A Study on a Morphological Identification of Sophora tonkinensis, Menispermum dauricum and Indigofera kirilowii (광두근(廣豆根), 북두근(北豆根) 및 화목람(花木藍)의 형태(形態) 감별에 관한 연구)

  • Objectives : A morphological classification among Sophora tonkinensis, Menispermum dauricum and Indigofera kirilowii was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. The cross-sections of Sophora tonkinensis and Indigofera kirilowii have piths, but Menispermum dauricum does not have any. 2. The lateral parts of Sophora tonkinensis cortex include the square crystal of calcium oxalate, but Indigofera kirilowii does not. Conclusion : The number of piths and the presence of the square crystal of calcium oxalate can be used to distinguished Sophora tonkinensis from Indigofera kirilowii.

  • Objectives : A morphological classification among Rheum palmatum Linne, Rheum undulatum Linne and Rumex crispus was made through microscopic observation. Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Results: 1. Stellate spots were found on the intersection of Rheum palmatum, composed of heteromorphy vascular bundles. 2. Stellate sponts were not found in Rheum undulatum and Rumex crispus, because they do not have heteromorphy vascular bundles. Conclusion : Rheum palmatum can be distinguished with Rheum undulatum and Rumex crispus, based on the presence of Stellate spots on its intersection.

  • Objectives : External form and internal intersection of Phragmites communis were observed by the parts of usage (leaves, stems, and roots). Methods : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Results : 1. The intersection of Phragmitis Folium was typically characterized as Gramineae plants, which was perfectly distinguished with Phragmitis Rhizoma and Phragmitis Caulis. 2. The cortex of Phragmitis Rhizoma has a large rectangular pore space, but Phragmitis Caulis does not. Conclusion : Phragmitis Folium, Phragmitis Caulis, and Phragmitis Rhizoma can be distinguished by microscope.

Degradation oof Triphenylmrthane Dyes by Citobacter sp. (Citrobacter sp.에 의한 Triphenylmethane계 색소의 분해)

  • The Optimal condition for degradation of crystal violet and other triphenylmethane dyes by Citrobacter sp. SK-3 isolated from the activated sludge of dye manufacturing factory was investigated. The optimal culture medium for the degradation of triphenylmethane dye was composed of minimum inorganic salt medium supplemented with 0.5% galactose, 0.1% beef extract, with the initial pH of 8.0 to 9.0. Under this condition, Citrobacter sp. SK-3 degraded 200 ppm of crystal violet completely within 24 hours. Citrobactre sp. SK-3 also degraded efficiently malachite green, pararosaniline, brilliant green, methyl violet, basic fuchsin and methyl red. Analysis of the degradation products of crystal violet through this layer chromatography and high performance liquid chromatography indicated that the methyl groups bound to crystal violet backborn were gradually demethylated to pentamethyl-, tetramethyl- and trimethylpararosaniline.

  • The phase of Cu,Zn,Al precursors strongly affects the activity of their final catalysts. Herein, the Cu,Zn,Al precursor was prepared by precipitation of $Al^{3+}$ onto primitive, amorphous Cu,Zn precipitate. This precursor turned out to be a phase mixture of zincian malachite and hydrotalcite in which the latter phase was less abundant compared to the co-precipitated precursor. The final catalyst derived from this precursor exhibited a little higher copper surface area and methanol synthesis activity than the co-precipitated counterpart. Therefore, the two precursor phases need to be mixed in an adequate proportion for the preparation of active $Cu/ZnO/Al_2O_3$ catalyst.

A Study on the Physical Properties of Seokrok and Noerok Used as Green Pigment (녹색안료로 사용되는 석록과 뇌록의 물리적 특성 연구)

  • The purpose of this study is to analyse the properties of Seokrok and Noerok that are used for restoration of heritage and arts. Malachite is main constituent mineral for Seokrok and Celadonite is Main component of Noerok. To evaluate the physical properties of pigment, A,B-class Seokrok and heated Seokrok that are sold in market were selected. To compare this results, Noerok sold in Japan were studied. In addition, we studied the pigments of Noerok. The heat treatment had no significant effect on the physical properties, except for the color-difference. The color-difference of Seokrok is larger than that of the Noerok sold in Japan. The $a^*$ values of Seokrok specimens are horizontal distribution, so it will expand the coloring ranges. The properties that are chromaticity, specific gravity and oil-absorption of Noerok are different from Seokrok. Noerok is suggested that achromatic color because the values of $a^*$ located near zero. Specific gravity of Noerok is smaller than Seokrok, but oil-absorption is larger twice. Noerok and Amnok, although ingredients are different, it is possible to use alternative because of similar physical characteristics. The result from this study expects to be used as useful referencing data for conservation and restoration of cultural heritage and understanding phenomena of the properties.

