This study was conducted to evaluate the sanitation level and use of dish cloths, chopping board, and drawer in the refrigerator in 50 university students' houses located in Seoul and Kyonggido. The survey questionnaire evaluated general information, frequency of the washing, cleaning method, drying method and the associated factors. The highest percentage of the cleaning and drying methods of dish cloths were “boiling(76.0%)”, and “dry in the kitchen or on the sink(78.0%)”, respectively. For the chopping board and drawer in the refrigerator, the highest percentage of cleaning method was “use detergent” and drying method was “dry in the kitchen”. The mean of coliforms for dish cloths, chopping board, and drawer in the refrigerator was 4.46${\pm}$1.95 log CFU/100 $cm^2$, 3.92${\pm}$1.64 log CFU/100 $cm^2$, 3.48${\pm}$2.01 log CFU/ 100 $cm^2$, respectively. Salmonella spp. were not detected from all samples. But E. coli were detected from dish cloths(4.0%) and S. aureus were detected from dish cloths(16.0%), chopping board(12.0%), and drawer in the refrigerator(8.0%). The results of microbiological test of kitchen utensils indicated that the sanitary conditions of dish cloths, chopping board, drawer in the refrigerator should be improve promptly.

Saccharomyces fermentati and Leuconostoc mesenteroides were isolated from a traditional kimchi, and then the Leu. mesenteroides was mutated to the acid-tolerant mutant Leu. mesenteroides M-l00. In the result of growth properties in MRS broth with various pHs adjusted with HCl and acid solution (latic acid:acetic acid=1:2), an acid-tolerant mutant Leu. mesenteroides M-100 showed more increased ability for growth than its wild strain at various temperatures. The strains were used as starters for the fermentation of kimchi. The experiments were performed with classified experimental groups (Group I, control kimchi; Group II, addition of YK-19 only; Group III, addition of M-100 only; Group IV, addition of mixture of M-100 and YK-19), and their pH, total acidity, reducing sugars content, organic acid productivity, organoleptic tests, and microfloral changes were compared. As a result, in pH and acidity, the optimal ripening period of Group IV was about 13.5 days (i.e. from the 8.5 to 22 days of fermentation). This result indicates that the optimal ripening period of Group IV was about 1.5 times longer than that of Group I. Furthermore, Group IV was edible to 28 days of fermentation. In addition, high contents of succinc acid was observed in Group IV. Group IV was also highly ranked on the organoleptic test. During the fermentation of kimchi, the number of Leuconostoc sp. in group I reduced after 7 days; however, the number of Leuconostoc sp. in Group II, III, and IV decresed after 14 days of fermentation. An especially high number of Leu. sp. was observed in Group IV, and this gave better flavor of kimchi than any other during the whole fermentation period. Citric acid, tartaric acid, succinic acid, fumaric acid, and lactic acid were detected in the kimchi, and a significant increase in the concentration of lactic acid during fermentation was observed in the all experimental groups.
The purpose of this study was to assess the clinical and microbiological effect of chlorhexidine rinse following scaling and root planing on periodontits. 10 patients with periodontal disease were selected for the study. They had not taken antibiotics for months and no history of dental treatment for 6 months before the study. They were good in general health. Patients received a scaling and root planing under local infiltration anesthesia, chlorhexidine rise group were subjected to twice a day 0.1% chlorhexidine rinse for a period 2 week. After initial clinical(plaque index, gingival index, probing pocket depth), microbiological and BANA tests were determined, each subject received a single session of scaling and root planing but no oral hygiene instructions. Clinical indices were measured, microbial parameters and BANA test were reassessed 1, 2 and 4 weeks after treatment. The results were as follows : 1. Plaque index, gingival index and pocket depth in chlorhexidine rinse group and control group were not significantly reduce during all weeks when compared chlorhexidine rinse group with control groups. Plaque index in chlorhexidine rinse group and control group were siginificantly reduced at 1, 2, 4weeks(P<0.05), gingival index and pocket depth wee ignificantly reduced at 2, 4weeks in both groups(P<0.05). 2. Perecntage of cocci and motile rods was significantly changed at 1, 2, 4weeks in chlorhexidine rinse group(P<0.05), control group was significantly changed at 4weeks in control group(P<0.05), intergroup difference was significantly at 2weeks in cocci and 4weeks in motile rods(P<0.05). 3. Percentage of non-motile rods in all group were not significantly changed when compared with those of baseline. 4. Percentage of spirochetes was significantly reduced at 4 week(P<0.05), control group was not significantly reduced during all weeks. 5. BANA test scores was significantly reduced during all weeks in chlorhexidine rinse group(P<0.05), control group was not significantly reduced during all weeks. The result showed that clinical and microbiological effect following scaling, root palning and chlorhexidine on periodontal disease.
Purpose: The purpose of this study was to investigate the clinical outcomes of non-carbapenem treatment for urinary tract infections (UTIs) caused by extended-spectrum β-lactamase (ESBL)-producing Escherichia coli (E. coli) in young children. Methods: We retrospectively reviewed the medical records of children under 2 years of age who were diagnosed and treated for UTIs caused by ESBL-producing E. coli from September 2014 to March 2020. Results: Forty-three children under 2 years of age were treated with non-carbapenem antimicrobials for UTIs caused by ESBL-producing E. coli without bloodstream infections. The overall clinical and microbiological success rates for empirical antimicrobial treatment were 90.7% and 97.7%. Three of the patients (7.0%) experienced a relapse of UTI within a month. An in vitro susceptibility test showed that two patients were sensitive and one was resistant to the antimicrobial treatments. Furthermore, there were no significant differences in the time to defervescence, clinical success, microbiological success, and relapse rate between the susceptible (n=13) and non-susceptible groups (n=30). Conclusion: In this study, the overall relapse rate of patients treated with non-carbapenem antimicrobials was 7.0%. The patients showed high success rates in the clinical and microbiological responses to the non-carbapenems regardless of the results of the in vitro antimicrobial susceptibility test. These results provide evidence that non-carbapenems may be viable alternative treatments for UTIs caused by ESBL-producing E. coli.
