Analytical method study for cephalexin with high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) applicable for residue studies in the whiteleg shrimp Litopenaeus vannamei (흰다리새우(Litopenaeus vannamei)에서 cephalexin의 잔류농도 연구를 위한 LC-MS/MS 분석법 연구)

  • Cephalexin, a semi-synthetic cephalosporin antibiotic, has long been used in fish aquaculture in various countries under legal authorization. The drug is thus widely available for use in other aquatic species except fishes like the crustacean whiteleg shrimp. This study aims to develop a sensitive method for laboratory residue studies to adopt in withdrawal period determinations. Through repeated trials from the existing methods developed for other food animal tissues, it was possible to achieve a sensitive high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method. The results showed that at a concentration of 0.1 mg/kg, the recovery rate was 81.79%, and C.V. value was 8.2%, which meet the recovery rate and C.V. recommended by Codex guideline. After satisfactory validation of analytical procedures, applicability to the shrimp tissue was confirmed in experimentally cephalexin-treated whiteleg shrimp. As a result, most muscle samples were detected below the limit of quantification (0.05 mg/kg) after day 3, and most hepatopancreas samples were detected below the limit of quantification after day 14. In particular, the limit of quantification 0.05 ppm with the presently developed method suggests sufficient sensitive over the current legal maximum residue limit of 0.2 mg/kg set for fishes.

Estimation of the Required Number of Fan Coil Unit for Surplus Solar Energy Recovery of Greenhouse (온실의 잉여 태양에너지 회수용 FCU 소요대수 검토)

  • In this study, previously reported surplus solar energy-related study result and current status of fan coil unit (FCU) for cooling and heating installed in the current sites were briefly examined and then a method to determine the number of FCUs required to recover surplus solar energy was schematically proposed to provide basic data for researchers and technical engineers in this field. The maximum, mean, and minimum outside temperatures during the experiment period were about $28.2^{\circ}C$, $4.4^{\circ}C$, and $-11.5^{\circ}C$, respectively. The horizontal surface solar radiation level outside the greenhouse was in a range of $0.8-20.5MJ{\cdot}m^{-2}$ and mean and total solar radiation were $10.8MJ{\cdot}m^{-2}$ and $1,187.5MJ{\cdot}m^{-2}$. The mean temperature and relative humidity in the greenhouse during the daytime were in a range of 18.8-45.5 and 53.5-77.5%. The total surplus solar energy recovered from the greenhouse during the experiment period was approximately 6,613.4MJ, which could supplement about 6.7% of the total heating energy 98,600.2 MJ. In addition, the number of FCUs installed for heating varies case to case, although similar FCUs are used. Thus, it is necessary to study the installation height, orientation and installation distance as well as the appropriate number of FCUs from the efficient and economical viewpoints. The required numbers of FCUs for surplus solar energy recovery were 8.4-10.9units and 6.1-8.0units based on air mass and circular flow rate that passed through the FCUs. Considering calculation methods and the risks such as efficiency and use environments of FCUs, it was found that about nine units (one unit per $24m^3$ approximately) needed to be installed. The required number of FCUs for surplus solar energy recovery was around one unit per $24m^3$ approximately.

Nutrient Recovery from Sludge Fermentation Effluent in Upflow Phosphate Crystallization Process (상향류 인 결정화공정을 이용한 슬러지 발효 유출수로 부터의 영양소 회수)

