Effects of Horsetail, Alfalfa, Ortie, Chêne and Aleppo oak as Potential Hemostatic Agents on Laboratory Coagulation Tests

  • This study investigated the effect of ethanol extracts of horsetail, alfalfa, ortie, chêne and aleppo oak on blood coagulation in vitro. Extraction was performed by the maceration method. Extracts were mixed with platelet and plasma, then prothrombin time (PT), activated partial thromboplastin time (APTT) and platelet aggregation tests were conducted. Alfalfa extract had a dose-dependent effect on the PT. Ortie, and horsetail, reduced the PT significantly compared to control group. Alfalfa, horsetail, and ortie reduced the APTT, but their effect was insignificant compared to the control group. The pooled extract showed the highest effect compared to the single extracts in a dose-dependent manner. Horsetail and alfalfa induced platelet aggregation in response to arachidonic acid but not in response to collagen. In the case of ortie, no aggregation occurred regarding the arachidonic acid, and incomplete was observed in response to collagen. Interestingly, blood clotting occurred immediately after adding the chêne, aleppo oak and the pooled extract, and therefore platelet poor plasma (PPP) and platelet rich plasma (PRP) became jelly. Generally, chêne and aleppo oak, as well as pooled extract, were more effective in inducing both primary and secondary coagulation pathways via shortening the PT and APTT, and induction of platelet aggregation.

  • This study aimed to explore the biodiversity of chemical compounds found in the bark of Guibourtia tessmannii from Gabon, commonly called Kévazingo, and evaluate their anti-termite activity to determine their potential values as a source of development of anti-termite products that can be valued in the fields of fine chemicals and wood preservation. Extraction of G. tessmannii bark powders was carried out using the cold maceration method with trichloroethylene, acetone, ethanol, and water. Phytochemical screening made it possible to highlight groups of chemical families present in the extracts. Anti-termite activity was tested on the wild termites "Cubitermes sp" of the genus Isoptera. The yield of the extracts were 17.11% for the buttress and 13.42% for the height at 6 m. Phytochemical tests revealed that alkaloids, polyphenols, sterols, tannins, reducing compounds, flavonoids, saponins, and anthraquinones were present in the extracts. Results of anti-termite activity indicated that anti-termite activity varied with the different parts of the bark studied, extraction solvent, and concentration (50/50) and (25/75) of the extracts used. The extracts at 50/50 concentration showed a slightly better anti-termite activity compared to the 25/75 concentration. In addition, the buttress Kévazingo or buttress showed the strongest anti-termite activity for the aqueous extract with a survival rate of 0% after 2 days.

  • This study aimed to analyze the anti-termite properties of tamanu (Calophyllum inophyllum L.) stem bark extracts against subterranean termites, specifically, Coptotermes curvignathus. The bark powder of C. inophyllum was extracted using different solvents, such as n-hexane, ethyl acetate, and methanol, using the maceration method. Anti-termite testing was performed using two paper disc methods: no- and two-choice tests. Whatman test paper was dripped with the extract solutions at concentrations of 4%, 6%, 8%, and 10% (w/v). Subsequently, the treated paper disc was placed into an acrylic tube, and the subterranean termite was added. The parameters utilized in the test included termite mortality and the weight loss of the test paper. The results revealed that the total extract yield of C. inophyllum stem bark was 30.24%. Furthermore, the extractive substances from C. inophyllum bark exhibited anti-termite activity. The most favorable outcomes were obtained with the n-hexane and ethyl acetate extracts at a concentration of 10%. The termite mortality and weight loss of the test paper were respectively 66% and 5.67% for the n-hexane extract and 66.67% and 6.19% for the ethyl acetate extract. In addition, the n-hexane extract contained friedelan-3-one, while the ethyl acetate extract contained 1,2-benzene dicarboxylic acid, dinonyl ester, and friedelan-3-one. The results suggested that these compounds are responsible for the observed anti-termite activity.

Studies on Brewing of Apple Wine -Apple wine containing lower concentration of alcohol- (사과주(果實酒)양조(釀造)에 관(關)한 연구(硏究) -저농도주정함유(低濃度酒精含有) 사과주에 관(關)하여-)

