개에 있어서 악성 림프부종 일례 (A Case of Malignant Lymphedema in a Dog)

  • A five-year-old, female Great Dane dog with edema, localized trauma, mild pain, and lameness of the right hind limb was referred to the Veterinary Medical Teaching Hospital of Chungbuk National University. This dog had a history of mammary tumor excisions 6 months ago. Abnormal changes were not seen in the values of complete blood count and serum biochemical tests. But pedal direct lymphangiography using aqueous-based radiographic agent showed the obstructed lymph flow in right popliteal lymph node. Based on these observations, the dog was suspected as lymphedema resulted from lymph drainage flilure without any other possibilities of inflammation or other causes. Although recommended chemotherapy and physiotherapy had been applied for resolvinr presented problems for one month, there was no improvement on edema of damaged region and any other clinical signs. Therefore, the necropsy was performed after euthanasia under agreement of the owner of patient. In histopatholofical examination, the most characteristic lesions in the mass of femoral region were diffuse edema fibrosis and neoplastic cells in the lymphatics. Also, the neoplastic cells were very similar to those found in the tumor mass of mammary gland, which had diagnosed as fibrosing carcinoma. These facts suggested that the cause of obstructed lymph flow was the neoplasia in lymphatics of the right hind limb. With these results, a diagnosis of malignant lymphedema was made in this dog.

Levels of Viral Glycoprotein Provide a Measure of Modulated Chemotherapeutic Effect

  • A chemosensitivity assay with small replicate Mm5mt/cl C3H mammary tumor cell cultures was developed to determine whether changes in viral antigen expression and release into culture fluids could be utilized as an in vitro measure of modulating drug effect. The 52,000 MW viral envelope glycoprotein (gp52) of the mouse mammary tumor virus (MMTV) was measured in culture fluids of control and drug-treated cultures while cell density was simultaneously determined by cell staining and OD 664 nm determination. While extra-cellular gp52 levels and cell density progressively increased over 72 hours for control cultures, declines in both parameters provided dual measures of effect for combination [N(phophonacetyl-L-aspartic acid)+5-fluorouracil], combination 〔N(phophonacetyl-L-aspartic acid )+5-fluoro-5'-deoxyuridine〕and single component treatment of this combination. At each treated time point, thesecombinations begin to produce a greater decline in both cell density and gp52 levels as compared to single drug treatments. These results indicate that N(phopho-nacetyl-L-aspartic acid) in combination can enhance the effectiveness of single drug.

Ductular mammary carcinoma in a lioness

  • Well bordered solid enlarged 16.7x9.7x8 cm in the size neoplasma was found in a 7-year-old lioness without any clinical signs of diseases in Chonju city zoo and surgically removed. Simple lobes were separated by pale gray yellowish fibrous tissue. The tumor parenchyma was yellowish with numerous cysts, necrotic foci and hemorrhages. Histologically neoplasma was composed of cell nests different in the size and shape with high mitotic activity. Slow invasive spreading on the border was observed. Formation of the narrow ductular lumina bordered the flattened cells leading to keratinization was seen in some places. It was noted by supplemental clinical investigations, that the tumor did not metastasized to other organs. This tumor is considered to be a sample of a malignant tumor without metastatic spreading and was note described in literature yet.

Expression of CD133, CD44, CK7, and OCT4 in Animal Cancers

  • Cancers are mainly sustained by a small pool of neoplastic cells, known as cancer stem cells or tumorinitiating cells. These cells possess the ability to self-renew and proliferate, and are thus able to form the tumor. In the present study cells that correspond to cancer stem cells in mammary and liver cancers in animals were identified by the expression of CD133, CD44, CK7, and OCT4 using immunochemistry. As a result, we found with CD133+ and CD44+ cancer stem cell-like phenotypes in mouse and canine hepatocellular carcinoma and canine mammary gland tumors. However, CK7+ and OCT4+ cells were not identified in animal mammary and liver cancer. CD133+ and CD44+ cells are wellknown stem cell lines and play key roles in development and metastasis in human cancer. These findings suggest that cancer stem cells are involved in animal tumorigenesis and may provide insight into mechanisms in cancer development as well as cancer diagnostics.

이하선에 발생한 유선 유사 분비성 암종 1예 (A Case of Mammary Analogue Secretory Carcinoma of the Parotid Gland)

  • Mammary analogue secretory carcinoma (MASC) has histologic similarities to not only acinic cell carcinoma but also other low grade cystadenocarcinoma, and has similar features to breast secretory carcinoma. MASC was not described through the existing classification system previously. But, MASC was distinguished from other salivary gland tumors by Skalova et al. in 2010, MASC has ets variant gene 6-neurotrophic tyrosine kinase, receptor, type 3 (ETV6-NTRK3) translocation. So far, there are 4 cases of MASC recognized in the head and neck region in Korea. One of the four is a tumor from the submandibular gland, and the other three are of the parotid gland. In this case report, we report a 40-year-old man with a MASC of the parotid gland, who presented with right infra-auricular mass.

