생삽주의 기능성을 분석한 결과, 추출 수율은 ethanol 추출물에 14.8%와 water 분획물에 17.7%를 나타내었다. Acetone 추출물과 butanol 분획물의 수소공여 능은 각각 72.9%와 74.2%로 높게 나타났으며, methanol 추출물과 butanol 분획물의 아질산염 소거 효과는 각각 95.0%와 79.2%로 가장 높게 나타났다. 생삽주의 용매별 추출물 중 methanol 추출물이 높은 항균 활성을 나타내었는데, Bacillus subtilis에 대해 20 mm clear zone을 나타내었으며, Pseu. aeruginosa에 대해 19 mm clear zone을 나타내었다. 항균 활성이 가장 높은 methanol 추출물을 계통분획하여 항균 활성을 확인한 결과, butanol 분획물이 높은 항균 활성을 나타내었는데, Bac. subtilis, V. parahaemolyticus에 대해 각각 18 mm clear zone을 나타냈다. Methanol 추출물과 butanol 분획물을 이용하여 최소저해농도를 확인한 결과 대체로 gram(+)균에 대해 2 mg/disc와 4 mg/disc에서 나타났구 gram(-)균에 대해서는 6 mg/disc에서 저해활성을 나타내었다.
Protopectinase(PPase)는 식물 세포벽의 protopectin을 분해하는 효소로서 식품의 가공에 이용될 수 있는 효소이다. PPase의 산업적 응용을 위하여 PPase를 생산하는 미생물 Bacillus subtilis IFO 12113의 배양여액에 acetone, methanol, ethanol을 각각 처리하여 PPase를 회수하는 연구를 수행하였다. Acetone의 경우, 배양여액 대 acetone을 1:1 (v/v) 비율로 첨가했을 때 효소는 59.2%가 회수되었으며 정제도는 1.7배 이었다. Methanol의 경우, 1:2(v/v) 비율로 처리하였을 때, 효소는 100% 회수되었으며 비활성도로 본 정제도는 4.0배 증가하였다. Ethanol의 경우에 1 :0.5 (v/v) 비율로 처리한 것을 원심분리시켜 침전물을 제거하고, 그 상등액에 최종 ethanol 비율을 1:1(v/v)까지 처리했을 때, 효소의 회수율은 68%를 나타냈으며 비활성도는 13.5배 증가하였다. 세균의 배양여액으로부터 PPase의 회수에는 methanol 처리가 가장 효율적이었으나, 산업적 응용을 위하여 정제효과까지 고려하면 ethanol처리가 가장 적절하였다.
초임계 유체 추출법에 의해 감초의 triterpenoid saponin(glycyrrhizin)을 추출하기 위해 50 MPa, $60^{\circ}C$와 3mL/min의 조건에서 보조용매와 soaking의 효과를 알아보았다. 순수한 초임계 이산화탄소만을 이용하여 추출한 결과 감초의 glycyrrhizin은 거의 추출되지 않았다. 99.9% methanol, ethanol과 isopropanol을 보조용매로 초임계 이산화탄소에 첨가하여 추출한 경우, 추출수율의 상승에는 큰 영향을 미치지 아니하였다. 시료를 물로 soaking하여 90% methanol을 보조용매로 사용하여 초임계 이산화탄소에 의해 추출한 경우, soaking 용매의 사용량이 증가할수록 추출수율은 증가하였고, 최고 21.68mg/g(회수율 53.25%)의 수율을 보여주었다. 한편 시료의 soaking 없이 70% methanol을 보조용매로 사용하였을 때는 유기용매추출법에 의한 수율보다 더 높은 수율을 나타내었고, 시료를 물로 soaking하여 70% ethanol을 보조용매로 초임계 이산화탄소에 첨가하여 추출하였을 때, 추출수율과 추출속도는 상승하였고, 시료의 90%(w/w)에 해당하는 물로 시료를 soaking한 경우 70% methanol을 보조용매로 사용하였을 때와 거의 동일한 추출수율을 얻을 수 있었다.
