콩 발효식품에 있어서 페놀물질의 분리와 이의 항산화성 (Antioxidative characteristics of isolated Crude Phenolics from Soybean Fermented Foods(Doenjang))

  • Antioxidative characteristics of doenjang(fermented soybeans paste) phenolics on the lipid oxidation systems were studied by the determination of the oxidative related activity including lipoxygenase (LOX) inhibition, metal chelating and free radical scavengning of doenjang phenolics. Manlikong variety containing the highest amounts of phenolic compounds among the soybean variety, was used for doenjang processing. Doenjang was prepared by the series of processes including soaking for overnight, cooking for 1hr at 12Lb, first fermentation (3 days at 30$\pm$2$^{\circ}C$) for the preparation of meju(soybean koji) after inoculation of Asp. oryzae, and further fermentation(60 days at 30$\pm$2$^{\circ}C$) for the ripening after addition of salt 13% to meju. In order to investigate the antioxidative activity of phenolics in doenjang, the doenjang phenolics was extracted with methanol form freeze dried defatted doenjang. Antioxidative effects of methanol extract on linoleic acid oxidation system were observed by the significantly decreased levels of peroxide and conjugated diene formation. In addition, methanol extract resulted in the inhibition of LOX activity. and also, metal(FeCl$_3$) chelation and free radical scavengning activities were increased with increasing concentration of methanol extract.

  • The new tridentate ligands of nitrogen donor atoms N,N-Bis(2-amino-ethyI)-benzyl-amine 2HCI(BABEA. 2HCI) and 2,6-Bis(amino-methyl)-pyridien. 2HCI(BAMP. 2HCI) were synthesized as their dihydrochloride salts and characterized by TA, IR. Mass and NMR spectroscopy. The protonation constants of the ligands and stability constants for Cd$^{2+}$, Pb$^{2+}$, Zn$^{2+}$and Cu$^{2+}$ ions were determined by potentiometric titration in aqueous solutions and compared with those of analogous ligands. The effect stability constants of ligands and metal ions for removal of heavy metals in aqueous solution were described.

  • The aim of present study was to examine the inhibitory effects of hydroxamic acid derivatives on the melanogenesis. We found that hydroxamic acid moiety was important for anti-melanogenic activity. Compounds 1a and 1b strongly inhibited melanin synthesis via deactivation of tyrosinase. Hydroxamic acid has metal ion chelating ability which is similar to that kojic acid, however, anti-tyrosinase mechanism of compounds 1a and 1b was different from that of kojic acid. Enzymatic Properties of Intracellular Adenosine Deaminase from Nocardioides sp. J-326TK

  • The properties of purified intracellular adenosine deaminase (adenosine aminohydrolase, EC 3.5.4.4) of Nocardioides sp. J-326TK isolated from soil have been studied. The enzyme deaminated adenosine and 2`-deoxyadenosine and the respective {TEX}$K_{M}${/TEX} values were 4.0×{TEX}$10^{-4}${/TEX} M and 5.0× {TEX}$10^{-4}${/TEX} M, but the enzyme was not active on 8-bromoadenosine, 6-methylaminopurine riboside, ATP, ADP, 2`-AMP, 3`-AMP, 5`-AMP, dAMP, cAMP, NAD, FAD, NADP and adenine. The enzyme activity was strongly inhibited by the addition of {TEX}$Hg^{2+}${/TEX} and {TEX}$Ag^{+}${/TEX}, {TEX}$Cu^{2+}${/TEX}, {TEX}$Co^{2+}${/TEX} and {TEX}$Mn^{2+}${/TEX} also inhibited the activity but much less extent. The effect of alkyl reagents, metal chelating reagents and certain other compounds on the enzyme activity were also examined. No reagent activated the enzyme. On the contrary, the enzyme reaction was slightly inhibited by o-phenanthroline and 6-benzyladenosine.

표고버섯의 Mitochondrial $F_1-ATPase$ 활성도에 미치는 금속이온의 역할에 관한 연구 (Study on the Role of Metal ions for the Activity of the Mitochondrial $F_1-ATPase$ in Lentinus edodes)

