전통 장류 유래 Bacillus subtilis SCJ4의 특성확인 및 통계학적 방법을 이용한 배양조건 최적화 (Identification of characterization and statistical optimization of medium constituent for Bacillus subtilis SCJ4 isolated from Korean traditional fermented food)

  • 순창군에서 제조된 전통 장류로부터 612종의 다양한 미생물을 분리하였고, 612종의 미생물 중에서 생리활성 분석 및 biogenic amine 생성여부를 조사하여 최종적으로 Bacillus subtilis SCJ4를 선별하였다. 최종 선별한 균주 SCJ4의 생화학적 특성과 16S rRNA 염기서열 분석에 의해 B. subtilis SCJ4로 명명하였으며, B. subtilis SCJ4의 산업적으로 사용이 적합한 최적배지조성을 확립하기 위하여 통계학적 방법인 반응표면 분석법을 사용하였다. 실험계획법은 배지조성을 예측하기 위해 Plackett-Burman design을 사용하여 모델을 설정하였다. 실험 모델을 통하여 선별된 배지 조성으로는 tryptone, peptone, $MgSO_4$로 예측되었으며, 배지조성에 대한 농도를 설정하기 위해 중심합성계획법을 사용하여 최적화를 수행하였다. 최종 예측된 배지 조성의 농도는 tryptone 15.35 g/L, peptone 12.235 g/L, $MgSO_4$ 3.5 g/L로 예측되었다. 이때 최대 균체량은 1.07497 g/L로 예측되었으며, 모델의 검증실험을 통하여 예측된 모델과 실제 결과가 동일함을 확인하였으며, 기본배양배지와 비교하였을 때 균체량이 0.8767 g/L에서 1.1222 g/L으로 약 28%증가하였다.

오디 식초의 품질 특성 및 생리활성 (Quality characteristics and physiological activities of mulberry (Morus alba) vinegar)

  • 본 연구에서는 고부가가치 오디 식초를 개발하기 위하여 식품 미생물인 효모와 초산균을 사용하여 오디를 발효하고, 품질의 표준화는 물론 우수한 생리기능성을 함께 갖춘 식초를 제조하고자 하였다. 오디의 최적 발효균주를 선발하기 위하여 균주를 달리하여 발효를 진행하고 품질 특성 및 생리활성을 측정하였으며, 오디 발효 시 알코올과 초산 생성능이 뛰어난 S. cerevisiae SRCM101756과 A. pasteurianus SRCM102419를 발효용 균주로 최종 선발하였다. 제조된 오디 와인에 선발된 초산균을 사용하여 오디 식초를 제조하고 품질 특성 및 생리활성을 분석하였다. 오디 식초는 발효 9일 차에 pH 2.98, 총산도 4.70%로 측정되었고, 𝛼-glucosidase 저해 활성은 100배 희석 시에 발효 전 13.22%에서 발효 후 19.19%로, 50배 희석 시에 발효 전 42.35%에서 발효 후 46.11%로 활성이 증가하는 것을 확인하였다. Angiotensin-converting enzyme 저해 활성을 측정한 결과, 25배 희석 시에 발효 전 44.82%에서 발효 후 63.88%로, 50배 희석 시에 발효 전 30.10%에서 발효 후 37.24%로 angiotensin-converting enzyme 저해 활성이 증가하였고, 발효 전에 비하여 발효 후 pancreatic lipase 저해 활성도 유의적으로 증가한 것으로 나타났다. 50배 희석 시료에서 발효 전 30.01%에서 발효 후 40.25%로, 25배 시료에서는 발효 전 42.51%에서 발효 후 55.33%로 그 활성이 증가하는 것을 확인하였다. 이와 같은 결과를 토대로 오디식초는 식품미생물을 이용한 발효를 통하여 생리활성이 증진된 고부가가치 식품이며, 향후 다양한 소재로도 활용될 수 있을 것으로 판단된다.

Production of cis, cis-Muconic Acid from Benzoic Acid via Microbial Transformation

  • For the production of cis, cis-muconic acid via biocatalytic conversion reactions from a toxic cosubstrate, benzoic acid, a fed-batch process using computer-controlled DO-stat feeding was developed. The mutant strain of Pseudomonas putida BM014 produced cis, cis-muconic acid from benzoic acid with high conversion yield. More than 32 g/L of cis, cis-muconic acid was accumulated in 42h and a productivity of 1.4g/(L.h)was achieved.