물-용매계에서 용매의 몰분율 변화에 따른 di-t-butyl nitroxide (DTBN)의 N hyperfine coupling 상수($a_N$)를 EPR spectroscopy에 의하여 측정하였다. 아세톤, dimethylsulfoxide (DMSO), methanol, ethanol 그리고 1-propanol과 같이 서로 다른 값의 극성도를 갖는 용액들을 용매로서 사용하여 연구하였다. 물-용매계에서 DTBN의 $a_N$ 값으로부터 용매화 평형에 대한 평형상수값을 계산하였고, 또한 몰전이 에너지($E_T$) 값을 용매의 흡수스펙트럼 실험결과로부터 계산하여 제시하였다. 물-용매계에서 용매의 몰분율에 대한 $a_N$ 값들을 도시한 결과 물-DMSO, 물-ethanol과 물-1-propanol계의 경우에는 직선으로부터 약간의 음의 편차가 관측되었으나, 물-아세톤과 물-methanol계의 경우에는 거의 직선적인 관계가 있음을 알 수 있었다. 물-아세톤계의 경우 ${\eta}{\rightarrow}{\pi}^{\ast}$ 전이에 의한 흡수파장 (${\lambda}$)은 아세톤의 몰분율과 비례하여 증가하였다. 물-아세톤과 물-DMSO계에서 ${\eta}{\rightarrow}{\pi}^{\ast}$ 전이에 의한 ${\lambda}$ 값들이 증가함에 따라 $a_N$ 값들은 감소하였으며 이들 값들은 온도에 의하여 거의 영향을 받지 않고 있어 DTBN 라디칼들이 이러한 용매내에서 안정된 전자구조를 가지고 있음을 알 수 있었다.
본 연구에서는 용매별(methanol, ethanol, acetone 및 water)로 추출한 canola meal의 항산화 활성을 조사하였다. 모든 canola meal 용매 추출물들은 첨가 농도가 증가할수록 항산화 활성이 점진적으로 증가하였다. 추출물 중에서 methanol 추출물이 가장 낮은 $EC_{50}$ (DPPH radical scavenging ability; 0.9 mg/mL, ABTS radical scavenging ability; 3.5 mg/mL, reducing power; 1.3 mg/mL and chelating ability; 8.5 mg/mL)과 가장 많은 항산화 물질 (total phenolics; $63.3\;{\mu}g$/mg, total flavonoids; $21.3\;{\mu}g$/mg)을 함유하는 것으로 나타났다. 보다 정확한 canola meal의 항산화 활성을 조사하기 위하여 항산화 활성이 높은 methanol 추출물을 sephadex LH-20 column을 이용하여 순차적으로 MF 1, MF 2 및 MF 3의 3개의 분획물을 얻었으며, 동일한 농도(1.5 mg/mL)에서 측정한 항산화 활성에서 DPPH radical scavenging ability는 MF 2(75.56%)에서, ABTS radical scavenging ability와 reducing power는 MF 3(31.17% and 1.60)에서, chelating ability는 MF 1(77.72%)에서 가장 높게 나타났다.
본 연구에서는 치아씨의 항산화 활성을 증가시키기 위하여 추출용매 종류와 용매농도, 추출시간과 추출온도를 달리하여 총 폴리페놀 함량과 DPPH 라디칼 소거능을 측정한 후 반응표면분석을 이용하여 최적조건을 수립하였다. 치아씨 추출조건은 에탄올과 메탄올용매 모두에서 추출용매 60%, 추출시간 130분과 추출온도 $20^{\circ}C$에서 총 폴리페놀 871.00mg%($R^2=0.9507$), 557.70mg%($R^2=0.9784$)로 가장 함량이 많았고, DPPH 라디칼 소거능은 72.14%($R^2=0.9675$), 52.79%($R^2=0.9524$)의 항산화력을 보였다. 용매종류에서는 에탄올 추출용매가 메탄올 추출용매보다 같은 조건에서는 월등히 높은 항산화력을 보여 에탄올이 치아씨 추출용매로 더 우수한 용매로 선정되었다. 또한 반응표면분석 결과, 용매추출 조건에서는 에탄올 용매에서는 용매추출 시간이나 온도보다는 용매농도에 가장 큰 영향을 받았고, 메탄올 추출조건에서는 용매추출 시간에서 가장 큰 영향력을 주었다. 반응표면분석 후 용매 농도, 시간과 온도의 최적 추출조건의 범위는 에탄올 추출조건은 에탄올농도 63%, 추출시간 100분과 추출온도 $18^{\circ}C$가 최적조건이고, 메탄올 추출조건에서는 추출농도 65%, 추출시간 120분과 추출온도 $16^{\circ}C$를 보였다.
