxin acts directly on the pig mammary gland to promote development of the alveoli and remodeling of the extracellular matrix.

A Study on the Advent of a Downsizing Society and the Modern Total Property Theory (축소형 사회의 도래와 현대총유론)

  • The Modern Total Property Theory is the social reform theory which emerged in the 2000s in Japan, which was suffering from the social issues similar to Korea - depopulation, aging society, centralization of metropolitan area, extinction risk of local provinces, and low growth. The theory aims to resolve social problems through the total property of spaces including land. The Modern Total Property Theory focuses on the issue of absolute land ownership. The absolute land ownership, which became the leverage of the economic growth after the Second World War, will not function properly in the future downsizing society. The existing ownership system functioned based on the premise that the value of land or buildings will be maintained or increase due to the existence of demand. However, large numbers of the ownerships that have no value will appear. However, since land or building cannot be touched even after they are let alone because they are bound by absolute ownership, the measure of turning the ownership of land or building into total property by establishing the subject of total property is being suggested. For this aim, the Modern Total Property Theory suggests that the bilateral relationship between state and individual be converted to the trilateral relationship among state, individual, and intermediate organization by inserting a middle clause in the modern law which is based on dual clauses - state and individual. Furthermore, since the Modern Total Property Theory is a macroscopic design for the reproduction of city and land and includes the legislation on the social policy level, it will provide the diagnosis and the prescriptions toward the social issues in Korea and adequate implications toward the reform direction of future social policies and judicial system.

  • In order to evaluate the bearing capacity behaviour of a monopod suction bucket foundation for an offshore wind tower at the western sea of Korea, a centrifuge load test and numerical analyses were performed. The monopod bucket foundation was designed to be installed in a silty sand layer. The model soil was prepared to simulate a target site by using soil samples having similar properties and controlling relative density. In-flight miniature cone penetration test and bender element array were used to confirm that the model soil had represented the target site conditions. The load - rotation curve of the centrifuge load test was analysed. A series of numerical analyses were performed to validate the experimental conditions. Self-weight of the model, distance to the boundary and elastic modulus of the soil layer were varied to study their effects on the load - rotation curves.

  • Ultrasonic imaging of 3-D structures for nondestructive evaluation must provide readily recognizable images with enough details to clearly show various flaws that may or may not be present. Typical flaws that need to be detected are miniature cracks, for instance, in metal pipes having aged over years of operation in nuclear power plants; and these sub-millimeter cracks or flaws must be depicted in the final 3-D image for a meaningful evaluation. As a step towards improving conspicuity and thus detection of flaws, we propose a pulse-echo ultrasonic imaging technique to generate various 3-D views of the 3-D object under evaluation through strategic scanning and processing of the pulse-echo data. We employ a 2-D Wiener filter that filters the pulse-echo data along the plane orthogonal to the beam propagation so that ultrasonic beams can be sharpened. This three-dimensional processing and display coupled with 3-D manipulation capabilities by which users are able to pan and rotate the 3-D structure improve conspicuity of flaws. Providing such manipulation operations allow a clear depiction of the size and the location of various flaws in 3-D.

Standard Penetration Test Performance in Sandy Deposits (모래지반에서 표준관입시험에 따른 관입거동)

  • This paper presents an equation to depict the penetration behavior during the standard penetration test (SPT) in sandy deposits. An energy balance approach is considered and the driving mechanism of the SPT sampler is conceptually modeled as that of a miniature open-ended steel pipe pile into sands. The equation consists of three sets of input parameters including hyperbolic parameters (m and ${\lambda}$) which are difficult to determine. An iterative technique is thus applied to determine the optimized values of m and ${\lambda}$ using three measured values from a routine SPT data. It is verified from a well-documented record that the simulated penetration curves are in good agreement with the measured ones. At a given depth, the increase in m results in the decrease in ${\lambda}$ and the increase in the curvature of the penetration curve as well as the simulated N-value. Generally, the predicted penetration curve becomes nearly straight for the portion of exceeding the seating drive zone, which is more pronounced as soil density increases. Thus, the simulation method can be applied to extrapolating a prematurely completed test data, i.e., to determining the N value equivalent to a 30 cm penetration. A simple linear equation is considered for obtaining similar results.
