Hepatoprotective Effects of Paecilomyces tenuipes Against Carbon Tetrachloride-induced Toxicity in Primary Cultures of Adult Rat Hepatocytes

  • Paecilomyces tenuipes (PT), one of the Ascomycetes family, has been used for medicinal purposes due to its broad pharmacological activities. The present study was undertaken to investigate the hepatoprotective effects of PT water extracts against $CCl_4$-induced hepatotoxicity in primary cultures of adult rat hepatocytes. When the extract of PT was directly added into the culture medium at 1, 2, and 5 mg/ml, the extracts not only reduce the $CCl_4$-induced elevation of aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase, and lipid peroxide, but also protect cultured hepatocytes from $CCl_4$-induced reduction of reduced glutathione, glutathione reductase, glutathione-S-transferase, glutathione peroxidase, catalase and superoxide dismutase. In addition, the effects of PT water extracts on cytochrome P450 enzymes were relatively marginal, indicating that the hepatoprotective effects of PT extract against $CCl_4$-induced toxicity might not be due to the inhibition of $CCl_4$ activation. In conclusion, the PT extracts were effective in protecting against $CCl_4$ induced hepatotoxicity in hepatocyte cultures, at least in part, by scavenging free radicals, and by modulating enzyme systems involved in cellular oxidative stress.

Genotoxicity Assessment of Gardenia Yellow using Short-term Assays

  • Gardenia yellow, extracted from gardenia fruit, has been widely used as a coloring agent for foods, and thus, safety of its usage is of prime importance. In the current study, short-term genotoxicity assays were conducted to evaluate the potential genotoxic effects of gardenia yellow. The gardenia yellow used was found to contain 0.057 mg/g of genipin, a known biologically active compound of the gardenia fruit extract. Ames test did not reveal any positive results. No clastogenicity was detected by a chromosomal aberration test, even on evaluation at the highest feasible concentration of gardenia yellow. Gardenia yellow was also shown to be non-genotoxic using an in vitro comet assay and a micronucleus test with L5178Y cells, although a marginal increase in DNA damage and micronuclei frequency was reported in the respective assays. Additionally, in vivo micronucleus test results clearly demonstrated that oral administration of gardenia yellow did not induce micronuclei formation in the bone marrow cells of male ICR mice. Taken together, our results indicate that gardenia yellow is not mutagenic to bacterial cells, and that it does not cause chromosomal damage in mammalian cells, either in vitro or in vivo.

울산 지역 보건소 이용 임신부의 아연 영양상태에 관한 연구 (The Study of Zinc Nutritional Status of Pregnant Women Visiting in Public Health Centers in Ulsan)

  • The study was designed to assess the zinc nutritional status by gestational age of pregnant women visiting in public health centers in Ulsan. The subjects were divided into 3 trimester by last menstrual period(LMP). Interview for dietary zinc intake and general characteristics of each subjects was given and biochemical analysis of blood and urine was performed. Serum zinc concentration and urinary zinc excretion were analyzed by Flame Atomic Absorption Spectrophotometer, and alkaline phosphatase(ALP) activity was analyzed by Bowers & McComb\\`s method with Schimadzu automatic analyser. Also urinary creatinine was analyzed by Hawk\\`s method. Mean intake of zinc was 6.61${\pm}$1.57mg and did not meet the RDA(44.1% of RDA) for pregnant women by gestational age. Zn intake of 3rd trimester was significantly increased but dietary zinc was almost supplied with cereal and grain (47.30%) which were reported with low zinc availability due to phytate. Mean concentration of serum Zn in 1st trimester was 86.4${\pm}$10.5$\mu\textrm{g}$/dl, was 72.4${\pm}$10.3$\mu\textrm{g}$/dl in trimester and 65.1${\pm}$10.8$\mu\textrm{g}$/dl in 3rd trimester and was declined significantly by gestational age during pregnancy. In was concluded that a decline in serum Zn by gestional age was not influenced by amount of Zn intake. However ALP activity and urinary zinc excretion increased significantly by gestational age. Zinc nutritional status of pregnant women was not confirmed yet due to the physiological changes during pregnancy. However, the pregnant woman may be in a marginal zinc deficient status because of low amount of Zn intake and low bioavailability of Zn from dietary sources. 관상동맥 회로술 치험 1예 (Aorto-Coronary Bypass Graft -A Case Report-)

