The contaminated soil at abandoned smelter areas present challenge for remediation, as the degraded materials are typically deficient in nutrients, and rich in toxic heavy metals and metalloids. Bioremediation technique is to isolate new strains of microorganisms and develop successful protocols for reducing metal toxicity with heavy metal tolerant species. The present study collected metal contaminated soil and characterized for pH and EC values, and heavy metal contents. The pH value was 5.80, representing slightly acidic soil, and EC value was 13.47 mS/m. ICP-AES analytical results showed that the collected soil samples were highly contaminated with various heavy metals and metalloids such as lead (183.0 mg/kg), copper (98.6 mg/kg), zinc (91.6 mg/kg), and arsenic (48.1 mg/kg), respectively. In this study, a bacterial strain, Bacillus cereus KM-15, capable of adsorbing the heavy metals was isolated from the contaminated soils by selective enrichment and characterized to apply for the bioremediation. The effects of heavy metal on the growth of the Bacillus cereus KM-15 was determined in liquid cultures. The results showed that 100 mg/L arsenic, lead, and zinc did not affect the growth of KM-15, while the bacterial growth was strongly inhibited by copper at the same concentration. Further, the ability of the bacteria to adsorb heavy metals was evaluated.
The sensitivity of a mehtanogen and sulfate-reducing bacterium isolated from a sea-based landfill site Cd$^{2+}$ and CU$^{2+}$ was studied. Methanogens and sulfate-reducing bacteria in leachates of the waste disposal site were enumerated using the MPN method. Methanobacterium thermoautotrophicum KHT, isolated from the leachate, could not grow at 0.5 mM Cd$^{2+}$ or 1.0 mM CU$^{2+}$. Desulfotomaculum sp. RHT, isolated from the same leachate, was able to insolubilization 3.0 mM Cd$^{2+}$ or 2.0 mM CU$^{2+}$ by production of hydrogen sulfide. When strains KHT and RHT were cultured together in the presence of the heavy metals, strain KHT could grow at high heavy metal concentrations after insolubilization of the metals by strain RHT. strain RHT.

Microbiologically influenced Corrosion (MIC) is one of the most deleterious effects of metal microbe interactions. When a fresh metal surface comes in contact with a non-sterile fluid, biofilm formation is ensued. This might result in the initiation of corrosion. The sites and materials where MIC is implicated are versatile. Industries such as shipping, power generation, chemical etc are reported to be affected. The rapid and unexpected failure of AISI type 304 stainless steel was investigated in the laboratory by simulation studies for a period of 4 months. Slime and water samples from the failure site were screened for corrosion causing bacteria. Both aerobic and anaerobic nora were enumerated and identified using PCR techniques. Pseudomonas sp. and Bacillus sp. were the most common aerobic bacteria isolated from the water and slime samples, whilst sulfate reducing bacteria (SRB) were the major anaerobic bacteria. The aerobic bacteria were used for the corrosion experiments in the laboratory. Coupon exposure studies were conducted using a very dilute (0.1%V/V) nutrient broth medium. The coupons after retrieval were observed under a Scanning Electron Microscope (SEM) for the presence of MIC pits. Compared to sterile controls, metal coupons exposed to Pseudomonas sp and Bacillus sp. showed the initiation of severe pitting corrosion. However, amongst these two strains, Psudomonas sp. caused pits in a very short span of 14 days. Towards the end of the experiment, severe pitting was observed in both the cases. The detailed observation of pits showed they vary both in number and shapes. Whilst the coupons exposed to Bacillus sp. showed widely spread scales like pits, those exposed to Pseudomonas sp. showed smaller and circular pits, which had grown in number and size by the end of the experiment. From these results it is inferred that the rapid and unexpected failure of 304 SS might be due to MIC. Pseudonwnas sp. could be considered as the major responsible bacteria that could initiate pits in the metallic structures. As the appearance of pits was different in both the tested strains, it was thought that the mechanisms of pit formation are different. Experiments on these lines are being continued.
휴폐광지역의 산성광산배수를 처리하기 위한 자연정화처리시설로서 소택지의 설치가 증가하고 있다. 이러한 소택지의 기질물질로서는 흔히 석회석, 참나무조각, 버섯퇴비 등이 벌크형태로 적용된다. 이러한 소택지의 정화효율은 시간이 지남에 따라 점차 저하되며, 특히 산성배수와의 화학반응에 의한 기질물질 층의 투수계수 감소에 주로 기인한 것으로 파악된다. 본 연구의 목적은 산성광산배수 처리시설에서 황산염환원균(SRB) 고정화 담체의 적용성을 평가하기 위한 것이다. 고정화담체는 유기물질인 버섯퇴비와 참나무 퇴비, pH 완충제인 석회석가루 등을 토양미생물을 접종한 m-CSB와 혼합하여 제작하였다. 본 실험에서 고정화담체는 벌크형태에 비하여 pH, 황산염제거율 및 중금속 제거율에서 효율이 우수함을 확인할 수 있었다.
