SYBR Green I-based Real-time PCR Assay and Melting Curve Analysis for Rapid Detection of Staphylococcus aureus from Raw Milks Samples (Real-time PCR을 이용한 원유시료 유래 황색포도상구균의 신속 검출)

  • The aim of this study was to develop a LightCycler-based real time PCR (LC-PCR) assay and to evaluate its diagnostic use for the detection of Staphylococcus aureus in raw milk samples. Following amplification of 113 bp of coa gene encoding an coagulase precursor specific for Staphylococcus aureus, melting curve and DNA sequencing analysis was performed to verify the specificity of the PCR products. Amplification of 209 bp gene encoding an altered penicillin-binding protein, PBP2a (mecA), melting curve analysis and DNA sequencing analysis was performed to verify methicillin resistance Staphylococcus aureus (MRSA). According to this study, 6 of 647 raw milk samples showed S. aureus positive and 2 of them showed a mecA positive and the detection limit was 10 fg of DNA. And we also isolated Staphylococcus chromogenes a causative agent of exudative epidermitis in pigs and cattle from 3 samples.

  • The microstructural changes of nylon 6 UDY, POY and FDY were compared after free-annealing through crystallinity, birefringence, and melting behavior analyses. Free-annealing was done at various temperatures $(120^{\circ}C\;,140^{\circ}C,\;160^{\circ}C,\;180^{\circ}C,\;200^{\circ}C)$ and times (15 min., 30 min., 60 min.) using vaccum oven. Crystallinity was measured by the density gradient column technique and birefringence was measured using a Nikon polarizing microscope with a quartz wedge and Senarmont compensator. Melting behavior was investigated on the basis of DSC melting corves. Crystallinites of specimens increased as the treatment temperature and time increased. Birefringence of UDY increased after annealing and increased as the treatment temperature increased. On the other hand, those of POY and FDY decreased after annealing. Especially, the changes of crystallinity and birefringence of treated POY were particularly lower than those of treated UDY and FDY. Melting peaks of untreated UDY, POY and FDY were different in the position and the shape, but little change was seen in melting peaks in spite of increasing the annealing temperature and time. UDY and FDY showed single melting peaks in all the specimens. But POY showed double melting peaks, which means the coexistences of crystals with different thermal properties.

  • An analysis is performed to investigate the influence of the buoyancy force and the thickness variation of melting layer in the containment that is filled with phase-change Material surrounding a cylindrical heating tube during melting process. The phase-change material is assumed to be initially solid at its phase-change temperature and the remaining solid at any given time is still at the phase-change temperature and neglecting the effect of heat transfer occuring within the solid. At the start of melting process, the thickness of melting layer is assumed to be a stefan-problem and after the starting process, the change of temperature and velocity is calculated using a two dimensional finite difference method. The governing equations for velocity and temperature are solved by a finite difference method which used SIMPLE (Semi Implicit Method Pressure linked Equations) algorithm. Results are presented for a wide range of Granshof number and in accordance with the time increment and it is founded that two dimensional fluid flow occurred by natural convection decreases the velocity of melting process at the bottom of container. The larger the radius of heating tube, the higher heat transfer is occurred in the melting layer.

  • Heat transfer phenomena during inward melting process of the phase change material were studied experimentally. N-docosane paraffin [$C_{22}H_{46}$] is used for phase change material and its melting temperature is $42.5^{\circ}C$. Experiments were performed for melting of an initially no-sub cooled or subcooled solid in a horizontal cylinder, in order to compare and investigate the radial temperature distribution, ratio of melting and melted mass, various energy components stored from the cylinder wall, figure of the melting front in the horizontal cylinder. The solid-liquid interface motion during phase change was recorded photographically. The experimental results reaffirmed the dominant role played by the conduction at early stage, by the natural convection at longer time during inward melting in the horizontal cylinder. Ratio of melting and melted mass are more influenced by wall temperature, rather than by the initial temperature of solid. The latent energy is the largest contributor to the total stored energy.

  • In the present investigation, experiments on the melting of a phase change material were performed to research heat transfer phenomena generated by means of conduction and natural convection in the vertical tube at inward melting. The phase change material used in the experiments is 99 percent pure n-Docosane paraffin which is measured melting temperature of $42.5^{\circ}C$, latent heat of 37.5 cal/g, heat conductivity of $0.1505W/m^{\circ}C$. Experiments were performed both in the no-subcooling which is initiating it at melting temperature of phase change material, and in the subcooling which means to initiate it under melting temperature of phase change material, in order to compare and investigate the horizontal temperature history, vertical temperature history, ratio of melting and melted mass, figure of the melting front in the vertical tube. In the experimental results, heat transfer from tube wall to phase change material were due to conduction at early stage and due to natural convection with the passage of time, and then occurred melting downward from surface by volumetric expansion. Natural convection affects temperature distribution in the tube, ratio of melting and melted mass, figure of the melting front and then progress rapidly in case of nosubcooling compared to subcooling.

