Purpose : The role of ghrelin, which promotes the secretion of growth hormone, was not well known until now. Recently it was found that the mutation of ghrelin gene is related to obesity and diabetes. This study is to find the screening method that can easily and effectively detect the polymorphism of Leu72Met in ghrelin gene of obesity patients and apply it to clinical usage. Methods : We compared PCR-RFLP, PCR-SSCP and ARMS methodologies for analyzing of the polymorphism of Leu72Met in ghrelin gene of obesity children, and also studied the merits and demerits of these methodologies. Results : In this study, we were able to find out the band of peculiar allele of Leu72Met in ghrelin gene using PCR-RFLP, PCR-SSCP and ARMS analyses. The polymorphism of Leu72Met in ghrelin gene determined by all above methodologies was in complete agreement. Compared to the PCR-RFLP and PCR-SSCP, ARMS analysis is simple, inexpensive and also consume less time. It is very sensitive to analyze the polymorphism and easy to understand the results of test. Conclusion : Though PCR-RFLP, PCR-SSCP and ARMS analyses were sensitive to analyze the polymorphism of Leu72Met in ghrelin gene, ARMS analysis appears to be more efficient than PCR-RFLP and PCR-SSCP. Therefore, we conclude that ARMS analysis is suitable to analyze the polymorphism of Leu72Met in ghrelin gene for large quantity of specimens.

BACKGROUND/OBJECTIVES: Although the association of body mass index (BMI) with metabolic syndrome (MetS) is well documented, there is little knowledge on the independent and joint associations of BMI and physical activity with MetS risk based on a continuous scoring system. This study was designed to explore the effect of physical activity on interactions between excess body weight and continuous metabolic syndrome (cMetS) in a nationwide survey of Iranian children and adolescents. SUBJECTS/METHODS: Data on 5,625 school students between 10 and 18 years of age were analyzed. BMI percentiles, screen time activity (STA), leisure time physical activity (LTPA) levels, and components of cMetS risk score were extracted. Standardized residuals (z-scores) were calculated for MetS components. Linear regression models were used to study the interactions between different combinations of cMetS, LTPA, and BMI percentiles. RESULTS: Overall, 984 (17.5%) subjects were underweight, whereas 501 (8.9%) and 451 (8%) participants were overweight and obese, respectively. All standardized values for cMetS components, except fasting blood glucose level, were directly correlated with BMI percentiles in all models (P-trend < 0.001); these associations were independent of STA and LTPA levels. Linear associations were also observed among LTPA and standardized residuals for blood pressure, high-density lipoprotein, and waist circumference (P-trend < 0.01). CONCLUSIONS: Our findings suggest that BMI percentiles are associated with cMetS risk score independent of LTPA and STA levels.
In this study, a diagnostic wind model, CALMET and a micrometeorological numerical model, ENVI-MET were used to analyze the wind field in and out of the site designated for the industrial complex around Buron-myeon, Wonju, Gangwon-do. The results of modeling with CALMET showed that the air flow in industrial complex was little affected by the surrounding terrain. And the result of wind field analysis with ENVI-MET showed there are turbulent air flows such as cavity and wake around structures in the industrial complex, which can cause high-air pollution. Therefore, it is necessary to design the industrial complex considering the wind path according to wind directions.
MET is one of the few physical therapy having its own criteria. Mr. Holten approached the Norwegian Health Authority in 1967 to get his MET System recognized as a treatment method of its own. He was granted his approval for his exercise system's specivic criteria connecteed with the treatment method. The apparatus should be designed that functional quality (arthrogenous, circulatory, respiratory, neuromuscular) in question is optimally influenced when the patient carries out exercise in a certain range against a graded resistance. The therapy reassesses the scheme of treatment at least every fifth session and the maximum number of patients being 5 person per hour. The important principles in medical exercise Therapy are stabilization of hyperfunction through the system of autostabilization and mobilization of hypofunction through automobilization. In MET excrcises are adjusted to the patient's reactions. MET equipment is therefore made to meet requirements for treating patients with painful pathological dysfunction in the musculo-skeletal system.

This research measures and compares on-site net radiation energy, air temperature, wind speed, and surface temperature considering various spatial characteristics with a focus on land use types in urban areas in Changwon, Southern Gyeongsangnam-do, to analyze the accuracy of an ENVI-met model, which is an analysis program of microclimate. The on-site measurement was performed for three days in a mobile measurement: two days during the daytime and one day during the nighttime. The analysis using the ENVI-met model was also performed in the same time zone as the on-site measurement. The results indicated that the ENVI-met model showed higher net radiation than the on-site measurement by approximately $300Wm^{-2}$ during the daytime whereas the latter showed higher net radiation energy by approximately $200Wm^{-2}$ during the nighttime. The temperature was found to be much higher by approximately $2-6^{\circ}C$ in the on-site measurement during both the daytime and nighttime. The on-site measurement also showed higher surface temperature than the ENVI-met by approximately $7-13^{\circ}C$. In terms of the wind speed, there was a significant difference between the results of the ENVI-met model and on-site measurement. As for the correlation between the results of the ENVI-met model and on-site measurement, the temperature showed significantly high correlation whereas the correlations for the net radiation energy, surface temperature, and wind speed were very low. These results appear to be affected by excessive or under estimation of solar and terrestrial radiation and climatic conditions of the surrounding areas and characteristics of land cover. Hence, these factors should be considered when applying these findings in urban and environment planning for improving the microclimate in urban areas.

