[ $PLC-{\gamma}1$ ] for Differentiating Adenocarcinoma from Reactive Mesothelial Cells in Effusions (체강 삼출액의 진단에 있어서 $PLC-{\gamma}1$ 면역 염색의 유용성)

  • Cytologic diagnosis of reactive or malignant effusion is sometimes difficult. Especially, differentiation of benign reactive mesothelial cells from malignant cells in body effusion is more difficult. Recently, immunohistochemistry has been used to diagnose difficult cases. Phospholipase $C(PLC)-{\gamma}1$ is one of the isoenzyme of the PLC which plays central role in signal transduction involving cellular growth, differentiation and transformation by phosphorylating many protein component. Increased expression of $PLC-{\gamma}1$ in human breast carcinoma, colorectal carcinoma and stomach cancers are reported. To evaluate the efficacy of positive $PLC-{\gamma}1$ immunostaining in the diagnosis of malignancy in effusions, paraffin-embedded cell blocks of pleural fluid and ascites from 10 patients(5 metastatic adenocarcinomas, and 5 reactive mesothelial cells) were immunostained with a monoclonal antibody to $PLC-{\gamma}1$. $PLC-{\gamma}1$ immuostained all the adenocarcinomas in cell block(5/5) with intense membrane pattern, however, none of the reactive mesothelial proliferations stained with the diagnostic membrane pattern. Thus, our study strongly supports the conclusion that $PLC-{\gamma}1$ immunopositivity is likely to become a useful adjunct for the diagnosis of malignancy in effusions.

Low Temperature Effects on the Strength and Fracture Toughness of Membrane for LNG Storage Tank (LNG 저장탱크용 멤브레인재(STS 304강)의 강도 및 파괴인성에 미치는 저온효과)

  • Tensile and fracture toughness tests of the cold-rolled STS 304 steel plate for membrane material of LNG storage tank were performed at wide range of temperatures, 11 IK(boiling point of LNG), 153K , 193K and 293K(room temperature). Tensile strength significantly increases with a decrease in temperature, but the yield strength is relatively insensitive to temperature. Elongation at 193K abruptly decreases by 50% of that at 293K, and then decreases slightly in the temperature range of 193K to 111K. Strain hardening exponents at low temperatures are about four times as high as that at 293K. Elastic-plastic fracture toughness($J_c$) and tearing modulus($T_{mat}$) tend to decrease with a decrease in temperature. The $J_c$ values are inversely related to effective yield strength in the temperature range of 111K to 293K. These phenomena result from a significant increase in the amount of transformed martensite in low temperature regions.

  • Timing of oviposition, nuclear maturation of oocytes, egg-sperm contact and brooding were observed in the giant fresh-water prawn Macrobrachium rosenbergii reared in the laboratory. When a pre-spawning molted female was transferred to the male, the female oviposited commonly at 5 to 10 hr after mating. The first polar body were extruded within 2-3 min after spawning. The egg chromosome progressed to release second polar body by 30 min and mitotic nuclear division occurred in 8 hr after spawning. Thumbtack-shaped spermatozoa were seen peneterating egg membrane with their spike directed forward. Transformation of spermatozoa and fertilization membrane could not be observed by light microscopy. The elapsed hours from oviposition to the end of brooding wee taken in 5-7 hr. Brood size for female 9 cm and 18 cm in body length was estimated to be 10,000~15,000 and 100,000 eggs, respectively and is proportional to the total length of the berried females. The ovigerous females reared at chlorinity of 2.21~4.25$\textperthousand$Cl. showed a normal egg development up to hatching.

