반도체 미세공정 기술을 이용한 Hollow형 실리콘 미세바늘 어레이의 제작 (Fabrication of Hollow-type Silicon Microneedle Array Using Microfabrication Technology)

  • Hollow-type microneedle array can be used for painless, continuous and stable drug delivery through a human skin. The needles must be sharp and have sufficient length in order to penetrate the epidermis. An array of hollow-type silicon microneedles was fabricated by using deep reactive ion etching and HNA wet etching with two oxide masks. Isotropic etching was used to create tapered tips of the needles, and anisotropic etching of Bosch process was used to make the extended length and holes of microneedles. The microneedles were formed by three steps of isotropic, anisotropic, and isotropic etching in order. The holes were made by one anisotropic etching step. The fabricated microneedles have $170{\mu}m$ width, $40{\mu}m$ hole diameter and $230{\mu}m$ length.

자동 미세침이 말초성 안면마비 환자의 증상 정도 및 안전성에 미치는 영향에 대한 임상 관찰 (Clinical Observation of Effect on Severity of Symptoms and Safety of Auto-microneedle Therapy in Patients with Peripheral Facial Paralysis)

  • Objectives : This study was to observe clinical application of auto-microneedle therapy system (AMTS) in patients with peripheral facial paralysis. Methods : 27 peripheral facial paralysis patients were observed after taking AMTS at Facial Palsy Center, Kyung Hee University Oriental Medicine Hospital from March 1, 2011 to January 9, 2012. We assessed the symptoms of facial paralysis with Yanagihara unweighted grading system, Sunnybrook facial grading system(SBGS) and facial disability index(FDI), and observed adverse events and total safety of the treatment. Results : The scores of facial palsy scales increased after AMTS in Yanagihara grading system and Sunnybrook facial grading system. AMTS-related adverse events were mild pain(5.9%) and fatigue(3.5%), which needed no extra treatment. The total safety evaluation was between 'safe' and 'nearly safe' level. There were no other serious adverse events. In addition, patients were satisfied with subjective improvement including facial tingling and numbness. Conclusions : AMTS can be applied as an adjunctive treatment for patients with peripheral facial paralysis due to its safety and clinical usefulness. It is easier to stimulate wide skin area in a short time. Further clinical research is required to investigate the effectiveness of ATMS in a more rigorous RCTs.

C57BL/6N 마우스 탈모모델에서 미세다륜침(MTS)과 황련해독탕 약침액 도포가 모발성장에 미치는 영향 (Effects of Microneedle Therapy System(MTS) and Hwangryeonhaedoktang Pharmacopuncture Solution on Hair Growth in an Alopecia Model of C57BL/6N Mouse)

  • Objective : This study was carried out to investigate the effect of Hwangryeonhaedoktang pharmacopuncture solution (HRHDT) and microneedle therapy system (MTS) on hair growth in an alopecia model of C57BL/6N mice.Methods : Six-week old mice were depilated and separated in 4 groups ; CON (saline), MXD (3% Minoxidil), MTS and HRHDT+MTS. The treatments were applied twice a week for 17 days. The hair growth was determined photographically. The hair density, thickness and length were identified by Folliscope and the weights of body and organs were measured. In dorsal skin tissue, the expression of hair growth-related gene and protein was analyzed by RT-PCR. In addition, the hair follicles in the dermis were observed by H&E staining.Results : The promotion of hair growth was observed in HRHDT+MTS and MTS compared to CON. The hair density, thickness and length were also improved in HRHDT+MTS and MTS compared to CON. The mRNA expression of IGF-1, PRL and PL and the protein expression of VEGF and IGF-1 were increased in HRHDT+MTS and MTS compared to CON. The hair follicles and hair root growth were improved in HRHDT+MTS and MTS compared to CON. In the above results, HRHDT+MTS were more effective than MTS.Conclusions : These results suggest that HRHDT and MTS have a hair growth activity and can be useful for the treatment of alopecia.

Methods Five-week old mice were depilated and separated in 4 groups ; CON (50% EtOH), MXD (5% Minoxidil), MTS and GLE + MTS. The treatments were applied twice a week for 3 weeks. The hair growth was determined photographically, the hair density, thickness and length were identified by Folliscope and the weights of body and organs were measured. In dorsal skin tissue, the expression of hair growth-related gene and protein was analyzed by RT - PCR or Western blot. In addition, the hair follicles in the dermis were observed by H&E staining. Results The promotion of hair growth was observed in GLE + MTS and MTS compared to CON. The hair density, thickness and length were also improved in GLE + MTS and MTS compared to CON. The mRNA expression of TGF-${\alpha}$, TGF-${\beta}1$, IGF-1, PRL and PL and the protein expression of VEGF and IGF-1 were increased in GLE + MTS and MTS compared to CON. The hair follicles and hair root growth were improved in GLE + MTS and MTS compared to CON. In the above results, GLE + MTS were more effective than MTS. Conclusions These results suggest that GLE and MTS has a hair growth activity and can be useful for the treatment of alopecia.

