It has been reported that hepatic microsomal cytochrome P450(CYP) 2E1 is responsible for the metabolism of chlorzoxazone(CZX) to 6-hydroxychlorzoxazone. The present study was undertaken to assess the possible interaction of rutaecarpine, an alkaloid originally isolated from the unripe fruit of Evodia rutaecarpa, with CZX. Male Spraque-Dawley rats were administered with 80 mg/kg/day of oral rutaecarpine for three consecutive days. Twenty four hr after the pre-treatment with rutaecarpine, the rats were treated with 20 mg/kg of intravenous CZX. Rat hepatic microsomes isolated from rutaecarpine-treated rats showed greater(50% increase) activity of p-nitrophenol hydroxylase(a marker of CYP2E1) when compared with the control rats. Compared with control rats, the AUC of CZX was significantly smaller(84% decrease) possibly due to significantly faster CL(646% increase) in rats pretreated with rutaecarpine. This could be, at least partially, due to induction of CYP2E1 by rutaecarpine.
Sulfites are used as food preservatives and excessive sulfite might disturb the body systems including the brain. Curcumin shows protective effects on the nervous system toxicity. The present study aimed to evaluate the protective role of curcumin in sulfite-induced anxiety in rats. Male rats were divided into five groups. The rats in groups I to V received distilled water (vehicle of sulfite, 1 mL/d), olive oil (vehicle of curcumin, 1 mL/d), curcumin (100 mg/kg/d), sulfite (25 mg/kg/d), and sulfite+curcumin, respectively, by daily gastric gavage for 8 weeks. At the end of 8 weeks the rats were tested in the elevated plus-maze for anxiety. The results showed that concomitant treatment of curcumin during sulfite consumption prevented the reduction of the time spent in the open arm and entrance to the open arm (the indexes of anxiety). Besides, an increase was found in motor activity of the rats in the sulfite+curcumin group compared to the sulfite-treated animals. Exposure of sulfite in rats can induce anxiety, and curcumin can act as an anti-anxiety agent.
This study was performed to investigate nutritional effect and the utilization possibility of rapeseed oil which could replace the imported edible oils and oil crops. The proximate compositions of a recommendable cultivar(Mokpo Dangyo 19) and a native kind(Asahi) and the characteristics of these rapeseed oils were analyzed. The animal experiment of these rapeseed oils was carried out during 8 weeks of growing periods after weanling. Forty male weanling Sprague-Dawley rats were randomly assigned to 3 diet groups of Dangyo 19 oil, Asahi oil and commercial Soybean oil. Mokpo Dangyo 19 variety contained more lipids than Asahi variety by 8% and there was no difference in physiochemical characteristics between Dangyo 19 oil and Asahi oil except that erucic acid was contained little in Dangyo 19 oil but 50% in Asahi oil. Body weight gain, FER(Feed efficiency ratio) and PER(Protein efficiency ratio) of rats fed Dangyo 19 oil were higher than those of rats fed Asahi oil and organ weights such as liver, kidney and epididymal fat pads weights of rats fed Dangyo 19 oil were significantly higher(P<0.05) thanthose of rats fed Asahi oil. the apparent digistibility of total diet and total lipid were higher in rats fed the diets containing Dangyo 19 oil than Asahi oil. The content of total lipid in heart in heart were not significantly different with dietary oil kinds. The content of total lipid in liver increased with age in all rapeseed oil groups.
This study was carried out to examine whether kimchi has hypolipidemic effect and to know how it exert lipid-lowering effect in rats. Male Sprague-Dawley rats were fed with kimchi-fee diet, or 3%, or 5%, or 10% kimchi diets for 6 weeks. Plasma cholesterol level was lowered in rats fed all concentrations of kimchi diets, and plasma triglyceride(TG) level was lowered in 10% kimchi diet group compared with that of control significantly(p<0.05). Th intake of kimchi lowered VLDL-cholesterol and VLDL-TG levels, whereas increased HDL-cholesterol level significantly(p<0.05). LDL-cholesterol level was lowered only in 5% kimchi diet group and LDL-TG level was lowered in all kimchi diet groups compared with those of control significantly (p<0.05). the intake of 5% and 10% kimchi diets also lowered the levels of hepatic cholesterol, TG, total lipid, and apolipoprotein B, whereas increased the levels of fecal total fat, cholesterol, TG, and apolipoprotein A-1 significantly(p<0.05). Triiodothyronine(T$_3$) level was elevated in rats fed kimchi diet, whereas thyroxine(T$_4$) level was not affected by kimchi treatment. These observations support that the intake of kimchin in rats loweres plasma and hepatic lipid levels by increasing the excretion of TG and cholesterol through feces, by the elevation of T$_3$ level, and by the altered lipoprotein metabolism.
