Applicability Evaluation of Deep Learning-Based Object Detection for Coastal Debris Monitoring: A Comparative Study of YOLOv8 and RT-DETR (해안쓰레기 탐지 및 모니터링에 대한 딥러닝 기반 객체 탐지 기술의 적용성 평가: YOLOv8과 RT-DETR을 중심으로)

  • Coastal debris has emerged as a salient issue due to its adverse effects on coastal aesthetics, ecological systems, and human health. In pursuit of effective countermeasures, the present study delineated the construction of a specialized image dataset for coastal debris detection and embarked on a comparative analysis between two paramount real-time object detection algorithms, YOLOv8 and RT-DETR. Rigorous assessments of robustness under multifarious conditions were instituted, subjecting the models to assorted distortion paradigms. YOLOv8 manifested a detection accuracy with a mean Average Precision (mAP) value ranging from 0.927 to 0.945 and an operational speed between 65 and 135 Frames Per Second (FPS). Conversely, RT-DETR yielded an mAP value bracket of 0.917 to 0.918 with a detection velocity spanning 40 to 53 FPS. While RT-DETR exhibited enhanced robustness against color distortions, YOLOv8 surpassed resilience under other evaluative criteria. The implications derived from this investigation are poised to furnish pivotal directives for algorithmic selection in the practical deployment of marine debris monitoring systems.

Characterization and predictive value of volume changes of extremity and pelvis soft tissue sarcomas during radiation therapy prior to definitive wide excision

  • Purpose: The purpose of this study was to characterize and evaluate the clinical significance of volume changes of soft tissue sarcomas during radiation therapy (RT), prior to definitive surgical resection. Materials and Methods: Patients with extremity or pelvis soft tissue sarcomas treated at our institution from 2013 to 2016 with RT prior to resection were identified retrospectively. Tumor volumes were measured using cone-beam computed tomography obtained daily during RT. Linear regression evaluated the linearity of volume changes. Kruskal-Wallis tests, Mann-Whitney U tests, and linear regression evaluated predictors of volume change. Logistic and Cox regression evaluated volume change as a predictor of resection margin status, histologic treatment response, and tumor recurrence. Results: Thirty-three patients were evaluated. Twenty-nine tumors were high grade. Prior to RT, median tumor volume was 189 mL (range, 7.2 to 4,885 mL). Sixteen tumors demonstrated significant linear volume changes during RT. Of these, 5 tumors increased and 11 decreased in volume. Myxoid liposarcoma (n = 5, 15%) predicted decreasing tumor volume (p = 0.0002). Sequential chemoradiation (n = 4, 12%) predicted increasing tumor volume (p = 0.008) and corresponded to longer times from diagnosis to RT (p = 0.01). Resection margins were positive in three cases. Five patients experienced local recurrence, and 7 experienced distant recurrence, at median 8.9 and 6.9 months post-resection, respectively. Volume changes did not predict resection margin status, local recurrence, or distant recurrence. Conclusion: Volume changes of pelvis and extremity soft tissue sarcomas followed linear trends during RT. Volume changes reflected histologic subtype and treatment characteristics but did not predict margin status or recurrence after resection.

  • Rapid detection of viable Salmonella in pasteurized milk is important to protect public health from food poisoning. Reverse transcriptase-polymerase chain reaction(RT-PCR) is recognized as a molecular genetical method to differentiate between live and dead bacteria The RT-PCR in this study was designed to detect specifically viable Salmonella in milk by using the primers whose nucleotide sequences were determined based on fimA gene which encodes the submit of type 1 fimbriae. Treatment of RNA preparation with RNase-free DNase was adequate enough to destroy DNA, which may otherwise be amplified in the RT PCR Seven strains of Salmonella were detected in the RT-PCR but Escherichia coli, Shigella sonnei, Citrobacter freundii, and Klebsiella pneumoniae were not. $10^7/ml$ and $10^6/ml$ of dead Salmonella which were heat-treated in milk were detectable by using the RT-PCR but $10^5{\sim}10/ml$ of the dead bacteria were not. The sensitivity of the RT-PCR in detecting viable Salmonella was 100 cells/ml.

Comparison Between ELISA and Gel-filtration Assay for the Guantitation of Airway Mucins

  • In this study, we developed immunoassay methods for the more convenient and effective detection of rat tracheal mucin and the results were compared with those of [$3^H$]glucosamine based gel-filtratioh method. A monoclonal anti-rat tracheal mucin antibody, mAbRT03, which specifically recognizes rat tracheal mucins, was used throughout in this study. To induce mucin secretion, varying concentrations of ATP (0-2 mM) were applied to the primary rat tracheal surface epithelial (RTSE) cell culture which had been metabolically radiolabeled with [$3^H$]glucosamine and the secretion of mucin was analyzed both by the immunoassay and the gel-filtration chromatography methods. For the immunoassay, the following two procedures were employed. 1) Simple ELISA; the culture spent media were directly coated onto the assay plate and the immunoreactivity with mAbRT03 was assessed from the standard curve generated with the purified rat mucin. 2) Inhibition ELISA; A known amount of the purified rat mucin was coated onto the assay plate and then ATP-stimulated culture spent media were added to inhibit the immunorelitivity with mAbRT03. The contents of mucin in the sample were calculated from the standard inhibition curve which was generated with the purified rat mucin. The assay results obtained from the immunoassays were identical with those from the gel-filtration methods. The present result indicates that ELISA can be substituted for the laborious, time-consuming gel-filtration assay in studying the regulation of airway mucin release from cultured airway epithelial cells.