Isoflavone was extracted with various concentration of aqueous methanol using whole hypocotyls as the starting material. Whole hypocotyls were preferred as the raw material because the residue could be easily removed from the solvent after the extraction process. Extraction yield was almost constant at the methanol concentration of $20-80\%$. Most of the isoflavone was extracted within 1 hr, and the extraction yield remained almost constant thereafter. When the concentration of methanol was $80\%$, the content of total solid was reduced due to the reduced extraction of contaminating protein as the result of protein insolubilization. Among resins tested, Diaion HP-20, Amberlite XAD-16, and Amberlite IRC-50 showed the highest capacity to absorb the compound. Open column chromatography with Diaion HP-20 showed that $80\%$ aqueous ethanol was most efficient as the eluting solvent with final recovery of the phytochemical being more than $95\%$. Maximum adsorption of the phytochemical occurred at the acidic pH 2-4. When the spatial velocity was increased to 15 and more, the degree of adsorption was decreased, whereas below spatial velocity of 15, the adsorption capacity of isoflavone to the resin was almost constant. The purity of the isoflavone purified by column chromatography was $78\%$.
Poly(ethylene terephthalate)(PET) filaments were treated in binary mixtures of organic solvents such as benzyl alcohol/perchloroethylene(BA/PER), benzyl alcohol/trichloroethylene(BA/TRI), benzyl alcohol/ethylene chloride(BA/EC), methanol/perchloroethylene (ME/PER), and methanol/trichloroethylene(ME/TRI). From the equilibrium swelling of PET, total and partial isotherms of PET for sorption of solvent mixtures and isotherms for distribution of the components between the phases were taken. Also the shrinkage and the crystallinity of PET treated with binary mixtures were investigated. The results obtained were summerized as follows: 1. All isotherms for distribution of the components between the phase deviated from the diagonal in system of PET-binary mixtures. Especially in the binary mixtures of ME/PER or ME/TRI, selectivo sorption of chlorinated hydrocarbon PER and TRI by PET occured. 2. The shrinkage of PET treated in binary mixrures was increased to compare with single solvent-treated, but the composition of binary mixtures corresponding to maximum values of shrinkage was not always agreed with the composition of binary mixtures corresponding to maximum values of shrinkage was not always agreed with the composition of binary mixtures exhibiting of the maximum swelling of PET. 3. The crystallinity of PET treated in binary mixtures generally increased than that of single solvent-treated.
Solvolysis rate constants of 2-phenylethyl-(2-$PhCH_2CH_2OCOCl$, 1) and 2,2-diphenylethyl chloroformate (2,2-$Ph_2CHCH_2OCOCl$, 2), together with the previously studied solvolyses of ${\alpha}$- and ${\beta}$-substituted chloroformate ester derivatives, are reported in pure and binary solvents at $40.0^{\circ}C$. The linear free energy relationship (LFER) and sensitivities (l and m) to changes in solvent nucleophilicity ($N_T$) and solvent ionizing power ($Y_{Cl}$) of the solvolytic reactions are analyzed using the Grunwald-Winstein equation. The kinetic solvent isotope effects (KSIEs) in methanol and activation parameter values in various solvents are investigated for 1 and 2. These results support well the bimolecular pathway with same aspects. Furthermore, the small negative values of the entropies of activation of solvolysis of 1 and 2 in the highly ionizing aqueous fluoroalcohols are consistent with the ionization character of the rate-determining step, and the KSIE values of 1.78 and 2.10 in methanol-d indicate that one molecule of solvent acts as a nucleophile and the other acts as a general-base catalyst. It is found that the ${\beta}$-substituents in alkyl chloroformate are not the important factor to decide the solvolysis reaction pathway.
As part of our endeavor to increase the productivity of steroid by enzymatic transformation of corticosteroids, attempts have been made to increase the solubility of steroids by using some organic solvents. When the solubility of steroids is the rate limiting factor in the steroid transformation, it was found that the use of solvents significantly improved the yield. Hydrocorisone as a substrate and 3-ketosteroid .DELTA.$^{1}$ dehydrogense as an immobilized whole cell enzyme were employed as the model system for this study. It was found that the yield of product, prodnisolone, goes through a maximum with an increase in the solvent concentration. At a high solvent concentration, the solvent showed a toxic effect and it causes a decrease in the product yield by the second order inhibition mechanism. Among the solvents evaluated, methanol and ethanol were found to be the best. These alcohols are not only good solvents but also showed minimal toxic effect. Based on the experimental results, it was concluded that the productivity of steroid can be increased by usign well selcted solvents systems for the enzymatic transformation of steroids.