  • Occlusive coronary artery disease presents a potential threat to a significant population in the United States. According to many case reports, the increasing incidence of coronary artery disease due to atherosclerosis is noted in Korean, recently. Operative vascular procedures have increased steadily in number over the past 20 years in the United States. There have been many isolated case reports about coronary artery surgery, but these had little clinical impact. Nowadays, major three coronary bypass surgery has developed principally at three cardiac centers in the United States since 1967. Among three coronary bypass operation, the aorta-coronary artery saphenous vein bypass graft was first demonstrated by Favolaro and Effler at the Cleveland Clinic in 1967. We experienced one case of coronary artery disease, which was treated successfully by aorta-coronary saphenous vein graft with mild hypothermia under extracorporeal circulation in May, 1977. Saphenous vein was removed from above the knee and was 2.5 mm in diameter. The left ventricle was not vented for the left ventricle was not overdistended. Temporary artificial pace-maker-Medtronic-was implanted for the prevention and treatment of post-operative arrhythmia and heart block in post-operative first day. He is a 57 year old male businessman who had been suffered from hypertension [200 mmHg in systolic pressure] since 4 years ago, who had intermittent conservative treatment at local clinic. He had been afflicted with severe chest pain with choking sensation for 50 days. This symptom was aggravated exposing cold weather, or cold water, but was respond to rest. Pre-operative ECG revealed no any other ischemic sign except sinus bradycardia. Significant S-T segment depression was noted at lead II, AVF after double 5 minutes exercise, indicating positive Master`s test. Serum cholesterol was slight elevated to 253 mg/dl. Final pre-operative diagnosis was made by coronary arteriogram, which showed about 1.0-cm segmental 90 % occlusive atherosclerotic lesion in the proximal part of right coronary artery above the origin of acute marginal artery. Left coronary artery revealed good patency and there was no collateral circulation between right and left coronary artery .Hospital course was not eventful. He was discharged with good result on the post-operative day. He has been free from chest pain for longer than 2 years. And also the arterial flow in the coronary bypass graft is auscultated with the pocket-sized ultrasonic velocity detector, which shows the patency of the coronary bypass graft good.

치과용 수복재의 용해성에 관한 분석연구 (A STUDY ON THE SOLUBILITY OF DENTAL RESTORATIVE MATERIALS)

  • The purpose of this experiment was to measure the leaking and solubility of commonly used dental restorative materials - Silux plus (CS), Hi-pol (CH), Clearfil F-II, Fissureseal (FS), Glass-Ionomer cement Fuji Type II (GI), Amalgam Cavex 68 (AM), Zinc Phosphate Cement (ZP) and gutta-percha (GP) and investigate the relation between the solubility and marginal leakage. Disc-shape specimens were fabricated with each material and dipped into deionized water, 0.01M lactic acid and 0.005M KOH solution, thus the total ionic concentrations in each solution was measured with ion chromatograph after 1, 3, and 7 days, respectively. For the solubility test, each specimen was immersed in 0.001M and 0.01M lactic acid for 24 hours, respectively and total weight loss was calculated. Also, Zn leaking through the margin of restorations was measured. The obtained results were as follows: 1. The amounts of eluted ion from the eight materials were most in 0.01M lactic acid and least in deionized water. 2. Of the eight materials, the fluoride release was greatest for glass ionomer cement (GI) in 0.01 M lactic acid after 7 days. 3. In analysis of the divalent cation, Mg was eluted most for zinc phosphate cement (ZP) and Ca for Clearfil F-II (CF) in 0.01M lactic acid after 7 days. 4. In analysis of transition metals, Cu and Zn were detected only. 5. The solubility rate of eight materials was greater in 0.01M lactic acid than in 0.001M for 24 hours, for zinc phosphate cement (ZP) the rate was greatest (5.4%) in 0.001M lactic acid, and amalgam least (0.01%). 6. The Zn concentration of restorative material with Z.P.C base was greater in 0.01M lactic acid than in 0.001M lactic acid.