  • This study aims to brew apple wine containing lower concentration of alcohol by fermentation and to retain $CO_2$ gas in apple wine, and investigation for the possibility of storage at room temperature was performed. A Saccharomyces sp. was proved to be acceptable for production of base wine as its higher fermentation rate at $20{\sim}25^{\circ}C$. However, B-2 was most reasonable for post-fermentation of apple wine as this strain strongly ferments sugars at low temperature $(4^{\circ}C)$. The yield of apple juice increased by maceration of apple pulps. The yield was about 5 % more than that of the unmacerated juice, whereas acid content was decreased by 10% compared with control. When stored apple wine containing 9% alcobol was introduced $1{\sim}3%$ sucrose at $7{\sim}8^{\circ}C$ for 100 days or more, the $CO_2$ pressure of apple wine in bottle shows $3kg/cm^2$ by bottle-pressure meter. It showed good storage of the wine at room temperature. $CO_2$ gas pressure in apple wine containing 6% alcohol, $5{\sim}10%$ hop extract, and 2% sugar was $2kg/cm^2$, he result also showed possibility of storage. Whereas 6% concentration of alcoholic apple wine without hop extract caused unusual fermentation during storage at the same condition. The desirable conditions for high quality apple wine should have $CO_2$ pressure of $2kg/cm^2$ or more and should be added $1{\sim}2% sugar to base wine. From these results, it can be concluded that the brewing of lower alcoholic apple wine is possible.

Ecological and Morphological Characteristics of Zostera caulescens Miki (Zosteraceae) in Korea (한국산 수거머리말(Zostera caulescens Miki.)의 형태 및 생태적 특성에 대한 연구)

  • Ecological characteristics on habitats and morphological features of the seagrass Zostera caulescens Miki in Korea were examined. The biogeographical distribution of Z. caulescens was confirmed in the south coast of the Korea. Zostera caulescens usually inhabits at the inner bay, where is sheltered from wave action and 3.0 to 6.5 m deep. The sediment in habitats is composed of very fine muddy sand or sandy mud. In morphology, Z. caulescens is easily distinguished from other species of the genus by the formation of vegetative canopy on the top of reproductive stems. The number of longitudinal ribs in testa was 24 revealed by scanning electron microscope (SEM) while the number of ribs in Z. marina has been often reported to be 16-20. Purplish anthocyanin spots were displayed on the surface when the testa was removed. We found that the size and shape of leaf epidermal cells in Z. caulescens were very different from those of Z. marina when the leaf cuticles were removed by maceration which could be useful characters for identifying Zostera species. The leaf of Z. caulescens displays two different casts of color when the fresh plant is closely observed. The margins of leaves appear brighter than the center of leaves due to the thickening by the development of paralleled venation in the middle of leaves. The comparison of two populations of Z. caulescens in Korea showed that they were considerably different in their shoot density and biomass. The shoot density and biomass at Gabae population was $367.3 m^{-2}\;and\;725.7g$ dry wt $m^P{-2}$ respectively, while those at Jukrim population were $112.5m^{-2}\;and\;392.0g\;dry\;wt\;m^{-2}$, respectively.

성스테로이드 호르몬과 해산 태생 경골어류의 성분화

  • 난생 경골어류의 성분화는 내인성 및 외인성 성스테로이드 호르몬의 영향을 받는 것으로 알려져 있지만, 해산 태생 경골어류의 성분화 과정에서 성스테로이드 호르몬의 작용은 아직 명확하게 밝혀져 있지 않다. 본 연구에서는 유용 양식어종의 하나인 해산 태생어류 조피볼락 Sebastes schlegeli를 대상으로 이 종의 성분화에 미치는 성스테로이드 호르몬의 영향을 조사하였다. 실험에 사용한 조피볼락은 출산후 일령 45일 전후의 성이 미분화한 개체들이었다. 실험어는 3개 실험군으로 나누어 (각각 3반복), 1개월간 각각 estradiol-l7${\beta}$ 또는 methyltestosterone을 처리하였고, 하나의 실험군은 호르몬 처리를 하지 않았다 처리기간 동안에는 각 실험군으로부터 무작위 sampling을 실시하여, 호르몬 처리에 따른 체내 성스테로이드 호르몬 변화를 조사하였으며, 각 실험군의생식소 분화과정을 조직학적으로 조사하였다. 성스테로이드 호르몬은 diethylether로 추출한후, rndioirmmunoassay 방법 (RIA)으로 분석하였다. 실험결과 대조구의 실험어는 1:1의 자연성비에 부합하는 성분화 결과를 나타내었지만, estradiol-17${\beta}$ 처리구는 자성화, methyltestosterone 처리구는 웅성화하는 경향을 나타내었으며, 이러한 성분화 shift는 외부에서 투여한 호르몬이 실재 내부호르몬양의 변화를 초래하여 이루어졌음이 RIA 분석에 의해 부분적으로 확인되었다. 본 연구의 결과는 해산 태생 경골어류의 성분화 역시 성스테로이 드호르몬의 작용에 의하여 매개 또는 조절되어진다는 사실을 제시한다. 하지만 태생어류에서 이 호르몬의 실질적 작용 메카니즘 및 명확한 작용시기에 대해서는 보다 많은 연구가 요구되어 진다.을 설계하는 것이 가능하였다.적(最適) 온도(溫度)는 $30^{\circ}C$, avicelase와 ${\beta}-glucosidase$의 최적(最適) pH는 5.0, CMCase는 pH 5.5 이었으며, 균사(菌絲) 생육(生育)은 pH 5.0에서 양호(良好)하였다. 배양(培養) 기간(期間)은 avicelase가 8일(日), CMCase가 10일(日), ${\beta}-glucosidase$는 16 일간(日間) 배양(培養)하였을 때 최대치(最大値)를 보였고, 균사(菌絲) 생육(生育)은 12일(日) 배양(培養)했을 때 가장 양호(良好)하였다.가한 반면, 중국인들은 고소한 향의 강도, 고소한 향의 기호도, 전체적인 맛에서 뚜렷하게 일본참기름을 우수하게 평가하였다.s의 항체(抗體)로 반응(反應)시킨 후 protein-A gold(15 nm)로 표식(標識)시킨 바 제일 바깥 상층(上層)의 keratinocyte에 있어서 세포막표면(細胞膜表面)을 따라 표식(標識)되어 세포막항원(細胞膜抗元)을 나타내었으며, 이와 같은 소견(所見)으로 미루어 정상피부(正常皮膚) 중층편평상피세포(重層扁平上皮細胞)에서도 동일(同一)한 소견(所見)을 관찰(觀察)할 수 있다고 본다.al remnants, Resorption of fetus로 관찰된 것이다.   • This study was carried out to enhance the stability and activity of polygalacturonase (PGase) purified from Kluyveromyces marxianus IFO 0288. Gums such as xanthan gum, guar gum, and locust bean gum were capable of increasing the catalytic stability and activity of the PGase. At $30^{\circ}C$, the rate constants for the inactivation of the PGase with xanthan gum, guar gum, and locust bean gum were estimated to be $0.0003min^{-1}$, below $0.0001min^{-1},\;and\;0.0001min^{-1}$ respectively, whereas control was estimated to be $0.0082min^{-1}$. The yield of the maceration reaction catalyzed by the PGase for the production of carrot single cells increased by 13% in the presence of guar gum, where the relative enzyme activity supplemented with guar gum was two-fold greater than that of the PGase alone.

