치수는 상아질로 둘러싸인 간엽조직으로 다양한 세포와 기저 물질들로 구성되어 있으며 혈관과 신경조직이 분포되어 있다. 치수의 염증은 조직의 분해를 야기하며 이는 Matrix Metalloproteinase에 의해 세포 외 기질의 분해가 촉진되어 병적인 과정을 거치게 된다. 이에 Lipopolysaccharide에 의한 MMP와 inflammatory cytokine의 유도와 peroxisome proliferator-activated receptors (PPAR)에 의한 염증매개 물질의 조절에 대해 알아보고자 하였다. 사람의 치수세포를 다양한 LPS농도에 노출시킨 후 24시간째 MMP-2, MMP-9의 변화를 보고 LPS에 의해 자극된 치수세포에서 ICAM-1, VCAM-1, $IL-1{\beta},\;TNF-{\alpha}$의 분비가 증가됨을 알 수 있었다. 또한 Adenovirus $PPAR{\gamma}\;(Ad/PPAR{\gamma})$와 $PPAR{\gamma}$ agonist인 rosiglitazone를 LPS로 자극된 치수세포에 처리하였을 때 48시간째 MMPs와 Adhesion molecules, cytokines의 감소를 확인하였다. 이로써 사람의 치수세포에서 $PPAR{\gamma}$가 가지는 항 염증효과에 대해 지속적 인 연구가 필요할 것으로 사료된다.

Matrix metalloproteinase-2 (MMP-2; 72-kDa gelatinase; 72-kDa type IV collagenase; gelatinase A) plays an important role in normal physiological processes and in many pathologic processes such as arthritis and metastasis of cancer. Tissue inhibitor of metalloproteinases-2 (TIMP-2) binds to proMMP-2 or mature MMP-2 at a 1:1 ratio and inhibits the catalytic activity of MMP-2. We demonstrated that the baculovirus/insect cell system does not have TIMP-2 activity. The human proMMP-2 free of TIMP-2 was expressed in the expression system and purified by one-step affinity chromatography using gelatin-Sepharose. The free proMMP-2 was autoactivated to the mature MMP-2 and autodegraded into smaller molecular weight forms in the absence of external activator. The activation and autodegradation of the proMMP-2 was much more rapid in the presence of 4-aminophenylmercuric acetate (APMA). Addition of TIMP-2 inhibits both APMA-induced activation and autodegradation of the free proMMP-2. However, an increasing concentration of TIMP-2 more readily inhibited activation of the free proMMP-2 than autodegradation. These results demonstrate that TIMP-2 plays roles in inhibition of both activation and autodegradation of the free proMMP-2 in addition to inhibition of the catalytic activity of MMP-2.
연구배경: 본 연구에서는 황색포도알균이 숙주세포 내로 침입하여 증식하는 정도를 관찰하고자 한다. 이때 세포 외 바탕 단백질의 변화가 수반될 것으로 가설을 설정하고, 이러한 변화에 영향을 미칠 것으로 생각되는 matrix metalloproteinase (MMP)의 발현과 역할에 대해 연구하고자 하였다. 방법: 황색포도알균은 $10^6{\sim}10^7CFU/ml$을 $10^5$개의 기관지상피세포인 BEAS-2B 세포에 2시간 동안 침입시킨다. 이후 세척으로 세포 밖에 있는 황색포도알균을 제거한 후, BEAS-2B 세포를 다양한 시간 동안(4, 6, 8, 12 시간) 배양한 후 황색포도알균의 집락수(CFU/ml)를 측정하였고, 단백질을 분리하여 세포 외 바탕단백질의 발현 정도와 MMP의 활성도를 측정하였다. 또한 MMP 억제제인 GM6001을 전처치한 후 황색포도알균을 세포에 침입시킨 후 세포 내에서의 균의 집락수 및 세포 외 바탕단백질의 변화를 관찰하였다. 결과: 황색포도알균의 집락을 측정한 결과 4시간과 12시간을 비교해 볼 때 MOI가 증가할수록, 감염시킨 시간이 길수록 숙주세포 내로 침입이 유의하게 증가하였다. BEAS-2B 세포에서 황색포도알균을 침입시킨 시간이 길수록, MOI가 증가할수록 MMP 2 및 MMP 9의 활성도와 dysadherin의 발현은 증가하였고, 이와는 대조적으로 E-cadherin의 발현은 감소하였다. MMP억제제인 GM6001을 전 처치 한 결과 황색포도알균의 세포 내 침입을 유의하게 감소시켰다. 결론: 황색포도알균이 기관지 상피세포 내로 침입할 때 dysadherin 및 E-cadherin 같은 세포 외 바탕 단백질의 변화를 동반하며, MMP 활성도가 균의 세포 내 침입에 관여하는 것으로 보인다.
