쌀겨의 발효는 온도상승이 되면서 열을 많이 발생하여 온도가 60-70도로 상승한다. 이 발효에서 분리된 Bacillus coagulans는 효소온욕시 사용되는 주요 미생물로서, 이를 응용하여 피부미용건강에 사용되어질 수 있다. 쌀겨발효 중 온도유지에 관련이 있는 major microflora의 분리 연구를 수행한 결과, 고초균이라 판단되는 균주 10여종을 분리하였으며 이를 분자분류학적인 연구를 수행한 결과 Bacillus coagulans로동정하였다. 본 균주는 쌀겨발효에서 온도상승에 관여하는 주요 미생물로서 이의 피부미용에의 응용이 가능하리라 판단된다.
굴(Crassostrea virginica)과 Weakfish(Cynoscion regalix)를 냉장(6$^{\circ}C$), 빙장($0^{\circ}C$), 부분동결저장(-4$^{\circ}C$) 및 동결저장(-2$0^{\circ}C$)온도에서 45일간 저장하면서 생균수 와 microflora의 변화를 조사하였다. 각 온도에서 저온저장중 굴로부터 255주, Weakfish로부터 240주, 합계 495주를 분리하여 microflora의 변화를 조사하였다. 저장직전의 생선에서 생균수는 굴이 4.9$\times$10/ sup 5/CFU/g, Weakfish가 $1.5\times$$10^4$CFU/$cm^2$였다. 저장직전의 굴에서는 Pseudomonas ll1III/IV가 67%, Vibrio가 20%를 차지하였다. Weakfish에서는 Acinetobacter가 40% Moraxella가 33%로서 주종을 이루었으며 Pseudomonas와 Vibrio는 아주 적은 비율을 차지하였다. 굴의 저온저장중 microflora는 저장온도에 큰 관계 없이 모든 저장온도에서 Pseudomonas lIII/IV-H가 전체 균주의 67.4%, Flavobacterium/Cytophaga가 9.3%, 다음으로는 Vibrio가 6.3%를 차지하였고 약 15%의 세균은 동정하지 못하였다. Weakfish의 저온저장중 microflora는 냉장, 빙장, 부분동결저장한 경우에 비호염성균인 Pseudomonas III/IV-NH가 전체 균주의 60~100%를 차지하였으며 동결저장한 Weakfish에서는 Moraxella가 전체 분리 균주의 40~60%를 차지하였다. 전체적으로 굴에서는 호염성 균주(pseudomonas III/ IV-H와 Vibrio)가 우세하였고 구균류가 분리되지 않았으나 Weakfish에서는 비호염균주(PseudomonasIII/IV -NH와 Moraxella가 우세하였으며 구균류가 4.6% 검출되었다. 저온저장한 굴과 Weakfish에서 Vibrio의 검출률은 굴에서 Weakfish보다 3배 높았으며 Listeria spp. 는 검출되지 않았으나 강한 용혈작용을 가진 균들이 각각 9주, 8주씩 분리되어 주의가 요망된다.
The effects of Bifidobacterium longum HY8001 supplement intake on the fecal microflora and fecal bacterial enzyme activity were studied in ten healthy human volunteers, before, during and after intake (respectively for 3 weeks). During intake of B. longum HY8001 supplement, fecal, $\beta$-glucuronidase and nitroreductase activities significantly decreased 44.6%(p<0.005) and 32.3%(p<0.01), respectively. Although numbers of major bacterial groups of fecal microflora were not affected by B. longum HY8001 intake for 3 weeks, the number of Bifidobacterium was significantly increased (p<0.05). This result indicates that intake of B. longum HY8001 might be potentially beneficial for the prevention and inhibition of colon cancer and improvement of human intestinal microflora composition.
