Due to the fast response to algae bloom issue in drinking water treatment plant, very fast determination methodology for algal toxin is required. In this study, column switching technique based online preconcentration method was combined with high resolution full scan mass spectrometer to save sample preparation time and to obtain fast and accurate result. After parameter optimization of online preconcentration, 1mL filtered sample was directly injected to trap column with switching valve system. Next, target toxins are eluted by 98% acetonitrile and analysed with 150 - 1,100 amu scan range at 50,000 resolving power. Method detection limit (MDL) for microcystin-LR, the most toxic isomer, was 0.1 ng/mL and others such as microcystin-YR, microcystin-RR and nodularin were 0.08, 0.03 and 0.04 ng/mL, respectively. This is the best improved sensitivities with 1mL volume in the literature. Furthermore, due to the use of ultra pressure HPLC (UPLC), the whole method run was completed in 4 min. Real sample applications for 173 sample including 55 surface water and 118 treatment plant samples for raw and treated water could be done within 16 hours. In our calculation, this methodology is roughly 80% faster than the previous manual solid-phase extraction with LC-MS/MS method.
Microcystins (MCs) produced by cyanobacteria are severe hepatotoxins for mammalian and protein phosphatase inhibitors. Irrigation water for grain and vegetables is often contaminated with cyanobacteria and microcystin during warm seasons. We assessed the effects of various concentrations (0, 0.01 to $10{\mu}gmL^{-1}$) of microcystin-LR (MC-LR) and microcystin-RR (MC-RR) exposure on Oryza sativa (rice) and Brassica oleraces var. italica (broccoli). The $EC_{50}$ of leaves and roots of rice was 0.9 and $1.1{\mu}gMC-LRmL^{-1}$, respectively. The no observed effect level (NOEL) of rice was less than $0.1{\mu}gmL^{-1}$ ($100{\mu}gL^{-1}$). The $EC_{50}$ of the stems and roots of broccoli was 8.7 and $7.2{\mu}gMC-RRmL^{-1}$, respectively. There was no difference in the germination rate of broccoli among microcystin-RR concentrations. After exposure to 0, 0.01 to $10{\mu}gmL^{-1}$ MC-RR for seven days, 14, 89 and 154 ng mg-1 (dry weight) MC-RR accumulated in B. oleracea. These $EC_{50}$ values showed that microcystin-LR and -RR affected the growth of rice and broccoli. These findings suggest that MC is carried into terrestrial ecosystems via irrigation, and that the biota of higher ecological niches can be influenced by MC through bioaccumulation. Therefore, a guideline for MC concentrations in irrigation water should be set using the NOEL.
This study was performed to evaluate the pollutants removal characteristics of two types of RBFs(Riverbank filtration, Riverbed filtration) intake facilities installed in Nakdong River and in Hwang River respectively. The capacity of each RBF is 45,000 m3/d for riverbank filtration intake facility and 3,500 m3/d for riverbed filtration intake facility. According to data collected in the riverbank filtration site, removal rate of each pollutant was about BOD(Biochemical Oxygen Demand) 52%, TOC(Total Organic Carbon) 57%, SS(Suspended Solids) 44%, Total coliforms 99% correspondingly. Furthermore, Microcystins(-LR,-YR,-RR) were not found in riverbank filtered water compared to surface water in Nakdong River. DOC(Dissolved Organic Carbon) and Humics which are precursors of disinfection byproduct were also reported to be removed about 59% for DOC, 65% for Humics. Based on data analysis in riverbed filtration site in Hwang River, removal rate of each contaminant reaches to BOD 33.3%, TOC 38.5%, SS 38.9%, DOC 22.2%, UV254 21.2%, Total coliforms 73.8% respectively. Additionally, microplastics were also inspected that there was no obvious removal rate in riverbed filtered water compared to surface water in Hwang River.
