Nitrite plays a major role in inhibiting the growth of foodborne pathogens, including Clostridium botulinum (C. botulinum) that causes botulism, a life-threatening disease. Nitrite serves as a color-fixing agent in processed meat products. However, N-nitroso compounds can be produced from nitrite, which are considered as carcinogens. Thus, consumers desire processed meat products that contain lower concentrations (below conventional concentrations of products) of nitrite or no nitrite at all, although the portion of nitrite intake by processed meat consumption in total nitrite intake is very low. However, lower nitrite levels might expose consumers to risk of botulism poisoning due to C. botulinum or illness caused by other foodborne pathogens. Hence, lower nitrite concentrations in combination with other factors such as low pH, high sodium chloride level, and others have been recommended to decrease the risk of food poisoning. In addition, natural compounds that can inhibit bacterial growth and function as color-fixing agents have been developed to replace nitrite in processed meat products. However, their antibotulinal effects have not been fully clarified. Therefore, to have processed meat products with lower nitrite concentrations, low pH, high sodium chloride concentration, and others should also be applied together. Before using natural compounds as replacement of nitrite, their antibotulinal activities should be examined.
This study aimed to evaluate the antimicrobial effects of Acanthopanax sessiliflorum fruit (ASF; Ogaza) extracts on Streptococcus mutans and Streptococcus sobrinus, which are agents that cause dental caries, and on Streptococcus mitis and Streptococcus salivarius, the microbial flora of the oral cavity. The ASF extracts obtained using 70% ethanol were fractionated in the order of ethyl acetate and n-Butanol, concentrated under reduced pressure, and lyophilized to give powdery solvent extracts. The antimicrobial activity of ASF extracts from each solvent was examined using the disk diffusion method. As a result, only those extracts obtained using an ethyl acetate solvent showed antimicrobial activity. These extracts were selected, and the minimum inhibitory concentration was measured by disk diffusion method at various extract concentrations. Results showed a minimum inhibitory concentration of 32 mg/ml. The viable cell count was measured to confirm the minimum bactericidal concentration. Results showed a minimum bactericidal concentration of 64 mg/ml. In the cytotoxicity test using normal human dermal fibroblast cells, the absorbance value of the test group was similar to that of the control group at 0.64, 1.28, and 6.4 mg/ml. The bacteria and their colonies were examined using a scanning electron microscope. Boundaries between the antimicrobial activity region and non-antimicrobial activity region were observed around the paper disk, which was immersed in the extract with 32 mg/ml concentration. Bacterial colonization was not observed in the area with antimicrobial activity. This finding suggests that ASF extracts can inhibit the growth of some microorganisms in the oral cavity, in addition to the effects of these extracts known to date. In particular, ASF extracts may be used as a preparation for preventing dental caries by adding the extract to the toothpaste or oral mouthwash.
곤충병원성 곰팡이 B. bassiana KK5균의 포자 발아 촉진제를 조사하기 위한 연구가 수행되었다. 탄수화물을 포함한 18 화합물을 B. bassiana 포자와 혼합하여 12시간 동안 water agar에서 배양하였다. 18종의 화합물 중 fructose, mannose 그리고 skim milk와 혼합된 포자의 발아율은 배양 12시간 후 무처리와 비교하여 촉진되는 것으로 조사되었다. 발아 촉진 효과가 확인된 1% fructose, mannose 또는 skim milk와 포자 혼합액을 진딧물에 살포 하였을 때, skim milk와 포자 혼합액의 처리에서 복숭아혹진딧물 3령 약충에 대한 살충증진 효과를 보였다.
