Green manure crops have been well recognized as the alternative for chemical fertilizer, especially N fertilizer, because of its positive effect on soil and the environment. Hairy vetch and green barley are the most popular crops for cultivation of rice in paddy field. This study was conducted to evaluate effects of hairy vetch and green barley on soil microbial community and chemical properties during short-term application (three years). For this study, treatments were composed of hairy vetch (Hv), green barley (Gb), hairy vetch + green barley (Hv+Gb), and chemical fertilizer without green manure crops (Con.). Hv+Gb treatment showed the highest microbial biomass among treatments. Principal component analysis (PCA) showed that PC1 (73.0 %) was affected by microbial biomass and PC2 (21.5 %) was affected by fungi, cy19:0/18:$1{\omega}7c$ (stress indicator). Combined treatment with hairy vetch and green barley could be more efficient than green manure crop treatment as well as chemical fertilizer treatment for improvement of soil microorganisms.

Climate change is more rapid in the Arctic than elsewhere in the world, and increased precipitation and warming are expected cause changes in biogeochemical processes due to altered microbial communities and activities. It is crucial to investigate microbial responses to climate change to understand changes in carbon and nitrogen dynamics. We investigated the effects of increased temperature and precipitation on microbial biomass and community structure in dry tundra using two depths of soil samples (organic and mineral layers) under four treatments (control, warming, increased precipitation, and warming with increased precipitation) during the growing season (June-September) in Cambridge Bay, Canada (69°N, 105°W). A phospholipid fatty acid (PLFA) analysis method was applied to detect active microorganisms and distinguish major functional groups (e.g., fungi and bacteria) with different roles in organic matter decomposition. The soil layers featured different biomass and community structure; ratios of fungal/bacterial and gram-positive/-negative bacteria were higher in the mineral layer, possibly connected to low substrate quality. Increased temperature and precipitation had no effect in either layer, possibly due to the relatively short treatment period (seven years) or the ecosystem type. Mostly, sampling times did not affect PLFAs in the organic layer, but June mineral soil samples showed higher contents of total PLFAs and PLFA biomarkers for bacteria and fungi than those in other months. Despite the lack of response found in this investigation, long-term monitoring of these communities should be maintained because of the slow response times of vegetation and other parameters in high-Arctic ecosystems.
The relationships among the contents of Pb, Zn, Cd, Cu and microbial biomass and activity of soil were investigated in Kyongbu, Yongdong, and Chungbu highways. The heavy metal concentrations were the highest in Kyongbu highway with high traffic density. The levels of these metals in soil in three highway sites were much higher than Mt. Kwank as control site. The highest concentrations of heavy metals were found in the upper layer of soil adjacent to the roadside. Dehydrogenase activity (DHA) and adenosine tri-phosphate (ATP) contents were generally lowest at distance of 0.5 m from the roadside. Simple regression analysis indicated that DHA and ATP contents were highly negatively correlated with Pb and Zn concentrations.
The long-term effects of soil management history on microbial communities are still poorly understood. Our objectives were to determine the impact of long-term application of soil amendments on microbial communities in rice paddy fields. The treatments selected were control where crops were grown without any nutrient application (CON); nitrogen-phosphorus-potassium (NPK); NPK plus compost (CNPK); NPK plus lime (LNPK); and NPK plus silicate (WNPK). The long-term addition of organic and inorganic amendments significantly changed soil chemical properties. The amount of organic carbon increased in the treatments with fertilizer and amendments over that in the soil without inputs. However, we could not observe the differences of bacterial population among the treatments, but the number of aerobic bacteria increased by the addition of amendments. Isolates from the rice paddy soils before irrigation were Dactylosporangium, Ewingella, Geobacillus, Kocuria, Kurthia, Kytococcus, Lechevalieria, Micrococcus, Micromonospora, Paenibacillus, Pedobacter, Pseudomonas, Pseudoxanthomonas, Rhodococcus, Rothia, Sphingopyxis, Stenotrophomonas, and Variovorax. Dominant genera were Arthrobacter, Kocuria, Kurthia, and Bacillus in the long-term field. Microbial biomass was the highest in the compost treatment (CNPK), and was the lowest in the CON. Dehydrogenase activity in soils treated with rice compost straw was the highest and the activity showed an increasing trend according to treatment as follows: CON < WNPK < NPK = LNPK < CNPK. These results demonstrate that soil management practice, such as optimal application of fertilizer and amendment, that result in accumulations of organic carbon may increase microbial biomass and dehydrogenase activity in long-term rice paddy soils.
Autotrophic nitrogen removal and its microbial community from a laboratory scale upflow anaerobic sludge bed reactor were characterized with dynamic behavior of nitrogen removal and sequencing result of molecular technique (DNA extraction, PCR and amplification of 16S rDNA), respectively. In the experiment treating inorganic wastewater, the anaerobic granular sludge from a full-scale UASB reactor treating industrial wastewater was inoculated as seed biomass. The operating results revealed that an addition of hydroxylamine would result in lithoautotrophic ammonium oxidation to nitrite/nitrate, and also hydrazine would play an important role for the success of sustainable nitrogen removal process. Total N and ammonium removal of 48% and 92% was observed, corresponding to nitrogen conversion of 0.023 g N/L-d. The reddish brown-colored granular sludge with a diameter of $1{\sim}2\;mm$ was observed at the lower part of sludge bed. The microbial characterization suggests that an anoxic ammonium oxidizer and an anoxic denitrifying autotrophic nitrifier contribute mainly to the nitrogen removal in the reactor. The results revealed the feasibility on development of high performance lithoautotrophic nitrogen removal process with its microbial granulation.

