Optimization of Culture Conditions for Erythritol Production by Torula sp.

  • The medium for erythritol production by Torula sp. in a 500-ml baffled flask was optimized to be 300 g/I sucrose, 10 g/I yeast extract, 3 g/I $KH_2PO_4$, and 10 mg/I $CuSO_4{\cdot}5H_2O{\;}at{\;}34^{\circ}C$ with initial pH of 5.5. Using this optimal medium, erythritol of 166 g/I was obtained after 140 h of cultivation, corresponding to 55.3% of the erythritol yield from sucrose with a productivity of 1.11 g/I/h. Optimal concentrations of carbbon and nitrogen sources in a fermentor were higher than that in a flask due to the higher oxygen supply of the fermentor. Employing the medium containing 300 g/I or 400 g/I sucrose for the determination of optimal C/N ratio, the C/N ratio was found to be more important than the nitrogen concentration for effective erythritol production, The optimal ratio of yeast extract to sucrose (g/g) was 20. The yield and productivity of erythritol were maximal in the medium containing 400 g/I sucrose and 20 g/I yeast extract. when dissolved oxygen in the culture was increased, the cell mass increased but the erythritol production was manimal in the range of 5 to 10% of dissolved oxygen. Under the optimal the rane of 5 to 10% of dissolved oxygen. Under the optimal culture condition of the fermentor, a final erythritol concentration of 200 gI was obtained after 120 h with a yield of 50% and the productivity was 1.67 g/I/h. Contamination of Sediments and Histological Alterations in Barfin Plaice Pleuronectes pinnifasciatus from Amursky Bay(Peter the Great Bay, East Sea/Sea of Japan)

  • In August-September 2001, 15 samples of bottom sediments were collected in the inner, middle and open parts of Amursky Bay near Vladivostok, Russia, and barfin plaice Pleuronectes pinnifasciatus was sampled from the inner and the middle locations of the bay. In the sediments from all three sites elevated concentrations of several heavy metals, i.e. Zn ($102-115{\mu}/g$ dry weight), Ni $(70-73{\mu}g/g)$ and Cu $(27-35{\mu}g/g)$ were discovered. The contents of oil hydrocarbons were very close to or slightly higher than the maximal normal environmental background level, $100{\mu}g/g$ dry weight. The sediments contained negligible amounts of hexachlorocyclohexane, while DDT concentrations were quite high (1.7-16.3ng/g dry weight). Generally, there were no substantial differences in the pollution levels of the locations studied and our results resembled those reported for Amursky Bay in the 1990s. Surprisingly, in 2001 'fiesh' DDT comprised 70-85% of the total DDT content in sediment from all the locations studied. In fish liver total DDTs concentrations were 212.8 and 122.54 ng/g wet weight for the inner and the middle locations, respectively, and 'fresh' DDT comprised 35 and 64% of DDTs, respectively. These results provide evidence of recent input of DDT from an unknown source into the ecosystem of Amursky Bay. Histopathological changes revealed in the plaice liver (vacuolization of hepatocytes, coagulative necrosis of hepatocytes, inflammatory reaction, and necrosis of epithelial cells of bile ducts) are probably connected with an intensive metabolism of DDT in the fish organism. No histological and histomorphometric differences were found in the state of the interrenal tissue. Similar condition of the liver and the interrenal tissue in barfin plaice sampled from the inner and the middle locations of Amursky Bay may be explained by the absence of great differences in the pollution levels of these sites.

Purification and Characterization of Thermotolerable Alkaline Protease by Alkalophilic Bacillus sp. No. 8-16 (알칼리성 Bacillus sp. No.8-16의 내열ㆍ알칼리성 단백질 분해효소의 정제와 특성)

  • Thermostable alkaline protease of alkalophilic Bacillus sp. No. 8-16 has been purified, and the properties of the enzyme investigated. The characteristic point of the organism used is especially good growth in alkaline and thermal condition. The alkaline protease of the strain No. 8-16 was purified from crude enzyme by acetone precipitation, CM-cellulose ion exchange chromatography, Sephadex G-100 and Sephadex G-75 gel filtration. Through the series of chromatograpies, the enzyme was purified to homogeneity with specific activity of 37 fold higher than that of the crude broth. Characteristics of the purified enzyme were as follow; $K_m$ value for the enzyme was 1.3 mg/ml, the alkaline protease showed a maximal activity at 7$0^{\circ}C$ and from the pH 6.0 through 12.0, and stable for 1 hr. at 6$0^{\circ}C$. The moleclar weight of the enzyme was estimated to be 33,000 by Sephadex G-100 gel filtration. The activity of the alkaline protease was inhibited by iodoacetic acid and Ag$^+$, Hg$^+$, PMSF (phenylmethylsulfonyl fluoride), and activated by $Ca^{2+}$ and Mn$^{2+}$.