Improved antimicrobial effect of ginseng extract by heat transformation

  • Background: The incidence of halitosis has a prevalence of 22-50% throughout the world and is generally caused by anaerobic oral microorganisms, such as Fusobacterium nucleatum, Clostridium perfringens, and Porphyromonas gingivalis. Previous investigations on the structure-activity relationships of ginsenosides have led to contrasting results. Particularly, the antibacterial activity of less polar ginsenosides against halitosis-related bacteria has not been reported. Methods: Crude saponins extracted from the Panax quinquefolius leaf-stem (AGS) were treated at $130^{\circ}C$ for 3 h to obtain heat-transformed saponins (HTS). Five ginsenoside-enriched fractions (HTS-1, HTS-2, HTS-3, HTS-4, and HTS-5) and less polar ginsenosides were separated by HP-20 resin absorption and HPLC, and the antimicrobial activity and mechanism were investigated. Results: HPLC with diode-array detection analysis revealed that heat treatment induced an extensive conversion of polar ginsenosides (-Rg1/Re, -Rc, -Rb2, and -Rd) to less polar compounds (-Rg2, -Rg3, -Rg6, -F4, -Rg5, and -Rk1). The antimicrobial assays showed that HTS, HTS-3, and HTS-4 were effective at inhibiting the growth of F. nucleatum, C. perfringens, and P. gingivalis. Ginsenosides-Rg5 showed the best antimicrobial activity against the three bacteria, with the lowest values of minimum inhibitory concentration and minimum bactericidal concentration. One major reason for this result is that less polar ginsenosides can more easily damage membrane integrity. Conclusion: The results indicated that the less polar ginsenoside-enriched fraction from heat transformation can be used as an antibacterial agent to control halitosis.

Three-Point Bending Fatigue Properties of Austenitic 304 Stainless Steel Sheets for Membrane (Membrane용 오스테나이트계 304 스테인리스강 판재의 3점 굽힘피로 특성)

  • Three-point bending fatigue properties of austenitic 304 stainless steel sheets were investigated at room temperature and LNG temperature($-162^{\circ}C$) in the strain range from 0.43 to $1.7\%$. The fatigue properties at $-162^{\circ}C$ were superior to those at room temperature due to the higher volume fractions of deformation-induced martensite. The cyclic hardening behavior owing to the deformation- induced martensite transformation was detected in both specimens. In room temperature testing, the mean load amplitude increased steadily with cycles, meaning that cumulative plastic incubation strain was required for martensite transformation. On the contrary, in $-162^{\circ}C$ tested specimen, the mean load amplitude increased rapidly within a few cycles due to the rapid transformation of martensite, and slightly decreased after the maximum is reached probably due to dynamic recovery.

  • Freezing of water in a polymer electrolyte membrane fuel cell (PEMFC) may cause severe problems in driving a fuel cell vehicle during the winter time. Characteristics of PEMFC which suffered low temperatures below zero degree was examined with the thermal cycles from 80 to $-10^{\circ}C$. With the thermal cycles, the cell performance was degraded due to the phase transformation and volume changes of water. Effects of freezing of water in PEMFC on the electrode structure and polarization resistance were examined by BET analysis, cyclic voltammetry, and AC impedance spectroscopy.

Enhanced Production of Itaconic Acid through Development of Transformed Fungal Strains of Aspergillus terreus

  • Metabolic engineering with a high-yielding mutant, A. terreus AN37, was performed to enhance the production of itaconic acid (IA). Reportedly, the gene cluster for IA biosynthesis is composed of four genes: reg (regulator), mtt (mitochondrial transporter), cad (cis-aconitate decarboxylase), and mfs (membrane transporter). By overexpressing each gene of the IA gene cluster in A. terreus AN37 transformed by the restriction enzyme-mediated integration method, several transformants showing high productivity of IA were successfully obtained. One of the AN37/cad transformants could produce a very high amount of IA (75 g/l) in shake-flask cultivations, showing an average of 5% higher IA titer compared with the high-yielding control strain. Notably, in the case of the mfs transformants, a maximal increase of 18.3% in IA production was observed relative to the control strain under the identical fermentation conditions. Meanwhile, the overexpression of reg and mtt genes showed no significant improvements in IA production. In summary, the overexpressed cis-aconitate decarboxylase (CAD) and putative membrane transporter (MFS) appeared to have positive influences on the enhanced IA productivity of the respective transformant. The maximal increases of 13.6~18.3% in IA productivity of the transformed strains should be noted, since the parallel mother strain used in this study is indeed a very high-performance mutant that has been obtained through intensive rational screening programs in our laboratory.