Genotoxicity Studies on Carrageenan: Short-term In Vitro Assays

  • Carrageenan is a naturally-occurring sulfated polygalactan which has been widely used in the dairy industry and a gelling agent in non-dairy products. In this study, four short-term in vitro genotoxicity assays were investigated to evaluate the potential genotoxic effects of carrageenan. The mutagenicity of carrageenan was evaluated up to a maximum dose of 5 mg/plate in Ames test. There was no increase in the number of revertant colonies compared to its negative control at any dose in all of strains tested. To assess clastogenic effect, the in vitro chromosomal aberration assay was performed using Chinese hamster lung cells. Carrageenan was not considered to be clastogenic in this assay at up to the highest feasible concentration which could be evaluated. The in vitro comet assay and micronucleus test results obtained on L5178Y cells also revealed that carrageenan has no genotoxicity potential, although there was a marginal increase in micronuclei frequencies and DNA damage in the respective micronucleus and comet assays. Taken together, our results indicate that carrageenan was not genotoxic based on four in vitro genotoxicity results.

In Vitro Antioxidant Activity Profiles of ${\beta}$-Glucans Isolated from Yeast Saccharomyces cerevisiae and Mutant Saccharomyces cerevisiae IS2

  • To explore the possible usefulness of ${\beta}$-glucans as natural antioxidants, the antioxidant profiles of ${\beta}$-glucan, extracted from Saccharomyces cerevisiae KCTC 7911, and water soluble and insoluble mutant ${\beta}$-glucan, isolated from yeast mutant S. cerevisiae IS2, were examined by five different in vitro evaluation methods: lipid peroxidation value (POV), nitric oxide (NO), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power, and ${\beta}$-carotene diffusion assay. The antioxidant activities of all ${\beta}$-glucans evaluated in POV test were comparable to or better than that of the known antioxidant, vitamin C. Remarkably, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan possessed 2.5-fold more potent activity than vitamin C at a dosage of 2 mg. Although vitamin C showed 100-fold greater activity than all ${\beta}$-glucans in NO and DPPH tests for measuring the radical scavenging capacity, all ${\beta}$-glucans revealed higher radical scavenging activity than the known radical scavenger, N-acetyl-L-cysteine (NAC), in DPPH test. The water insoluble mutant ${\beta}$-glucan had 2.6- and 5-fold greater antioxidative activity than water soluble ${\beta}$-glucan in NO and DPPH tests, respectively, showing that all ${\beta}$-glucans were able to scavenge radicals such as NO or DPPH. While all ${\beta}$-glucans revealed lower antioxidant profiles than vitamin C in both reducing power activity and ${\beta}$-carotene agar diffusion assay, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan did show a marginal reducing power activity as well as a considerable ${\beta}$-carotene agar diffusion activity. These results confirmed the potential usefulness of these ${\beta}$-glucans as natural antioxidants.

The Effects of Smoking Cessation and Antioxidant Vitamins on Oxidative Stress

  • In this study, the effects of smoking cessation and relative antioxidant activities on the oxidative stress were determined by using in vitro method. Thirty healthy smokers who were free of any disease and smoked more than 1 pack per day for the past 10 years participated in this study. For smoking cessation, smokers were asked to wear nicotine patch (21mg nicotine/ patch) everyday for 30 days and then to replace at the same time of the day. Smoking cessation program in conjunction with nicotine patch replacement was also conducted every week, one hour/each session, for 4 weeks. Canthaxanthin, $\beta-carotene$, and $\alpha-tocopherol$ were added into red blood cells at pre and post smoking cessation. As indicators of oxidative stress, hemoglobin degradation, lipid peroxidation, and percent hemolysis were determined at both pre and post smoking cessation. After 30 days of smoking cessation, the subjects gained an average of 5 pounds, varying 2 to 8 pounds, by suggesting that behavioral problems rather than nicotine itself are more important for gaining weight in ex-smokers. The total hemoglobin concentrations in blood were similar in pre and post smoking cessation, but smoking cessation resulted in a decrease in the percentage of methemoglobin from 0.96% to 0.85% Smoking cessation also caused to decease malondialdehyde (MDA) values ($26.7{\pm}7.8$ vs. $23.6{\pm}4.5$ (without oxidation), $179.3{\pm}21$ vs. $161.2{\pm}28$ nmol/ml (with oxidation) (p<0.05)), not percent hemolysis. Various antioxidants with smoking cessation significantly decreased MDA values(p<0.05), in contrast to marginal decrease of MDA in smoking cessation only. Three antioxidants used in this stu study were similarly effective in inhibiting MDA production, but relative effectiveness of canthaxanthin or $\alpha-tocopherol$ was greater than that of $\beta-carotene$ (p<0.05), in case of oxidation induced. The percent hemolysis was greatly decreased when antioxidants were added into the blood of ex-smokers (p<0.05) but no statistical significance in relative effectiveness of antioxidants was observed.