동물은 고기, 우유, 및 달걀과 같은 중요한 산물을 생산한다. 현대사회의 도시화과정 및 고도 성장은 보다 많은 동물성 산물을 요구하고 작은 농장에서 밀집화된 사육이 이루어지게 되는 원인이 되었다. 결과적으로 가축 분뇨생산량이 증가되었으며, 그에 따른 악취문제도 필연적인 결과로 나타났다. 분뇨로 인한 악취문제를 해결하지 못할 경우 축산업의 지속적인 발전과 환경개선을 이루는 큰 장애가 될 것이다. 축산환경으로 생산되는 악취는 휘발성지방산, 암모니아와 휘발성아민, 인돌과 페놀, 그리고 황화합물 네 종류로 분류된다. 질소성분의 절반이상은 뇨로 배설되기 때문에 동물을 사육하는 과정에서 저단백 사료와 합성아미노산을 급여시키고, 소화효소나 미생물제제 혹은 생균제를 사료에 첨가시켜줄 경우 질소원의 배출량을 감소시켜줄 수 있는 중요한 방법이 될 것이다. 따라서, 축산환경의 개선과 축산분야의 생산성을 향상시켜주기 위하여 사료 및 보조사료를 급여하는 다양한 시도가 진행되고 있다.
Stimulator of interferon gene (STING) is induced by various inflammatory agents, such as lipopolysaccharide and microbial pathogens, including virus and bacteria. In this study, we obtained a full-length cDNA of a STING homolog from olive flounder using rapid amplification of cDNA ends PCR technique. The full-length cDNA of Paralichthys olivaceus STING (PoSTING) was 1442 bp in length and contained a 1209-bp open reading frame that translated into 402 amino acids. The theoretical molecular mass of the predicted protein sequence was 45.09 kDa. In the PoSTING protein, three transmembrane domains and the STING superfamily domain were identified as characteristic features. Quantitative real-time PCR revealed that PoSTING expressed in all the tissues analyzed, but showed the highest level in the spleen. Temporal expression analysis examined the significantly upregulated expression of PoSTING mRNA after viral hemorrhagic septicemia virus (VHSV) stimulation. In contrast, no significant changes in the PoSTING expression were detected in Edwardsiella tarda-challenged group compared to the un-injected control. The expression of P. olivaceus type I interferon (PoIFN-I) was also highly upregulated upon VHSV challenge. These results suggest that STING might be involved in the essential immune defense against viral infection together with the activation of IFN-I in olive flounder.
Reproductive disorders in cows cause economic loss in livestock farms. Reproductive diseases, such as follicular cyst, luteal cyst, endometritis, pyometra, and repeat breeding cause infertility. Among these diseases, endometritis and pyometra are uterine infections that are leading causes of infertility. This study was performed to investigate the causative agents of uterine diseases using bacterial culture. Bacteria were obtained from the reproductive organs (vagina, uterine cervix, and uterine horn) of dairy cow diagnosed with endometritis or pyometra, and cultured on blood agar. The colonies obtained from cultivation for 24 hours were passaged. To identify the bacteria, the colonies grown in passaged culture Gram stained and applied to an automatic biochemical microbial identification system. Escherichia coli were commonly detected in vagina, uterine cervix, and uterine horn of dairy cows diagnosed to pyometra. The cows having endometritis showed not only Escherichia coli but also Pantoea spp. and Klebsiella spp. strains. Dairy cows that were infected with Escherichia coli in uterus caused mastitis or digestive disease. These results suggest that sanitary feeding and management beforehand are needed to prevent bacterial infections.
A sundry system is one of popular systems for composting livestock manure, of which main honest is to utilize unlimited, clean, and free solar radiation. A sundry system with a composter of two horizontal screw-type concrete ducts at different height, was constructed and operated for three days for each test in May, 1993, to evaluate its composting performance. Four treatments of the mixture ratio of swine manure and saw dust (manure : sawdust= 1 : 1.25, 1 : 1, 1 : 0.7, 1 : 0.5) were implemented to evaluate the effect of the mixture ratio on degradation of the composting materials of a sundry system with a screw-type composter. Maximum temperature of the composting materials was over 5$0^{\circ}C$ at D1 or D2 (one or two days after operation starts) for each test. Mean C/N ratio and water contents of the materials were reduced by more than 15 and 20%, respectively. Microbial density of each test showed a typical variation with the lapse of the composting time. Mesophilic microorganism seemed to play more important role on degradation of the materials than thermophilic. A sundry system with a screw-type composter can be considered as a feasible system on basis of maturity data. The conclusion was completely reverse from that of Choi et at., although both adopted a sundry system. A further study is recommended to pursue the cause of better performance of the screw-type composter, whether it was due to affirmative weather or more efficient composter.