시아노박테리아 같은 수중 미생물에 대한 2차대사물질에 대한 연구는 육상식물이나 미생물에 관련된 연구방법을 응용하고 있으며 아직까지 체계화 되어있지 않아 시아노박테리아를 보다 효율적으로 조사하기 위한 새로운 연구기술의 모색이 절실히 필요하다. 이 연구에서는 의학적인 관점에서 시아노박테리아를 조사하기 위한 유용한 접근 방법을 모색하였고 체계화시켰다. 균주마다 특성화된 최적 배양을 하였고 PCR 증폭을 이용한 분자생물학적인 방법과 HPLC를 통한 정성분석으로 의학적으로 의미있는 NPs를 생산하는 유망 균주를 선별하였다. 선별된 균주에서 나온 추출액은 SPE와 preparative HPLC를 거쳐 분리 정제되어지고, 정제된 물질들은 질량분석기에 의해 분자량과 구조가 결정되어 졌으며, 생활성 테스트에 의해서 그 생물학적인 활성을 정함으로써 의학적인 가능성이나 활용성에 대해서 고찰하였다. 이번 시험에서 98개의 실험균주 중 46개의 균주가 NRPS 또는 PKS 관련 gene을 함유하고 있었으나 HPLC를 이용한 정성분석에서 단지 5개의 균주가 상당히 의미있는 관련 단백질을 생산하고 있음이 알려졌다. 즉, 41 균주에서 관련 gene은 존재하였지만 그 발현이 미약하였고 또는 프로모터의 활성이 이루어지지 않았다. 이와같은 휴먼 gene의 활성화는 자외선 조사 또는 건조를 이용한 자극과 배양액의 성분 조절로 이루어질 수 있고 이를 통해 흥미있는 결과를 이끌어낼 수 있다(13, 14). 이번 연구에서는 HPLC를 통한 정성분석은 Biomass에 대해서 이루어졌다. 그러므로 배양액으로 유출되는 extracellular substances에 대한 분석은 행해지지 않았다. 시아노박테리아가 NRPS/PKS 관련 유전자를 함유하고 있을 때, 특히 biomass에서 NPs 이 검출되지 않을 경우 배양액 안에서의 존재 가능성 대해서 조사할 필요성이 요구되어진다. 시아노박테리아는 대표적인 photoautotroph 미생물로써 $CO_2$를 탄소성분과 에너지원으로 사용한다. 그러므로 E. coli 같은 미생물보다 배양시 경제적이고 또한 다른 종류의 미생물이 탄소영양분의 부족과 cyanobacterial NPs의 생물학적 환성에 의해서 공생하기 어려워 옥외배양을 통한 대량생산이 가능하다. 그러나, 생산되는 cyanobacterial NPs의 높은 구조적 안정성까지 포함하여 시아노박테리아는 미래의학산업의 중요한 천연소스임에 틀림이 없지만 개체 분화의 시간이 4$\sim$10시간 정도로 다른 미생물에 비해 길고 배양시 광원의 필요성 등 한계 요소를 지니고 있다(15). 그러므로 최근에는 개체 분화가 빠른 시아노박테리아나 또는 다른 미생물에 대해서 cyanobacterial NRPS/PKS gene의 heterologous expression 이 연구되어지고 있다(16).
본 연구에서는 유전체 배리어 방전 플라즈마를 이용한 상수원의 이취미 및 독성물질 분해에 대하여 조사하였다. 이취미 물질로 지오스민(geosmin)과 2-methyl isoborneol (2-MIB)을 사용하였고, 독성물질로는 microcystin-LR (MC-LR), microcystin-RR (MC-RR), microcystin-YR (MC-YR), 그리고 anatoxin-a를 사용하였다. 플라즈마 반응기의 유입기체에 따른 분해 효율(반응시간 150 s 기준)은 지오스민의 경우 산소(100%) > 건조공기(96%) > 질소(5%) 순이었으며, 2-MIB의 경우에도 산소(100%) > 건조공기(94%) > 질소(2%) 순이었다. 이 결과는 이취미 물질이 주로 플라즈마 방전에 의해 생성된 산화성 성분, 특히 수명이 긴 오존에 의해 분해된다는 것을 나타낸다. 산소를 사용했을 때 지오스민과 2-MIB는 150 s 이내, microcystin류는 10 s 이내, anatoxin-a는 30 s 이내에 모두 분해되었다. 실제 호소수를 사용한 경우 증류수에서보다 이취미 및 독성물질의 분해효율보다 높게 나타났다.