1. 질병명 : Interstitial pneumonia 2. 본질명의 개요 역사 및 역학 Michael R Elwell, Joel F Mahler, G N Rao: “ Have You Seen This\ulcorner" ; Inflammatory Lesions in the Lungs of Rats. Toxicologic Pathology, 25: 529-531, 1997. Male and female F344 rats, approximately 19 weeks old, from prechronic toxicity studies performed for NTP/NIEHS over a period of several years at different laboraories located throughout the US. The rats were supplied by 2 different production colonies located in the eastern and western areas of the US. Gross findings ㆍ In some rats the lesions were noted as pale or tan foci in the lungs Microscopic findings ㆍ A prominent increase in perivascular lymphocytes ㆍ A variable increase in the amount of peribronchiolar lymphoid tissues ㆍ Frequently an inflammatory cell exudate within the alveolar spaces ㆍ Focal hyperplasia of alveolar type 2 cells Similar lung lesions were not observed in B6C3F1 mice concurrently on study with affected rats. Similar lung lesions were not observed in F344 rats at the end of 2-year NTP studies. Virus, mycoplasma, bacterial serology, bacterial culture, protozoal identification: negative EM: ㆍ No virus particles were identified. ㆍ Rod shaped bacteria were observed in the alveolar spaces. ㆍ Bacteria were not observed in the bronchi/ bronchioles of rats with alveolar organism. The present study examined the characteristics of behavior Induced by dopamine agonists following treatment with 6-hydroxydopamine(6-OHDA) unilaterally into left striatum in rats. 6-OHDA was administered at doses of 8,16 and 24 $\mu\textrm{g}$/$\mu\textrm{l}$(in 0.1% ascorbic acid) into dopaminergic neurons in left striatum of 7 weeks old rat under anesthetic. Locomotor activity was significantly decreased at 1 week following 6-OHDA-administration in 7 weeks old rats. The contralateral circling behavior was induced by apomorphine(5 mg/kg, i.v.) after 1 week following 6-OHDA(24$\mu\textrm{g}$/$\mu\textrm{l}$) treatment, and was further increased by repeated administration of apomorphine at 2, 3 and 4 weeks. The contralateral circling behavior was also induced by lisuride and 1-dopa in a dose dependent manner, but not by SK & F 82526 in 7 weeks old rats treated with 6-OHDA. The contralateral circling behavior was significantly higher in 21 weeks old rats but significantly lower In 35 weeks old rats when compared with 7 weeks old rats. The contralateral circling behavior induced by apomorphlne did not differ significantly in 7 and 35 weeks old male and female rats. These results suggest that 6-OHDA treatment into left striatum causes remarkable destrurtion of intrastriatal dopaminergic netcons leading to dopaminergic receptor supersensitivity. Thus, the contralateral circling behavior in duces by apomorphine may be used as indicator for neurodegenerative diseases.
Objectives: This study was performed to analyse the effects of Sweet Bee Venom(SBV-purified melittin supported by G&V Co., the major component of honey bee venom) on the respiratory system in rats. Methods: All experiments were conducted at Biotoxtech Company, a non-clinical studies authorized institution, under the regulations of Good Laboratory Practice(GLP). Male rats of 5 weeks old were chosen for this study and after confirming condition of rats was stable, SBV was administered in thigh muscle of rats in 0.175, 0.35 and 0.7 mg/kg dosage. And checked the effects of SBV on the respiratory system using the whole body plethysmography. And respiratory rate, tidal volume and minute volume of rats were checked after administered SBV (melittin). Results: 1. In the measurement of respiratory rate, there were not observed any significant differences compared with control group. 2. In the measurement of tidal volume, there was not observed any significant differences compared with control group. 3. In the measurement of minute volume, 0.35mg/kg dosage group showed significant differences compared with control group. But we estimated that this result was caused by individual differences. Conclusions: Above findings suggest that SBV seems to be safe treatment in the respiratory system of rats. And further studies on the subject should be conducted to yield more concrete evidences.