  • Ge-Sb-Se계 칼코게나이드 유리의 Melting 조건변화에 따른 특성변화를 연구하였다. Glass melting 조건(homogenization-temperature, homogenization-time, annealing) 에 따라 제작된 칼코게나이드 유리 bulk를 FT-IR, XRD, SEM 등의 분석장비를 이용하여 특성을 분석하였다. Homogenization temperature가 높을수록 석영관 급냉 시 발생되는 mechanical stress와 내부응력차로 인해 칼코게나이드 유리 깨짐현상이 증가하였으며 조성비와 melting 조건에 따라 XRD분석에서 확인되지 않는 미소결정이 SEM 분석결과 관찰되었다. 본 연구를 통해 칼코게나이드 유리의 melting 조건에 따른 경향성을 확인할 수 있었다.

An Experimental Study of Accelerating Phase Change Heat Transfer

  • The present paper investigated the effect of ultrasonic vibrations on the melting process of a phase-change material (PCM). Furthermore, the present study considered constant heat flux boundary conditions unlike many of the previous researches adopted constant wall temperature conditions. Therefore, in the present study, modified dimensionless parameters such as Ste* and Ra* were used. Also, general relationships between melting with ultrasonic vibrations and melting without ultrasonic vibrations were established during the melting of PCM. Experimental observations show that the effect of ultrasonic vibrations on heat transfer is very important throughout the melting process. The results of the present study reveal that ultrasonic vibrations accompany the effects like agitation, acoustic streaming, cavitation, and oscillating fluid motion. Such effects are a prime mechanism in the overall melting process when ultrasonic vibrations are applied. They enhance the melting process as much as 2.5 tildes, compared with the result of natural melting. Also, energy can be saved by applying ultrasonic vibrations to the natural melting. In addition, various time-wise dimensionless numbers provide conclusive evidence of the important role of ultrasonic vibrations on the melting phenomena.

  • A numerical study is made on the melting process of an unconstrained ice inside an isothermal ice-ball capsule. The unmelted ice core is continuously ascending on account of buoyancy forces. Such a buoyancy-assisted melting is commonly characterized by the existence of a thin liquid film above the ice core. The present study is motivated to present a full-equation-based analysis of the influences of the initial subcooling and the natural convection on the fluid flow associated with the buoyancy-assisted melting. In the light of the solution strategy, the present study is substantially distinguished from the existing works in that the complete set of governing equations in both the melted and unmelted regions are resolved in one domain. Numerical results are obtained by varying the wall temperature and initial temperature. The present results reported the transition of the flow pattern in a spherical capsule, as the wall temperature was increased over the density inversion point. In addition, time wise variation of the shapes for the liquid film and the lower ice surface, the time rate of change in the melt volume fraction and the melting distance at symmetric line is analyzed and is presented.

  • In order to improve the production process of low cost and high purity Vanadium, this study was done to reduce $V_2O_5$ into V-Al master alloy by Aluminothermic Reduction, followed by refining of V-Al master alloy electron beam melting. As melting time was increased in electron beam melting of V, the contents of interstitial impurities and Al, Fe were decreased but the contents of Si, Mo and W were increased due to lower vapor pressure of these elements than that of matrix V. Consequently, it was profitable that melting of V was done for 180 seconds. In addition, with number of melting, the purity of V did not significantly vary, because volatile impurities in V were removed mostly during the first step of melting. As a result of V refining by electron beam melting, high purity Vanadium of 3N(99.91wt%) was acquired including interstitial impurities total contents of which were maximum 400ppm.

Single Crystal Growth of YBa2Cu3Ox by Flux Method (Flux법에 의한 YBa2Cu3Ox 단결정 육성에 관한 연구)

  • Single crystals of YBa2Cu3Ox superconductor were grown by means of the flux method. The effectof starting material, cooling rate, melting time, and melting temperjature were evaluated as influencing paraemters. The larger single crystals of YBa2Cu3Ox were obtained with Y2BaCuOy powder as a starting material than with YBa2Cu3Ox powder. The optimum range of synthetic condition for single crystal growth was as follows ; 2-5$^{\circ}C$/hour of cooling rate, 2-5 hour of melting time and melting temperature at 106$0^{\circ}C$. The obtained size of single crystal was 2mm in average and the largest one was 5mm in maximum.