Parasporin-4, A Novel Cancer Cell-killing Protein Produced by Bacillus thuringiensis

  • Bacillus thuringiensis was isolated as a pathogen of the sotto disease of silkmoth larvae about a hundred years ago. Since then, this bacterium has attracted attentions of not only insect pathologists but also many other scientists who are interested in its strong and specific insecticidal activity. This has led to the recent worldwide development of B. thuringiensis-based microbial insecticides and insect-resistant transgenic plants, as well as a landmark discovery of par asp orin, a cancer cell-specific cytotoxin produced by B. thuringiensis. In this review, we describe examination of interaction between inclusion proteins of B. thuringiensis and brush border membrane of insects using a surface plasmon resonance-based biosensor, identification and characterization of parasporin-4, the latest parasporin produced by the B. thuringiensis A1470 strain, and an effective method for preparing the parasporin-4 from inclusion bodies expressed in the recombinant Escherichia coli cells.

유류분해 미생물의 특성 및 제제화 가능성 평가

  • The various microbial tests were performed to determine bioremediation agent capacity for eight strains isolated from the oil contaminated regions. Two tests for isolated strains were conducted such as cell hydrophobicity and emulsifying activity. The biodegradation of SHM (saturated hydrocarbon mixture) and AHM (aromatic hydrocarbon mixture) with the strains also was carried out. The strains having higher cell hydrophobicity and emulsifying activity degraded petroleum oil effectively. The degradation capacity for SHM was represented more than 90% in YS-7 and WLH-1 of isolated strains, and KH3-2 were capable of degrading AHM. Especially, WLH-1 as yeast was shown more than two or three times in the degradation capacity of automobile engine lubricants and the biomonitoring results of contaminated soil for residual oil degrading test showed that the hydrocarbon biodegradation was increased in the second treatment by this strain.

Leuconostoc mesenteroides CBI-110에 의한 Alternan의 생산 (Production of Alternan by Leuconostoc mesenteroides CBI-110)

  • Alternan known as a kind of dextran is a linear and cyclic polysaccharide which is synthesized with sucrose by alternaansucrase (EC2.4.1.140) of Leuconostoc mesenteroides. But this polysaccharide is different from dextran in its structure and other properties. For the purpose of applying alternan for the food, cosmetic, and pharmaceutical industries, we isolated an excellent alternan-porducing strain, Leu. mesenteroides CBI-110. The optimum culture conditions were studied to improve the yield, and the activity of alternansucrase was increased up to 33U/ml in the optimum culture media. The concentrations of $Mn^{2+}$ and $Cu^{2+}$ ions were 0.01% and 0.03%, respectively, for the effective production of alternan. Finally, we obtained 25.6g/L of alternan.

Microbial Production and Characterization of Superparamagnetic Magnetite Nanoparticles by Shewanella sp. HN-41

  • A facultative dissimilatory metal-reducing bacterium, Shewanella sp. strain HN-41, was used to produce magnetite nanoparticles from a precursor, poorly crystalline iron-oxyhydroxide akaganeite ($\beta$-FeOOH), by reducing Fe(III). The diameter of the biogenic magnetite nanoparticles ranged from 26 nm to 38 nm, characterized by dynamic light scattering spectrophotometry. The magnetite nanoparticles consisted of mostly uniformly shaped spheres, which were identified by electron microscopy. The magnetometry revealed the superparamagnetic property of the magnetic nanoparticles. The atomic structure of the biogenic magnetite, which was determined by extended X-ray absorption fine structure spectroscopic analysis, showed similar atomic structural parameters, such as atomic distances and coordinations, to typical magnetite mineral.

식물 뿌리썩음병을 유발하는 Fusarium solani에 대한 Pseudomonas 속의 생물학적 방제기작 (the mechanism of Biological Control of Pseudomonas spp.Against Fusarium solani Causing Plant Root-Rot Disease)

  • The 543 microbial strains were isolated from the rhizosphere in ginseng root rot-suppressive soil. From these, isolate KGM-100 was finally selected for the screening of powerful antagonistic bacterium for the soil-born Fusarium solani causing root rot of many important crops. The isolate KGM-100 was identified as Pseudomonas aeruginosa. Antibiotics produced by the P. aeruginosa KGM-100 were partially characterized as heat-stable and low-molecular weight antibiotics. The strain also produced siderophore, which was assumed to be pseudobactin. Pot test showed approximately 30-50% biological control effects when Fusarium solani-infected soil was treated with the P. aeruginosa KGM-100.

Streptomyces Showing Antifungal Activities against Six Plant Pathogenic Fungi

  • Screening tests against six plant pathogenic fungi were performed in order to develop biopesticides. Actinomycetes were used to discriminate Bacillus thuringiensis for wide use as a microbial pesticide. From more than 100 actinomycetes tested, twelve strains showed potent antifungal activities. We report in vivo screening results from fermentation broths of these twelve strains and identification of the strain taxa.
