Effect of a dual trigger on oocyte maturation in young women with decreased ovarian reserve for the purpose of elective oocyte cryopreservation

  • Objective: The aim of this study was to determine whether co-administration of a gonadotropin-releasing hormone (GnRH) agonist and human chorionic gonadotropin (hCG) for final oocyte maturation improved mature oocyte cryopreservation outcomes in young women with decreased ovarian reserve (DOR) compared with hCG alone. Methods: Between January 2016 and August 2019, controlled ovarian stimulation (COS) cycles in women (aged ≤35 years, anti-Müllerian hormone [AMH] <1.2 ng/mL) who underwent elective oocyte cryopreservation for fertility preservation were retrospectively analyzed. Results: A total of 76 COS cycles were triggered with a GnRH agonist and hCG (the dual group) or hCG alone (the hCG group). The mean age and serum AMH levels were comparable between the two groups. The duration of stimulation, total dose of follicle-stimulating hormone used, and total number of oocytes retrieved were similar. However, the number of mature oocytes retrieved and the oocyte maturation rate were significantly higher in the dual group than in the hCG group (p=0.010 and p<0.001). After controlling for confounders, the dual-trigger method remained a significant factor related to the number of mature oocytes retrieved (p=0.016). Conclusion: We showed improved mature oocyte collection and maturation rate with the dual triggering of oocyte maturation in young women with DOR. A dual trigger appears to be more beneficial than hCG alone in terms of mature oocyte cryopreservation for young women with DOR.

  • Lipid composition of human milk samples (ten colostrum and 10 mature) was analyzed by quantitative thin-layer chromatography with flame ionization detector. Six kinds of lipid components existed in human milk. Among them, triglyceride which was most abundant lipid component showed significantly lower levels in colostrum than in mature milk. Other lipid components have no significant differences between colostrum and mature milk.

  • Backgorund: WEHI-231 B cell line is a representative model for $IgM^+$ mature B cells. To understand the signaling differences between mature and immature B cells, we compared the responsiveness of WEHI-231 and Bal 17 B cell lines to BCR cross-linking. Methods: The extents of tyrosine phosphorylation, ligand-induced internalization, and activation-induced cell death upon BCR cross-linking were compared in two cell lines. Results: Despite a higher expression of BCR, cross-linking of BCR on WEHI-231 cell evoked a weaker level of tyrosine phosphorylation and BCR endocytosis than Bal 17 cells. Furthermore, the endocytosed BCR could not enter the lysosomal compartment and stayed as peripheral spots in WEHI-231 cells. Conclusion: WEHI-231 cell showed preferred BCR-mediated signaling pathways leading to a reduced capability of antigen presentation as well as the enhanced apoptosis in comparision with Bal 17 cells. These results might reflect the signaling differences between mature and immature B cells.

Rapid Propagation of Pelagonium Inquinans Via Organogenesis from Mature Leaf Explants

  • A method for plant regeneration via organogenesis from Pelagonium inquinans leaf disc has been developed. Mature leaf explants were collected from field-grown plants and used for the induction of adventitious shoot regeneration on Murashige and Skoog (MS) medium supplemented with 3% (w/v) sucrose plus plant growth regulators. Maximum shoot organogenesis, with $11.8{\pm}1.5$ shoots (98.6%) per leaf disc, was obtained with $2\;mg/l$ $N^6-benzyladenine$ (BA) and $0.5\;mg/l$ ${\alpha}-naphthyleneacetic$ acid (NAA) in 30 days. For rooting, the in vitro proliferated and elongated shoots were excised into 1.5-2 cm in length microcutting, which were plated individually on an half-strength MS (1/2MS) medium supplemented with 2% (w/v) sucrose plus various concentrations of indole-3-butyric acid (IBA). Shoots rooted with a frequency of 100% following culture on 1/2MS medium containing $0.5\;mg/l$ IBA.

