Effect of Acute Ethanol Intoxication on Hepatic Rhodanese Activity in Rats with Extrahepatic Cholestasis

  • Liver and serum rhodanese activities were determined in acute ethanol intoxicated rats with extrahepatic cholestasis induced by common bile duct ligation (CBD) to manifest the biochemical background of alcohol drinking hazard under the hepatobiliary disease. Liver cytosolic and microsomal rhodanese activities and these Vmax values in CBD ligated rats with acute ethanol intoxication were found to be decreased much more than that in CBD ligation alone. However, the difference of Km value on above hepatic enzyme was not found between the experimental groups. On the other hand, serum rhodanese activity in CBD ligated rats with acute ethanol intoxication was greater increased more than that in CBD ligation alone. These results indicate that the biosynthesis of the hepatic rhodanese decreases and the serum rhodanese activity increases in cholestasis combined with acute ethanol intoxication, reflecting damage of aggravated hapatocytic membrane. Accordingly, the resulting data supported the fact that alcoholic drinks were enzymologically harmful to the hepatobiliary disease.

Conjugated Linoleic Acid Supplemented to Dietary Fat Has an Antioxidant Activities, but It Depends on the Type of Fat in Diet (쥐에서 식이에 보충한 Conjugated Linoleic Acid가 식이지방 종류에 따라 항산화작용에 미치는 영향)

  • The study was designed to observe an antioxidant activities of conjugated linoleic acid(CLA) in rat liver by determining the activities of antioxidative enzymes(superoxide dismutase, glutathione peroxidase, catalase) and the levels of tocopherol and thiobarbituric acid reactive substance(TBARS). Male Sprague Dawley rats at weeks-old were divided into groups according to the types of dietary fat(beef tallow and fish oil) and then each group was subdivided into groups depending on CLA supplement. All rats were fed experimental diet containing 12% total fat including 1% CLA by weight for 30 weeks. CLA supplemented to beef tallow diet did not have significant effect on the level of TBARS and tocopherol. The level of TBARS was significantly increased in fish oil diet(highly unsaturated fat diet), but its level was significantly reduced by increasing SOD and GSH-Px activities when CLA was supplemented to fish oil diet so that CLA showed a sparing action of tocopherol in tissue. CLA did not have significant effect on peroxisomal catalase activities, but its activity was significantly increased when TBARS production was high in the fish oil diet. CLA could be incorporated into phospholipid of microsomal membrane, and interfered the conversions of C18 : 0 into C18 : 1 and C18 : 2 into C20 : 4 in liver. In conclusion, CLA had an antioxidant activities depending on the type of fat in diet. Therefore, it could be recommended to use CLA when highly unsaturated fat was used in meal preparation.

Effect of Dietary Conjugated Linoleic Acid on Plasma Levels of Glucose and Lipids and Hepatic Lipogenic Enzyme Activity in Otsuka Long Evans Tokushima Fatty Rats (OLETF 비만쥐에서 CLA첨가 식이가 혈장의 포도당과 지질농도 및 간조직의 Lipogenic Enzyme 활성도에 미치는 영향)

  • The study was designed to observe whether the conjugated linoleic acid supplemented to diet could reduce plasma levels of glucose and lipids which were increased in 27-weeks old Otsuka Long Evans Tokushima Fatty(OLETF) rats. Twenty male OLETF rats of 7 weeks old were fed an experimental diet containing 4.5%(w/w) total fat including 1% CLA and six of twenty rats were sacrified at 6 weeks feeding. The rest of OLETF rats was divided into 2 groups, one group was continuously fed for 14 weeks more the same experimental diet containing 1% CLA and the other group was fed control diet which eliminated CLA. CLA did not significantly reduce food intake and body weight gain in OLETF obese rats. Plasma triglyceride and total cholesterol level were significantly increased at older age of OLETF obese rats, but CLA could significantly reduce plasma cholesterol and triglyceride increased in obese rats. However, CLA was not strong enough to reduce the increased plasma glucose level and hepatic lipogenic enzyme acitivies. CLA was mostly deposited in epididymal fat pad and could be incorporated into hepatic microsomal membrane and did interfere the conversion of C18 : 0 into C18 : 1 in liver. In conclusion, CLA could have anti-atherogenic effect by reducing plasma cholesterol and triglyceride which was increased in genetically obese rats, but CLA(1%) was not good source of dietary fatty acid to reduce body fatness and plasma glucose which was increased by obese gene in older rats.

