Objective: Pullulan derivatives (PD) can be used to make self-assembled hydrogel nanoparticles which are responsive to ionic strength. The aim of this study is to evaluate the potential of PD as a retaining carrier of radioisotope inside tumors. Materials and Methods: Four types of PD were evaluated which included pullulan acetate (PA), succinylated PA (SPA), PA-DTPA and SPA-DTPA conjugates. They were radiolabeled with Tc-99m. Labelling efficiencies were determined at 30 min, 1, 2, 4 and 12 hours after radiolabeling. CT-25 colon cancer cells were subcutaneously injected into Balb/c mice. After 2 weeks of subcutaneous injection, Tc-99m-labelled PD (Tc-99m-PD) were injected into the tumors. Whole body images of mice were obtained at 30 min, 1, 2, and 12 hr after intratumoral injection. All twenty mice were grouped into four groups by largest diameter; control A (largest diameter = 5 mm, n = 5), control B (largest diameter = 10 mm, n = 5), pullulan A (largest diameter = 5 mm, n = 5), pllulan B (largest diameter = 10 mm, n = 5). Dynamic images were obtained for 1 hour after intratumoral injection. Static images were obtained at 1 hr, 2 hr, 3 hr and 4 hr after intratumoral injection with Tc-99m pertechnetate and Tc-99m-PA. Target-to-background ratios and retention rates were calculated. Results: Labeling efficiencies of PA, SPA, PA-DTPA and SPA-DTPA were $94.5{\pm}5.9%,\;97.8{\pm}3.5%\;94.2{\pm}3.8%,\;and\;92.5{\pm}6.2%$, respectively (p>0.05). Percent retention rates (%RR) of PA and PA-DTPA were significantly higher than those of control, however, those of SP-DTPA and SPA became similar to control at 4 and 12 hr, respectively. %RR of pullulan A and pullulan B at 1, 4 and 8 hr is significantly higher than that of control (p < 0.05). However, %RR between pullulan A and pullulan B were similar. Conclusion: The lonic strength dependent PD-nanoparticles are retained in the tumor. No difference of %RR according to tumor size was noted. Therapeutic application of PD labelled with beta- or alpha- emitting radionuclides can be expected.
Water quality parameters were surveyed in Lake Andong. Turbidity, temperature, secchi disc transparency (SD), phosphorus, nitrogen and chlorophyll a concentration were measured at dam site from July 1993 to December 1998. Minimum transparency in summer was only about 2 meters in 1993 and 1994, but it decreased to about 1 meter in 1997 and 1988. Total phosphorus concentration of the epilimnion increased slightly from $11{\sim}30\;mgP/m^3$ in 1993 to $18{\sim}42\;mgP/m^3$ in 1998. Total nitrogen concentration of the epilimnion decreased slightly from $1.81{\sim}2.96\;mgN/L$ in 1993 to $1.48{\sim}2.57\;mgN/L^3$ in 1998. TN/TP weight ratio decreased from $82{\sim}281$ in 1993 to $21{\sim}143$ in 1998 due to the increase of phosphorus concentration and the decrease of nitrogen concentration. Dissolved inorganic phosphorus concentration and the decrease of nitrogen concentration. Dissolved inorganic phosphorus and nitrate nitrogen concen tration of the epilimnion were in the range of $0.9{\sim}5.3\;mgP/m^3$ and $1.36{\sim}1.68\;mgN/L$, respectively. Chlorophyll a concentration in summer was in the range of $11.0{\sim}19.1\;mg/m^3$ in 1994, 1996 and 1997, but it decreased to $2.3{\sim}6.5\;mg/m^3$ in 1998. Trophic state of Lake Andong can be classified as mesotrophic to eutrophic from TP, TN and chlorophyll a concentration.
