Cytological Modification of Sorghum Leaf Tissues Showing the Early Acute Response to Maize Dwrf Mosaic Virus

  • Sorghum leaf tissues showing the early acute response of systemic infection with maize dwarf mosaic virus (MDMV) strain A, contained unusual virus-induced cytological modifications including cell wall thickenings and protrusions, intercellular vesicles termed as "paramural bodies", modified plasmodesmata, abnormal plastids, and cylindrical inclusion bodies. Abnormal cell wall, some of which associated with paramural bodies, was frequently contained modified plasmodesmata. Various abnormal plastids were located within infected cells of leaf tissues showing the early acute response. The most important changes in chloroplast seen in the tissues are the presence of small vesicles, deformation of membranes, reduction in granal stack height, disappearance of osmiophilic globules and degeneration of stuctures. The cytological modification was not occurred in nucleus but a group of degenerated mitochondria with abnormal membranes attached to cylindrical inclusion bodies were observed. It was hard more or less to prove the relationship clearly between virus and cellular organelles in virus replication.plication.

Comparison of Cytoplasmic Inclusions Induced by Maize Dwarf Mosaic Virus Strain A and B (Maize Dwarf Mosaic Virus strain A와 B에 의해 유도된 세포질 봉입체의 비교분석)

  • Comparative ultrastructural studies of sorghum (Sorghum bicolor L. Moench) cultivar (cv.) HOK and cv Pioneer 8680 leaf cells separately infected with maize dwarf mosaic virus (MDMV) strain A and B, respectively, revealed the formation of numerous cylindrical inclusions in both cross and longitudinal sections. The MDMV-A and -B were distinguished by the presence or absence of specific inclusion bodies in the cytoplasm. Electron microscope revealed the presence of pinwheels, bundles, scrolls, and laminated aggregates in Pioneer 8680 sorghum leaf cells infected with MDMV-B while no laminated aggregates were found in cells of HOK sorghum leaf cells infected with MDMV-A. Differences in the ultrastructure of cylindrical inclusions between two strains of MDMV, especially with respect to laminated aggregates, have been morphologically indexed to classify potyviruses into subdivision. The presence of laminated aggregates may be assigned to subdivision III while the absence of laminated aggregates is assigned to subdivision I. These variations of structures associated with cylindrical inclusions appeared virus-specific inductions and may be represented the morphogenesis of inclusion bodies following development of infections.

Resistance of Corn to Maize Dwarf Mosaic Virus: Genetic Analysis by Diallel Cross (옥수수의 위축모자이크바이러스에 대한 저항성: 이면교배에 의한 유전분석)

  • The studies were conducted to test for combining ability and to evaluate resistance to maize dwarf mosaic virus (MDMV) by diallel crosses of corn inbreds. For the genetic analysis of the resistance, a diallel set of crosses without reciprocals was made using the eight corn inbreds which had different degrees of resistance to MDMV. Twenty eight $F_1$ hybrids showed different symtom severity. the highest value is 3.63 and the lowest value is 1.87 in disease ratings (1-4). General combining ability (GCA) for resistance to MDMV was highly significant, but specific combining ability (SCA) was not significant. Two inbreds, A632 and KS15 showed negative GCA effects, indicating that these parents were good general combiners and that resistance to MDMV increased in hybrid combinations. Hyrid A632 x KS5, showed the highest negative SCA effect and several combinations showed negative SCA effects. The analysis of parent-offspring covariance (Wr) and array variance (Vr), suggest that there may be many dominant genes in the resistant inbreds and many recessive genes in the susceptible inbreds.
