This study was designed to investigate the effects of silk fibroin powder (Mw 500) on oxidative stress and membrane fluidity in liver membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10g) were fed basic diet (control group), and experimental diets (SEP-2.5 and SFP-5.0 groups) added 2.5 and 5.0 g/kg BW/day for 6 weeks. Cholesterol levels resulted in a significant decrease (12.1% and 9.0%, respectively) in the liver mitochondria and microsomes of SEP-5.0 group compared with control group. Membrane fluidity as significantly increased (16.1% and 16.5%, 5.8% and 17.4%) in the liver mitochondria and microsomes were significantly inhibited (16.1% and 18.3%, 8.1% and 15.1%, respectively) at the SFP-2.5 and SEP-5.0 groups compared with control group. Induced oxygen radicals (BOR) in liver mitochondria and microsomes were significantly inhibited (16.1% and 18.3%, 8.1% and 15.1%, respectively) at the SFP-2.5 and SEP-5.0 groups compared with control group. Induced oxygen radicals (IOR) in liver microsomes were significantly inhibited (17.0% and 26.6%, respectively) at the SFP-2.5 and SFP-5.0 groups compared with control group, but IOR in liver mitochondria was significantly inhibited about 12.3% at the SWP-400 group only compared with control group. Lipid peroxide (LPO) levels were significantly decreased (8.3% and 18.0%, 13.4% and 18.4%, respectively) in the liver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were dose-dependently decreased (5.4% and 11.6%, 19.0% and 24.4%, respaectively) in the iver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. These results suggest that administration of SFP may play an effective role in attenuating an oxidative stress and increasing a membrane fluidity in liver membranes.

Sprague-Dawley(SD) male rats(160$\pm$10 g) were fed basic diet(control group), and experimental diets(SWP-200 and SWP-400 groups) added 200 and 400mg/kg BW/day for 6 weeks. There were no significant differences in cholesterol levels of brain memberanes by administration of silkworm powder (SWP). Membrane fluidities were significantly increased(21.5% and 30.8%, respectively) in brain mitochondria of SWP-200 and SWP-400 groups compared with control group, but significant difference between brain microsomes could not obtained. Basal oxygen radicals (BORs) in brain mitochondria and mircrosomes were significantly inhibited(8.5% and 16.5%, 16.8%and 24.8%, respectively) by SWP-200 and SEP-400 groups compared with control group. Induced oxygen radicals(IORs) in brain mitochondria were significantly inhibited(16.6% and 21.4%, respectively)by sWP-200 and SWP-400 groups compared with control group, but IOR in brain microsome were significantly inhibited about 16.0% by SWP-400 groups only compared with control group. Lipid peroxide(LPO) levels were significantly decreaed(14.8%and 22.4%, respectively) in brain mitochondria of SWP-200 and SWP-400 groups compared with control group, but LPO level was significantly decreased about 16.0% in brain microsome of SWP-400 group only. Oxidized protein(OP) levels were remarkably decreased(about 14.8% and 16.5%, respectively) in brain mitochondria of SWP-200 and SWP-400 groups, but OP level was significantly decreased about 13.0% in brain microsome of SWP-400 group only compared with control group, Theses results suggest that administration of in brain microsome of SWP-400 group only compared with control group. These results suggest that administration of SWP may play effective role in attenuating an oxidative stress and increasing a membrane fluidity in brain membranes.
The response of Spirulina (Arthrospira) platensis to high salt stress was investigated by incubating the cells in light of moderate intensity in the presence of 0.8 M NaCl. NaCl caused a decrease in photosystem II (PSII) mediated oxygen evolution activity and increase in photosystem I (PSI) activity and the amount of P700. Similarly maximal efficiency of PSII (Fv/Fm) and variable fluorescence (Fv/Fo) were also declined in salt-stressed cells. Western blot analysis reveal that the inhibition in PSII activity is due to a 40% loss of a thylakoid membrane protein, known as D1, which is located in PSII reaction center. NaCl treatment of cells also resulted in the alterations of other thylakoid membrane proteins: most prominently, a dramatic diminishment of the 47-kDa chlorophyll protein (CP) and 94-kDa protein, and accumulation of a 17-kDa protein band were observed in SDS-PAGE. The changes in 47-kDa and 94-kDa proteins lead to the decreased energy transfer from light harvesting antenna to PSII, which was accompanied by alterations in the chlorophyll fluorescence emission spectra of whole cells and isolated thylakoids. Therefore we conclude that salt stress has various effects on photosynthetic electron transport activities due to the marked alterations in the composition of thylakoid membrane proteins.
Objectives : Alcohol-induced oxidative stress has been known to injure various tissues or organs. This stress is related with free radicals which are produced as the result of long-term alcohol consumption. Malonyldialdehyde(MDA) is produced by the interaction of free radicals and cell membrane lipids, and indicates the degree of lipid peroxidation indirectly. The purpose of this study was to investigate the relationship between red blood cell(RBC) membrane lipid peroxidation by free radicals, and associated hepatic injuries and hematologic changes. Methods : Thirty-three subjects diagnosed as alcohol dependence according to DSM-IV diagnostic criteria were evaluated within 72 hours after discontinuing alcohol drinking. Clinical characteristics were evaluated by CAGE questionnaire and Korean Michigan Alcoholism Screening Test(MAST). RBC membrane MDA level was measured as the marker of RBC membrane lipid peroxidation. Aspartate aminotransferase(AST), alanine aminotransferase(ALT) and gamma-glutamyltransferase(GGT) were used as the biochemical markers of liver damage due to alcohol ingestion. The alcohol-induced hematologic change was assessed by mean corpuscular volume(MCV). Results : The results were as follows. Clinical characteristics were not different between two groups having normal and abnormal levels of AST, ALT, GGT or MCV. The levels of MDA were not correlated with the clinical characteristics and serum levels of AST, ALT and GGT. However, there was a significant correlation between the levels of MDA and the value of MCV(p=0.017). Conclusions : These findings suggest that oxidative stress in alcohol dependence may not be reflected in liver enzyme markers such as AST, ALT and GGT, but may be reflected in MCV.