In order to study ecology of microorganisms during Takju brewing, microflora changes were examined fromm the start to the sixth day of Takju fermentation in 24 hours intervals. Takju made from rice, flour and dried sweet potato in a liter volume open container at the laboratory and a sanple of Takju brewing factory were studied for their microflora and their changes during fermentationl together with a sample of Kokja. Results obtained were as follows ; 1. The followings were the identified microorganisms in Kokja. The molds ; Absidia spinosa, Aspergillus parasiticus. The yeasts ; Candida melinii, Candida Solani, Hansenula anomala. The bacteria ; Luctobacillus casei, Leuconostoc mesenteroides, Bacillus subtilis, Bacillus pumilus. 2. Torulopsis inconspicua, Lactobacillus casei, Leuconotoc mesenteroides, Bacillus subtilis, Bacillus pumilus were isolated from main mash of laboratory-made Takju samples. The yeast, Torupsis inconspicua which was not present in Kokja and, probably of a contaminant yeast, dominated the yeast flora of Takju mash of rice, flour and sweet potato of labotatory brewing. The laboratory brewing lost also always showed large population of lactic acid bacteria flora. 3. None of the wild yeasts which were present in Kokja appeared in Takju mashes. The Kokja appears to be of no use as the yeast source for Takju fermentation. Also the Kokja appears to be of not so effective amylolytic and proteolytic enzyme sources considering the microflora characteristics. Probably the major role of Kokja in Takju fermentation may be to contribute in taste formation. 4. Inoculation of Sacharomyces cerevisiae into the mash to the level of $10^7$ ml at the start of fermentation greatly changed the ecological aspects eliminating conditions of rather slow rising of natural contaminant yeast populaiton and fermentation which might give rise to prosperity of lactic acid and Bacillus bacteria that would be avoidable. 5. Examination of microflora of the large factory scale Takju fermentation showed the quite similar pattern of microflora and their changes to that of the cultured yeast-inoculated laboratory batch Takju fermentation. The cultured yeast dominated as the only predominant microflora, and the lactic acid bacteria flora were completely suppressed and aerobic bacteria, greatly. Probably this may be the regular microflora pattern of normal Takju fermentation. The role of lactic acid bacteria and aerobic bacteria in Takju fermentation may not be clear yet from this experiment alone.
한국인의 장내세균중의 유해효소생성 균주를 분리하여 장내환경 개선연구에 활용하고자 ${\beta}-glucuronidase$와 $7{\alpha}-dehydroxylase$의 유해효소를 분비하는 장내세균으로 알려진 Clostridium속 균주를 분리하였다. 동일인의 장내세균을 2회에 걸쳐서 검색한 후 최다 우점종이외의 장내세균은 동일인이라도 식이등 외부환경에 의하여 변화될 수 있음을 알았다. 이어서 Neomycin-Nagler 선택배지를 사용하여 Clostridium spp.를 14종 분리하여 이들 균주가 유해효소를 생산하는지를 조사하였다. 일차 선발균주중 Clostridium sp. Isolate-11은 ${\beta}-glucuronidase$를 강력히 생성하여 0.021 unit/mg protein의 활성을 보여주었으나 이들 분리 균주중에서 $7{\alpha}-dehydroxylase$의 역가를 TLC상에서 검출할 수 있는 균주는 발견하지 못했다. Isolate-11를 배양생리학적으로 동정한 결과 이 균은 Clostridium scatologenes인 것으로 잠정동정하였다.
수침조건에 따라 수침찹쌀과 수침액의 미생물학적 변화와 효소활성 및 구조적 특성을 살펴본 결과 수침액의 미생물상에서, 수침초기에는 젖산균이 우세하였고, 수침기간이 길어짐에 따라 미생물 균수가 급속히 증가하면서 젖산균과 Bacillus spp.이 우세 균총으로 관찰되다가 수침이 계속되면서 모든 미생물이 감소하는 경향을 보였으며 그 중에서 Bacillus spp.이 가장 많이 관찰되었다. 수침액의 $\alpha$-amylase는 수침온도와 수침기간이 증가함에 따라 지속적으로 활성이 증가하였고, protease 활성은 수침기간이 길어짐에 따라 활성이 증가한 후 감소하는 경향을 보였다. 수침 찹쌀의 $\alpha$-amylase는 수침온도가 높을수록 감소하였고, $\beta$-amylase와 glucoamylase는 수침 1일에 증가하여 수침 21일까지 일정하였다. 수침찹쌀의 미세구조를 살펴보면, 세포벽이 떨어져 나간 흔적이 보이고 다각형의 찹쌀전분입자들이 노출되면서 표면이 거칠어 보였다. 또한 찹쌀전분 입자 표면의 미세한 구멍이 수침 11일 이후 관찰되어 효소작용의 결과와 일치하는 경향을 나타냈다.