  • Oral fluid has become increasingly popular as an alternative specimen in the field of driving under the influence of drugs (DUID) and work place drug testing. In this study, an analytical method for the detection and quantification of ${\Delta}^9$-tetrahydrocannabinol (THC) and its metabolite, 11-nor-9-carboxy-${\Delta}^9$-tetrahydrocannabinol (THC-COOH) in oral fluid by SPE and GC-MS was established and fully validated. The stability of THC and THC-COOH in oral fluid during storage was also determined by examining the THC and THC-COOH concentration changes depending on time and container materials. Oral fluid samples were kept over 21 days at room temperature, $-4^{\circ}C$ and $-20^{\circ}C$ in two different specimen collection tubes; glass and polypropylene tubes. Three replicates for each condition with different temperature and types of a container were analyzed at five different time points over 21 days. When oral fluid samples were stored in glass tubes, the loss of both THC and THC-COOH was less than 10% at all room temperature, $-4^{\circ}C$ and $-20^{\circ}C$. However, in polypropylene tubes, the loss of both THC and THC-COOH increased significantly over the study period. In particular, the concentration of THC decreased more rapidly than that of THC-COOH at room temperature and the maximal percentage of THC lost was 90.3% after 21 days. The result indicates that it would be necessary to collect oral fluid samples in glass containers and cool the samples until analysis in order to prevent the degradation of analytes.

The Effect of the Patellofemoral Pain Syndrome on EMG Activity During Step up Exercise (스텝업 운동이 무릎넙다리 통증증후군을 가진 축구선수의 근활성도에 미치는 영향)

  • The purpose of this study was to examine EMG activities and VMO/VL ratio of the vastus medialis oblique, and vastus lateralis during step up exercise according to ankle and knee positions in soccer players with patellofemoral pain syndrome. Methods: Subject(patellofemoral pain syndrome, PFPS: n=8 and without PFPS, non PFPS; NPFPS: n=8) perfomed step up exercise at each knee and ankle position(knee flexion $30^{\circ}$, $60^{\circ}$, and $90^{\circ}$, ankle internal rotation $30^{\circ}$, neutral, and external rotation $30^{\circ}$) while EMG activity was collected. The EMG signals were expressed by the % maximal voluntary isometric Contraction(%MVIC) values. Statistical analysis consisted of two way repeated measures analysis of variance with post hoc analysis. Results: Main results were as follows: 1) EMG of VMO, and VL was tend to be lower in PFPS compared to NPFPS. 2) EMG of VMO and VL with knee flexrion $60^{\circ}$ was significantly higher the results with knee flexion $30^{\circ}$, and $90^{\circ}$. VMO and VL with ankle external rotation $30^{\circ}$ was significantly higher the results with internal rotation $30^{\circ}$ and neutral position. Conclusion: Considering the EMG activity was reduced due to the to the PFPS and that performing step up with knee flexion $60^{\circ}$ with ankle external rotation $30^{\circ}$ position may provide the most effective condition for patients with patellofemoral pain syndrome.

The Effect of Head Posture Change on Initial Occlusal Contact in Temporomandibular Disorder Patient (측두하악 장애환자에서 두부자세 변화가 초기 교합접촉에 미치는 영향)

  • The purpose of this study was to evaluate an effect of change on head posture initial occlusal contacts with measuring the distances between initial occlusal contacts and maximum intercuspal position at different head posture in TMDs patient. For this study, 24 patients from age 13 to 36 were selected, they were examined health history taken, patients who have sign and symptoms of TMDs were examine before the study. For the normal group, 21 adults from age 23 to 25 were selected. They have normal or class I molar relationship, and have no other prosthetic restorations. Difference on distance between initial occlusal contact and maximum intercuspal position with mandibular kinesiograph$(MKG^R)$(K6 diagnostic system, Myo-tronic Inc, USA) in upright, supine, 45$^{\circ}$ extension, 30$^{\circ}$ flexion position of the head were measured. The Frankfort horizontal plane was used as a reference plane. The results were as follows : 1. There were significant differences between initial occlusal contacts of the normal and patient group on upright position and 30$^{\circ}$ flexion of the head(p<0.05, p<0.01) 2. The position of the initial occlusal contacts have a tendency to place anterior and inferior to maximal intercuspal position in upright position and 30( flexion of the head as well as posterior and inferior in supine position and 45$^{\circ}$ extension of the head in the normal and patient groups. 3. There were significant differences among the initial occlusal contacts between uptight and supine position; upright and 45$^{\circ}$ extension of the head(p<0.05); supine position and 30$^{\circ}$ flexion of the head, .and 30(flexion and 45$^{\circ}$ extension of the head in the patient group(p<0.01) The result have shown that after treatment on the supine position, it may be necessary to check occlusal contact on the upright position as well ass flexion of the head. It may need careful adjustment in occlusal condition on upright position of TMDs patient.

