MicroRNA-296-5p Promotes Invasiveness through Downregulation of Nerve Growth Factor Receptor and Caspase-8

  • Glioblastomas (GBM) are very difficult to treat and their aggressiveness is one of the main reasons for this as well as for the frequent recurrences. MicroRNAs post-transcriptionally regulate their target genes through interaction between their seed sequence and 3'UTR of the target mRNAs. We previously reported that miR-296-3p is regulated by neurofibromatosis 2 (NF2) and enhances the invasiveness of GBM cells via SOCS2/STAT3. In this study, we investigated whether miR-296-5p, which originates from the same precursor miRNA as miR-296-3p, can increase the invasiveness of GBM cells. It was observed that miR-296-5p potentiated the invasion of various GBM cells including LN229, T98G, and U87MG. Through bioinformatics approaches, two genes were identified as miR-296-5p targets: caspase-8 (CASP8) and nerve growth factor receptor (NGFR). From results obtained from Ago2 immunoprecipitation and luciferase assays, we found that miR-296-5p downregulates CASP8 and NGFR through direct interaction between seed sequence of the miRNA and 3'UTR of the target mRNA. Knockdown of CASP8 or NGFR also increased the invasive ability of GBM cells, indicating that CASP8 and NGFR are involved in potentiation of invasiveness by miR-296-5p. Consistent with our findings, CASP8 was downregulated in brain metastatic lung cancer cells, which have a high level of miR-296-5p, compared to parental cells, suggesting that miR-296-5p may be generally associated with the acquisition of invasiveness. Collectively, our results implicate miR-296-5p as a potential cause of invasiveness in cancer and suggest it as a promising therapeutic target for GBM.

MicroRNA Expression in Leukemia Cell Line(K562 cell) Using Rhus Verniciflua Stokes (건칠(乾漆)을 이용한 K562 만성 골수성 백혈병 세포주에서의 MicroRNA 발현 규명)

  • Objective : The purpose of the study was to identify expression profiling of miRNAs associated with cancers after treating allergen-removed Rhus Verniciflua Stokes and allergen-removed Rhus Verniciflua Stokes fumigaed Angelica gigas on leukemia cell lines. Methods : miRNA expression has been analyzed using miRNA array method through denaturation and hybridization after isolating the total RNA from leukemic cell line treated with 100 ㎍/㎖ of aRVS and aRVS-A each. Microarray expressions were interpreted as 'significant' on miRNAs when decreased less than 0.5 fold or increased more than 1.5 fold compared with the control group. Results : Among 158 miRNAs in total, 32 miRNAs were significantly presented in miRNAs expression. miRNA has been activated with a variety of genes for predicted targets, and the overexpressed miRNAs were categorized according to proliferation and metastasis of cancer in this study. The findings were reported that seven miRNAs (let-7b, miR-193a-5p, 296-3p, 26a, 22, 124a, 92b) showed significant expressions on proliferation and growth, seven miRNAs (miR-193a-5p, 26a, 200c, 183, 124a, 198, 210) presented meaningful expressions on invasion and metastasis, two miRNAs (let-7b, miR-210) were highly expressed on angiogenesis, five miRNAs (let-7b, miR-26a, 181d, 181c, 296-5p) related with apoptosis, and six miRNAs (let-7b, miR-200c, 183, 370, 124a, 191) were associated with prognosis of cancer and early diagnostic factors for cancer. Conclusion : The mechanism of miRNA takes a role in diagnosis, treatment, and prognotic factors for cancer as well. This study suggested that further detailed research on overexpression of specific miRNA should be carried out continuously in the future.