해양으로부터 polysaccharide를 생산하는 세균을 분리하여 동정한 결과 Zoogloea sp.(KCCM 10036) 로 밝혀졌으며, 이 해양유래 Zoogloea sp.는 셔로 다 른 두종류의 다당류인 수용성다당류(WSP)와 세포 결합다당류(CBP)를 통시에 생산하여 지금까지 보 고된 관련 미생물과는 다른 특이성을 보였다. 이들 중 WSP는 원심분리 시킨 배양액의 상등액으로부터 acetone 침전법에 의해 분리하였고, CBP는 침전물 로부터 acetone 빛 cetylpyridinium chloride 처 리 에 의 해 분리하였다. WSP와 CBP는 모두 단백 질을 함유하였으나, 이 중 CBP가 보다 높은 단백질 함유 율을 나타내었다. WSP를 다량 생산하기 위한 최척 탄소원은 fructose였고, CBP의 경우는 glucose였 다. Polysaccharide 생산촉진인자로 biotin, ampicillin, 그리고 surf actant를 각각 배지에 첨가하였을 경우, 각각의 농도가 $0.2mg/\ell, 0.4mg/\ell,0.6mg/\ell,$에서 생산량이 가장 높은 값을 보였다. 1%의 glucose를 탄소원으로 하여 fermentor 배양하였을 때, $9.1g/\ell$ 의 WSP와 $2.5g/\ell$ 의 CBP가 생산되었다.
Coix lacryma-jobi (Hanjeli) is known to posses anti-microbial properties. Therefore, phytochemical compounds of C. lacryma-jobi have been studied to produce novel antimicrobial agents as treatments against antibiotic-resistant bacteria.The objective of this study was to determine the phytochemical composition and antibacterial activity of the C. lacryma-jobi oil against Escherichia coli, Staphylococcus aureus, and Bacillus subtilis. The phytochemical composition of the oil was determined via gas chromatography mass spectrophotometry (GC-MS). Moreover, agar disk and agar well diffusion were employed to screen the antibacterial activity of the oil. An agar well diffusion test was implemented to determinate MIC's (minimum inhibitory concentrations). Dodecanoic acid, tetradecanoic acid, 2,3-dihydroxypropylester, 1,3-dioctanoin, N-methoxy-N-methyl-3,4-dihydro-2H-thiopyran6-carboxamide, propanamide, 5-Amino-1-(quinolin-8-yl)-1,2,3-triazole-4-carboxamide, and pyridine were identified in the C. lacryma-jobi oil. The MIC value of the oil was 0.031 g/L and the MBC of the oil was 0.125 g/L effective in all test bacteria. Dodecanoic acid displayed inhibitory activity against gram-positive and gram-negative bacteria. Therefore, our research demonstrated C. lacryma-jobi (Hanjeli) oil exhibited antibacterial activity against E. coli, S. aureus, and B. subtilis. These research suggest that C. lacryma-jobi root oil could be used for medicinal purposes; however clinical and in vivo tests must be performed to evaluate its potential as an antibacterial agent.
사과나무에서 발생하는 탄저병에 대한 길항미생물을 찾기 위하여 자연계로부터 유용미생물을 분리하고 사과 탄저 병원균에 대한 길항력 검정과 균주를 동정한 결과는 다음과 같다. 자연계로부터 얻은 1,000여종의 미생물중에서 탄저병원균에 대하여 길항력이 우수한 미생물을 1차적으로 11종 선발하였으며, 이중에서 가장 길항력이 뛰어난 미생물 CH1141을 최종적으로 선발하였다. 길항미생물 CH1141은 분리한 탄저병원균에 대하여 65%의 높은 생장억제력을 보였다. 길항력이 우수한 CH1141의 형태적 성질, 배양적 특성 및 생리 생화학적 성질 등을 조사하여 비교 검토한 결과 Bacillus subtilis와 유사한 균으로 동정되었다.