Study on the Manufacturing Technology of Mural Tomb in Goa-dong of Daegaya Period (대가야 고아동 고분벽화 제작기술에 관한 연구)

  • Rigorous analysis was performed to identify the structure and materials of the murals to study techniques used on mural tombs of ancient Daegaya era(6th century). The murals were painted by applying mortar on the walls and the ceiling after building a stone chamber and creating ground layers on mortar layers. Mud was applied on most of the mortar layers on four sides of the walls except the ceiling. Sand was not used in mortar but was made of materials with pure calcium substances. In addition, shells in irregular sizes with incomplete calcination were mixed; and the mortar's white powder was inferred as lime obtained by calcination of oyster shells. Kaolinite($Al_2Si_2O_5(OH)_4$) was used in the ground layer, Cinnabar(HgS) was used for red pigment, Malachite($Cu_2CO_3(OH)_2$) for green and Lead white($PbCO_3{\cdot}Pb(OH)_2$) for white. Mud plaster was applied on the mortar and was composed thinly and densely using clayey of particle size smaller than that of medium sand. It was assumed that the finishing was for repair after long time had passed since the mortar layer came off. Using lime made with oyster shells as mortar is unprecedented in ancient Korean mural tombs and its durability was very poor, suggesting that Gaya's mortar production technique was relatively behind compared to that of Koguryo's in the same era.

On Improvement of Garlic Productivity by Inactivation of Virus in Garlics (마늘 Virus 불활성화에 의한 생산성 향상에 관하여)

  • The effect of heat or chemotherapeutant treatment on the mosaic virus infected garlic(Allium sativum L.) scales and that of chemotherapeutant added to the culture medium were summarized as following. The treatment of the virus infected garlic scales at $37\~57^{\circ}C$ for 35 days to one hour in water or in air shelved no effort of inactivating the virus. Although treatment of the garlic scales at $62\~72^{\circ}C$ for 90 to five minutes reduced the mosaic symptom on the leaves of the garlic plants grown after the heat treatment, it reduced the growth vigor of the plants so greatly that complete inactivation of the virus in garlics was not feasible. The mosaic symptom on the leaves of garlic plant was reduced when the infected garlic scales were grown after 24 hours soaking in $10\~50\;ppm$ Malachite Green, 2,4-Dichlorophnoxy Acetic Acid, or in $20\~100\;ppm$ Quinhydron. These chemotherapeutants, however, inhibited the growth of garlic plant at the high concentration. Garlic scales soaked in $10\~50\;ppm$ Naphthyl Acetic Acid showed the least mosaic symptom without its complete extiction on the garlic leaves. When incorporated into the modified Murashige-Skoog's medium, $0.5\~l.5\;ppm$ Naphthyl Acetic Acid could inactivate the mosaic virus in newy developed garlic plants showing no mosaic symptom on the leaves, no inclusion bodies and intact nuclei in the leaf tissue cells.

Occurrence and Mineralogical Properties of Green-Blue Inorganic Pigments in Korea (국내 녹색-청색계열 무기안료의 산출과 광물학적 특성)

  • Traditional inorganic pigments applied to dancheong, buddhist painting, and wall painting were produced from natural minerals which were later replaced by synthetic pigments, resulting in the loss of the recipe to prepare mineral pigments. This study examined the domestic occurrence and mineralogical characteristics of green and blue mineral pigments required for the conservation of cultural heritage. Cuprous green-blue mineral pigments were found as the weathering products of waste dumps and ores of abandoned Cu-Pb-Zn sulfide mines. Mineralogical analyses using X-ray diffraction and scanning electron microscopy identified diverse hydrous copper sulfate pigments of green (brochantite and devilline) and blue color (linarite, bechererite, and schulenbergite) with minor green pigments of antlerite and atacamite commonly associated with cerussite, smithsonite, anglesite, and cuprite. Noerok, a green silicate pigment, replaced the fractured basalt lava. Celadonite was responsible for the green color of Noerok, closely associated with opal in varying ratio. Glauconite, green silicate pigment, was identified in the Yellow Sea sediments. Malachite and azurite, the most important green and blue pigments of Korean cultural heritage, were not identified in this study.

A Preliminary Survey Result of Cu Occurrence in Tsogttsetsii Area, Mongolia (몽골 촉트체치 지역 동 산출지 예비조사결과)

  • Tsogttsetsii area, an intrusive complex associated with Cu mineralization, is located in the South Gobi, Mongolia. We performed the cross geochemical and extended exploration survey in Tsogttsetsii area. Cu mineralization in Tsogttsetsii area is porphyry Cu type related with alkali granite intruded in Late Carboniferous to Early Permian. In the concentrated occurring to malachite appears extensively prophylitic alteration zone having a chlorite and epidote. As results of the survey, Cu contents of potable XRF and of chemical composition for altered rocks ranges 1.08 to 18.3% in the 30 points and 1.08 to 32.9% in the 13 points, respectively. Ore minerals identified in XRD analysis and polarizing microscope that samples of copper oxides were composed mainly of malachite, azurite, permingeatite and cuprite and the other minerals are pyrite, chalcopyrite, pyrargyrite, dickite, calcite, chlorite and epidote. Mineralization can be considered occurring to selectively some granite of the surrounding aplite and faults in the only upper part coming up the hydrothermal solution of the remaining residual magma after the aplite intrusion.
