Study on Bond Used in Shoes Manufacturing Industry (신발제조업체에서 사용되는 접착제에 관한 연구)

  • ln order to acquire the fundamental data for the organic solvents in bond and to contribute the health improvement of workers in deprtment of organic solvents in shoes manufacturing industries. The authors surveyed the contents of organic solvent in adhesive and determined the amount of volatilization of organic solvent by time and temperature with gaschromatography from March to September 1990. The results were as follows; 1) The kinds for organic solvents in bond were 9 that was Toluene, C-Hexane, N-Hexane, C-Hexanon, Aceton, Methyl Ethyl Keton, Dimethyl Formamide, Etyly Acetate. 2) Toluene and Methyl Ethyl Keton among the organic solvents in adhesive were over 80.18%. 3) The amount of volatilization of Methyl Cyclohexanone and Aceton by time and temperature were the most level than other compounds.

  • Preparation of optically pure (3R, 4R)-4-acetoxy-3-[(1R)-1-hydroxyethyl]azetidin-2-o ne derivatives, which can be employed as starting materials for synthesis of carbapenem and penem antibiotics, was established in high efficiency from 6-amino-penicillanic acid (6-APA). 6-APA was diazotized and brominated to give 6, 6-dibromopenicillanic acid and its methyl ester was metalated with methylmagnesium bromide and condensed with acetaldehyde. The product, methyl 6-bromo-6-(1-hydroxyethyl)penicillanate was reduced with Zn-$NH_4Cl-NH_4OH$-acetone efficiently to give methyl 6-(l-hydroxyethyl)-penicillanate, which was protected either with ${\beta},{\beta},{\beta}$ -trichloroethoxycarbonyl group or with t-butyldimethylsilyl group. The thiazolidine rings of these compounds were cleaved by treatment of mercury(II) acetate in acetic acid and permangante in acetone in sequence to afford the desired optically pure final products.

  • Sensitive and safe method for visualization of DNA in agarose gels using visible dye is described. To improve the sensitivity, we studied a counterion-dye staining method using methyl orange as a counterion-dye which contributes to reduce excessive background staining by indoine blue. Dye concentrations, PH of staining solution, mixing molar ratio of two dyes, and staining times were optimized for the counterion-dye staining. By the staining with a mixed solution of 0.005% indoine blue and 0.00165% methyl orange in 10% ethanol 0.2M sodium acetate, 8 ng of the 3 kb DNA in an agarose gel was detected within 1hr. (omitted)

  • The transesterification reaction between diethanolamine and methyl-methacrylate was kinetically investigated in the presence of various metal acetate catalysts at $120^{\circ}C$. The quantity of methylmethacrylate reacted in the reaction flask was measured by gas chromatography and liquid chromatography, and the reaction rate was investigated by measuring of the quantity of products and reactnts under various catalysts. The transesterification reaction was carried out in the first order reaction kinetics with respect to the concentration of diethanolamine and methylmethacrylate, respectively. The apparent rate constant was found to obey first-order kinetics with respect to the concentration of catalyst. The linear relationship was shown between apparent rate constant and reciprocal absolute temperature, and by the Arrhenius plot, the activation energy has been calculated as 11.08 Kcal with zinc acetate catalyst, 17.99 Kcal without catalyst. The maximum reaction rate was appeared at the range of 1.4 to 1.6 of electronegativity of metal ions and instability constant of metal acetates.

  • Aspergillus sp. F184 was isolated from soil for the development of new xanthine oxidase inhibitor. This xanthine oxidase inhibitor was sequentially purified by filtration, HP-20 adsorption column chromatography, ethyl acetate extraction, silica gel column chromatography and crystallization, and was named as YUX 104. YUX 104 was identified to be 5,6-epoxy-2-hydroxy-3-methyl-2-cyclohexene-1,4-dione(terreic acid) by NMR and mass spectroscopic sudies.

Analysis of Residual Solvents in Food Packaging Materials Using Solid Phase Microextraction Method (Solid Phase Microextraction법을 이용한 식품포장재 중의 잔류용제 분석)

  • Solid phase microextraction (SPME) was used for the determination of 6 standard solvents (methanol, isopropanol, methyl ethyl ketone, ethyl acetate, cyclohexane, toluene) in food packaging materials. SPME method is a solvent-free sample preparation technique in which a fused silica fiber coated with polymeric organic liquid is introduced into the headspace above the sample. SPME method using fiber coated polydimethylisiloxane (PDMS) was compared with static headspace (SHS) method used as a reference. It was found that the optimal adsorption condition using PDMS-SPME method was 2$0^{\circ}C$ for 15 minutes for the standard solvents. Detection limits, linearity, reproducibility and recovery of both SHS and PDMS-SPME methods have been determined using 6 standard solvents. Both methods were characterized by high reproducibility and good linearity. Using SHS methods, the mean recovery of the 6 standard solvents was ranged from 75.5% to 105.8% with a mean relative standard deviation (RSD) of 0.3% to 4.8%. With PDMS-SPME method, the mean recovery of the 6 standard solvents was ranged from 86.7% to 108.3% with a mean RSD of 0.4% to 2.5%. The detection limits of both methods were the same for toluene, cyclohexane and methyl ethyl ketone; those of PDMS-SPME method were higher than those of SHS method for methanol, isopropanol and ethyl acetate. PDMS-SPME fiber shoed excellent adsorption for non-polar solvents such as toluene, while it showed relatively low adsorption for polar solvents such as methanol.