Zinc Status Assessment by Analysis of Mononuclear Cell Metallothionein mRNA Using Competitive-Reverse Transcriptase-Polymerase Chain Reaction

  • Marginal Zn deficiency is prevalent through the world and yet human zinc status has not been properly assessed due to the lack of a reliable diagnostic indicator. One potential possibility for zinc status assessment using Zn-binding protein, metallothionein (MT)-mRNA, has been proposed. The purpose of the present study was aimed to show whether measurement of mononuclear cell (MNC) MT mRNA, using a competitive-reverse transcriptase-polymerase chain reaction (competitive-RT-PCR) assay, could indicate zinc status in human subjects. In this study, MNC MT-mRNA expression was measured using a competitive-RT-PCR to compare before and after 14 days of zinc supplementation (50 mg Zn/das zinc gluconate). RT-PCR oligonucleotide primers which were designed to amplify both a 278 bp segment of the human MT-2A cDNA and a 198 bp mutant competitor cDNA template from MNCs, were prepared. MT-2A mRNA was normalized by reference to the housekeeping gene, $\beta$-actin, mRNA for which was also measured by competitive-RT-PCR. There was considerable inter-individual variation in MT-mRNA concentration and yet, the mean MT-2A mRNA level increased 4.7-fold after Zn supplementation, as compared to before Zn supplementation. This MT-2A mRNA level was shown as the same pattern and, even more sensitive assay, compared to the conventional plasma and red blood cells (RBCs) Zn assessment in which plasma and RBCs zinc levels increased 2.3- and 1.2-fold, respectively (p<0.05). We suggest that MT competitive-RT-PCR can be a useful assessment tool for evaluating human zinc status.

Bioconversion of Lignocellulosic Materials with the Contribution of a Multifunctional GH78 Glycoside Hydrolase from Xylaria polymorpha to Release Aromatic Fragments and Carbohydrates

  • A bifunctional glycoside hydrolase GH78 from the ascomycete Xylaria polymorpha (XpoGH78) possesses catalytic versatility towards both glycosides and esters, which may be advantageous for the efficient degradation of the plant cell-wall complex that contains both diverse sugar residues and esterified structures. The contribution of XpoGH78 to the conversion of lignocellulosic materials without any chemical pretreatment to release the water-soluble aromatic fragments, carbohydrates, and methanol was studied. The disintegrating effect of enzymatic lignocellulose treatment can be significantly improved by using different kinds of hydrolases and phenoloxidases. The considerable changes in low (3 kDa), medium (30 kDa), and high (> 200 kDa) aromatic fragments were observed after the treatment with XpoGH78 alone or with this potent cocktail. Synergistic conversion of rape straw also resulted in a release of 17.3 mg of total carbohydrates (e.g., arabinose, galactose, glucose, mannose, xylose) per gram of substrate after incubating for 72 h. Moreover, the treatment of rape straw with XpoGH78 led to a marginal methanol release of approximately 17 ㎍/g and improved to 270 ㎍/g by cooperation with the above accessory enzymes. In the case of beech wood conversion, the combined catalysis by XpoGH78 and laccase caused an effect comparable with that of fungal strain X. polymorpha in woody cultures concerning the liberation of aromatic lignocellulose fragments.