Objectives: Microcystin (MC) produced during cyanobacterial blooms is a worldwide problem presenting a serious health threats to humans and ecosystems. During July through October of 2013, the Ilwol Reservoir experienced a high biomass of phytoplankton (maximum $211.7mg/m^3$ of Chlorophyll-a) containing the toxigenic cyanobacterium Oscillatoria sp. The aim of this study is to analyze MC concentration in the reservoir water, as well as in representative fish species (Carassius cuvieri, Carassius auratus, Channa argus). We also evaluated the human health risk of exposure to MCs accumulated in the fish. Methods: Concentrations of MCs in the water and fish samples were analyzed by liquid chromatography with a triple quadrupole tandem mass spectrometer (LC/MS/MS) and enzyme-linked immunosorbent assay (ELISA). Results: The total levels of four MC variants, including MC-LR, MC-RR, MC-YR and MC-LA were below the WHO drinking water guideline limit (1 ug MC-LR per liter) both for the dissolved and particulate fraction present in the water samples. The mean MC concentrations in the livers of all species were significantly higher than in the gills (p < 0.01) and muscles (p < 0.05). The values of estimated daily intake of MCs in muscles, the edible part of the fish, would be only $0.005-0.015{\mu}g/kg{\cdot}day$, much lower than WHO's provisional tolerable daily intake of $0.04{\mu}g/kg{\cdot}day$. Conclusion: This study suggests that, owing to the spatial distribution or temporal variation of MC, there is a need for careful monitoring of cyanotoxin in reservoir water and aquatic animals to protect public health.
호소, 하천 등에 다량의 영양염류의 유입과 수문학적, 지리학적, 생물학적 요소 등으로 인해 남조류가 대량 발생하게 되며 대표적 독성물질인 마이크로시스틴-LR(MC-LR)이 증가한다. MC-LR이 포함된 지표수를 적절하게 처리하기 위하여 정수처리공정에서는 고도산화공정을 적용하고 있다. 다양한 고도산화공정 중 특히 UV, $UV/H_2O_2$, $UV/O_3$, $UV/TiO_2$ 등에 대한 연구는 꾸준히 되어왔다. 기존의 UV램프의 짧은 교체주기, 수은 폐기물 발생, 큰 열 손실 등의 단점을 보완한 UV-LED를 MC-LR제거에 적용하였다. MC-LR 초기농도 $100{\mu}g/L$을 280 nm의 파장인 LED-L ($0.024mW/cm^2$)와 LED-H ($2.18mW/cm^2$)를 이용하여 산화시켰을 때 각각 최대 약 30%, 95.9%의 MC-LR 제거율을 나타냈다. LED-H를 조사 시 자연유기물 변화는 휴믹물질, UVD, SUVA가 감소하는 경향을 보였고 방향족 유기물이 지방족 유기물로 분해되어 저분자 물질이 되었다. $LED-H/H_2O_2$($H_2O_2$: 1, 2, 5, 10 mg/L)산화반응에 의한 MC-LR제거율은 LED-H 단독에 의한 MC-LR 제거율과 유사하였다. 남조류가 발생한 낙동강 원수를 대상으로 LED-L산화를 적용하여 수질분석을 통하여 특성변화를 확인하였다. DOC 및 TOC의 변화는 거의 없었으나 SUVA와 $UV_{254}$의 감소로 인하여 유기물의 분해되었으며 조류유래물질인 용존 및 총 MC-LR, geosmin, 2-MIB농도가 시간이 지남에 따라 서서히 감소하였다.