마이크로니들을 이용한 FITC-BSA의 경피투과 거동 (Transdermal Permeation Behavior of FITC-BSA using Microneedle)

  • Penetration rate of large molecule through skin is very low due to the barrier effect of stratum corneum. Novel microneedle treatment device with roll was designed for transdermal delivery of large molecular drugs such as vaccine and protein drugs. The permeation rates of FITC labelled bovine serum albumin (FITC-BSA) as a model protein were determined using modified Franz diffusion cell and hairless mouse skin which were treated by hydrogel or solution containing FITC-BSA. Fluorescent spectrophotometer was used to analyze the concentration of FITC-BSA. Microscope using fluorescent filter was used to capture the image and location of FITC-BSA in the skin. We confirmed that permeation rate of BSA was increased with the treatment by microneedle and was increased by the increasing frequency of treatment. Furthermore, the permeation rate observed from hydrogel treated skin was significantly higher than that from solution treated skin.

생체의학에 적용 가능한 테이퍼형태의 금속성 마이코로니들 어레이의 개발 (A Development of Tapered Metallic Microneedle Array for Bio-medical Application)

  • 본 연구에서는 유리기판을 통한 SU-8의 이중층을 후면 노광을 통하여 테이퍼지고 속이 빈 형태의 마이크로니들 배열구조물을 만드는 새로운 방법을 제안하였으며 테이퍼지고 속이 빈 형태의 원형축의 Buckling현상에 관한 해석해를 구하였다. Pyrex 7740을 유리기판으로 사용하고 이중층 구조의 SU-8 막을 후면 노광으로 금형 구조물을 제작하는 공정을 개발하였다. $200\;{\mu}m$ 높이의 SU-8기둥들에서 $4.37{\sim}5^{\circ}$범위의 테이퍼 각도를 $400\;{\mu}m$ 높이의 SU-8기둥에서는 $3.08{\sim}4.48^{\circ}$범위 내의 테이퍼 각을 보여주고 있다. 후면 유체저장소와 가로세로 각각 10개의 테이퍼 형태로 전기도금된 니켈 마이크로니들 어레이를 유리 기판을 통해 이중층 구조의 SU-8막을 후면 노광하고. 니켈을 전기도금 함으로서 실현시켰다. $200\;{\mu}m$$400\;{\mu}m$ 높이의 벽두께 $10{\mu}m-20{\mu}m$ 및 내경 $33.6{\mu}m-101{\mu}m$인 마이크로니들 어레이를 제작하였다. 또한 $400\;{\mu}m$ 높이의 벽두께 $20\;{\mu}m$ 및 내경 $33.6\;{\mu}m$$3.08^{\circ}$의 테이퍼 각 마이크로니들의 임계 버클링힘은 1.8N이었다. 이 해는 차후의 테이퍼 형태의 마이크로니들의 설계시 많은 도움을 주리라 생각한다.

광간섭 단층 영상기술을 이용한 생체 내 microneedle 삽입 구조 영상 (High-resolution imaging of microneedles in biological tissue with optical coherence tomography)

  • Optical coherence tomography (OCT) allows non-invasive, cross-sectional optical imaging of biological tissue with high spatial resolution and acquisition speed. In principle, it is analogous to ultrasound imaging, but uses near-infrared light instead of ultrasound, measuring the time-delay of back-scattered light from within biological tissue. Compared to ultrasound imaging, it exhibits superior spatial resolution (1~10 um) and high sensitivity. Therefore, OCT has been applied to a wide range of applications such as cellular imaging, ophthalmology and cardiology. Here, we describe a novel application of OCT technology in visualizing microneedles embedded in tissue that is developed to deliver drugs into the dermis without the injection mark in the human skin. Detailed three-dimensional structural images of microneedles and biological tissues were obtained. Examining structural modification of microneedles and tissues during insertion process would enable to evaluate performance of various types of microneedles in situ.

미세다륜침을 이용한 인삼추출물의 피부개선 효과연구 (A Study on the Skin Improvement Effect of Ginseng Extract with Microneedle Therapy System)

  • Objectives : The purpose of this study was to find out the efficacy of ginseng on the skin by comparing the difference effect between only MTS (microneedle therapy system) and the ginseng solution application after MTS treatment. Methods : The present author performed once a week and total 3 times of MTS treatment. The treatment time took about 5~10 minutes and the depth of the needle was adjusted according to the participant's skin condition. In the control group, skin toner was applied after MTS and in the experimental group, ginseng extract dissolved toner was applied. One week after each treatment, the change of skin condition was measured. Two weeks after the end of the clinical trial, the skin condition was checked again. Results : In the case of t-zone oil, there was a significant decrease in the test group and in the case of pores and pigmentation, there was a significant decrease in both the test group and the control group, but the degree of significance was greater in the test group. In the case of skin tone, there was an increase, but no significance. Conclusion : The MTS treatment itself showed good effects on t-zone oil, pores, pigmentation, and skin tone. But the effect was much better when the ginseng extract solution was applied to the skin after MTS treatment. This study suggests that ginseng solution increases the efficacy of MTS.