Background Radioiodine ($^{131}I$), a major component of nuclear fallout and a valuable therapeutic agent for thyrotoxicosis and thyroid cancer, has been regarded as a mutagen or a carcinogen without any convincing evidence. To evaluate the genotoxicity of radioiodine ($^{131}I$) we performed a micronuclei test in mice bone marrow. Materials and methods : Mice (ICR strain, $25{\sim}30 g$) were divided to 4 groups: control, group 1 (0.17 mCi/kg, usual therapeutic dose for thyrotoxicosis), group 2 (1.67 mCi/kg, usual therapeutic dose for thyroid cancer), and group 3 (16.67 mCi/kg, usual accumulated dose causing bone marrow suppression). $^{131}I$ was administered intraperitoneally. Ten mice of each group were sacrificed at days 1 and 3. Bone marrow were smeared and stained with May-Grunwald Giemsa method. One thou-sand polychromatic erythrocytes (PCE) and normochromatic erythrocytes (NCE) were counted under the light microscope, and the number of micronucleated PCEs were recorded. Results : The frequency of micronuclei in PCE (and NCE in parenthesis) in the control group was $0.25{\pm}0.07$ ($0.23{\pm}0.07$)% in day 1 and $0.24{\pm}0.07$ ($0.21{\pm}0.07$)% in day 3. Those in group 1 was $0.27{\pm}0.1$ ($0.23{\pm}0.09$)% in day 1 and $0.28{\pm}0.07$ ($0.25{\pm}0.06$)% in day 3. Micronuclei was noted in $0.29{\pm}0.08$ ($0.26{\pm}0.09$)% in day 1 and $0.31{\pm}0.05$ ($0.29{\pm}0.06$)% in day 3 in group 2, and in $0.32{\pm}0.06$ ($0.25{\pm}0.09$)% in day 1 and $0.33{\pm}0.08$ ($0.3{\pm}0.06$)% in day 3 in group 3. There was no difference in the frequency of micronuclei between each groups (p> 0.05). Conclusion : Radioiodine ($^{131}I$) did not cause any genotoxicity in mice bone marrow even at the large dose (16.67 mCi/kg).

BACKGROUND: The purpose of this study was to evaluate ecological function of small-scale pond and movement characteristics of mudfish (Misgurnus mizolepis) during midsummer drainage period. METHODS AND RESULTS: In situ experiments were performed in the paddy field with mudfish under the condition of midsummer drainage from 13 July to 29 July 2010. The mudfish used in this experiment is approximately 1,000 individuals with a cut tail. Mudfishs were released in the rice field before midsummer drainage and caught again in the small-scale pond and the paddy field after midsummer drainage. Results showed that the abundance of mudfish was higher in drainage canal than small-scale pond at the early stage of midsummer drainage, because flow was formed toward the drainage canal. In that time, 3% of the total marked mudfish were captured at outlet of drainage canal. As the paddy was drying, 5% of total marked mudfish moved to the small-scale pond during midsummer drainage period. Contrary to the general hypothesis, the marked mudfish was not found in holes in paddy field.of total caught in the small-scale pond ingested mainly animal prey, and it's frequency of empcy stomach was 10%.oOn the other hand, all m total collected in the paddy field showed empcy stomach. It was apparent from the experiment that m total are eeldng normally in the small-scale pond, while m total are not eat properly in paddy field. CONCLUSION(s): As the paddy was drying, mudfish moved to the small-scale pond during midsummer drainage period but mudfish was not found in holes in paddy field. It can be concluded that small-scale provides a shelter and prey to mudfish in the midsummer drainage period.