The matrix metalloproteinases (MMPs) are a family of enzymes responsible for degrading connective tissue. MMPs catalyze the breakdown of collagen from the extracellular matrix, leading to wrinkle formation and accelerated skin aging. Furthermore, ultraviolet irradiation causes increased expression of certain MMPs. In the extracellular matrix turnover, MMPs are interacting with endogenous regulators named tissue inhibitors of metalloproteinases (TIMPs). Using peptide substrate assays, it has been demonstrated that TIMP-MMP complexes interact highly specifically with $K_{i}$ values of 10$^{-9}$ -10$^{-16}$ M. Therefore applications for TIMP as inhibitor of collagen degradation are suggested for cosmetic anti-aging products to prevent wrinkle formation and loss of elasticity. To date four TIMP proteins (TIMP-1, TIMP-2, TIMP-3 and TIMP-4) have been identified which show a high degree in sequence similarity. The production of human TIMP-2, a 194-residue nonglycosylated protein, was performed by fed-batch culture of Escherichia coli. TIMP-2 accumulated in the bacterial cells in an insoluble form as inclusion bodies. The inclusion bodies were solubilized and the protein refolded to yield the native TIMP-2 in the active form. The integrity of the protein was confirmed by mass analysis, Edman sequencing and gel shift experiments with authentic samples. The inhibitory activity of the refolded and purified TIMP-2 was demonstrated with MMP-1 and MMP-2 assays using synthetic fluorogenic peptide substrates.s.
목 적 : 뇌수막염에서 MMP-9은 뇌혈관 내피하 기질막을 분해하여 혈관-뇌장벽을 파괴하는데 주된 역할을 담당하며 TIMP-1은 MMP-9의 전효소와 복합체를 형성하여 MMP-9의 작용을 선택적으로 억제하는 인자로 알려져 있으나 이들의 상호작용은 아직 밝혀진 바가 별로 없다. 최근 세균성 뇌수막염 환자의 뇌척수액에서 MMP-9과 TIMP-1 측정에 대한 연구가 있었고, 특히 세균성 뇌수막염에서 신경학적 합병증의 예후인자와 관련하여 MMP-9의 역할에 대한 보고들이 있었으나 아직 무균성 뇌수막염 환자를 대상으로 한 보고는 별로 없었고 특히 국내에서 보고된 바는 없었다. 이에 저자들은 무균성 뇌수막염 환아의 혈액과 뇌척수액에서 MMP-9과 TIMP-1의 농도를 측정하여 대조군과 비교하고, MMP-9과 TIMP-1 및 다른 뇌수막염 관련인자들 사이의 상관관계를 비교 분석하고자 하였다. 방 법: 2002년 6월부터 7월까지 강북삼성병원 소아과 내원환자 중 발열과 뇌막자극증상을 보인 40명의 환아들을 대상으로 뇌척수액 검사를 시행하여, 무균성 뇌수막염 소견을 보인 25명을 뇌수막염군, 정상소견을 보인 14명을 대조군으로 하였다. 입원당일 혈액과 뇌척수액을 채취하여 혈액에서 백혈구수를 측정하고, 뇌척수액에서 백혈구수와, 당, 단백농도 및 뇌압을 측정하였다. 나머지 혈액 및 뇌척수액 검체는 실온에서 10분간 2,000g으로 원심분리하여 영하 70도에서 보관후 sandwich ELISA 방법을 이용하여 각각의 검체에서 일시에 MMP-9과 TIMP-1의 농도를 정량하였다. 결 과 : 뇌척수액 MMP-9과 TIMP-1 농도는 뇌수막염군에서 의미있게 증가되었고(P<0.05), 혈청 MMP-9과 TIMP-1 농도는 두 군간에 차이가 없었다. 뇌척수액 MMP-9과 TIMP-1은 서로 의미있는 양의 상관관계를 보였으며($r_s=0.42$, P<0.05), 뇌척수액 MMP-9/TIMP-1 비율은 대조군에 비해 무균성 뇌수막염군에서 의미있게 증가되었다(P<0.05). 뇌척수액 MMP-9과 TIMP-1은 뇌척수액 총백혈구수와 양의 상관관계를 보였다($r_s=0.43$, P<0.05, $r_s=0.48$, P<0.05). 뇌척수액 TIMP-1은 뇌척수액 단백 농도와 양의 상관관계를 보였다($r_s=0.43$, P<0.05). 결 론 : 무균성 뇌수막염 환아의 급성기 뇌척수액에서 MMP-9과 TIMP-1이 의미있는 증가를 보였고, 뇌척수액 TIMP-1은 정상군에서 상대적으로 높은 농도를 보이고 있어 MMP-9과 TIMP-1은 각각 뇌혈관장벽의 파괴와 유지에 관여한다고 생각된다.