Transporting clinical samples for microbiological testing requires a proper transport medium that guarantees the survival of microorganisms. Therefore, the aim of the study was to determine the ability of Amies Transport Medium (ATM) to maintain the viability of microorganisms in clinical specimens and its suitability as a transport medium for microbiological testing. This study evaluated the performance of swab provided by KS Co., Ltd. for three groups of bacteria comprising aerobic and facultative anaerobic bacteria, anaerobic bacteria, and fastidious bacteria, according to the Clinical and Laboratory Standard Institute (CLSI) 8.11.2. The ATM stability test was conducted by dividing the medium into two groups based on the product expiration date of use. All tested media, A and B (the date of manufacture and expiration date are different) showed ≥5 CFU, and there was no significant difference in the result values of Category A and Category B with different serial numbers for each test. The results of this experiment when cross-checked with the guidelines suggest that ATM is a suitable transport medium for microbiological testing, as it maintains the viability of microorganisms and is suitable for overgrowth trials. In addition, compared to the number of CFUs at the origin, the number of CFUs did not increase by more than 1 log after storage. These results have important implications for the development of transport media that can guarantee the survival of microorganisms in clinical specimens.
Rotting bacteria were isolated from decayed root rot of ginseng (Panax ginseng C.A. Meyer), cultured purely, and it's pathogenicity was confirmed by reinoculation test. Two strains (E3, E7) were selected to be tested in more detailed study with respect to their morphological, cutural and biochemical characters. The strains causing red rot of ginsneg were identified as Erwinia carotovora.
The folic acid contents of 31 kinds of the common food stuffs harvested in Korea has been determined by microbiological assay with Streptococcus faecalis "R" ATCC 8043 as the test microorganism. The results of the determination are as shown in followed table.wed table.
Various antimicrobial drug screen tests have been used in order to ensure food safety. However, the conventional screen tests, the Swab Test on Premises(STOP, USA), the Calf Antibiotic and Sulfa Test(CAST, USA) and the European Economic Community 4-plate Test(FPT, EU) are not sufficiently rapid or sensitive enough to detect low levels of sulfa drugs in meat. We developed a new screen test kit for the determination of the antimicrobial residues in meat called the Bacillus megaterium Disk Assay(BmDA). A comparison of BmDA with the older screen tests showed BmDA was as good as the older ones with several advantages. The new test kit is faster-it can be read in 4∼6 hours instead of 16∼18 hours. Moreover, BmDA can discriminate sulfa drugs from other antimicrobial drugs because p-aminobenzoic acid countacts the inhibiting action of sulfa drugs. Minimum detectable levels of sulfa drugs were significantly improved at the lever of 0.025*0.1 pp, compared with the level of 1.0 ppm in FPT. A comparison of BmDA with the older screen tests in HPLC confirmed meat samples exceeded the Korean tolerance value of 0.1 ppm showed BmDA was the most sensitive in the microbiological screen tests. As the microbiological screen tests have already known, a person familiar with simple laboratory techniques should have no difficulty in using it to detect antimicrobial residues in meat. This would be a simple, economic method of antimicrobial residues detection which might be succesfully used by many laboratories.
A restriction endonuclease, Kpn I has been isolated from Klebsiella pneumonia. Cells were broken by sonication. After ultracentrifugation the supernatant containing Kpn I activity was further purified by Sepharose-6B gel filtration, DEAD-Cellulose, Heparin-Agarose, and Aminohexyl-Agarose column chromatography. Final enzyme preparation was essentially free of contamination exonuclease and phosphatase, as judged by ligation-recut test. Total activity of the enzyme recovered from 10 grams of cells was $4.7\times 10^5$ units.
The objective of the present study was to evaluate the clinical and microbiological effects of scaling and root planing combined with local application of 2% minocycline gel to patients with moderate to advanced chronic adult periodontitis. 27 healthy patients with moderate to advanced chronic adult periodontitis were enrolled in the study. The quadrants that had 2 or more teeth with $5{\sim}8mm$ probing pocket depth and radiographic evidence of alveolar bone loss were selected and divided into test side and control side according to the splitmouth design. All patients received standardized oral hygiene instructions at the beginning of the study. Subsequently scaling and root plaining was performed on all remaining teeth until 0 week. The 2% minocycline gel was applied to periodontal pocket at 0, 1, 2, 3week in the test side. The normal saline was irrigated subgingivally for about 30 seconds in the control side. The clinical and microbiological analysis was carried out at 0, 4, 8, and 12weeks. The results of this st udy were as follows; 1.2% minocycline gel delivered subgingivally as an adjunct to scaling and root planing provided benefit in reducing sulcular bleeding index and pocket depth than the use of normal saline. 2. The relative proportion of cocci and non-motile bacteria was increased in the test and control groups with time, and there was no statistically significantdifference between two groups. 3. The proportion of spirochetes was slowly reduced in the control group, but, inthe test group, they were remarkably reduced from the 4th week, and there was a statistically significant difference between two groups. 4. In both groups, the relative proportion of motile rods was notably decreasedat the beginning of the study, and remained until 12th week in the test group,but, in the control group, they were slowly increased from the 4th weekand finally similar to that of the initial examination. In conclusion, local application of 2% minocycline gel may be effective in the clinical and microbiological aspects as an adjunct to scaling and root planing in periodontal disease sites.