Purpose: The aim of this study was to evaluate the normal values of regional cerebral blood flow (rCBF) and cerebrovascular reserve (CVR) in normal children to aged volunteers using Tc-99m HMPAO, Materials and Methods: Thirty four right-handed normal volunteers (20 males, 14 females, mean age $40.3{\pm}24.9$ years, range 4 to 82 years) were underwent rost/acetazolamide (ACZ) brain SPECT using Tc-99m HMPAO and the sequential injection and subtraction method. rCBF was estimated on the basis of a semiquantitative approach by means of right/left ratio, region/cerebellum and region to whole brain ratios in (rental, parietal, temporal, and occipital lobes, basal ganglia, thalami, and cerebellum. CVR was measured by means of % perfusion increase calculated as % mean count change compared to rest rCBF in each regions. Results: Mean values of right to left ratios range from 1.004 to 1.018, rCBF was highest in cerebellum and lowest in basal ganglia and thalami. Frontal and temporal rCBF decreased while occipital and thalamic rCBF increased according to age. No sexual difference of rCBF was noted. Mean CVR was $29.9{\pm}12.9%$. Mean CVR significantly increased to late teens, and declined thereafter. After 6th decade, CVR in both frontal lobes, left parietal lobe and right basal ganglia decreased significantly with advancing age. There was no sexual difference of CVR. Conclusion: Quantitative assessment of CVR was possible by ACZ Tc-99m HMPAO brain SPECT. It revealed that rCBF and CVR changed according to age in normal Korean volunteers. There was no sexual difference.
Purpose: The purpose of this study is to evaluate migration of technetium-99m hexamethylpropylene amine oxime ($^{99m}Tc$-HMPAO) labeled immature and mature dendritic cells (DC) in the mouse. Methods: DC were collected from bone marrow (BM) of tibiae and femurs of mice. Immature and mature DC from BM cells were radiolabeled with $^{99m}Tc$-HMPAO. To evaluate the functional and phenotypic changes of DC from radiolabeling, the allogeneic mixed lymphocyte reaction (MLR) and fluorescence-activated cell sorting (FACS) analysis were performed before and after labeling with $^{99m}Tc$-HMPAO. Migration of intravenously injected DC (iv-DC) was assessed by serial gamma camera images of mice with or without subcutaneous tumor. Percent injected dose per gram (%ID/g) was calculated in lungs, liver, spleen, kidneys, and tumor through dissection of each mice after 24 hours of injection. Results: Labeling efficiency of immature and mature DC were $60.4{\pm}5.4%\;and\;61.8{\pm}6.7%$, respectively. Iv-DC initially appeared in the lungs, then redistributed mainly to liver and spleen. Migration of mature DC to spleen was significantly higher than that of immature DC ($38.3{\pm}4.0%\;vs.\;32.2{\pm}4.1%$ in control group, $40.4{\pm}4.1%\;vs.\;35.9{\pm}3.8%$ in tumor group; p<0.05). Migration to tumor was also significantly higher in mature DC than in immature DC ($2.4{\pm}0.3%\;vs\;1.7{\pm}0.2%$; p=0.034). Conclusion: Assessment of migration pattern of DC in mice was possible using $^{99m}Tc$-HMPAO labeled immature and mature DC. Migration of mature DC to spleen and tumor was higher than that of immature DC when they were i.v. injected.
The limnological characteristics of a coastal lagoon were studied in Lake Hyangho, one of a series of brackish lagoons along the eastern coast of Korea. Phytoplankton community structure, physical factors, and chemical factors were surveyed from May 1998 through November 2002 on a two-month interval basis. Temperature, salinity, Secchi disc transparency, TN, TP, organic matter content of sediment, chlorophyll a concentration, dominant phytoplankton species, and phytoplankton cell density were measured. Salinity gradient was formed between the overlying freshwater stream water and the permeated seawater at the bottom. The chemocline was persistent at the depth of 2 ${\sim}$ 5 m that caused discontinuities of salinity, DO, and temperature profiles. The inversion of vertical temperature profiles with higher temperature in deeper layer was observed in early winter. Secchi disc transparency was very low with the range of 0.1 to 1.1m. TP, TN, and Chl. a concentration in the epilimnion was 0.011 ${\sim}$ 0.238 mgP $L^{-l}$, 0.423 ${\sim}$ 2.443 mgN $L^{-l}$, and 0.7 ${\sim}$ 145.2 mg $m^{-3}$, respectively. Sediment was composed of silt and coarse silt. COD, TP, and TN content of dry sediment were 19.7 ${\sim}$ 73.3 mg$O_2\;g^{-1}$, 0.61 ${\sim}$ 1.32 mgP $g^{-l}$ and 0.64 ${\sim}$ 0.88 mgN $g^{-l}$, respectively. Dominant phytoplankton species were chlorophytes (Ankistrodesmus falcatus) and cyanobacteria (Oscillatoria sp. and Merismopedia tennuissima). The total cell density was in the range of 560 ${\sim}$ 35,255 cells $mL^{-l}$.