  • The nutrient recovery in phosphate crystallization process was investigated by using laboratory scale uptlow reactors, adopting sequencing batch type configuration. The industrial waste lime was used as potential cation source with magnesium salt($MgCl_2$) as control. The research was focused on its successful application in a novel integrated sludge treatment process, which is comprised of a high performance fermenter followed by a crystallization reactor. In the struvite precipitation test using synthetic wastewater first, which has the similar characteristics with the real fermentation effluent, the considerable nutrient removal(about 60%) in both ammonia and phosphate was observed within $0.5{\sim}1$ hr of retention time. The results also revealed that a minor amount(<5%) of ammonia stripping naturally occurred due to the alkaline(pH 9) characteristic in feed substrate. Stripping of $CO_2$ by air did not increase the struvite precipitation rate but it led to increased ammonia removal. In the second experiment using the fermentation effluent, the optimal dosage of magnesium salt for struvite precipitation was 0.86 g Mg $g^{-1}$ P, similar to the mass ratio of the struvite. The optimal dosage of waste lime was 0.3 g $L^{-1}$, resulting in 80% of $NH_4-N$ and 41% of $PO_4-P$ removal, at about 3 hrs of retention time. In the microscopic analysis, amorphous crystals were mainly observed in the settled solids with waste lime but prism-like crystals were observed with magnesium salt. Based on mass balance analysis for an integrated sludge treatment process(fermenter followed by crystallization reactor) for full-scale application(treatment capacity Q=158,880 $m^3\;d^{-1}$), nutrient recycle loading from the crystallization reactor effluent to the main liquid stream would be significantly reduced(0.13 g N and 0.19 g P per $m^3$ of wastewater, respectively). The results of the experiment reveal therefore that the reuse of waste lime, already an industrial waste, in a nutrient recovery system has various advantages such as higher economical benefits and sustainable treatment of the industrial waste.

  • Internal combustion engines typically expel 30%-40% of the energy supplied by fuel to the environment through their exhaust system. Therefore, further significant improvements in the thermal efficiency of IC engines are possible by recovering the waste heat from the engine exhaust gas. With this fact in mind, a numerical simulation was carried out to investigate the potential of using thermoelectric generation with an internal combustion engine for waste heat recovery. Physical parameters such as the exhaust temperature and mass flow rate were evaluated in the exhaust system, and the optimum location for inserting a thermoelectric generator (TEG) into the system was determined. The TEG will be located in the exhaust system and will use the energy flow between the warmer exhaust gas and the external environment. The optimum position of the temperature distribution and the TEG performance were predicted through numerical analysis. The experimental results obtained showed that the power output significantly increases with the temperature difference between the cold and hot sides of the TEG.

Development of Analytical Method and Monitoring of Veterinary Drug Residues in Korean Animal Products

  • This study was conducted to determine the residual amount of veterinary drugs such as meloxicam, flunixin, and tulathromycin in animal products (beef, pork, horsemeat, and milk). Veterinary drugs have been widely used in the rearing of livestock to prevent and treat diseases. A total of 152 samples were purchased from markets located in major Korean cities (Seoul, Busan, Incheon, Daegu, Daejeon, Gwangju, Ulsan and Jeju), including Jeju. Veterinary drugs were analyzed by liquid chromatography-tandem mass spectrometry according to the Korean Food Standards Code. The resulting data, which are located within 70-120% of recovery range and less than 20% of relative standard deviations, are in compliance with the criteria of CODEX. A total of five veterinary drugs were detected in 152 samples, giving a detection rate of approximately 3.3%; and no food source violated the guideline values. Our result indicated that most of the veterinary drug residues in animal products were below the maximum residue limits specified in Korea.

  • GC/MS was used to find an optimum experimental condition for the screening of organic acids in urine. Urinary organic acids were isolated through the liquid-liquid extraction method (LLE) to examine the influence of pH and the effect of including the back extraction and oximation processes respectively on the extraction. When pH was adjusted to 0.5 during the extraction without oximation process, relatively higher recovery rate and the smallest relative standard deviations (0.3-12.4%) were obtained. By removing the interference, the addition of back extraction made possible surer identification of organic acids with retention time of 15-16 minutes. Under this condition, we obtained the content distribution of urinary organic acids in healthy Korean children (n=16) by establishing the calibration curves for 51 standard organic acids.

  • The unique study of TiO2 sol-gel modified carbon paste electrode (CPE) nanocomposites have been developed for electrochemical sensor detecting fipronil pesticide compound. We develop the easy synthesized TiO2 via a sol-gel method and modified in CPE which applied electrochemical system as cyclic voltammetry (CV) because the concentration is proportional with current peaks. We discover the TiO2 optimal mass used of 0.1 g which is compared with 0.7 g carbon and 0.3 mL paraffin. It has high-current anodic (Ipa) of 1.13×103 μA and high-current cathodic (Ipc) -0.96×103 μA in scan rate of 0.5 V/s. The limit of detection (LOD) of fipronil has been determined of 34.0×10-5 μM in percent recovery of 0.8%. Its high-stability for lifetime TiO2-CPE nanocomposites was expressed for 13 days which mean that can be used for detecting fipronil pesticide.