Impact of Self-Esteem and Job Stress on Job Satisfaction of the Emergency Medical Technicians (응급구조사의 자아존중감, 직무스트레스가 직무만족도에 미치는 영향)

  • The purpose of this study was to investigate self-esteem and job stress which might influence job satisfaction of the emergency medical technicians. The partcipants were 135 emergency medical technicians in hospital. Data were collected from August 15 to September 10, 2009. The collected data was analyzed with the SPSS win 15.0 version. The average self-esteem score was 3.47, job stress 3.16, job satisfaction 3.14. The score for job satisfaction. showed significant difference motivation of desire(F=3.169, p=.016) and region(F=3.414, p=.011). Self-esteem had significant positive correlation to job satisfaction(r=.296, p=.000). Job stress had significant negative correlation to job satisfaction(r=-.327, p=.000). Self-esteem and job stress were significant predictor and accounted for 16% of the variance in job satisfaction of the emergency medical technicians. Future job satisfaction management program for the emergency medical technicians should be considered their self-esteem and job stress.

A Study on Self-Esteem, Health Promoting Behavior and the Quality of Life of the patients undergoing Hemodialysis (혈액투석환자의 자아존중감, 건강증진행위와 삶의 질과의 관계연구)

  • This study was conducted to investigate the relationship between self-esteem, health promoting behavior and the quality of life of the patients undergoing hemodialysis. The subjects were 86 patients undergoing hemodialysis in H hospital. The data were collected using a questionnaire. Data collection was done from January 11th to June 16th, 1999. The analysis of data was done by use of descriptive statistics, t-test, ANOVA, Pearson Correlation Coefficient and Multiple regression using the SPSS/PC+ program. The results are as follows: 1. The mean of the level of self-esteem was 25.02, the level of health promoting behavior, 120.04 and the level of QOL, 128.09. 2. Test for hypothesis: Hypothesis 1. "The higher the level of self-esteem of the patients undergoing hemodialysis, the higher the level of quality of life will be." was supported(r=.296, p<.01). Hypothesis 2. "The higher the level of health promoting behavior, the higher the level of quality of life will be." was supported (r=.628, p<.001). 3. Health promoting behavior explained 39.5percent of QOL($R^2$=.395, F=27.040, p<.001). 4. Monthly income(F=3.85, p<.01) and marital status(F=4.64, p<.05) were significantly related to the quality of life In conclusion, this study showed that self-esteem and health promoting behavior may be important factors that can improve the quality of life of the patients undergoing hemodiaysis. This study identified that nursing plans should include these factors to help physical, psychological and social adaptation of the patients undergoing hemodialysis.

Characterization and Enhanced Production of Enterocin HJ35 by Enterococcus faecium HJ35 Isolated from Human Skin

  • A strain named as HJ35 was isolated from the skin of sixty-five men and fourteen women for acne therapy, in order to find an effective antimicrobial agent against Propionibacterium acnes. Isolate HJ35 was identified as Enterococcus faecium based on 16 rDNA sequence and produced enterocin HJ35 having antimicrobial activities against most lactic acid bacteria, En­terococcus spp., Staphylococcus aureus, S. epidermidis, Clostridium perfringens, some bacilli, Mi­crococcus flavus, Listeria monocytogenes, L. ivanovii, Escherichia coli, Pseudomonas fluorescens and Propionibacterium acnes, in the modified well diffusion method. Especially, enterocin HJ35 showed a bactericidal activity against Propionibacterium acnes P1. The antimicrobial activity of enterocin HJ35 was disappeared completely with the use of protease XIV. But enterocin HJ35 activity is very stable at high temperature (up to $100^{\circ}C$ for 30 min), in wide range of pH (3.0${\~}$9.0), and by treatment with organic solvents. The apparent molecular mass of enterocin HJ35 was estimated to be approximately 4${\~}$4.5 kDa on detection of its bactericidal activity after SDS-PAGE. In batch fermentation of E. faecium HJ35, enterocin HJ35 was produced at the mid­log growth phase, and its maximum production was obtained up to 2,300 AU/mL at the late stationary phase. By employing fed-batch fermentation, the enhanced production of enterocin HJ35 was achieved up to 12,800 AU/mL by feeding with 10 g/L glucose or 6 g/L lactate.