Objective : The aim of this present study is to evaluate the inhibitory effect of allergen removed Rhus verniciflua (ARV) on Matrix Metalloproteinase-9 (MMP-9), Matrix Metalloproteinase-2 (MMP-2) which is considered to have a clinically important role in tumor metastasis. Methods : The inhibitory effects of standardized extract of ARV on the MMP-2, MMP-9 were investigated by spectrofluorometer while the inhibitory effects on the active MMP-2, pro MMP-2, pro MMP-9 were investigated by zymography. Antimetastatic effect of standardized extract of ARV was investigated in vitro on human fibrosarcoma cell (HT1080)'s invasion through Matrigel. Results : The standardized extract of ARV showed inhibitory effects on the active MMP-2 (IC50, $1.01{\mu}g$/ml), active MMP-9 (IC50, $2.5{\mu}g$/ml) depending on concentrations which was determined by spectrofluorometer. The standardized extract of ARV showed inhibitory effects on the active MMP-2, pro MMP-2, pro MMP-9 depending on concentrations which was determined by zymography. However its inhibitory effect on pro MMP-9 was relatively weaker rather than active MMP-2, pro MMP-2. The standardized extract of ARV showed inhibitory effects in vitro on human fibrosarcoma cell (HT1080)'s invasion through Matrigel according to concentration. Conclusions : These results indicate that standardized extract of ARV has antimetastatic effect through inhibit again MMP-2, MMP-9. Also its inhibitory effect is more powerful on active MMP-2, pro MMP-2 than on active MMP-9, pro MMP-9. It is necessary to conduct further studies on other MMP families, TIMP, and each component of standardized extract of ARV.
Fibroblast growth factor-2 (FGF-2) is known to modulate numerous cellular functions in various cell types, including cell proliferation, differentiation, survival, adhesion, migration, and motility, and also in processes such as wound healing, angiogenesis, and vasculogenesis. FGF-2 regulates the expression of several molecules thought to mediate critical steps during angiogenesis. This study examines the mechanisms underlying FGF-2-induced cell migration, using human umbilical vein endothelial cells (HUVECs). FGF-2 induced the nondirectional and directional migration of endothelial cells, which are inhibited by MMPs and plasmin inhibitors, and induced the secretion of matrix metalloproteinase-3 (MMP3) and MMP-9, but not MMP-l and MMP-2. FGF-2 also induced the secretion of the tissue inhibitor of metalloproteinase-l (TIMP-I), but not of TIMP- 2. Also, the pan-PKC inhibitor inhibited FGF-2-induced MMP-9 secretion. It is, therefore, suggested that FGF-2 induces the migration of cultured endothelial cells by means of increased MMPs and plasmin secretion. Furthermore, FGF-2 may increase MMP-9 secretion by activating the PKC pathway.
Objective: This study aimed to explore the expression of tissue factor (TF), protease activated receptor-2 (PAR-2), and matrix metalloproteinase-9 (MMP-9) in the MCF-7 breast cancer cell line and influence on invasiveness. Methods: Stable MCF-7 cells transfected with TF cDNA and with TF ShRNA were established. TF, PAR-2, and MMP-9 protein expression was analyzed using indirect immunofluorescence and invasiveness was evaluated using a cell invasion test. Effects of an exogenous PAR-2 agonist were also examined. Results: TF protein expression significantly differed between the TF cDNA and TF ShRNA groups. MMP-9 protein expression was significantly correlated with TF protein expression, but PAR-2 protein expression was unaffected. The PAR-2 agonist significantly enhanced MMP-9 expression and slightly increased TF and PAR-2 expression in the TF ShRNA group, but did not significantly affect protein expression in MCF-7 cells transfected with TF cDNA. TF and MMP-9 expression was positively correlated with the invasiveness of tumor cells. Conclusion: TF, PAR-2, and MMP-9 affect invasiveness of MCF-7 cells. TF may increase MMP-9 expression by activating PAR-2.