  • The E2 protein of HCV (hepatitis C virus) is thought to have a potential role in the development of subunit vaccines and diagnostics. To express it by the insect cell/baculovirus expression (Bacu) system, we constructed a recombinant Autographa californica nuclear polyhedrosis virus (AcIL3E2), determined the most appropriate expression conditions in terms of host cell line and culture medium, and characterized the expressed HCV E2 protein. A culture system using Trichoplusia ni BTI-TN5Bl-4 cells and SF 900IISFM medium expressed a relatively high level of HCV E2 protein. It was revealed that its glycosylation properties and subcellular localization were almost the same as the ones in the mammalian cell expression system previously reported, suggesting the recombinant HCV E2 protein derived from our Bacu system can be utilized for development of a subunit vaccine and diagnostics. Interestingly, HCV E2 protein was not degraded at all even at 43 h post-heat shock in the heat shock-induced necrotic cells, probably due to its integration into the microsomal membrane, indicating that heat shock can be employed to purify HCV E2 protein.

Effects of Butanol Extract of Aralia elata on Lipid Peroxidation (두릅나무 부탄올 추출물이 지질 과산화에 미치는 영향)

  • It is well known that lipidperoxide, formed in vivo, induced the denaturation of enzyme and destruction of cell membrane to acute injury of tissue. Aralia elata have physiological activates, the improvement of lipid metabolism, antidiabetic activity etc., which was thought to have the relationship to lipid peroxidation. The anti-lipidperoxidative effect of Aralia elata have not yet established. In this study, we examined the anti-lipidperoxidative effects of Aralia elata (Butanol fraction) on CCI$_{4}$ induced lipidperoxidation in rats, and elucidated the anti-lipidperoxidative mechanism. In rat liver homogenate intoxicated with CCI$_{4}$ (0.5 ml/100g), BuOH fraction of Aralia elata (80 mg/Kg/day) exhibited 85.41% anti-lipidperoxidative effect but in serum 69.63% inhibitory effects, respectively. In mitochondrial and microsomal fraction showed inhibition of 55.85% and 69.30%, respectively. In order to elucidate the mechanism of anti-lipidperoxidation effects of Aralia elata, enzymatic (NADPH dependent) and non-enzymatic (Ascorbic acid catalyzed) reaction, in vitro, were performed. In enzymatic reation, Aralia elata exhibited 59.43% anti-lipidperoxidation effects, but in non-enzymatic reaction exhibited 43.27% inhibition. Therefore, it is noteworthy that antioxidative power of them may mainly results from the inhibition by enzymatic reaction.

  • Carbon tetrachloride (CCI$_4$) induces the hepatotoxicity due to the reactive free radicals generated by cytochrome P-450 (CYP-450) enzyme, which result in destabilization of cellular membrane. Diltiazem, a calcium channel blocking agent, has been known to suppress the CYP-450 enzyme activities. To study the effect of diltiazem in $CCl_4$-treated rats, we measured the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and glutathione S-transferase (GST), contents of bilirubin, albumin, total protein, cholesterol, triglyceride, blood urea nitrogen, creatinine malondialdehyde and calcium. Also we conducted liver histopathologic examinations. Diltiazem, when administered 1 hour prior to CCI$_4$ treaeent, significantly reduced the activities of ALT and AST, the contents of microsomal malondialdehyde and calcium in liver and microsome as compared with those of $CCl_4$-treated rats. In addition, histopathologic examination showed that diltiazem prevented the development of centrilobular necrosis induced by CCI$_4$ in liver tissue. Our results suggested that diltiazem could inhibit the formation of free radicals and the influx of calcium. Therefore diltiazem may be applied to suppress the liver damage caused by $CCl_4$.

  • Objective : This study was undertaken to determine if Scutellaria baicalensis Georgi (SbG) extract exerts protective effect against oxidant-induced cell injury in renal proximal tubular cells. Methods : The cell injury was evaluated by lactate dehydrogenase (LDH) release in rabbit renal cortical slices and lipid peroxidation was estimated by measuring malondialdehyde (MDA). t-Butylhydroperoxide (tBHP) was used as a model of oxidant. Results : tBHP at 1 mM increased LDH release and lipid peroxidation, which were prevented by SbG in a dose dependent manner over concentration range of 0.001-0.1%. SbG provided the protective effect against oxidant-induced reduction in PAH uptake by renal cortical slices and microsomal Na+-K+-ATPase activity. SbG attenuated tBHP-induced depletion of reduced glutathione. 0.2 mM $HgCl_2$ increased LDH release and lipid peroxidation, which were completely prevented by 0.05% SbG. Conclusion : SbG prevents oxidant-induced impairment in membrane transport function.