soil microbial activities and diversities in a newly reclaimed soil. Soil chemical properties, population of microbe, microbial biomass, and properties of microbial community were investigated under 4 different treatment (animal manure compost+green manure, chemical fertilizer, and without fertilizer). The experiment was conducted for 3 years from 2012 to 2014. The most of chemical properties in the animal manure compost+green manure treatment were increased continually compare to chemical fertilizer and without fertilizer. The population of bacteria and fungi were higher in the animal manure compost+green manure treatment, however, there was no difference on actinomyces. Soil microbial biomass C content was higher in the animal manure compost+green manure treatment than in chemical fertilizer and without fertilizer. Biolog examination showed that catabolic diversities of bacterial communities were higher in the treatment of animal manure compost+green manure. It was showed that principle component analysis of the Biolog data differentiated the organic matter amended soils from NPK and control. These results indicated that application of animal manure compost+green manure had a beneficial effect on soil microbial properties.
This study was conducted to investigate the effect of different surface soil management practices on soil microflora in volcanic ash soils of citrus orchard. Soil samples were collected from citrus orchards of clean cultivation, grass sod, and grass mulch system in May and September 1997. Soil chemical properties, populations of various microorganisms, enzyme activities, microbial biomass C were analyzed. Average soil pH were 4.7, and average nitrogen and organic matter contents were 6 and $140.2g\;kg^{-1}$, respectively. Aerobic bacteria were distributed at $26,2-47.3{\times}10^6cfu\;g^{-1}$ level. Among the aerobic bacteria Pseudomonas spp., Rhizobium spp., and thermophilic Bacillus spp. were dominant in most of the investigated orchard soils. Density of actinomycetes were low at $1.8-84.6{\times}10^5cfu\;g^{-1}$ level. Fungi were distributed at $26.4-182.1{\times}10^5cfu\;g^{-1}$ level and the density was higher in grass mulch and sward sites. In september, phosphomonoesterase activity was high at $239.6{\mu}g\;PNP\;g\;soil^{-1}\;h^{-1}$ in clean cultivated citrus orchards. Soil cellulase activity were higher at $602.6{\mu}g\;GE\;g\;soil^{-1}$\;24\;h^{-1}$ in grass sward cultivation than any other soil management practices. Soil microbial biomass C was higher in grass mulch cultivated orchards.
To develop indicators for soil health evaluation, biological characteristics of native soils from the different parent materials were studied. Survival rate of fluorescent Pseudomonas in soils was the lowest as 0.1% while those of thermophilic Bacillus and alkaliphilic bacteria were over the 90% by the soil drying stress. There was positive relationship between soil microbial biomass and organic carbon exudated from the microbial biomass by the treatment. The average air-drying effect of soils was 39.7% with ranges of 9.7~95.0%. The propagules of mesophilic Bacillus and Gram negative bacteria were increased by the re-wetting of dried soils. Soil pH affected positively to the recovering rate of microbial number. Average recovering rate of microbes was 65.3%, and there was positive relationship between microbial biomass recovery and fluorescent Pseudomonas population.
In order to understand the role of heterotrophic protists in the coastal waters off Inchon, abiotic and biotic factors were measured from January 1992 to February 1993. Microbial carbon biomass (mean212.9$^{\pm}$119.1 $^{\mu}$gC/1) was composed of 4.2% bacteria, 0.3% cyanobacteria, 12.l% autotrophic nanoflagellates, 6.6% heterotrophic nanoflagellates, 5.8 heterotrophic ciliates and 71.0% diatom and Mesodinium spp. The carbon biomass of heterotrophic protists (heterotrophic nanoflagellates and ciliates) was highest in October 1992 (mean 37.8$^{\pm}$22.5 $^{\mu}$gC/1), and was low in August 1992 (mean 21.2$^{\pm}$10.8 $^{\mu}$gC/1) and in February 1993 (mean 19.5$^{\pm}$6.4 $^{\mu}$gC/1). However, the contribution of heterotrophic protists to total microbial carbon biomass was higher in January 1992 and February 1993 (about 21%) when the phytoplankton was dominated by nanoplankton than in August and October (about 9%) when large diatoms occurred in large numbers. This study suggests that in Kyeonggi Bay heterotrophic protists might play a more important role as prey for zooplankton and as consumers of bacteria & small phytoplankton in less productive seasons (especially winter) than in productive seasons (autumn), and that the classic trophic pathway from diatoms through copepods to fish might be dominant nearly every season.
Fertilization effects on changes in soil $CO_2$ flux and organic C in switchgrass (Panicum virgatum L.) land managed for biomass production were investigated. The mean daily soil $CO_2$ flux in the manure treatment was 5.63 g $CO_2-C\;m^{-2}\;d^{-1}$, and this was significantly higher than the mean value of 3.36 g $CO_2-C\;m^{-2}\;d^{-1}$ in the control. The mean daily $CO_2$ fluxes in N and P fertilizer treatments plots were not different when compared to the value in the control plots. Potentially mineralizable C (PMC), soil microbial biomass C (SMBC), and particulate organic C (POC) were highest at the 0 to 10 cm depth of the manure treatment. Potentially mineralizable C had the strongest correlation with SMBC (r = 0.91) and POC (r = 0.84). There was also a strong correlation between SMBC and POC (r = 0.90). Our results indicated that for the N and P levels studied, fertilization had no impact on temporal changes in soil organic C, but manure application had a significant impact on temporal changes in soil $CO_2$ evolution and active C constituents such as PMC, SMBC, and POC.