Hemostatic Dysfunction in a Dog with Mammary Gland Carcinoma

  • Hypercoagulability is the most common coagulopathy seen in dogs with neoplasia, whereas a hypocoagulable state is relatively rare. A 16-year-old spayed female miniature Schnauzer presented with bilateral epistaxis, bilateral hindlimb swelling, and lameness. Previously, the dog was diagnosed with intermediate grade solid carcinoma after mastectomy, and then showed generalized ecchymoses on the abdomen and bilateral hindlimbs on presentation. Laboratory tests revealed a hypocoagulable state, including thrombocytopenia, hypofibrinogenemia, and delayed prothrombin and activated partial thromboplastin time. Thromboelastography demonstrated a prolonged K time with a decreased alpha angle and low maximal amplitude, reflecting a decrease in clot strength. The hypocoagulable state of disseminated intravascular coagulopathy was speculated based on the presence of an underlying tumor, clinical symptoms, and laboratory results of the hypocoagulable state. This case shows the incidence of hemostatic dysfunction as a paraneoplastic syndrome in a dog with mammary gland carcinoma.

마우스를 이용한 생체내 실험에서의 플라보피리돌의 방사선민감화 효과 (In Vitro Radiosensitization of Flavopiridol Did Not Translated into In Vivo Radiosensitization)

  • 목 적: 이전의 암세포주를 이용한 실험실내 연구에서 플라보피리돌은 암세포의 방사선에 의한 아포토시스를 증가시키는 것을 알 수 있었다. 이번 연구에서는 마우스를 이용한 생체내 실험에서 플라보피리돌의 방사선민감화 효과를 알아보고자 하였다. 대상 및 방법: 마우스 유방암 세포주인 EMT-6를 Balb/c 마우스에 피하주사하여 종양을 만든 후 플라보피리돌 단독 치료군, 방사선 단독 치료군, 방사선과 플라보피리돌 병합 치료군 및 대조군으로 나누어 종양의 성장 속도를 비교 하였다. 플라보피리톨은 2.5 mg/kg을 하루 2회 복강내에 주사하였고, 방사선은 1일 1회, 회당 4 Gy를 조사하여 총 28 Gy를 조사하였다. 각 치료군에서의 종양 성장 곡선을 구하여 비교하였다. 마우스로부터 채취한 종양으로 파라핀 절편을 만틀어 TUNEL 및 면역조직화학염색을 시행하였다. 결 과: 종양 성장을 비교하였을 때 대조군보다 방사선 단독 치료군과 방사선과 플라보피리돌 병합 치료군에서 종양 성장이 지연되는 것을 볼 수 있었다. 그러나 대조군과 플라보피리돌 단독 치료군 사이에서는 종양 성장에 차이가 없었고, 방사선 단독 치료군과 방사선과 플라보피리돌 병합 치료군 사이에서도 차이가 없었다. TUNEL 염색으로 아포토시스율을 비교했을 때 각 치료군 사이에 차이가 없었으며, 면역조직화학염색으로 Ku70 발현을 비교했을 때에도 각 치료군 사이에 차이가 없었다. 결 론: 플라보피리돌은 마우스 유방암 모델에서 방사선민감화 효과를 나타내지 않았다.

백서에서 발생시킨 유방암의 방사선 치료시 온열요법이 미치는 영향에 관한 실험적 연구 (An Experimental Study on The Effect of Hyperthermia on Radiation Therapy of Mammary Carcinoma of Rat)

  • The renewed interest in the use of hyperthermia in cancer therapy is based on radiobiological and clinical evidence indicating that there may be significant thereapeutic advantages with the use of hyperthermia alone or combined with irradiation plus heat. Authors performed the experiment using the chemically induced mammary carcinoma of rats to observe the difference in temperature changes between tumor and normal tissue during heat, and to compare the response of the tumors to radiation alone and to radiation plus hyperthermia. The results were as follows 1. Temperature of tumors was significantly higher than in the normal tissue during heating and the difference was about $1.5^{\circ}C$. 2. $TCD_{50}$ in radiation alone and hyperthermia immediately following radiation was 1,282 rad and 795 rad, respectively and TER value was 1.81.

수술적으로 절제한 유선종양 79례 (Mammary Gland Tumors Treated by Surgical Excision : A Retrospective Study in 79 Dogs)

  • Mammary gland tumors are the most common neoplasms occurring in female dogs. The treatments of mammary gland tumors are surgery, chemotherapy, hormonal therapy and radiational therapy, but surgical removal remains widely accepted treatment option for mammary gland tumors. The purpose of this study is to evaluate clinical outcomes of dogs which are performed surgical excision. Medical records were reviewed for dogs(79 cases) with mammary gland tumors treated surgically at Veterinary Medical Teaching Hospital in Seoul National University from 2001 to 2005. While 49 cases(62.0%) were benign, 30 cases(38.0%) were malignant tumors. The mean age of these dogs was 10.4 years old(range $1{\sim}16$ years). The maximal diameter of malignant tumors were various, whereas most of the benign tumors were smaller than 3cm(36 cases, 73.5%). In 12 cases(15.2%), regional lymph nodes were enlarged and lymph nodes of 3 cases had resected surgically. The performed techniques were total mastectomy, regional mastectomy, simple mastectomy, lumpectomy and unilateral mastectomy in order. Twenty-eight cases(35.4%) had postoperative complications consisted of recurrence of tumors, necrosis, dehiscence, delayed healing and edema of limbs. The recurrence rates of benign and malignant mammary gland tumors were 8 cases(16.3%) and 6 cases(20.0%). Metastasis rate was 7 cases(8.9%). Although postoperative complications were no remarkable difference in recurrence rates among surgical techniques in this study, other complications such as edema of limbs, necrosis, dehiscence and delayed healing were remarkable difference as surgical techniques. Therefore, this result suggest that choice of appropriate surgical techniques should be determined according to each patient's physical status and characteristics of tumors.
