Allozyme Variation and Population Structure of Carex okamotoi (Cyperaceae), a Korean Endemic Species (한국 내 국부적으로 분포하는 지리사초의 알로자임 변이와 집단구조)

  • The genetic diversity and population structures of fourteen Carex okamotoi (Cyperaceae) populations in Korea were determined using genetic variation at 25 allozyme loci. The Carex okamotoi species is native to Korea. It is endemic to three mountains (Mt. Taeback, Mt. Sobak, and Mt. Noreong) where it is found at 700~1,500 m above sea level. The percentage of polymorphic loci was 40.0%. Genetic diversity at the species level and at the population level was low ($H_{ES}$=0.106; $H_{EP}$=0.094), and the extent of the population divergence was relatively low ($G_{ST}$=0.082). Measurement of deviation from random mating ($F_{IS}$) within the 14 populations was 0.238. An indirect estimate of the number of migrants per generation was 2.78 (Nm=2.78). Analysis of fixation indices revealed a substantial heterozygosity deficiency in some populations and at some loci. Mean genetic identity between populations was 0.986.

  • Starch gel electrophoresis was used to estimate genetic diversity and population structure of Pyrola fauriena H. Andr. in Korea. The percentage of polymorphic loci within enzymes was $57.1\%$. The values of genetic diversity at the species level and at the population were higher than average values for herbaceous with similar life history traits (Hes : 0.149; Hep = 0.134, respectively), whereas the extent of the population divergence was relatively low $(G_{ST}=0.082)$. $F_{IS}$, a measure of the deviation from random mating within the 12 populations, was 0.298. An indirect estimate of the number of migrants per, generation (Nm = 2.81) indicates that gene flow is moderate among Korean populations of the species. Analysis of fixation indices revealed a substantial heterozygosity deficiency in some populations and at some loci. This indicates that some populations sampled may have been substructured largely due to rhizotamous spread and decrease of population sizes.

  • Enzyme electrophoresis was used to estimate genetic diversity and population structure of Chimaphila japonica Miq. in Korea. The percent of polymorphic loci within the enzymes was 48.7%. Genetic diversity at the species level and at the population level was high (Hes=0.278 ; Hep=0.222, respectively), whereas the extent of the population divergence was relatively low ( $G_{ST}$ =0.079). $F_{IS}$ , a measure of the deviation from random mating within the 7 populations, was 0.355. An indirect estimate of the number of migrants per generation (Nm=2.61) indicates that gene flow is high among Korean populations of the species. In addition, analysis of fixation indices revealed a substantial heterozygosity deficiency in some populations and at some loci. Factors contributing to the high levels of genetic dive-rsity found in the entire species of C. japonica include wide distribution, long-lived perennials, ability to regenerate due to rhizomatous spread, outcrossing induced by animal vectors, and occasional pollen dispersal by wind.

Genetic Variation of the Beet Armyworm, Spodoptera exigua (Hubner), Populations in Korea Using Polymorphic Allozymes (다형 동위효소를 이용한 국내 파밤나방(Spodoptera exigua (Hubner)) 집단의 유전변이)

  • Genetic variation of the beet armyworm, Spodoptera exigua (Hubner), was analyzed by polymorphic allozymes. Field populations were subdivided by different hosts, geographical locations, and seasons. Estimated average heterozygosity ($0.443\pm$0.013) indicated high genetic variation in all field populations of S. exigua. There were significant inbreeding effects deviated from Hardy-Weinberg equilibrium in each of subpopulations. These significant nonrandom matings were caused by within-subpopulations probably due to sampling errors, but not by mating isolation among subpopulations. Wrights ($F_{ST}$ ) and Neis (D) genetic distances indicated little genetic differentiation among subpopulations, though some southern local subpopulations (Haenam and Sachon) were relatively different of northern subpopulations (Andong and Kunwi). Estimated number of migrants per generation was 5.9 among host subpopulations, 10.6 among geographical subpopulations, and 31.8 among seasonal subpopulations. These genetic analyses suggest that Korean S. exigua subpopulations have little genetic differentiation mostly due to their significant migratory capacity.

Genetic Diversity and Population Structure of Liriope platyphylla (Liliaceae) in Korea (한국내 맥문동의 유전적 다양성과 집단 구조)

  • Genetic diversity and population structure of eleven Liriope platyphylla (Liliaceae) populations in Korea were determined using genetic variation at 20 allozyme loci. The percent of polymorphic loci within the enzymes was 55.9%. Genetic diversity at the species level and at the population level was high(Hes = 0.178; Hep = 0.168, respectively), whereas the extent of the population divergence was relatively low ($G_{ST}$ = 0.064). $F_{IS}$, a measure of the deviation from random mating within the 11 populations, was 0.311. Total genetic diversity values ($H_T$) varied between 0.0 and 0.535, giving an average over all polymorphic loci of 0.323. The interlocus variation in within population genetic diversity ($H_S$) was high (0.305). An indirect estimate of the number of migrants per generation (Nm = 3.66) indicates that gene flow is high among Korean populations of the species. In addition, analysis of fixation indices revealed a substantial heterozygosity deficiency in some populations and at some loci. Mean genetic identity between populations was 0.988. It is highly probable that directional toward genetic uniformity in a relatively the homogenous habitat is thought to be operated among Korean populations of L. platyphylla.