  • 표고버섯의 mitochondria에 금속 chelating agent인 10 mM EDTA 및 10 mM o-Phe을 포함하는 10 mM Tris-HCl 완충용액(pH 7.5)으로 48시간 동안 각각 투석처리 하였을 때 mitochondria 내의 철 이온 함량이 투석처리를 하지 않은 대조구에 비하여 각각 74% 및 68% 감소되며, mitochondrial $F_1-ATPase$의 활성도는 대조구에 비하여 각각 56% 및 49%의 활성을 상실하였다. EDTA 투석에 의하여 활성을 상실한 metal-free mitochondrial $F_1-ATPase$$0.5\;mM\;Fe^{3+}$$0.05\;mM\;Mg^{2+}$에 의하여 각각 81% 및 70%의 활성이 회복되었으며, $Fe^{2+}$에 의해서는 활성이 회복되지 않았다. 또한 이 효소는 $0.5\;mM\;Fe^{3+}$$0.05\;mM\;Mg^{2+}$이 공존할 때 95%의 재활성화를 나타내었으며, $Fe^{2+}$$Mg^{2+}$의 공존 효과는 없었다. o-Phe으로 투석한 효소도 각 이온에 대하여 EDTA로 투석한 효소와 유사한 결과를 나타내었다. 그러므로, 표고버섯 내의 mitochondrial $F_1-ATPase$는 그 활성을 나타내는 데 있어서 $Fe^{3+}$$Mg^{2+}$을 필요로 함을 알았다. 기질 ATP에 대한 효소의 Km값은 1.67 mM이나, 효소를 가장 크게 활성화시키는 $0.5\;mM\;Fe^{3+}$$0.05\;mM\;Mg^{2+}$이 존재할 때의 Km값은 0.65 mM로서 기질 친화성이 증가되었다.

국내산 Black Chokeberry와 Blueberry의 총 폴리페놀, 총 플라보노이드 함량 및 생리활성 비교 (Comparison of Total Polyphenols, Total Flavonoids, and Biological Activities of Black Chokeberry and Blueberry Cultivated in Korea)

  • 본 연구에서는 국내에서 생산되는 블랙초크베리와 블루베리의 기능적 특성을 비교 조사하기 위하여 총 폴리페놀 함량, 총 플라보노이드 함량, DPPH radical 소거능, superoxide 음이온 소거능, ABTS radical 소거능, 금속 킬레이트 효과, 환원력, 아질산염 소거능 등을 측정하였다. 총 폴리페놀 함량은 블랙초크베리가 117.20 mg/g, 블루베리가 42.26 mg/g이었으며 총 플라보노이드 함량은 블랙초크베리가 32.50 mg/g, 블루베리가 26.39 mg/g으로 나타났다. 생리활성 측정 결과 블랙초크베리는 본 실험에서 조사한 기능성에서 탁월한 활성을 나타내었는데 추출물의 농도가 증가함에 따라 전반적으로 활성이 증가하는 경향을 보였으며, 특히 DPPH radical 소거능, ABTS radical 소거능, 금속 킬레이트 효과, 아질산염 소거능은 5 mg/mL의 농도에서 90% 이상의 활성을 나타냄으로써 양성대조군으로 사용한 ascorbic acid의 활성과 대등하였다. 블루베리도 추출물의 농도증가에 따라 활성이 증가하는 경향을 나타내었으나 블랙초크베리의 0.2~0.7배에 해당하는 낮은 활성을 나타내어 블랙초크베리보다 기능성이 떨어지는 것을 알 수 있었다. 블랙초크베리와 블루베리 추출물의 총 폴리페놀 함량, 총 플라보노이드 함량과 생리활성 간의 상관관계를 조사한 결과, 총 폴리페놀 및 총 플라보노이드 농도가 증가할수록 생리활성이 증가하는 경향을 나타내었고 이들 간에는 높은 양(+)의 상관성이 있음을 보여주었다.

에탄올아민이 금속에 의한 종이의 노화 방지에 미치는 영향 (Effect of Ethanolamine Species on Paper Aging by Metals)

  • It is known that ethanolamines play a critical role for deacidification of paper sized by alum-rosin. However, amines also are effective as a chelating agent of metal. The present work was focused on whether amines could scavenge metals and prevent from the aging of paper. Metals such as alum, copper(II) and iron(III) was added to paper, and the paper treated with amines was aged in a thermo-hygrostat for 3-6 days. In the case of paper added to alum, the amines efficiency against paper aging was good in the oder of triethanolamine, diethanolamine and monoethanolamine attributable to the intensity of basicity and steric effect. Even in the case of paper treated with copper(II) chloride, iron(III) chloride, and copper(II) chloride, the significant preservation efficiency was shown by ethanolamine during accelerated aging. This outcome pinpoints the fact that ethanolamine can prevent paper aging not only from acid by neutralizing acid contained in paper but also from metals by producing of complexes with metals. These consequences above convince that ethanolamine makes it possible for mass deacidification for paper which contains acid and metals. Future studies should be conducted concerning whether, in reality, the treatment of its gas mode, in a single or multiple applications, has significant effect on lessening paper aging.