  • Intracranial teratomas are diagnosed mostly in young population and usually involve midline structure. We report a case of mature teratoma in an adult patient with unusual location in cerebellar hemisphere. A 47-year-old woman presented with severe headache and nausea. Computed tomography and magnetic resonance imaging demonstrated a posterior fossa lesion with cerebellar hemispheric location not involving midline. Histological examination of surgical specimen showed fully matured representative tissues of the three germ layers confirming teratoma. This is a rare example of mature teratoma with unusual age of the patient and location.

The relationships between body-cathexis and clothing image preferences in male college students (남자대학생의 신체만족도와 의복이미지 선호의 관계연구)

  • The purpose of this study was to investigate the relationships between body-calthexis and clothing image preferences in male college students. The sample included 28 male college students and an instrument was developed based on the previous studies. The statistical analyses used for this study were factor analysis cluster analysis and t-test. The result of factor analysis showed that body-cathexis consisted of four areas of body parts: face/head upper body middle body and lower/total body. Clothing image preferences consisted of 'strong vs weak' 'soft vs hard' 'young vs mature' 'mannis vs feminine' and 'distinguished vs undistinguished' images. Cluster analysis revealed that male college students are segmented into two groups. and the two groups differed in regard to clothing image preferences such as 'strong vs weak' 'young vs mature' and 'mannish vs feminine' images. In addition the two consumer segments were different concerning body-cathexis for middle body and all body areas combined. The consumers who preferred feminine weak and mature clothing images were more satisfied with their middle area of the bodies and all body areas combined.

  • A sequential sub-cellular study of the epicarp of Nerium indicum has been performed. Outer epidermis of the ovary wall is covered externally with a thin cuticle. Cytoplasm possesses most of the cell organelles in the ovary stage itself. Outermost zone of the pericarp is the epicarp, developing from the outer epidermis. In the developing fruit, cell organelles are found with its maximum intensity. In mature fruit, the epicarp becomes multilayered due to additional development of few collenchymatous cells close to the outermost layer. Epicarpic cell possesses large central vacuole, around which a thin layer of cytoplasm is present. Number of cell organelles are considerably reduced in the mature fruit. In the ovary stage starch grains are electron transparent, while in the mature fruit it is fruit it is electron transluscent.

  • The metabolic enzymes, nitrate reductase, amylase and peroxidase and the Photorespiratory enzyme, Slycolate oxidise in Nicotiana tabacum varieties were studied at various growth stages. The enzyme activities of young leaves with rapid growth were different from those of old ones with stationary growth. In young leaves, activity of nitrate reductase was higher than that in mature ones and amylase activity was fairly constant in all stages. Activities of glycolate oxidise and peroxidase were found to be significantly lower in young leaves than in mature ones. Activity of glycolate oxidase in mature middle leaves was 2.45 times higher than that of young ones and inhibited by 36% when the enzyme was treated with 0.16 mM isonicotinic hydrazide.

  • The aim of this experiment was to evaluate the levels of hormones (progesterone, IGF-I and IGFBP-3) in blood plasma, growth, carcass traits and their interactions of sexually immature (n = 18) and sexually mature (n = 17) gilts. To calculate average daily weight gain (ADG), gilts were individually weighed at the beginning of the trial and at slaughter (110${\pm}$10 days old). Blood concentrations of progesterone, IGF-I and IGFBP-3 were determined by RIA. The right hot carcass sides were dissected and the individual basic parts from carcasses were weighed to record the carcass traits. IGFBP-3, ADG and carcass traits were not affected by pubertal maturation. Compared to sexually immature gilts, mature gilts had higher blood concentrations of progesterone and IGF-I. High correlations were noted between levels of some hormonal substances, productive performance and carcass traits of sexually immature and mature gilts.

  • Detachment rate of mature fruits of the Lycium chinense Mill with various frequency, amplitude, and shaking time using an experimental vibratory system was analyzed. The experimental vibratory system was designed such that the frequency, amplitude, and shaking time could be controlled easily. The detachment rate increased as the frequency increased at constant amplitude and increased as the amplitude increased at constant frequency. The contents of the detached immature fruit, however, also increased as the frequency and amplitude increased, which was undesirable for the detachment efficiency considering the only mature 900 rpm and amplitude of 35 m for the shaking time of three seconds.