  • Human microsomal prostaglandin E synthase-1 (mPGES-1) is a membrane associated protein that catalyzes the conversion of prostaglandin $H_2$ ($PGH_2$) into prostaglandin $E_2$ ($PGE_2$). In this study, the expression of human mPGES-1 in E. coli was significantly enhanced by modifying the utility of specific codons and the recombinant mPGES-1 was efficiently purified to homogeneity. The $K_m$ and $V_{max}$ of the purified enzyme were determined and the trimeric state characterized by chemical cross-linking with glutaraldehyde. The purified mPGES-1 was used for the screening of a chemical library of bioactive or drug compounds to identify novel inhibitors, and oxacillin and dyphylline were identified as moderately inhibiting mPGES-1 with $I_{C50}$ values of 100 and 200 ${\mu}M$, respectively. As these compounds competitively inhibited the catalysis of $PGH_2$, their binding sites appeared to be located near the $PGH_2$ binding pocket.

  • Drug-metabolizing system which has the important role in drug metabolism is localized in smooth endoplasmic reticulum of hepatocytes and is composed of NADPH, NADPH-cytochrome $P_{450}$ reductase, cytochrome $P_{450}$ and others. It is well known that the enzyme system is induced by phenobarbital and methylcholanthrene. Lipid peroxidation is reaction of oxidative deterioration of polyunsaturated lipids. Formation of lipid peroxides in liver microsome has been found to produce degradation of phospholipid, which are major components of microsomal membrane. The relationship between the formation of lipid oxides and the activities of drug-metabolizing enzyme in the liver of rats was reported by several investigators. In this study the effect of riboflavin tetrabutylate, an antioxidant on lipid peroxidation, specially the relationship between lipid peroxidation and drug-metabolizing enzyme system was investigated. In addition the effect of vitamin A derivatives, such as retinoic acid and retinoid on the enzyme was also observed. Results are summarized as followings. 1) The pretretment with riboflavin tetrabutylate inhibited completely the lengthened sleeping time due to $CCl_{4}$ treatment. 2) The increase of TBA value was prevented by the pretreatment with riboflavin tetrabutylate. 3) The pretreatment with riboflavin tetrabutylate also prevented the decrease of drug-metabolizing enzyme caused by $CCl_{4}$. 4) Both retinoic acid and retinoid remarkably decreased the activity of aminopyrine demethylase. Pretreatment of riboflavin tetrabutylate, however, prevented inhibitory effect of retinoic acid on the enzyme activity.

The Effect of Juglandis Semen Herbal Acupuncture on Alterations of Tubular Transport Function in Rabbits with Mercury-Induced Acute Renal Failure (호도약침(胡桃藥鍼)이 수은(水銀)에 의한 급성신불전(急性腎不全) 가토(家兎)의 신세요관(腎細尿管) 물질이훈계(物質移勳系) 장애(障碍)에 미치는 영향(影響))

  • Objective : This study was undertaker to determine if Juglandis Semen herbal acupuncture (JSA) exerts protective effect against alterations in membrane transport function in rabbits with mercury-induced acute renal failure. Methods : Nephrotoxicity was induced by subcutaneous administration of Hg(a single dose of 10mg/kg) and JSA was performed at both sides of Shenshu($(BL_{23})$, Sinsu) for 7 days. Results: The administration of Hg at a subcutaneous single dose of 10 mg/kg caused a reduction in GFR to 12% of the basal value and an increase in fractional $Na^+$ excretion to 8.9-fold, indicating generation of acute renal failure. When JSA were given for 7 days prior to Hg administration, such changes were significantly attenuated. The fractional excretion of glucose and phosphate was increased to approximately 102- and 35-fold, respectively, in rabbits treated with Hg alone. The increase in rabbits treated with Hg following ISA are significantly lower than that in animals treated with Hg alone. Uptakes of glucose and phosphate in purified isolated brush-border membrane and $Na^+-K^+-ATPase$ activity in microsomal fraction were inhibited in rabbits treated with Hg alone, suggesting that impairment in proximal reabsorption of glucose and phosphate is resulted from a direct damage of membrane transport carriers and disruption of the normal $Na^+$ gradient. Such changes were prevented by JSA. Conclusion These results indicate that the administration of Hg causes impairment in reabsorption of solutes in the proximal tubule via the generation of reactive oxygen species. JSA provides the protection against the Hg-induced impairment in proximal reabsorption, and its effect may be resulted from its antioxidant action.