In order to estimate the availability of ruminally-protected Lys (RPLys) and Met (RPMet), quantitative changes of free Lys and Met in the total abomasal outflow and feces were determined in three Holstein heifers with a T-shaped duodenal cannula. RPLys was prepared by coating Lys with fat and RPMet with a pH-sensitive polymer. RPLys and RPMet containing 30 g of each amino acid were supplied together to heifers, and total abomasal outflow was collected hourly from the T-shaped duodenal cannula for a 48 h period. Collected abomasal outflows were fractionated into liquid and solid phases to measure free Lys and Met concentration. At 2 wk after total abomasal outflow sampling, heifers were supplied RPLys and RPMet together again, and total feces were collected every 12 h for a 3 d period t estimate excreted RPLys and RPMet in feces. The amounts of Lys and Met recovered from the liquid fractions of abomasal outflow were estimated to be $23.9{\pm}8.3%$ and $68.6{\pm}3.6%$ (p=0.008) of RPLys and RPMet ingested, respectively. Final intestinal disappearances of Lys and Met were estimated to be $49.5{\pm}2.6%$ and $78.2%{\pm}6.5%$ (p=0.015) of ingested RPLys and RPMet, respectively.

Objectives : The aim of this study was to examine the effects of brain-derived neurotrophic factor (BDNF) genetic polymorphism and job stress on the severity of alcohol drinking. It was hypothesized that individuals with the Met/Met BDNF genotype would be more vulnerable than those carrying the Val allele. Methods : Participants were 133 healthy Korean adults (mean age $28.2{\pm}1.1$). Job stress and the severity index of drinking were investigated through self-reported questionnaires. BDNF (rs6265) gene was genotyped. Results : There was no significant association between job stress and the severity of alcohol drinking. Although the severity of alcohol drinking was not associated with BDNF genetic polymorphism, there was a significant difference in men according to genotype and job stress. Men with homozygous BDNF Met allele were more severe in alcohol drinking when job stress was high, less severe in alcohol drinking when job stress was low than those carrying the Val allele (F = 4.47, p = 0.038). Also higher level of job stress was correlated with higher severity of alcohol drinking in men homozygous for BDNF Met allele (rs = 0.620, p = 0.005). Conclusions : These findings suggest the possibility that Met allele could have differential susceptibility, with men homozygous for BDNF Met allele being more susceptible to both more adverse and less adverse environmental influences.
Objective : This study concernes the usefulness of $^{11}C-methyl-L-and$ D-methionine[Met]-positron emission tomography[PET] for glioma grading and detection of recurrence in gliomas, compared with fluorine-18, 2-fluoro-deoxyglucose[FDG]-PET. Methods : Eighty patients underwent Met-PET study for evaluation of glioma : 37 astrocytomas [WHO grade II, 3; III, 8; IV, 26]. 27 oligodendrogliomas [WHO grade II, 16; III, 11]. and 12 suspicious recurrent gliomas. All images were taken within 2 weeks before operation. For suspicious recurrent cases on magnetic resonance images, both FDG-PET and Met-PET were performed. Results : In astrocytoma, Mean maximum standard uptake value[SUV] of region of interest[ROI] was not different between WHO grades [p=0.108]. but ROI/normal contralateral tissue SUV [T/N] ratio was statistically different between WHO grades [p=0.002]. T/N ratio was more closely related to visual scale than maximum SUV of ROI [p<0.001 and p=0.107 respectively]. In oligodendroglioma, there was no statistical difference between WHO grades in view of maximum SUV and T/N ratio. For recurrent gliomas, sensitivity of FDG-PET and Met-PET was 25% and 100%, while specificity of FDG-PET and Met-PET were 100% and 80%, respectively. Conclusion : Met-PET might be an appropriate tool for tumor grading in astrocytoma and be more sensitive for detection of recurrence in gliomas than FDG-PET.
The growth characteristics of sulfur-oxidizing bacteria, Acidithiobacillus thiooxidans AZ11, MET, and TAS were investigated in mineral salt media supplemented with elemental sulfur of 1$\sim$50 g $L^{-1}$. The sulfur oxidation rates of A. thiooxidans. MET and TAS increased highly with increasing sulfur concentration up to 10 g L$^{-1}$, but the rates increased slowly in sulfur concentration over 10 g L$^{-1}$. A. thiooxidans AZ11 showed the parallel increase of sulfur oxidation rate until sulfur concentration increased up to 40 g L$^{-1}$. The maximum sulfur oxidation rates (V$_{max}$) of AZl1, MET and TAS were 1.88, 1.38 and 0.43 g S L$^{-1}$ d$^{-1}$, respectively. The maximum specific growth rates (${\mu}_{max}$) of AZ11, MET, and TAS were 0.33 d$^{-1}$, 0.30 d$^{-1}$ and 0.45 d$^{-1}$, respectively. Although MET and TAS couldn't grow at sulfate concentration of 40 g L$^{-1}$, AZ11 could grow in the presence of 58 g L$^{-1}$ sulfate, the final oxidation product of elemental sulfur.
Purpose: PMV, PET, and similar thermal comfort indices and microclimate modeling have recently become actively used to evaluate thermal comfort. This study will look at pedestrian roads with diverse spatial characteristics on university campus using the ENVI-met model as the base for onsite measurement. Method: The PET was used as the thermal comfort index. The first microclimate measures were collected on September 20, 2014, and the second microclimate measures were collected on June 1, 2015. The ENVI-met model was used at the same time. Result: As a results, Onsite measurement results differed depending on the PET spatial characteristics. The location associated with the most discomfort had a PET of $47.8^{\circ}C$. The spatial characteristics of this place included a with no shade. The most comfortable location had shade, and the PET was $24.6^{\circ}C$. When the ENVI-met model and onsite measurements were compared, similar patterns were found, but with a few differences at specific points; this was due to the limitation of using input materials such as trees, buildings, and covering materials with the ENVI-met model. This factor must be thoroughly considered when analyzing modeling results.
