  • In the course of evaluation of hepatoprotective components against alcohol-induced toxicity from Aloe spp., the methanol extract was found to cause a significant inhibition of rat liver cytosolic alcohol dehydrogenase activity. Systematic fractionation of active tractions monitored by bioassay led to isolation of four compounds; aloe-emodin, aloenin, ethylidene-aloenin and ${\beta}-sitosterol$, which were estimated as active principles for inhibition of c-ADH and c-ALDH activities in vitro.

  • For the biological survey, effects of Ailanthi Cortex Radicis, the root bark of Ailanthus altissima(Simaroubaceae) on epoxide hydrolyzing enzymes were checked. The methanolic extract and its chloroform fraction were shown to activate the liver metabolizing enzyme system including epoxide hydrolase system which was monitored by activities of transaminase, lactate dehydrogenase, alkaline phosphatase and epoxide hydrolase system in bromobenzene treated rats. But they showed no effect on glutathione S-transferase activity.

사염화탄소에 의해 유발된 간독성에 대한 상백피 추출물의 간보호효과 (Protective Effects of Extracts of Mori Cortex Radicis on Carbon Tetrachloride-induced Hepatotoxicity)

  • In order to evaluate the protective effects of extracts of Mori cortex radicis on carbon tetrachloride-induced hepatotoxicity, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, lactic dehydrogenase, malondiadehyde values and glutathione S-transferase activity were measured in ICR mice. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride-treatment were markedly increased than normal control but those levels were significantly decreased by the treatment of butanol fraction of Mori cortex radicis mathanol extract. Glutathione S-transferase activity was decreased by carbon tetrachloride than control, but that also inhibited by the treatment of butanol fraction of Mori cortex radicis methanol extract. These results demonstrate a possible hepatoprotective role of extract role of extract of Mori cortex radicis against ${CCl_4}-induced$ hepatoxicity in vivo.

황금(黃芩)의 항산화 효과 (Anti-Oxidative Effects of Scutellariae Radix)

  • Objectives : This study was carried out to determine if Scutellariae Radix have protective effect against cell injury induced by various toxic agents in rat kidney slices. Methods : Water(SWe) and methanol(SMe) extracts were prepared for this experiment. Cell injury was estimated by measuring lactate dehydrogenase(LDH). Lipid peroxidation was examined by measuring malondialdehyde. Results : SMe prevented the LDH release by CCl4, menadione, tert-butyl hydroperoxide and mercury treatment in vitro in kidney slices, but SWe prevented the LDH release by CCl4 and mercury. SMe also prevented reduction in GSH by CCl4 and lipid peroxidation induced by mercury. Conclusions : Thus, SMe may have more powerful efficacy on anti-oxidative effects when compared with SWe. And further studies have to be followed concerned with procedure of extraction of SMe and its change of effects.

Indole, Indole-3-calbinol 및 Benzofuran이 간장 microsome과 cytosol의 약물대사 효소 활성도에 미치는 영향 (Differential Effects of Indole, Indole-3-carbinol and Benzofuran on Several Microsomal and Cytosolic Enzyme Activities in Mouse Liver)

  • 이물질(xenobiotics) 대사에 관여하는 간장 microsome과 cytosol 효소 활성에 indole, indole-3-carbinol 및 benzofuran이 미치는 영향을 검색하기위하여 마우스에 이들 약물을 각각 5 mmole/kg씩 10일간 투여하여 다음 몇 가지의 성적을 얻었다. Benzofuran은 microsome 효소인 aniline hydroxylase, 7-ethoxycoumarin O-deethylase, p-nitrophenol UDPGA-transferase, epoxide hydrolase와 cytosol 효소인 glutathione S-tranferase, NADH : quinone reductase, UDP-glucose dehydrogenase의 활성도를 증가시켰다. 그러나 benzofuran과는 구조적으로 furan ring내의 N원소가 O원소로 치환되었을 뿐 주된 구조가 유사한 indole과 indole-3-carbinol 투여로는 UDPGA-transferase와 NADH: quinone reductase의 활성도 증가를 볼 수 없었으며, 특히 indole은 NADPH : cytochrome C reductase만을 증가시킨데 비하여 구조상 indole에 carbinol (methanol)기가 붙은 indole-3-carbinol은 수종의 mixed function oxidase와 아울러 특히 epoxide hydrolase의 활성도 역시 증가시켰다. 이러한 결과는 benzofuran과 indole-3-carbinol에 의한 epoxide hydrolase 활성도 증가의 기전의 일부를 설명할 수 있을 것으로 생각된다.