각종 Flavonoids의 라디칼 봉쇄능과 ACE 활성 억제능에 관한 연구 (Studies on the Radical Scavenging Effects and the Inhibitory Effects on ACE Activity of Several Flavonoids)

  • 천연에서 생산되는 식물성 천연항산화성 물질인 flavonoid의 지질 산화 억제능과 라디칼 봉쇄능 및 금속착체 형성능을 비롯하여 ACE 저해능을 조사하고 그 결과를 보고하였다. 즉, flavonoid와 linoleic acid를 반응시킨 결과 linoleic acid의 산화를 크게 억제하는 것으로 나타났으며 특히, 산화의 초기에 억제 효과가 큰 것으로 나타났다. 이들의 DPPH에 대한 수소 공여능을 조사한 실험에서 공시한 대부분의 flavonoid가 양질의 효과를 나타내었으나 naringin과 kaempferol은 봉쇄능이 약한 것으로 나타났으나 daidzein과 morin, quercetin 등은 아주 우수한 radical 봉쇄능을 가지는 것으로 나타났다. 한편, rutin의 금속 착체 형성능을 조사한 실험에서는 C $u^{2+}$$Mg^{2+}$에 대한 결합능이 있는 것을 확인할 수 있었다. 그리고 ACE 저해능을 조사한 실험에서는 모든 시료에서 ACE 활성을 저해하는 결과를 나타내었다. 특히, isoflavone인 daidzein과 genistein에서 저해능이 가장 큰 것으로 나타났다.

Evaluation of Antioxidative Activity of Agrimonia pilosa-Ledeb Leaves on Non-lipid Oxidative Damage

  • Present study was conducted to evaluate the anti oxidative activity of the Agrimonia pilosa-Ledeb leaves on non-lipid oxidative damage. The antioxidative activity of methanolic (MeOH) extract of the Agrimonia pilosa-Ledeb leaves on non-lipid oxidation, including liposome oxidation, deoxyribose oxidation, protein oxidation, chelating activity against metal ions, scavenging activity against hydrogen peroxide, scavenging activity against hydroxyl radical and 2'-deoxyguanosine (2'-dG) oxidation were investigated. The MeOH extract of the Agrimonia pilosa-Ledeb leaves exhibited high anti oxidative activity in the liposome model system. Deoxyribose peroxidation was inhibited by the MeOH extract of the Agrimonia pilosa-Ledeb leaves and MeOH extract of the Agrimonia pilosa-Ledeb leaves provided remarkable protection against damage to deoxyribose. Protective effect of MeOH extracts of the Agrimonia pilosa-Ledeb leaves on protein damage was observed at $600{\mu}g$ level (82.05%). The MeOH extracts of the Agrimonia pilosa-Ledeb leaves at $300{\mu}g$ revealed metal binding ability (32.64%) for hydrogen peroxide. Furthermore, the oxidation of 2'-deoxyguanosine (2'-dG) to 8-hydroxy-2-deoxyguanosine (8-OH-2'dG) was inhibited by MeOH extracts of the Agrimonia pilosa-Ledeb leaves and scavenging activity for hydroxyl radical exhibited a remarkable effect. From the results in the present study on biological model systems, we concluded that MeOH extract of the Agrimonia pilosa-Ledeb leaves was effective in the protection of non-lipids against various oxidative model systems.

붉나무 껍질 에탄올 추출물의 추출특성과 항산화 활성에 관한 연구 (Extraction Characteristics and Antioxidant Activity of Ethanol Extract of Rhus javanica Bark)

  • 붉나무의 유용성에 대한 기초자료를 마련하고자 붉나무 수피를 에탄올로 추출하여 추출물의 몇 가지 이화학적 특성과 항산화 활성을 측정하였다. 추출물의 가용성 고형분의 함량은 73.5 mg/100g, 당도는 $17.8^{\circ}Brix$로 측정되었으며, 총방향족 화합물, 총플라보노이드 함량 및 총페놀성 화합물의 함량은 각각 흡광도 0.508, 49.88 mg/100g 및 296.6 mg/100g으로 측정되었다. 추출물의 환원력은 대조구로서 추출물의 가용성 고형분과 동량의 ascorbic acid의 27.5 %로 측정되었으나, 철환원력은 ascorbic acid와 유사하게 측정되었다. 추출물의 DPPH라디칼 소거능은 95.16 %로서 ascorbic acid와 거의 유사하였고, 금속이온 봉쇄능은 ascorbic acid의 74.73 %보다 높은 81.58 %로 측정되었다. 그리고 pH 2에서 추출물의 아질산염 소거능은 51.76 %로 측정되었으며, Rancimat test에 의한 대두유 산화 억제효과가 있음을 확인하였다.
