최근 척수손상 백서에서 인체지방조직유래 중간엽 줄기세포 이식 및 성장호르몬의 투여가 신경회복에 미치는 영향 (The Effect of Human Adipose Tissue Derived Mesenchymal Stem Cells and Growth Hormone on the Recovery of Neurological Deficits due to Experimental Spinal Cord Injury in Rat)

  • Purpose: Human adipose tissue-derived mesenchymal stem cells(hATSCs) can be differentiated into multiple mesenchymal lineages, including bone, cartilage, and muscle. And growth hormone play important roles in the normal growth and development of the CNS. In this study, we explored whether the transplanted hATSCs and growth hormones could improve functional recoveries from rats with contusive spinal cord injury. Methods: We divided 30 female rats, which were subjected to a weight driven implant spinal cord injury, into 3 groups with 10 rats each; Group A as a control group, group B with hATSCs transplantation on injured region, and group C with hATSCs transplantation and GH administration for 7 days. Then, we researched their neurologic functional recoveries before and 2, 4, and 8 weeks after transplantation using Basso-Beattie-Bresnahan (BBB) locomotor rating scale. And we checked Y-chromosome positive cells by FISH(Fluorescent in situ hybridization) to identify the survival of transplanted mesenchymal stem cells. Results: After 4 weeks of transplantation, the group B and group C showed significant improvement of neurologic function on BBB locomotor rating scale in comparison with the group A(Group A: $13.1{\pm}0.58$, Group B: $14.6{\pm}0.69$, Group C: $14.9{\pm}0.56$). Moreover, the group C displayed meaningful recovery of neurologic function after 8 weeks in comparison with group B (Group B: $15.7{\pm}0.63$, Group C: $16.5{\pm}1.14$). The group A, the control one, improved for 5 weeks after injury, and had no more recovery. On the other hand, Group B and C showed the improvement of neurologic function continuously for 9 weeks after injury. Conclusion: In this study, we found out that hATSCs transplantation have an effect on neurologic functional recovery of spinal cord injured rat and GH injection seems to bring the synergistic results on this good tendency.

인간 유방암 줄기세포에서 레몬잎 메탄올 추출물의 항암 효능 (Anticancer Activities of the Methanolic Extract from Lemon Leaves in Human Breast Cancer Stem Cells)

  • 본 연구에서는 레몬 잎 메탄올 추출물(MLL)의 인간 유방암 줄기 세포인 MCF-7-SC에 대한 항암 활성을 조사하였다. MLL이 MCF-7-SC에서 apoptosis를 유도하였으며, 이를 apoptotic body의 형성, sub-G1 phase 및 annexin V-positive 세포와 Bax/Bcl-2 ratio의 증가, caspase-9과 caspase-3의 활성화 및 PARP의 절 단을 통하여 확인하였다. 동시에 MCF-7-SC에서 MLL은 acidic vesicular organelles의 형성, LC3-II의 축적 증가, Akt/mTOR/p70S6K의 활성 억제 등을 통하여 autophagy를 유도하였다. Epithelial-mesenchymal transition (EMT)는 세포가 전이 상태를 획득하기 위한 중요한 과정이며, 이 기작은 암세포가 전이되는 것을 억제함에 있어서 중요한 표적이 된다. 낮은 농도에서의 MLL은 epithelial 마커 단백질인 E-cadherin이 증가와 mesenchymal 마커 단백질인 Snail과 Slug의 발현 감소를 통해 EMT 과정을 저해함으로써 MCF-7-SC에서 항전이 활성을 나타내었다. 본 연구에서는 레몬 잎 메탄올 추출물이 농도 의존적으로 유방암 줄기세포에 대해 세포 독성과 항전이 활성을 나타내고 있으며, 따라서 레몬잎은 항암 소재로서의 개발 가능성이 높은 식물이라고 사료된다.