The effect of mulberry (Morus alba L.) leaf extract(MLE) on oxidative stress and membrane fluidity in brain membranes of SD rats fed with 100 and 300 mg/kg BW/day were carried out for 6 weeks. Cholesterol accumulations resulted in a consistent decreases (4.6% and 5.6%, respectively) in brain mitochondria and microsomes of MLE-300 group compared with control group. Membrane fluidities were dose-dependently increased (2.2% and 5.1%, 5.0% and 15.2%) in brain mitochondria and microsomes of MLE-100 and MLE-300 groups compared with control group. Basal oxygen radicals(BORs) in brain mitochondria and microsomes were significantly inhibited (15.7% and 25.1%, 9.0% and 12.4%, respectively) by MLE-100 and MLE-300 groups compared with control group. Induced oxygen radicals(IORs) in brain mitochondria and microsomes were significantly inhibited (8.9% and 13.1%, 16.5% and 23.2%, respectively) by MLE-100 and MLE-300 groups compared with control group. Lipid peroxide (LPO) levels were significantly decreased (8.5% and 18.1%, 7.6% and 12.3%) in brain mitochondria and microsomes of MLE-100 and MLE-300 groups compared with control group. Oxidized protein (OP) levels were dose-dependently decreased (4.3% and 14.2%, 10.0% and 10.9%, respectively) in brain microsomes of MLE-100 and MLE-300 groups compared with control group. These results suggest that MLE may play an effective role in an attenuating an oxidative stress and increasing a membrane fluidity in brain membranes.
In general, the design of membrane structures takes three steps. The first is shape finding analysis which is determination of initial equilibrium geometry with uniform stresses. The second step involve the computation of the stress-deformation to get completed membrane under various load conditions. The third step is to divide the membrane structures into several plan strips from the initial equilibrium states. This procedure is needed because of the initial shape has usually undevelopable curved surface and is called as "cutting patterns generation". By introducing this work, the deformation due to the initial stress is removed and approximate cutting patterns are generated. In this approach, however, material properties is not considered, therefore the error between the design stresses and actual stresses during the fabrication of plan strips should be occurred. In this paper, actual equilibrium shape analysis procedure for HP shape models is presented. The deviations of stresses between the design stresses and actual stresses are estimated.
In general, the cutting pattern of the membrane structures is determined by dividing the complicated curved 3-D surface into several 2-D plane strip by using flattening technique. In this procedure, however, some discrepancies ore occurred between actual stresses of equilibrated state and designed uniform stresses because the material properties are not considered. These deviations can cause the critical structural problems, wrinkling or overstress, and thus a optimization process should be considered. In this paper, a new analytical method for determining an optimum cutting pattern considering material properties is presented. Here, iterative procedure is introduced to decrease the errors caused in numerical process. The optimization method proposed can diminish the deviations occurred by material properties and numerical errors, simultaneously. As a results, it is shown that the final stress distributions for the HP shell model are sufficiently near to design stress distributions, and it can be concluded that this method can be used to obtain the optimized cutting pattern of membrane structures.
The purpose of this paper is to develope the evaluation technique to find proper elastic constants that characterize the material nonlinearity of structural membrane. The stress-strain curves of membrane material show strong nonlinearity. But generally the analysis is carried out under the assumption on material linear and geometrical nonlinear method. Because, it is very difficult to evaluate proper tangential stiffness. This paper use multi-step-linear approximation method taking the concept of effective stress for the evaluation of stiffness of membrane material, and then compare the results between linear and nonlinear analysis. Also. it shows better results than linear method

The response mechanism of simply supported two-way reinforced concrete (RC) slabs under fire was numerically studied from the view of stress redistribution using the finite element software ABAQUS. Results show that: (1) Simply supported two-way RC slabs undergo intense stress redistribution, and their responses show four stages, namely elastic, elastic-plastic, plastic and tensile membrane stages. There is no cracking in the fire area of the slabs until the tensile membrane stage. (2) The inverted arch effect and tensile membrane effect improve the fire resistance of the two-way slabs. When the deflection is L/20, the slab is in an inverted arch effect state, and the slab still has a good deflection reserve. The deformation rate of the slab in the tensile membrane stage is smaller than that in the elastic-plastic and plastic stages. (3) Fire resistance of square slabs is better than that of rectangular slabs. Besides, increasing the reinforcement ratio or slab thickness improves the fire resistance of the slabs. However, an increase of cover thickness has little effect on the fire resistance of two-way slabs. (4) Compared with one-way slabs, the time for two-way slabs to enter the plastic and tensile cracking stage is postponed, and the deformation rate in the plastic and tensile cracking stage is also slowed down. (5) The simply supported two-way RC slabs can satisfy with the requirements of a class I fire resistance rating of 90 min without additional fire protection.