  • The preparation method of the mortar-plastic composites(M P C) were studied with styrene, methyl methacrylate and vinyl acetate as monomers. Radiation, thermal-catalytic and radiation-catalytic methods were used as curing methods. Almost all of the above monomers and methods were possible to use for preparing M P C Although thermal-catalytic method was excellent to get M P C in a short time, the tensile strength of the product was less than those obtained by radiation method. It was possible to prepare the M P C which included up to about 10% plastics and was strengthened about ten times on the properties of acid resistandes and tensile strength comparing with the control. The improvement of the properties is much superior to concrete-plastic composite(C P C).

Inhibitory Effects of Ixeris Dentata on the Mutagenicity of Aflatoxin $B_1$, N-methyl-N연-nitro-N-nitrosoguanidine and the Growth of MG-63 Human Osteosarcoma Cells (씀바귀 추출물들의 돌연변이 유발 억제 및 MG-63 암세포 성장 저해 효과)

  • Ixeris dentata was extracted with methanol and then the methanol extract was further fractionated to hexane, chloroform, ethyl acetate, butanol and aqueous fraction. The methanol extract of lxeris dentata had the strong antimutagenic effect on the aflatoxin B1(AFB1) and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Ames mutagenicity test and SOS chromotest. Among the solvent extracted fractions from the methanol extract, the chloroform fraction exhibited the greatest antimutagenic effect suppressing the mutagenicity of AFB1 with inhibition rate of 74 percent. The methanol extract of Ixeris dentata also revealed the inhibitory effect on the growth of MG-63 human osteosarcoma cells after 6 days of breeding at 37℃. The chloroform fraction and the ethyl acetate fraction from the methanol extract of lxeris dentata were most effective and inhibited the growth of MG-63 cells by 97 and 93 percent, respectively. It is suggested that the inhibitory effects of lxeris dentata on the mutagenicity and the growth of MG-63 human osteosarcoma cells are strong in the lipid soluble fractions.

  • The effect of nuruks and crude amylolytic enzyme on free amino acid and volatile components of brown rice vinegar prepared by static cultures was investigated. Five groups consisted of AV (100% nuruk without crude amylolytic enzyme), BV (75% nuruk; 25% crude amylolytic enzyme), CV (50% nuruk; 50% crude amylolytic enzyme), DV (25% nuruk; 75% crude amylolytic enzyme) and EV (100% crude amylolytic enzyme without nuruk). Free amino acid content in AV vinegar (132.06 mg%) was lower than the others (184.56-191.22 mg%). Acetic acid, 3-methyl butyl acetate, acetoin and isoamyl alcohol were major volatile components as analyzed using gas chromatography-mass spectrometry after headspace solid-phase microextraction. Acetic acid in AV and EV samples represented 67.56% and 55.53% of total GC peak area, respectively. E-nose provided different patterns in each case showing variation in sensory properties.

Influence of Heating Conditions on the Formation of Acrylamide and Other Products in Asparagine-Glucose Model Reaction System (Model reaction system에서 가열조건이 acrylamide 및 기타 화합물들의 생성에 미치는 영향)

  • The Formation of acrylamide was studied in Maillard model reaction systems based on asparagine-glucose. The mixture of asparagine and glucose in equal molar ratio, and then heated at 125, 150, 175 and $200^{\circ}C$ for 10, 20 and 30 minute, respectively. The reaction products were extracted with ethyl acetate and methanol, and then isolated and detected on FFAP capillary column and HP-5MS 5% phenyl methyl siloxane column by using GC/MS. Acrylamide was detected only from methanol extracts and on FFAP capillary column, at retention time 23.53 min., and the detection limit was 4.6 ng. Acrylamide content mainly increased with increasing temperature and processing time till $175^{\circ}C$, therefore, maximal acrylamide formation occurred at $175^{\circ}C$ for 10 minute ($116{\mu}g/g$), while, above $175^{\circ}C$, higher temperatures or prolonged processing times caused a decrease of acrylamide levels, finally disappeared at $200^{\circ}C$ for 30 minute. Three major compounds were identified as 1,3-dihydroxypropanone, 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyrane-4-one and 5-hydroxymethylfurfural, and three minor compounds also as 5-methylfurfural, 2-acetylpyrrole and N,N-dimethylcyclohexamine, from ethyl acetate or methanol extracts on FFAP or HP-5MS capillary column.