The effect of ultraviolet (UV) radiation on the characteristics of algae-derived dissolved organic matter (DOM) was examined by comparing the biodegradability and DOM fraction distribution of algal DOM before and after UV exposure. Algal DOM from two axenic cultures of Microcystis aeruginosa and Oscillatoria agardhii were irradiated for 24 h at a UV intensity of 42 W/$m^2$. A complete degradation of algal DOM during the UV exposure did not occur, remaining at constant concentrations of dissolved organic carbon(DOC). After UV exposure, however, microbial degradations were reduced by 17% in M. aeruginosa and 53% in O. agardhii, respectively, and decomposition rates also were two times lower in UV exposed algal DOM. In addition, the chemical compositions of algal DOM altered substantially after UV radiation exposure. The proportions of hydrophilic bases (HiB; protein-like DOM) decreased considerably in both algal DOM sources after UV exposure (16.8% and 20.0% of DOM, respectively), whereas those of hydrophilic acids (HiA; carboxylic acids-like DOM) increased as much as the decrease of the HiB fraction. Capillary ion electrophoresis (CE) analysis showed that several carboxylic acids increased significantly after UV exposure, further confirming an increase in HiA fractions. The results of this study clearly indicate that algal DOM can be changed in its chemical composition as well as biodegradability without complete degradation by UV radiation.

In order to provide a basis for studying the molecular mechanism of pharmacological action of local anesthetics and to develop a fluorescence spectroscopic method which can detect the microviscosity of native and model membranes using intramolecular excimerization of 1,3-di(l-pyrenyl)propane (Py-3-Py), we examined the effect of lidocaine HCl on the microviscosity of model membranes of phosphatidylcholine fraction extracted from synaptosomal plasma membrane vesicles (SPMVPC). The excimer to monomer fluorescence intensity ratio (I'/I) of Py-3-Py in liquid paraffin was a simple linear function of $T/{\eta}.$ Based on this calibration curve, the microviscosity values of the direct probe environment in SPMVPC model membranes ranged from $234.97{\pm}48.85$ cP at $4^{\circ}C$ to %19.21{\pm}1.11$ cP at $45^{\circ}C.$ At $37^{\circ}C,$ a value of $27.25{\pm}0.44$ cP was obtained. The lidocaine HCl decreased the microviscosity of SPMVPC model membranes in a concentration-dependent manner, with a significant decrease in microviscosity value by injecting the local anesthetic even at the concentration of 0.5 mM. These results indicate that the direct environment of Py-3-Py in the SPMVPC model membranes is significantly fluidized by the lidocaine HCl. Also, the present study explicitly shows that an interaction between local anesthetics and membrane lipids is of importance in the molecular mechanism of pharmacological action of lidocaine HCl.
The autochthonous and allochthonous organic carbon loading were measured in Lake Soyang, to estimate the amount of carbon loading into the lake and the contribution of their sources to tile lake's carbon loading. Autochthonous carbon loading was estimated from phytoplankton primary production with the extracellular organic carbon (EOC). Allochthonous loading was determined by measuring dissolved organic carbon (DOC) and particulate organic carbon (POC) concentration in the main inflowing Soyang River. Both autochthonous and allochthonous organic carbon loading were high during the svmmer, from July to September, and accounted for 43.2% and 71.7% of the annual loading, respectively. Primary productivity was elevated up to $1,000\;mgC\;m^{-2}\;d^{-1}$ during summer and lowest in winter. EOC production from phytoplankton was also large in summer, resulting in a high DOC concentration in the lake water. Primary production of phytoplankton and allochthonous organic matter loading from the watershed contributed to 53.6% and 46.4% of total loading, respectively. The EOC production accounted far $4.4{\sim}21.2%$ of POC primary production, implying that EOC production of phytolankton must be considered in estimation of primary production.