  • For a locus with two alleles (IA and IB), the frequencies of the alleles are represented by $$p=f(I^A)={\frac{2N_{AA}+N_{AB}}{2N} },\;q=f(I^B)={\frac{2N_{BB}+N_{AB}}{2N}}$$ where NAA, NAB and NBB are the numbers of IA IA, IA IB and IB IB respectively and N is the total number of populations. The frequencies of the genotypes expected are calculated by using p2, 2pq and q2. So in this paper, we consider the method of whether some genotypes is in Hardy-Weinburg equilibrium. Also we calculate the probability generating function for the offspring number of genotype produced by a mating of the ith male and jth female under a diploid model of N population with N1 males and N2 females. Finally, we have conditional joint probability generating function of genotype frequencies.

Study on a Suppressor System for Segregation-Distorter Action in Natural Populations of Drosophila melanogaster in Korea (한국산 초파리의 자연집단에 있어서의 SD 요소에 대한 억제요인에 대하여)

  • In order to see if any suppressor system for the SD action was involved in natural populations of D. melanogaster samples from the populations of nine localities in Korea, Choonchun, Yusoo, Namhai, Shinchon(Seoul), Kwangjoo(Kyunggi), Koonsan, Kwangjoo(Chunnam), Jejoo and Pusan were analyzed by using the mating scheme for locating the suppressor on chromosome pairs. And also two kinds of recombinant SD lines R-1, the American line and $R(SD^NH -1)-1$, the Japanese one were used in order to see any difference of the response of the suppressor for differently originated SD. The results of the experiment are given below. (1) The suppressor system was involved in all lines of natural populations from nine localities of Korea. (2) Most of the suppressors were found to be located on the X chromosome and only a few lines from three populations showed to carry the suppressors on the second chromosome and on the third or fourth chromosoem. (3) The response of the suppressor for differently originated SD lines showed no significant difference.

한국산 검정망둑(Tridentiger obscurus)과 민물검정망둑(T.brevispinis)의 분류학적 고찰

  • Morphological and isozyme analyses were used to examine closely related species of Tridentiger obscurus and \ulcorner: brevispinis, a pair of cryptic species which has intricate problem with regard to species boundary, and to clarify the taxonomic position. In this study, it was revealed from their 17 allopatric streams that there are differences of band ornamentation on the lateral side and three genetic markers between two species. However, genetic relationship between them (S = 0.813, D = 0.192) was within the intraspecific level of similarity. However, at the sympatry of two species no hybrid was found except at Bangjuk stream where the electrophoretic hybrids with slight deficiency of heterozygosity were observed. It is shown that at sympatry there is a finer microenvironmental subdivision between two species at all populations including Bangjuk stream. To manifest the extent of assortative mating, four syrnpatric subpopulations at Bangjuk stream were analyzed for departure from Hardy-Weinberg equillibrium by F-statistics. All subpopulations were deviated significantly from the expectation under random mating. The evidences presented here supports the recognition of Tridentiger obscurus and T. brevispinis as semispecies in the late stage of reproductive isolation

Population Structure of the Gibberella fujikuroi Species Complex Associated with Rice and Corn in Korea

  • Several species belonging to the Gibberella fujikuroi species (Gf ) complex are commonly associated with rice and corn, not only causing serious diseases, but also producing fumonisins, a group of mycotoxins harmful to animals and humans. To characterize the population structure of the putative fumonisin-producing Gf complex in Korea, we obtained 276 candidate isolates from rice and corn harvested in 2009 and 2010 by diagnostic polymerase chain reaction with several specific primer sets. Phylogenetic trees were constructed using multilocus sequences (combined RPB2 and EF1A, totaling 1.6 kb) from these isolates. Among the 135 isolates from rice, F. fujikuroi (teleomorph: G. fujikuroi; 59.3%) and F. proliferatum (G. intermedia; 13.3%) were predominant, followed by F. concentricum (5.9%). Additionally, twenty-five (18.5%) rice isolates belonged in a distinct subclade of F. commune, a non-member of the Gf complex. In contrast, F. verticillioides was the most predominant species (38.3%) among the 141 corn isolates, and followed by F. fujikuroi (27.7%), F. proliferatum (14.9%), F. subglutinans (7.1%), and F. concentricum (2.8%). A single mating type (MAT1-1) was found predominantly among the Gf complex isolates examined. Possible distinct subclades were detected within the populations of F. fujikuroi and F. proliferatum; however, this needs further confirmation. This is the first reported population-level characterization of putative fumonisin-producing Gf complex associated with rice and corn in Korea.

Speciation of the Two Species of the Genus Hyla (Anura) in Korea (한국산 청개구리속 2조의 종분화에 관한 연구)

  • A comparison between two species of Hyla japonica and H. suweonesis in Korea was carried out to clarify their morphological, genetic and ecological differences, to estimate presumed divergent time between them, and to elucidate their probable process and mechanism of speciation. The results are as follows; 1.The breeding season of H. japonica was earlier than that of H. suweonensis about one and half months. 2.H.japonica was distributed all over the country, while H. suweonensis was restricted in Kyonggi plains. 3.On the extemal morphometric characters, differences were hardiy seen between two species. But on the discriminant function analysis, there was apparent differences between them. 4.Genic variation based on 24 loci showed that H. japonica populations were about two fold more variable than that of H. suweonensis populations. 5.Genetic relatedness between two species was rather remote (S =0.520, D=0.634) 6.Their presumed divergent time was estimated to be about three and a quarter mfllon years. It is presumed that H. suweonensis was originated from H. japonica during the period of late pleocene and early pleistocene. 7.Premating isolating mechanisrns (breeding season, mating call, and microhabitat segregation) may be operating to prevent gene ifow between them.