이 연구의 목적은 KURT(KAERI underground research tunnel) 지하수 내에 금속이온을 환원시키는 미생물의 존재 여부를 확인하고 배양하여, 이들의 활동에 따른 철과 망간 환원의 관찰과 환원물의 광물학적 특성을 연구함으로써, 금속환원미생물에 의한 산화상태로 존재하는 철과 망간의 환원과 광물 상전이 가능성을 확인하는 것이다. KURT 지하수 내 금속을 환원하는 미생물은 전자공여체로 포도당, 초산, 젖산, 개미산, 피루브산을, 전자수용체로 Fe(III)-citrate를 사용하여 농화배양 하였으며, 16S rRNA 분석을 통해 종 다양성을 확인하였다. 농화배양된 금속환원미생물에 의한 철과 망간의 환원과 생광물화작용을 알아보기 위해 전자공여체로 포도당, 초산, 젖산, 개미산, 피루브산을, 전자수용체로 철수산화물인 아카가나이트(akaganeite, ${\beta}$-FeOOH)와 망간산화물(manganese oxide, ${\lambda}-MnO_2$)을 이용하여 금속환원 실험을 실시하였다. 미생물 활동에 의해 형성된 환원물의 광물학적 특성은 SEM, EDX, XRD 분석을 통해 확인되었다. 연구 결과 KURT 지하수에서 금속을 환원하는 혐기성 미생물로는 Fusibacter, Desulfuromonas, Actinobacteria, Pseudomonas sp. 등이 확인되었고, 이 미생물들은 체외에서 철과 망간을 환원하여 이들 광물의 상전이를 확인하였다. 철(Fe)은 $Fe^{3+}$을 포함한 아카가나이트(${\beta}$-FeOOH)에서 $Fe^{2+}/Fe^{3+}$를 포함한 자철석($Fe_3O_4$)으로 환원되었고, 망간(Mn)은 $Mn^{4+}$를 포함한 망간산화물(${\lambda}-MnO_2$)에서 $Mn^{2+}$을 포함한 능망간석($MnCO_3$)으로 환원되었다. 이러한 지하 140 m의 KURT 지하수에서 서식하는 미생물들에 의해 철과 망간이 환원됨은 다른 중금속과 핵종원소의 환원 가능한 환경이 조성되었을 뿐 만 아니라, 미생물에 의하여 환원된 철의 재산화에 의해서도 주변 핵종원소가 환원될 수 있음을 의미한다. 따라서 이러한 직 간접적인 산화-환원 반응에 의해 KURT 지하수 내에서는 금속환원미생물들이 유해금속물질을 침전시켜 이동성을 줄일 수 있을 뿐만 아니라 고준위 폐기물에서 유해물질의 유출시 핵물질의 확산을 막는데 중요한 역할을 할 수 있을 것으로 사료된다.
Microbiologically influenced corrosion (MIC) of carbon steel gas pipeline in soil environments was investigated at field and laboratory MIC is very severe corrosion and it is not easy to distinguish this corrosion from Inorganic corrosion because of its localized, pitting-type character Therefore, it is important to provide proper assessment techniques for the prediction, detection, monitoring and mitigation of MIC. It is possible to predict the MIC risk, i.e., the activity of sulfate-reducing bacteria (SRB) through the analysis of soil environments. Chemical, microbiological and surface analysis of corrosion products and metal attacked could reveal the possibility of the occurrence of MIC. Various electrochemical and surface analysis techniques could be used for the study of MIC. Among these techniques, thin-film electrical resistance (ER) type sensors are promising to obtain localized corrosion rate of MIC induced by SRB. It is also important to study the effect of cathodic protection (CP) on the MIC In case of coated pipeline, the relationship between coating disbondment and the activity of SRB beneath the disbanded coating is also important.
Copper is used in various applications in environments favoring and enabling formation of biofilms by naturally occurring microbes. Copper is also the chosen corrosion barrier for nuclear waste in Finland. The copper canisters should have lifetimes of 100,000 years. Copper is commonly considered to be resistant to corrosion in oxygen-free water. This is an important argument for using copper as a corrosion protection in the planned canisters for spent nuclear-fuel encapsulation. However, microbial biofilm formation on metal surfaces can increase corrosion in various conditions and provide conditions where corrosion would not otherwise occur. Microbes can alter pH and redox potential, excrete corrosion-inducing metabolites, directly or indirectly reduce or oxidize the corrosion products, and form biofilms that create corrosive microenvironments. Microbial metabolites are known to initiate, facilitate, or accelerate general or localized corrosion, galvanic corrosion, and intergranular corrosion, as well as enable stress-corrosion cracking. Sulfate-reducing bacteria (SRB) are present in the repository environment. Sulfide is known to be a corrosive agent for copper. Here we show results from corrosion of copper in anoxic simulated ground water in the presence of SRB enriched from the planned disposal site.

The purpose of this study was to investigate the potential of zeolite-supported sulfatereducing bacteria (SRB) in enhancing the removal of Cu2+, Ni2+, and Cr6+ in contaminated seawater. Our results show that SRB-immobilized zeolite carriers can enhance the removal of heavy metals. In addition, heavy metals were generally better removed at conditions of 37°C. Cu2+, Ni2+, and Cr6+ were effectively removed by 98.2%, 90.1%, and 99.8% at 100 parts per million concentration of the heavy metals, respectively. These results indicate that SRB-zeolite carriers hold great potential for use in the removal of cationic heavy metal species from marine environment.
The biotechnology including genetic engineering is expected to be applied to various fields of wastewater treatments in order to promote biological reaction rate, to grade up the effluent quality and to advance the stability of microorganisms against temperature, pH and toxic substances. The current topics in Japan on application of biotechnology to wastewater treatment will be reviewed at the beginning of the presentation. Next, the research of Biochemical Engineering Laboratory is to be presented, especially focused on the following subjects ; (1) Application of genetic engineering to the investiation of heavy metal uptake by the resistant bacteria of Hg Cd or Zn. (2) Relationship between sulfate reducing bacteria and wastewater treatment, offensive odor and corrosion of sewer tranks.