  • 최근 들어 국내에서도 홍수에 의한 재산과 인명피해를 최소화하기 위해 지역특성에 알맞은 홍수지도를 제작 중에 있으며, 주로 LiDAR를 이용하여 홍수지도 지형자료를 구축하고 있다. 그러나 이에 많은 시간과 비용이 소요되기 때문에 이미 전국적으로 구축된 수치지형도를 이용한 홍수지도 지형자료 구축의 가능성과 타당성을 검증의 필요성이 제기되고 있다. 이에 따라 본 연구에서는 수치지형도를 이용한 홍수지도제작시 가장 핵심이 되는 정확한 지형자료구축을 위해서 Breakline처리기법과 하천종횡단자료 연계방안을 중심으로 구리시 일부지역을 대상으로 DEM을 시범제작 하였다. 수치지형도와 LiDAR를 이용하여 구축한 DEM을 검수점을 통한 RMSE오차를 이용하여 수직위치정확도를 비교분석하였다. 분석을 위하여 선행연구(홍수지도제작, 2002)에서 검수점으로 사용한 9점과 제방부분의 실측한 9점을 본 연구의 검수점으로 사용하였다. 비교 결과 RMSE가 수치지형도 DEM의 경우 ${\pm}2.658m$이고, LiDAR DEM의 경우 ${\pm}3.430m$로 나타났다. 두 자료 모두 알려진 오차범위보다 훨씬 높은 오차가 발생함을 알 수 있었다. 향후 수치지형도를 이용한 효율적인 홍수지도 지형자료 구축을 위해서는 수치지형도의 신속하고 지속적인 갱신이 필요할 것이며, 현재 도시지역에만 구축되어 있는 1:1,000 수치지형도를 주변 하천지역까지 확대해서 구축해야 할 필요성이 있을 것으로 사료된다. 아울러 도입초기단계의 LiDAR 측량기술의 보급과 관련모델의 기술의 발달로 인하여 정확도 높은 DEM이 요구된다면 고밀도의 방대한 양의 LiDAR 자료로 DEM을 구축하는 것이 바람직하다고 사료된다.및 이용대책을 수립하는 데 활용될 수 있다., $1,000-2,000{\mu}g\;g^{-1}$), 나머지 177종 식물은 살초활성을 나타내지 않았다. 높은 살초활성을 나타낸 식물로부터 분리되는 천연활성물질들은 향후 새로운 제초제 개발을 위한 모화합물 제공, 상호대립억제 작용성을 갖는 작물 품종 개발 혹은 개량에의 활용, 그리고 친환경 유기농업용 잡초방제에의 활용이 기대된다.내(皮膚上皮組織內)의 Langerhans cell내(內)의 Birbeck granule에 비(比)해 수적(數的)으로 현저히 감소(減少)되어 있었다. 그러나 Thy-1 양성(陽性) dendritic cell에서 볼 수 있는 dense-core 과립(顆粒)은 별변화(別變化)없이 쉽게 관찰(觀察)될 수 있었다. 조직배양(組織培養)을 한 견(犬)의 keratinocyte에 대(對)해 사람 pemphigus vulgaris의 항체(抗體)로 반응(反應)시킨 후 protein-A gold(15 nm)로 표식(標識)시킨 바 제일 바깥 상층(上層)의 keratinocyte에 있어서 세포막표면(細胞膜表面)을 따라 표식(標識)되어 세포막항원(細胞膜抗元)을 나타내었으며, 이와 같은 소견(所見)으로 미루어 정상피부(正常皮膚) 중층편평상피세포(重層扁平上皮細胞)에서도 동일(同一)한 소견(所見)을 관찰(觀察)할 수 있다고 본다.al remnants, Resorption of fetus로 관찰된 것이다. STUDIES ON THE TISSUE CULTURE OF PANAX GINSENG