Cobalt chloride에 의해 유도되는 상피-중간엽 이행에서의 국소부착 단백질의 인산화의 역할 규명 (Role of FAK Phosphorylation in Cobalt Chloride-Induced Epithelial-to-Mesenchymal-Like Transition)

  • 본 연구는 인간 폐암세포의 저산소 상태를 재현하기 위한 $CoCl_2$의 처리 조건을 최적화 하였고, 최적화 된 저산소 상태에서 인간 폐암세포의 암화 과정 및 기전을 규명하였다. 인간 폐암세포, A549와 H460에 500 ${\mu}M$ $CoCl_2$를 24시간 처리하였을 때 저산소 상태의 대표적인 전사인자, HIF-$1{\alpha}$의 발현이 증가함을 확인하였고 인간 폐암세포들의 성장에는 전혀 영향을 미치지 않음을 확인하였다. 또한 $CoCl_2$를 처리한 인간 폐암 세포에서 상피-중간엽 이행(epithelial-to-mesenchymal-like transition)의 대표적인 마커인 E-cadherin 발현의 감소와 ${\alpha}$-SMA의 증가를 확인하였고, 세포-세포 간 junction 부위가 깨어짐을 E-cadherin 형광염색 실험을 통하여 확인하였다. 더 나아가 $CoCl_2$를 처리한 인간 폐암 세포에서 상피-중간엽 이행의 분자적 기전을 밝히기 위해 세포벽에 존재하는 인테그린(integrin)의 발현을 웨스턴 블랏팅과 FACS분석을 통하여 알아본 결과, $CoCl_2$를 처리한 인간 폐암세포에서 인테그린 ${\beta}3$발현의 증가를 확인하였다. 뿐만 아니라, $CoCl_2$를 처리한 인간 폐암세포에서 인테그린 ${\beta}3$의 하부 신호전달 물질인 국소부착 카이네이즈(FAK)의 인산화가 증가함을 확인하였다. 상기의 결과로서, 국소부착 카이네이즈의 인산화를 저해함으로써 인간 폐암세포가 악성세포로 전이되는 것을 저해할 수 있을 것으로 기대 되어진다.

산소 농도의 변화와 물리적 손상이 제대혈 중간엽 줄기세포의 증식에 미치는 영향 (Effect of Oxygen Concentration, Physical Trauma on Proliferation of Umbilical Cord Blood-derived Mesenchymal Stem Cells)

  • Human umbilical Mesenchymal Stem Cell(uMSC) has been known as one of major component to regenerate connective tissues such as bone, cartilage, fat and others. The effect of low(5%), normotensive(20%) oxygen and freezing-thawing damage on proliferation of uMSC were investigated. low oxygen concentration culture of uMSC resulted in enhanced proliferation significantly($p$ <0.05) than 20% of oxygen culture. After the freezing-thawing injury to uMSC, 5% oxygen culture showed marked proliferation of uMSC than that of 20% oxygen($p$ <0.05) in the 5th passage of uMSC. Expression of antioxidant enzymes such as superoxide anion 1 and glutathione peroxidase 1 appeared marked in 20% oxygen cultured uMSC, which suggest oxidative stress could induce less proliferation of uMSC. Above findings would suggest proliferation of uMSC in 5% of oxygen will give more yields.

Anti-metastatic mechanism of mountain cultivated wild ginseng in human cancer cell line

  • Objective : Ginseng is one of most widely used herbal medicine. Ginseng showed anti-metastasis activities. However, its molecular mechanisms of action are unknown. So we want to report the wild ginseng repress which plays key roles in neoplastic epithelial-mesenchymal transition process. Methods : Treatment of the human colorectal carcinoma LOVO cells and human gastric carcinoma SNU601 cells with the increased concentrations of cultivated wild ginseng extracts resulted in a gradual decrease in the AXIN2 gene expression. Results : Metastasis-suppressor genes, maspin and nm23 was not affected by the treatment of ginseng extracts in LOVO cells. Moreover, the mountain cultivated wild ginseng or mountain wild ginseng are similar in their inhibitory effects on the expression of AXIN2 gene, but are substantially stronger than cultivated ginseng. Conclusion : We described the novel mechanism of wild ginseng-induced anti-metastasis activity by repressing the expression of AXIN2 gene that plays key roles in epithelial-mesenchymal transition process.

폐 과오종의 세침흡인 세포학적 소견 - 1례 보고 - (Fine Needle Aspiration Cytology of Pulmonary Hamartoma - Report of a Case -)

  • Pulmonary hamartoma is the most common benign tumor of the lung but rarely encountered in routine cytologic work-up. We recentely experienced a case of fine needle aspiration cytology of pulmonary hamartoma in a 65-year old male patient. The characteristic cytopathologic features were 1) The mesenchymal components with cuboidal epithelial cells in a clean background, 2) The presence of fibromyxoid mesenchymal component composed of fibroblasts in a loose fibrillary stroma, and 3) Antler-liker fibromyxoid cluster having the rimming of epithelial cells which correspond to the epithelial glowing in cleft-like fashion in pulmonary hamartoma.