This study was conducted to investigate the effect of dietary supplementation of vitamin A on the membrane property and ultrastructure in ethanol-administered rat livers. Male Sprague-Dawley rats weighing of 130 ~150g were fed with experimental diets for 7 weeks. The diets contained different types of vitamin A which were $\beta$-carotene, retinyl acetate and retinoic acid. After feeding theexperimental diets for 7 weeks, a dose of 3.0g ethanol (30%, W/V)/kg B.W was injected to rats intraperitoneally. Control rats received 0.9% saline containing isocaloric sucrose instead of ethanol. Plasma membrane fluidity of liver decreased in rats fed with vitamin a -Deficient diet with ethanol as compared to that of control rats. Fluidity change of liver plasma membrane that ethanol had induced was influenced by dietary supplementation of vitamin A, but not influenced by the type of supplemented vitamin. A . The ultrastructural changed of hepatic mitrochondria were observed in some rats such as vitamin A-deficient rats with ethanol. Inadequate consumptionof vitamin A contributed to ultrastructural changes such as swelled mitochondria occurred by ethanol-induced hepatotoxicity. Although accurate mechanism involved in the plasma membrane-stabilizing effect of vitamin A is still unclear, dietary supplementation of vitamin A such as retinyl acetate is neede to modulate this change. The direct involvement of membrane property on the cell damage caused by ethanol treatment remains to be established.
This study was conducted to investigate the subacute intravenous toxicity of rHu-EPO in Sprague-Dawley rats. rHu-EPO was daily administered to male and female rats for 30 days with different dose levels(100, 500, 2,500 unit/ kg). After the administration period of 30 days, 5 males and 5females rats per each dose group were assigned for recovery period of additional 30 days. There were no clinical signs compared with control group but slight decrease in spontaneous motor activities and locomotions were observed at high dose groups of males and females. In males, feed consumption was reduced at 500 unit/kg and body weight gain was retarded. In the administration sites of tail, focal congestion was observed in a few animals treated with rHu-EPO. No deaths were occurred during the course of study. In hematological examination. a significant increase of hemoglobin and hematocrit was observed in the males and females rats receiving high dose of rHu-EPO. rHu-EPO administration for 30 days showed a marked decrease in glucose concentration. At the highest dose groups, there was a significant incerase in the weights of spleens in both sexes. but this was considered to be related to its pharmacological activity. These results indicate that rHu-EPO does not induce any significant toxic effect on Sprague-Dawley rats for 30 days.
The present study was designed to investigate the serum cholesterol lowering effect oj the phytosterol derivative (LPSS) on high cholesterol (HC) diet-induced hypercholesterolemia in male weaning Sprague-Dawley (SD) rats. Rats were fed with HC diet containing 1 % cholesterol and 0.5% cholic acid for 1 week. After 1 week, the LPSS oil suspension (0.32 g/kg B. W.) was orally administered to the rats fed with either basal diet or HC diet groups for 7 days. In addition, the LPSS powder (0.14%) mixed with basal diet or HC diet was Jed to the rats for 7 days. Serum total cholesterol and LDL-cholesterol contents were not altered by administration of the LPSS oil suspension with basal diet. However, they were significantly decreased by administration of the LPSS oil suspension with HC diet at day 14. Also, they were significantly decreased by the LPSS powder mixed with basal diet or HC diet at day 9, 11, 14. HDL-cholesterol contents were not altered by the LPSS oil suspension or LPSS powder. These results indicated that the phytosterol derivative(LPSS) might decrease serum total cholesterol and LDL-cholesterol contents in rats.