  • Unlike the tissue culture in animals and human being, in higher plants various parts of the plant are cultured for varied purposes, and they are named variously depending on which parts are used as explants or what purposes they are cultured for. Followings are some of the names of culture used frequently: organ culture, tissue culture, callus culture, single cell culture, meristem culture, mericlone culture, ovary culture, ovule culture, embryo culture, endosperm culture, anther culture, pollen culture, protoplast culture, etc.. As the names of the culture indicate, in some kinds of culture the explants used for culture are actually not tissues, but organs, single cells, or protoplasts. It seems, however, convenient to call all of the above-mentioned cultures grossly as tissue culture. Several kinds of tissue culture were attempted using Panax ginseng as material and some of the results were summarized below. 1. Callus culture After dormancy of the sed was broken, whole embryo or parts (hypocotyl, cotyledon and epicotyl) of partly grown embryo were cultured in the media supplemented with growth regulators. Rapid swelling occurred in a few weeks, but most of the swelling was observed only in the basal part of epicotyl, changes in the other parts of embryo appearing in much later stages. The swelling or increase in size, however, was resulted not from the divisions of cells, but from the mere expansion of cell. Real calli were formed about two months after inoculation of explants. Callus tissues developed from cortex, pith, and vascular bundle in the cases of hypo- and epicotyl, from mesophyl tissue in the case of cotyledon. Shoots developed more easily from cotyledons regardless of whether they are detached from or attached to the embryo proper. 2. Culture in the Knudson C medium When cotyledons, detached from or attached to the embryo proper, were cultured in the growth regulator-free Knudson C medium comprision only several kinds of mineral compounds and sucrose, shoot primordium or callus developed profusely and finally plantlets were produced directly from shoot primordium or indirectly through callus. In this medium epidermal cells as well as mesophyl cells of the cotyledon became meristematic and divided, changing into multinucleate cells or multicellular bodies, developing eventually into either shoot primordia or calli. 3. Anther culture Anthers were cultured in the media supplemented with various growth regulators applied singly or in combinations. Callus was formed mostly in the connective tissue of anther. Cells of anther wall layers changed in appearance, but no division occurred. Microspores of all stages in development were not changed, ruling out the possibility that microspore-originated callus might be formed. 4. Isolation of protoplast Protoplasts were isolated from young root, leaf, and epicotyl, using 0.7M D-mannitols as osmoticum and using macerozyme and cellulase respectively for maceration and digestion of the cell wall. Production in large number of naked intact protoplast was rather difficult as compared with other plant species. Fusion of protoplasts occurred infrequently mainly due to the fewer number of naked protoplasts in the solution.

Determination of Microbial Community as an Indicator of Kimchi Fermentation (김치발효의 지표로서 미생물군집의 측정)

  • Attempts were made to define the characteristics of microbial community as an indicator of Kimchi fermentation. Determination of communities was carried out by simple Gram-stain, followed by direct microcopic counts. In room-temperature $(15^{\circ}C)$ fermentation, microbial succession was occurred in the order of communities of Gram-positive bacteria, yeasts and Gram-negative bacteria. It was characteristic that Gram-positive bacterial community was developed during the production of lactic acid, yeasts community was developed to cause rancidity, and Gram-negative bacterial community was relevant to maceration (or softening) as well as rancidity. The fluctuation of apparent Gram-negative reaction group might be used as a criterion of death or aging of Gram-positive bacterial populations. In low-temperature fermentation $(5^{\circ}C)$, however, it was found that yeasts and Gram-negative bacterial communities did not developed but only Gram-positive bacterial community did. It follows from these results mentioned above that maturity of Kimchi depends on the development of Cram-positive bacterial community. Thus, the size and occurrence of microbial community are avaiable for an indicator of Kimchi fermentation, and also determination of community could be a useful method to predict the maturity.