Fecal microflora of 12 breast-fed(BF) and 15 formula-fed(FF) infants were investigated at 1 year after birth and the results were compared to those that had previously been obtained from the same subjects at 1 week after birth, and before and after weaning. At 1 year the two lactation groups showed no significant differences both in frequencies and numbers of each major bacterical species, except eubacteria, which showed higher frequency in FF infants. Bifidobacteria appeared as dominant species in 50% BF infants, heres bacteroides in 73.3% FF infants. Fecal pH was lower, though insignificant, in BF infants(6.5$\pm$0.4) than in FF infants(6.8$\pm$0.5). In BF infants, the number of bifidobacteria, clostridia, and E. coli deceased from birth up to 1 year, whereas the number of bacteroides similar trends to BF infants except that the number of bifidobacteria increased, but insignificantly. This research showed that the floral differences resulted not from the age, but from the types of feeding and that breast feeding and that breast feeding could be better than formula feeding in the BF infants had more stable floral composition and bowels with lower pH, which can play a protection role against pathogen infection.
As a result of screening the medicinal herbs which selectively control human intestinal microflora, water extract of Akebia quinata Decaisne was proved to have a strong inhibitory activity against the growth of Clostridium pefringens, a major harmful intestinal bacterium. The anti-bacte-rial activity was stable under the thermal treatment at 100$\circ$C for 120 min and in a range of pH 1 to 11. In addition, the water extract of Akebia quinata Decaisne showed the antibacterial activities against five different strains of Clostridia including C. perfringens. On the contrary, the extract did not inhibit the growths of Bifidobacterium, Lactobacillus, Escherichia coli, Enterococcus faecalis. The extract, however, suppressed markedly the growth of Bacteroides fragilis and Staphylococcus aureus in vitro. Alike in the mixed culture inoculated with human feces as starter, in vivo tests using rats showed that the extract tends to increase the numbers of Bifidobacteria and Lactobacilli in the intestinal microflora of rats, whereas those of Clostridia were attenuated.
As a basic study for quality improvement of Korean soybean paste and soybean sauce, we investigated on microflora of soybean paste and soybean sauce fermentation. Major Microorganisms were isolated from the sample pastes and sauces, and identified systematically. Selected Microorganisms were identified by MIS whole cell fatty acid analysis by gas chromatography. Identification results showed that Bacillus licheniformis, bacillus pumilis and Bacillus subtilis were dominant in soybean paste and Staphylococcus vitulus, Bacillus subtilis, Bacillus pumilus, and Lactobacillus fermentum were dominant in soybean sauce. It seemed that these Microorganisms played an important role in soybean paste and soybean sauce fermentation and could be used for the further studies such as protease and amylase activities.
Major hazards existed in slaughterhouse are pathogenic microorganisms originated from intestinal microflora of slaughtered animals. This study was intended for the identification of microbial contamination sources during pork slaughtering in small plants. Total aerobic bacteria, Coliform group, Salmonella spp, Listeria monocytogenes, and Campylobacter jejuni/coli were isolated from the surface sample of pork carcasses. Contamination level among different sampling points of ham, belly and neck did not showed statistical differences. Therefore, the mixed sampling from belly and neck of carcass could be effective for microbiological monitoring. Isolation rates of pathogenic microorganisms showed Salmonella spp 20.9%, Listeria monocytogenes 10.5%, and Campylobacter jejuni/coli 8.1% from 296 sampling points. High prevalence rate of Salmonella spp indicated that the contamination of intestinal microflora occurred due to unsanitary processing control, which required HACCP system in small plants. It was recommended that the prerequisite program should be a key factor for a successful HACCP system implementation especially in small size slaughterhouse.