Effect of Kyungohkgo on Aerobic Capacity and Anti-fatigue in High School Soccer Players (경옥고 섭취가 고등학교 축구선수의 운동수행능력 향상 및 피로 회복에 미치는 영향)

  • This study was designed to investigate the effects of Kyungohkgo to improve aerobic capacity and eliminate exercise-induced fatigue in high school soccer players. Twenty four subjects were participated and randomly assigned into two groups [KG, Kyungohkgo group (n=12); PG, Placebo control group (n=12)]. Two groups were completed treadmill exercise protocol using graded exercise test at before and after experimental treatment of 4 weeks. The $VO_2$max and endurance time were measured by gas analysis and heart rate (HR) was measured by polar system at pre, post 0, post 5, post 15, post 30 and post 60 minutes. Blood samples were collected to analyze blood components. 1. The $VO_2$max was significant increased in the group of after intake Kyungohkgo compared to the group of after intake placebo (p<.05). 2. The HR was significant decreased in the group of after intake Kyungohkgo compared to the group of after intake placebo during recovery time at post 5 mins(p<.05), 30 mins(p<.01), 60 mins(p<.01). 3. Weight, body mass index, percent body fat, anaerobic threshold, endurance time, blood lactate concentrate, lactate dyhydrogenase, creatine kinase, serum glutamic oxaloacetic transaminase, serum glutamic pyruvic transaminase, other energy sources(total-cholesterol, triglyceride, high density lipoprotein-cholesterol, low density lipoprotein-cholesterol, creatinine) and electrolyte (Na, K, Cl) were shown no significant differences between groups. These results suggested that Kyungohkgo can be used as ergogenic aids to improve aerobic capacity and eliminate exercise-induced fatigue.

Acute porcine reproductive and respiratory syndrome outbreaks in immunized sow herds: from occurrence to stabilization under whole herd vaccination strategy

  • Outbreaks of porcine reproductive and respiratory syndrome virus (PRRSV) in vaccinated sow herds from occurrence to stabilization were monitored and analyzed in terms of serology and reproductive performance. Three different conventional pig farms experienced severe reproductive failures with the introduction of a type 1 PRRSV. These farms had adopted mass vaccination of sows using a type 2 PRRSV modified live vaccine (MLV). Therefore, to control the type 1 PRRSV, an alternative vaccination program utilizing both type 1 and type 2 MLV was undertaken. Following whole herd vaccinations with both types of MLV, successful stabilization of PRRS outbreaks was identified based on serological data (no viremia and downward trends in ELISA antibody titers in both sows and suckling piglets) and recovery of reproductive performance. Additionally, through comparison of the reproductive parameters between outbreak and non-outbreak periods, it was identified that PRRSV significantly affected the farrowing rate and the number of suckling piglets per litter at all three pig farms. Comparison of reproductive parameters between periods when the different vaccination strategies were applied revealed that the number of piglets born in total and born dead per litter were significantly increased after the introduction of the type 1 PRRS MLV.

Quantitative Evaluation of Radix Astragali through the Simultaneous Determination of Bioactive Isoflavonoids and Saponins by HPLC/UV and LC-ESI-MS/MS

  • The three major active isoflavonoids (calycosin-7-O-β -glucoside, isomucronulatol 7-O-β-glucoside, formononetin) and two main saponins (astragaloside I, astragaloside IV) in an extract of Radix Astragali were determined using rapid, sensitive, reliable HPLC/UV and LC-ESI-MS/MS methods. The separation conditions employed for HPLC/UV were optimized using a phenyl-hexyl column (4.6 × 150 mm, 5 μm) with the gradient elution of acetonitrile and water as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 230 nm. The specificity of the peaks was determined using a triple quadrupole tandem mass spectrometer equipped with an electrospray ionization (ESI) source that was operated in multiple reaction monitoring (MRM) in the positive mode. These methods were fully validated with respect to the linearity, accuracy, precision, recovery and robustness. The HPLC/UV method was applied successfully to the quantification of three major isoflavonoids in the extract of Radix Astragali. The results indicate that the established HPLC/UV and LC-ESI-MS/MS methods are suitable for the quantitative analysis and quality control of multi-components in Radix Astragali.
