콩 발효식품으로부터 분리한 페놀물질획분의 성분분석과 항산화 작용 특성 (Composition and Antioxidative Characteristics of Phenolic Fraction Isolated from Soybean Fermented Food)

  • Doenjang(fermented soybean paste) was prepared by the series of processes including soaking, cooking, first fermentation(3 days at 30$\pm$2$^{\circ}C$) for the preparation of meju(soybean koji) after inoclulation of Aspergillus oryzae, and further fermentation (60 days at 30$\pm$2$^{\circ}C$) for the ripening after addition of salt 13% to meju. the crude phenolics extracted from defatted soybean and doenjang were fractionated onto the neutral phenolics(isofavonoids) and acidic phenolics(phenolic acids), respectively. Composition and antioxidative characteristics of phenolic fractions were determined. The neutral phenolic fractions contained genistin, genistein and daidzein; on the other hand, acidic phenolic fractions had syringic acid and seven other components. The content of genistin in doenjang dramatically decreased at the early stage of fermentation, whereas the content of genistein rather increased. In addition, the content of syringic acid of acidic phenolic fractions were increased during fermentation. These changes in individual phenolic components affected the antioxidative activity of neutral phenolics or acidic phenolics. antioxidative activity of phenolic compounds were evaluated during soybean fermentation. The antioxidative and free radical scavenging activity of neutral phenolic fractions and acidic phenolic fractions on linoleic acid autoxidation were also investigated.

제조방법이 다양한 시판 재래 및 양조 간장의 관능적 특성 (Sensory Characteristics of Different Types of Commercial Soy Sauce)

  • Soy sauce, a basic ingredient in Asian cuisine, is made of fermented soybeans, salt, water, and barley or wheat flour. The sensory characteristics of soy sauce are not only determined through its main ingredients but also by various flavor compounds produced during the fermentation process. This study was conducted to identify the sensory attributes of five different commercial soy sauce samples that differ in Meju types (traditional Meju or modified Meju) and usage types. Thirty three sensory attributes, including appearance (1), odor (16), flavor (14), and mouthfeel (2) attributes, were generated and evaluated by eight trained panelists. The collected data were analyzed by analysis of variance and principal component analysis. There were significant differences in the intensities of all sensory attributes among the soy sauce samples. Soy sauce made with traditional Meju had high intensity of fermented fish, beany, musty odors and salt, Cheonggukjang, fermented fish flavors. Whereas two soy sauces made with modified Meju were strong in alcohol, sweet, Doenjang, roasted soybean flavors. Two soy sauces for soup made with modified Meju had medium levels of briny, sulfury, fermented odors and bitter, chemical flavors and biting mouthfeel characteristic.

메주 유래의 Syncephalastrum racemosum PDA 132-2가 생산하는 Protease의 정제 및 특성 (Purification and Characteristics of Protease Produced by Syncephalastrum racemosum PDA 132-2 from Korean Traditional Meju)

  • Protease related mold was isolated and selected as a starter culture for commercial production of meju. Isolated microorganism was identified as Syncephalastrum racemosum PDA 132 2. To obtain basic data about protease for production of soybean peptides and application of the strain in meju fermentation, we extrated and purified protease and charateristics of the enzyme were investigated. The optimum condition for the production of enzyme was pH 4.0, 30oC, 5 days. The protease was purified 19.7 folds by gel filtration and ion exchange chromatography and specific activity was 12.4unit/mg. The purified enzyme was 34kDa in size, thiol protease(100% inhibited by PCMB), and was acidic protease(stable between pH 2.0~5.0). Vmax of the enzyme was 2.14 g/min which was lower(1/50) than that of by Asp. wentti and B. subtilis.

재래식 메주에서 분리한 유산균들의 각종 효소활성 및 기능성 (Catabolic Enzyme Activities and Physiological Functionalities of Lactic Acid Bacteria Isolated from Korean Traditional Meju)