가공 인상의 항피로효과 (Effect of Heat Processed Ginseng on Anti-Fatigue)

  • Processing of traditional herbal medicine is one of the pharmaceutical technique in oriental medicine. Most frequently used processing method in oriental medicine are roasting and steaming. In this studies, to elucidate the pharmacological transformation of traditional herbal medicine by means of processing them, Ginseng Radix (root of Panax ginseng, Araliaceae) was used as a sample. Processed ginseng radix (SGR, Sun Ginseng) was prepared by steaming of roots of white ginseng (GR) for 3 hours at $120^{\circ}C$. The biological activities of methanol extract of GR and SGR were investigated. According to DPPH radical scavenging effects, and inhibitory effects of xanthine oxidase and AAPH induced hemolysis, PGR exhibited more effective than those of GR in vitro. And, the antifatigue effect of GR and SGR were investigated using a weight-loading forced swimming test by monitoring swimming times and prolonged intensity exercise model rats by measuring blood biochemical parameters. GR and SGR were significantly prolonged swimming times in 8% body weight ratio loaded mice. Also, they had the inhibitory effects on the decrease of blood glucose levels, the elevation of serum creatinine, lactic acid and free fatty acid, and lactic dehydrogenase activities in forces swimming rats with 1% of the body weight attached to the neck for 3 hours. SGR was more excellent than GR on these effect. Also, these effects were transformed to the n-butanol fraction of methanol extract of SGR. From these results, it can be considered that SGR has antifatigue effect.

Mouse hepatoma 세포를 이용한 농산부산물로부터 quinone reductase활성물질의 탐색 (Screening of Quinone Reductase Inducers from Agricultural Byproducts Using Mouse Hepatoma Cell Line)

  • Quinone reductase(QR)를 포함한 2상효소계를 활성화시키는 성분들은 많은 동물실험에서 발암물질의 세포내 작용을 억제함으로서 항종양효과를 나타내는 것으로 보고되어 있다. 본 연구에서는 대표적인 농산부산물로서 미강, 밀기울, 탈지대두박, 두유박, 참깨박, 들깨박등 6종의 시료에 대한 암예방효과를 갖는 물질의 존재여부를 탐색하기 위하여, mouse hepatoma cell line(Hepalclc7 cells) 을 이용하여, quinone reductase활성유도 여부를 측정하였다. 참깨박과 들깨박의 80%메탄올 추출물은 0.5mg/ml 농도에서 강력한 QR 유도활성을 나타냈으며, 같은 농도에서 다른 시료들은 거의 QR 효소활성을 증가시키지 않았다. 한편 QR효소활성을 유도하는 성분을 찾아내기 위하여 일차적으로 TLC를 수행한 결과, 참깨박과 들깨박의 메탄올 추출물 가운데 사용한 전개용매(n-butanol : n-propanol : 2N ammonium hydroxide(10 : 60 : 30)에서 가장 빨리 이동하는 분획(Rf=0.70)이 유효성분을 함유하고 있음을 확인하였으며, 현재 활성성분의 동정이 진행중에 있다.

Glutamate에 의한 산화적 스트레스로부터 신경세포를 보호하는 제비꽃 추출물의 영향 (Protective Effect of Neuronal Cell on Glutamate-induced Oxidative Stress from Viola mandshurica Extracts)

  • 본 연구에서는 신경독소일 뿐만 아니라 흥분성 신경전달물질로 잘 알려져 있는 glutamate 세포독성이 산화적 손상과 관련하고 있고, 여기에 방어효과를 보이는 제비꽃 추출물에 관하여 연구하였다. MTT reduction assay를 통하여 glutamate의 세포독성을 확인하였고 ascorbic acid와 같은 대표적인 항산화제를 처리한 후 광학 현미경을 이용한 형태학적 변화를 관찰하였다. N18-RE-105 세포주에 최종 농도 20mM의 glutamate를 처리 하면 40.8% 의 생존율을 보이는데 반하여 ascorbic acid 500 ${\mu}M$ 최종농도로 처리하였을 때 85.3%의 세포 생존율을 확인할 수 있었다. 그리고 신경세포 보호효과를 가지는 제비꽃을 methanol, ethanol, acetone 추출한 뒤 MTT reduction assay를 이용하여 활성을 확인하였으며 그 중 acetone 추출물을 최종농도 50, 100 ${\mu}g/ml$를 처리 시 76.8%, 79.4%로 가장 높은 세포 생존율을 확인할 수 있었다. 이 결과는 N18-RE-105 세포주의 형태학적 변화와 LDH release assey에서도 일치하는 결과를 확인하였다.

Hepatoprotective Effects of Waxy Brown Rice Fermented with Agrocybe cylindracea

  • The methanol extract of waxy brown rice fermented with Agrocybe cylindracea was prepared. The extract was then freeze dried and fed to rats at the level of 0.5, 1.0, 2.0 g/kg body weight for 14 days, followed by the treatment with carbon tetrachloride for three consecutive days to induce hepatotoxicity. After sacrificing the rats, the enzyme activity of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), and ${\gamma}$-glutamyl transpeptidase (${\gamma}$-GTP) in serum was determined. Biochemical analysis on serum for albumin, total protein, triglyceride, and total as well as HDL-cholesterol were carried out along with a histopathological study of liver tissues. Based on these data, we suggest that the waxy brown rice cultured with A. cylindracea may exert hepatoprotective activity against hepatotoxicity caused by chemicals such as carbon tetrachloride.
