Trisoxazole Macrolide from a Marine Sponge Sarcotragus Species

  • Bioassay-directed fractionation of the lipophilic extract of a marine sponge Sarcotragus sp. led to the isolation of a known trisoxazole containing macrolide, mycalolide B (1). Its structure was identified by NMR and MS analyses. This is the first report on the isolation of macrolide from a sponge of the genus Sarcotragus (Order: Dictyoceratida).

  • A new marine sponge, Antho (Acarnia) seogwipoensis n. sp., of the family Microcionidae, was collected from Seogwipo-si, Jeju-do, Korea, about 100 m in depth using a gill net on 1969. The genus Antho Gray, 1867 including Demospongiae, Poecilosclerida, Microcionidae, is a large group of sponges. About 100 species in Antho were reported from worldwide. The genus Antho contains five subgenera: Antho, Acarnia, Isopenectya, Jia, and Plocamia. Among them, about 30 species in Acarnia were described in world sponge. A new sponge's body shape is branching, size up to 124 mm wide, 213 mm high, 3-8 mm thick in branch and 7-9 mm thick in stalk. Antho (Acarnia) seogwipoensis n. sp. is similar to A. (A.) novizelanicum Ridley and Duncan, 1881 based on their spicules type and skeletal structure, but differs in the spicules dimension and growth form. This new species is branched growth form and have three kinds of toxa.

  • A new sphingosine (1) was isolated from the MeOH extract of a marine sponge Haliclona (Reniera) sp. by bioactivity-guided fractionation. The 1D and 2D NMR, and MS spectroscopic analyses were used to establish the planar structure of 1. The stereochemistry of the compound was defined on the basis of modified Mosher's method, comparison of optical rotation and NMR data with those of the reported. Compound 1 was mildly cytotoxic to a panel of five human solid tumor cell lines.

  • A new glycerol ether has been isolated from a marine sponge Stelletta sp. By bioactivity-guided fractionation. The structure was established on the basis of NMR and MS analyses. The compound was evaluated for cytotoxicity against a small panel of five human tumor cell lines and exhibited significant cytotoxicity.

  • The ethanolic extracts of two shallow water sponge species collected from the Aegean Sea afforded thirteen terpenoids in total, two of which were determined to be new. The structure elucidation of the terpenoids was carried out by spectroscopic techniques and comparison with related authentic compounds. The terpenoids have also been assayed for antibacterial activity. This is the first report about the metabolites isolated from the marine sponges of the Aegean Sea in Turkey.

Two New Scalaranes from a Korean Marine Sponge Spongia sp.

  • Intensive chemical investigation of Korean marine sponge Spongia sp. has led to the isolation of two new scalaranes. The planar structures of the new compounds 1 and 2 were determined through 1D and 2D NMR spectral data analysis, while the relative stereochemistry of the compounds was determined based on the analysis of $^1H-^1H$ coupling constants and NOESY spectroscopic data. Compounds 1 and 2 did not display any significant biological activities on farnesoid X-activated receptor (FXR) in co-transfection assay.

ARDRA와 DGGE를 이용한 Halichondria panicea 해면의 공생세균 다양성 (Bacterial diversity of the Marine Sponge, Halichondria panicea by ARDRA and DGGE)