  • 전통 재래식 메주 3종을 수집하여 호기성 생균수, 유산균수, 효모 및 곰팡이의 수를 측정한 결과 각각 $10^7\sim10^8$ CFU/ g, $10^6\sim10^8$ CFU/g, $10^7\sim10^8$ CFU/g의 분포를 나타내었고, 일반적으로 메주의 발효에 가장 크게 관여하고 있는 효모 및 곰팡이이 외에도 유산균 역시 메주의 발효에 깊이 관여하고 있는 주요 균주인 것으로 나타났다. 재래식 메주로부터 몇 종의 유산균을 분리한 후, 3종의 우수균주를 선별하여 동정을 실시한 결과, Leuconostoc mesenteroides, Lactobacillus plantarum, Lactococcus lactis로 밝혀졌으며, 이들을 각각 Lm-SMm, Lp-SMm, Ll-GAm로 명명하였다. 3종 유산균의 효소 활성을 측정한 결과, 모두 protease, lipase, $\alpha$-amylase 활성을 나타내었고 특히 Ll-GAm이 상대적으로 높은 효소 활성을 나타내었다. DPPH를 이용하여 항산화 효과를 측정한 결과, Lm-SMm, Lp-SMm, Ll-GAm은 각각 45%, 48%, 60%의 free radical 소거 효과를 나타내었고, MTT assay를 이용하여 HT-29 인체 결장암 세포 성장억제 효과를 측정한 결과 각각 45%, 67%, 70%의 성장억제 효과를 나타내었다. 따라서 전통메주로부터 분리한 우수한 유산균은 장류제조 산업에 있어서 적용성이 있을 것으로 생각되어지며, 메주 제조에 있어서 스타터로 사용되었을 때 장류제품의 품질을 향상시킬 수 있을 것으로 기대된다.

  • 재리식 메주, 고오지, 나토 메주, 고오지와 나토의 혼용 메주로 담금한 된장의 일반성분 유리당 및 지방산 조성은 다음과 같다. 조단백질은 10.3~14.6%로 재래식 메주 사용의 된장과 나토된장이 높았고 고오지 된장이 낮았다. 수분은 46.9~54.5% 범위로 숙성과정 중 다소 증가하였고 나토된장이 많았다. 에틸알코올은 숙성 75일 까지는 고오지와 나토의 혼용 된장에서 높았으나 숙성 90일에 고오지 된장이 2.8%로 최대치를 보였다.총당과 환원당은 숙성기간의 경과에 따라 감소하였으며 이들 당 함량은 고오지 된장, 고오지와 나토의 혼용된장, 재래식 메주사용의 된장, 나토된장의 순으로 높았다. 된장의 유리당으로 glucose, galactose, mannose, arabinose가 동정되었으며 이중 glucose, galactose가 타유리당 보다 많았다. 숙성 90일 후에는 glucose나 총 유리당 함량은 재래식 메주로 담근 된장에서 높았고, galactose는 고오지 된장에서 높았다. 된장의지방산으로 palmitic, stearic, oleic, linoleic, linolenic, arachidonic acid가 분리되었고, 이 중 oleic acid가 38.5~46.9%로 시험된장 모두에서 많은 비율로 나타났다. 그러나 숙성기간이나 시험구에 따른 지방산 조성 비율은 뚜렸한 차이가 없었다.

고오지 종류에 따른 식혜 고추장의 숙성 중 이화학적 특성 변화 (Changes in Physicochemical Characteristics of Kochujang Prepared with Different Koji during Fermentation)

  • 전통 고추장의 품질개선과 균일화를 위하여 메주 종류를 달리하여 고추장을 제조하고 발효시키면서 이화학적 성분의 변화를 관찰하였다. 고추장의 품질지표인 아미노산성 질소는 P-2 고오지 단독 첨가구(KP2), P-1 고오지 단독 첨가구(KP1), 전통메주와 P-2 고오지 혼합 첨가구(MKP2)의 함량이 높아 고오지 첨가로 고추장 품질 개선의 가능성이 제시되었다. 고추장 발효 중 산도와 pH는 메주 종류에 따른 뚜렷한 차이는 없었다. 고추장의 신맛의 척도가 되는 유기산은 모든 시료에서 citric acid의 함량이 가장 높았고 그 다음으로 malic acid, oxalic acid, succinic acid, acetic acid, lactic acid 순이었다. 총 유기산 함량은 담금 초 보다 후기에 높았고 메주 종류에 따른 유의적 차이는 없었으나 발효 100일 후 P-2 고오지 첨가구의 함량이 높았다. 고추장 발효 중 검출된 유리당은 maltose, fructose, glucose로 P-1 고오지를 일부 또는 전체 첨가한 시료(MKP1, KP1)에서 그 함량이 높아 고추장의 단맛을 증가 시키는데 고오지 첨가가 유리할 것으로 확인되었다. 고추장의 주요 유리아미노산은 aspartic acid, glutamic acid, alanine, phenylalanine, argine 등이었고 총 유리아미노산 함량은 아미노산성 질소의 함량변화와 유사한 경향으로 고오지 함량이 많을수록 높았다. 숙성이 완료된 고추장의 관능평가 결과 색택, 냄새, 맛, 전체적 기호도에서 처리구간의 유의적 차이는 없었으나 전통메주와 P-1 고오지 혼합첨가구(MKP1)와 전통 메주와 P-2 고오지 혼합첨가구(MKP2)에서 종합적 기호도가 높았다.