Effect of different lateral occlusion schemes on peri-implant strain: A laboratory study

  • PURPOSE. This study aims to investigate the effects of four different lateral occlusion schemes and different excursions on peri-implant strains of a maxillary canine implant. MATERIALS AND METHODS. Four metal crowns with different occlusion schemes were attached to an implant in the maxillary canine region of a resin model. The included schemes were canine-guided (CG) occlusion, group function (GF) occlusion, long centric (LC) occlusion, and implant-protected (IP) occlusion. Each crown was loaded in three sites that correspond to maximal intercuspation (MI), 1 mm excursion, and 2 mm excursion. A load of 140 N was applied on each site and was repeated 10 times. The peri-implant strain was recorded by a rosette strain gauge that was attached on the resin model buccal to the implant. For each loading condition, the maximum shear strain value was calculated. RESULTS. The different schemes and excursive positions had impact on the peri-implant strains. At MI and 1 mm positions, the GF had the least strains, followed by IP, CG, and LC. At 2 mm, the least strains were associated with GF, followed by CG, LC, and IP. However, regardless of the occlusion scheme, as the excursion increases, a linear increase of peri-implant strains was detected. CONCLUSION. The peri-implant strain is susceptible to occlusal factors. The eccentric location appears to be more influential on peri-implant strains than the occlusion scheme. Therefore, adopting an occlusion scheme that can reduce the occurrence of occlusal contacts laterally may be beneficial in reducing peri-implant strains.

Anthraquinone Productivity by the Cultures of Adventitious Roots and Hairy Roots from Curled Dock (Rumex crispus) (소리쟁이 (Rumex crispus)의 부정근과 모상근 배양에 의한 Anthraquinone 생산성)

  • In order to survey anthraquinone productivity from in vitro root culture, transformed hairy roots of Rumex crispus were induced from leaf segments by infection with Agrobacterium rhizogenes strain $A_4$ and compared with adventitious roots. The optimum condition of adventitious root formation from leaf segments was 5 $\mu$M NAA added to MS medium. Mannopine was detected in the extract of hairy roots by paper electrophoresis, but not in adventitious roots. Secondary root tips of both adventitious roots and hairy roots elongated without lateral root branching in hormone free MS medium, but primary root tips showed more rapid growth with extensive lateral root branching. MS basal medium was the best for growth of the adventious roots and hairy roots for anthraquinone content. Adventitious root tips and hairy root tips cultured in liquid MS medium supplemented with 0.05 $\mu$M NAA and 0.1 $\mu$M kinetin (contained 5% sucrose) showed the maximal growth and anthraquinone content. Anthraquinone content of hairy roots was increased by the culture periods, but was reduced after 25 days of culture.

  • The development of direct receiver techniques is expected to be a solution for future wideband or multi-band wireless systems based on software defined radio. In this Paper, we study the regeneration of I and Q signals for the SDR based direct conversion receiver, so that we can handle a wide bandwidth and maintain maximal flexibility in system utilization. After modeling the basic system considering the real wireless communication environment, and studying the impact of imperfect phase imbalance on the performance of a direct conversion receiver, we propose a suboptimal I and Q signal regeneration algorithm for the system. The proposed algerian regenerates I and Q signals using a real time early-late compensator which effectively estimates phase imbalances and gives feedback in a directreceiver. The proposed algorithm is shown to mitigate the impact of AWGN and improves performance especially at low SNR channel condition. According to the computer simulation, the BER performance of the proposed system is at least about 4 dB better than conventional systems under $45{\~}55$ degrees random phase errors.

Effect of Non-elastic Taping on Ankle Dorsiflexion and Activity of the Triceps Surae Muscles While Vertical Jumping (수직점프 시 비탄력 테이핑이 발목 발등굽힘과 하퇴삼두근의 근활성도에 미치는 영향)

  • PURPOSE: There are several standard interventions for managing Achilles tendinitis, including eccentric exercise and calf muscle stretches, orthoses, electrotherapy, and taping. However, no study has determined the effect of non-elastic taping on deloading the Achilles tendon while vertical jumping. Therefore, this study determined the effect of non-elastic taping on ankle dorsiflexion and the triceps surae muscle activity while vertical jumping in healthy subjects. METHODS: The study recruited 17 participants. A motion analysis system was used to measure the angle of ankle dorsiflexion and wireless surface electromyography was used to measure the soleus and gastrocnemius activities while vertical jumping. Non-elastic taping was applied on randomized leg side. All subjects performed maximal effort vertical jumps without and with non-elastic taping, with three trials for each condition. The mean peak dorsiflexion and muscle activities during the three trials were calculated and paired t-tests were used to compare the mean values without and with non-elastic taping. Significance was defined as (p<.05). RESULTS: The maximum angle of ankle dorsiflexion and activity of the gastrocnemius muscle decreased significantly when non-elastic tape was applied (p<.05), while there was no significant difference in the soleus activity between no-taping and taping (p>.05). CONCLUSION: We introduce non-elastic taping as a method to decrease maximum ankle dorsiflexion and gastrocnemius activity while vertical jumping.