  • 제주도에서 채집한 해양 해면 Halichondria panicea의 공생세균 군집구조를 배양에 의한 ARDRA와 비배양에 의한 DGGE 분석 방법에 의하여 조사하였다. 16S rRNA gene-ARDRA 분석을 위해 변형된 Zobell 배지와 Marine agar를 이용하여 120균주를 선별하고 제한효소, HaeIII와 MspI을 사용하여 ARDRA type을 구분하였다. ARDRA type으로부터 유래한 16S rRNA gene 염기서열 분석 결과, 알려진 세균 종과 96% 이상의 유사도를 나타내었으며 Alphaproteobacteria, Gammaproteobacteria, Bacteroidetes, Firmicutes 등 3문 4강이 관찰되었다. 그 중 Alphaproteobacteria가 우점하였다. 같은 해면, H. panicea의 DGGE 분석을 위해 total genomic DNA로부터 16S rRNA gene를 증폭하여 DGGE fingerprinting을 수행한 결과 14개의 밴드가 관찰되었다. 각 밴드의 16S rRNA gene 염기서열은 알려진 세균의 염기서열과 100%의 유사성을 나타내었으며 대부분의 염기서열은 uncultured bacteria에 속하였다. DGGE 분석으로부터 미생물의 군집은 Alphaproteobacteria, Gammaproteobacteria, Acidobacteria, Actinobacteira, Bacteroidetes, Cyanobacteria, Chloroflexi 등 6문 7강으로 나타났다. ARDRA와 DGGE 방법에 의해 Alphaproteobacteria, Gammaproteobacteria, Bacteroidetes가 공통적으로 발견되었으나 전체적인 공생세균의 군집구조는 분석방법에 따라 차이를 나타내었다. 배양에 의한 방법보다 비배양 방법에서 더 다양한 세균군집구조를 나타내었다.

Aureivirga callyspongiae sp. nov., Isolated from Marine Sponge Callyspongia elegans

  • A Gram-negative, aerobic, motile by gliding, and rod-shaped marine bacterium, designated CE67T was isolated from the marine sponge Callyspongia elegans on Biyang-do in Jeju Island. The CE67T strain grew optimally at 25℃, pH 7.5, and in the presence of 2-3% (w/v) NaCl. Phylogenetic analysis based on 16S rRNA gene sequence showed that strain CE67T was related to the genus Aureivirga and had the highest 16S rRNA gene sequence similarity to the Aureivirga marina VIII.04T type strain (96.3%). The primary fatty acids (>10%) of strain CE67T were iso-C15:0 (35.3%) and iso-C17:0 3OH (21.8%). The polar lipid profile of strain CE67T contained phosphatidylethanolamine, unidentified aminolipids, and unidentified lipids. The predominant menaquinone was MK-6. The DNA G+C content was 29.1 mol%. Based on the polyphasic taxonomic analysis, strain CE67T was determined to be a representative novel species of the genus Aureivirga for which we propose the name Aureivirga callyspongiae sp. nov., whose strain type is CE67T (=KCTC 42847T=JCM 34566T).

Suberitenone B: A Novel Inhibitor of Cholesteryl Ester Transfer Protein (CETP) with an Unprecedented Skeletal Class from the Antarctic Sponge Suberites sp.

  • Cholesteryl Ester Transfer Protein (CETP) mediates the transfer of cholesterol ester and triglyceride between high-density lipoprotein (HDL) and other low-density lipoproteins, therefore, it might influence HDL levels. The levels of HDL is closely related to the atherogenic diseases in human and there were several reports that the trasgenic mice expressing CETP had much worse atherosclerosis than non-expressing control one. Therefore, selective inhibitors of CETP have the potential to be used as antiatherosclerotic agents. Continued screening for potent inhibitors of CETP led to the isolation of Suberitenone B from marine sponge. Suberitenone B, sesterterpenoids of a new skeletal class have been isolated from the sponge Suberites sp. collected from King George Island the Antartic. The structure of the metabolite has been determined by NMR experiments and chemical methods.

천연물 연구에서의 Peroxide 성분 (Peroxide Constituents in the Natural Product Research)

  • Peroxides in natural products have been recently received a considerable attention due to their various biological and pharmacological properties. Nearly 300 peroxides have been isolated and structually characterized from natural sources, mainly as constituents of Compositae and marine sponge, and occur randomly in about 10 other plant families. Among peroxides studied, sesquiterpene endoperoxide, quinghaosu, has been already clinically applied as a new antimalarial drug. Based on the peroxides reported, structural classification, natural distribution and biological and pharmacological activities are reviewed. Color reagent and spectroscopic identification of peroxide are also described.