  • Bekesy measured the sound transformation system of the middle ear 49 years ago. According to his reports, a ratio between the size of ear drum and the size of oval window is 17 : 1, and the lever function of the ossicles is physiologically 1.3 : 1. Therefore, the hearing might be aggravated to 27.5㏈ in the case of the vanishment of 3 ossicles. In 1952, Wullstein reported 5 types of tympanop-lasty and the fourth type among them was especially named for the sound-protection. The oval window is only exposed by the sound pressure and the round window is not exposed. According to the application by this idea, the post-operative hearing might be improved until 27.5㏈. Mean while, in 1942, Onchi verified through his experiment that the results of Bekesy's measurement was not completely conformed to Onchi result. Bekesy measured the sound pressure on the stapes plate of the oval window, on the other hand, Onchi measured the sound pressure on the surface of the perilymph of the oval window after removing the stapes plate(Fig. 1).(Figure omitted) The difference of their experiment is recognized that the impedance of the stapes plate exists or not (Fig. 1). Both Audiogrums are compared as Fig. 2. The result of IV type of tympanoplasty is success ful in 54% as the Table 1. (Table Omitted) The reason of unsatisfactory is caused by the thick and unmovable window-membrane and by the closing of air passage to the round window. The closing of the air passage to the round window is occurred by the adhesion between the grafting membrane and the surface of promontorium. In order to preserve this adhesion, I produce to transplantate the mucous membrane of the lip to the bone surface of tympanic cavity after removing the granulation tissue of the tympanic cavity and to form a membranous canal for the sake of air passage (Fig. 3). (Figure Omitted) The post-operative hearing by this method is shown as Fig. 4, 5. In other words, the post-operative sound pressure entered into the cochlea directly, by way of the oval window only, not by way of the round window, as a theorie of the sound protection. Effects of Tobacco-Specific Carcinogen on Protein Kinase C Isoforms (흡연특이성 발암물질이 특정 Protein Kinase C Isoform에 미치는 영향)

  • Cigarette smoking is the leading cause of the lung cancer. However, mechanism of action underlying the carcinogenesis in the lung still remains to be elucidated. The present study attempted to look into the carcinogenic potential of tobacco-specific nitrosamine, NNK (4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone) and the effects of protein kinase C (PKC) isoforms in an immortalized human epithelial cell model. Material and Method: Immortalized human epithelial cells were exposed with NNK and examined for its carcinogenic potential as measured by saturation density, soft-agar colony formation, and cell aggregation assay. The specific isoform of PKCs involved in the cellular transformation was analysed through western blot with monoclonal antibody and measured separately in cytosolic fraction and membrane fraction. Result: Human epithelial cells exposed with NNK showed prominent carcinogenic potential in saturation density, soft agar colony formation, and cell aggregation assay. PKC isoform analysis results are as follows: PKC- $\alpha$ showed significant translocation of protein levels from cytosolic fraction to membrane fraction, as analyzed by immunoblot. PKC- $\varepsilon$ showed a dose-dependent increase of translocation. PKC- λ was not affected by NNK treatment. Conclusion: The study demonstrated that there was a certain specificity in the patterns of isoform induction following chemical carcinogen exposure. Thus, it is suggested that identification of specific isoform be a clue to find target molecules in the carcinogenesis.

  • The purpose of this paper is to find minimum surface shape of pneumatic structure using the finite element method. The pneumatic membrane structure is a kind of large deformation problem and very flexible composite material, which mean geomatric nonlinearity. It is not to resist for compression and resultant moment. As the displacement due to internal pressure is getting bigger, it should be considered the direction of forces. It becomes non-linear problem with the non-conservative force. The follower-force depends on the deformation and the direction of force is normal to each element. The solution process is obtained the new stiffness matrix (load correction matrix) depending on deformation through each iterated step. However, the stiffness matrix have not the symmetry and influence on the time of covergence. So in this paper Newton-Rhapson method for solving non-linear problem and for using symmetic matrix, the load direction is changed in each iterated step using the transformation matrix.