Development of Kanjang (Traditional Korean Soy Sauce) Supplemented with Glasswort (Salicornia herbacea L.)

  • Five types of meju were prepared from 100% defatted soybean (DFSG0), a mixture of 90% DFS and 10% glasswort (DFSG1), a mixture of 80% DFS and 20% glasswort (DFSG2), a mixture of 70% DFS and 30% glasswort (DFSG3), and a mixture of 60% DFS and 40% glasswort (DFSG4). Five types of kanjang were separately prepared from the 5 types of meju by ripening in brine for 6 months. The contents of certain minerals (Mg, Ca, Fe, Mn, and Zn), organic acids (citric acid, malic acid) and the antioxidative effects in the kanjang were increased in proportion to the glasswort content in the meju. However, the free amino acid contents in the kanjang were reduced in proportion to the glasswort content in the meju. DFSG1- and DFSG2-kanjang did not show distinct differences from DFSG0-kanjang based on aroma, flavor, and taste that were compared simply by panel tests. The bacterial and fungal community in the fermented meju and kanjang was not affected by the addition of glasswort to the meju-making process. Bacteria belonging to the Lactobacillus and Bacillus genera and the Lactobacillus family predominated, and yeasts belonging to the Saccharomyces genus and fungi belonging to the Aspergillus genus predominated in the fermented meju and kanjang. In conclusion, the glasswort was a supplement that nutritionally improved the kanjang (except for free amino acid contents) but didn't influence the growth of microorganisms that are responsible for the fermentation of meju and kanjang.

청국장 제조(製造)에 관(關)한 연구(硏究) (Studies on the Manufacturing of Chungkukjang)

  • 장메주의 Protease activity는 54시간 발효시(醱酵時)에 가장 높았고 (Fig. 1) 유리아미노태질소(態窒素)는 $36{\sim}42$시간에서 가장 많이 유리되었으며 총산(總酸)은 24시간에서 제일 많았다. 수분(水分)이 많은 청국장은 적은 것에 비(比)하여 아미노태질소(態窒素)의 유리양이 많았고 그후에는 감소(減少)되었으나 5일까지는 거의 같게 증가되어 peak를 이루었다. 청국장의 총산(總酸)은 20일까지 계속 증가했고 그후에는 서서히 증감(增減)했으며 수분(水分)이 많은 것이 작은 것보다 총산(總酸)이 현저히 많았다.(Table 2) 숙성도(熟成度)와 총산량(總酸量)으로 보아 청국장의 수분함량(水分含量)은 55%가 좋았다.

메주에서 분리된 Scopulariopsis 속의 분리균 (The fungal isolates of Scopulariopsis collected from Korean home-made Mejus)

  • From five years' previous work, the fungal isolates of Scopulariopsis were reported to be important flora at the late stage of meju fermentation. Mainly, the white or pale brown powders of spore mass of these fungi were observed on the surfaces of rectangular mejus, and to be an important sign for well-done Korean traditional home - made mejus. Out of the five isolates previously collected and stored, two kinds of Scopulariopsis isolates were identified as S. brevicauli and S. fusca. The microscopic differences between two were found to be branching patterns of annellophore and ornamentations of spore wall (warty and smooth). However, the intermediate form between two ornamentations of spore wall were also observed in our isolates. This observation was consistent with other result made from the protein electrophoresis. The isolates of Scopulariopsis were considered to be similar or superior to those of Aspergillus species, as compared with production of protease and amylase related enzymes. Thus, these isolates were speculated to be important fungi in Korean traditional home - made meju fermentation and also in production of protease and amylase.

Aspergillus cumulatus sp. nov., from Rice Straw and Air for Meju Fermentation

  • A new species named Aspergillus cumulatus sp. nov. is described in Aspergillus section Aspergillus (Eurotium state). The type strain (KACC $47316^T$) of this species was isolated from rice straw used in meju fermentations in Korea, and other strains were isolated from the air in a meju fermentation room. The species is characterized by growth at a wide range of water activities and the formation of aerial hyphae on malt extract 60% sucrose agar (ME60S) that resemble a cumulus cloud. Furthermore, A. cumulatus produces yellow ascomata containing small lenticular ascospores (5.1-5.7 ${\mu}m$) with a wide furrow, low equatorial crests, and tuberculate convex surface. The species is phylogenetically distinct from the other reported Aspergillus section Aspergillus species based on multilocus sequence typing using rDNA-ITS, ${\beta}$-tubulin, calmodulin, and RNA polymerase II genes.