Analysis of Vitellogenin Gene Expression in Synechogobius hastus (Gobiidae) (풀망둑 난황전구단백질 유전자발현 추적기법)

  • In an effort to develop the tools for monitoring the contamination of xenoestrogen in the aquatic environment of Korea, reverse transcription-polymerase chain reaction (RT-PCR) analysis of vitellogenin (VTG) mRNA expression were optimized in Synechogobius hastus. Based on the partial VTG cDNA sequence VTG mRNA level in livers from male fishes was analyzed by RT-PCR. As an internal control beta actin mRNA was amplified. 3 ${\mu}g$ of total RNA was reverse transcribed in 20 $\mu$l reaction using murine leukemia virus 〔MuLV〕 reverse transcriptase. Subsequent PCR using the 1 ${\mu}g$ of cDNA resulted in linear increase in PCR product of VTG in female liver cDNA from 10 to 30 cycles of amplification. On the contrary, in male, PCR product first detected at 28 cycles of amplification and linearly increased during 38 cycles of amplification, suggesting that male S. hastus expresses minute amount of VTG mRNA which is $2^{-18}$ equivalent of female. In conclusion, the optimized protocol of VTG mRNA expression in the liver of male S. hastus will be promising the environmental monitoring the xenoestrogen contamination in the western coast and estuaries in Korea.

Expression of Leptin and Its Receptor in Rat Ovary (흰쥐 난소내 Leptin 및 Leptin 수용체의 발현)

  • Leptin, the product of the obese gene, is produced by adipose tissue and is known to be a hormone concerned with regulation of appetite and metabolism. Recent reports have shown that leptin is associated not only with obesity but also with female reproduction, but it has not yet been ascertained whether leptin acts directly on the ovaries or indirectly via the hypothalamus or pituitary pathway. The object of this study is to determine the expression of leptin and its receptor in the ovaries of 3 and 8 weeks old rats by immunohistochemistry and RT-PCR. In the ovaries of 3 and 8 weeks old rats, leptin was stained in the theca cells and portions of granulosa cells of atretic follicles, whereas leptin receptors was stained in interstitial cells and ova of preantral follicles. The RT-PCR results showed that leptin receptor mRNA was expressed in the ovaries of both immature and adult rats, while leptin mRNA was not. In conclusion, leptin mRNA was not expressed in the ovaries, however, leptin was detected by immunohistochemistry. Compared to leptin itself, leptin receptors in the ovaries were ascertained by both RT-PCR and immunohistochemistry. These results suggest that leptin is related to the regulation of the physiological functions of the ovaries.

Using reverberation time to evaluate the amount of scattered sound energy from a tree (잔향시간지표를 이용한 나무의 음향확산성 평가)

  • In urban spaces surrounded by buildings, trees could disperse sound energy, which affect sound level distribution and street canyon reverberation. Therefore, this paper examines the amount of scattered sound energy from a tree in open field by means of a reverberation time (RT). Five trees of different species and crown sizes were considered. The influential factors include crown size and shape, foliage condition, and source-receiver distance. The results show that RT is proportionally increased with the increase of tree crown sizes, which is the most determining factor. The maximum RT measured was 0.34 sec at 4000 Hz for the studied trees in leaf. The presence of leaves increased RT at high frequencies, typically by 0.14 sec at 4000 Hz. With increasing source-receiver distance within 40 m, RT was slightly changed.

  • Substantial evidences have been accumulated about the hormone-like effects of exogenous substances such as pesticides and industrial chemicals during past years. The effects of these substances on the endocrine system are believed to be either enhancing or reducing of various endocrine action. It is necessary to identify putative causal agents by the batter system and to assess their ability to disrupt the endocrine system. A variety of in vitro and In vivo approaches have been used to determine the androgenic effects of environmental chemicals. To establish the method for assessment of the putative endocrine disruptors with androgenic activity, we carried out the cell proliferation assay by MTS method after treatment with the various concentration of testosterone in LNCaP cells (human prostatic cancer cell line) and also observed the expression of androgen-related genes by quantitative RT-PCR. In the cell proliferation assay, the results showed that the grouth of LNCaP cells increased within level of at least 10pM testosterone. We measured by quantitative RT-PCR method on the effects of testosterone on mRNA expression of androgen receptor (AR), prostate-specific antigen (PSA), bone morphogenetic protein (BMP) and BMP receptor (BMPR) In LNCaP cells. The results demonstrated that mRNA expression of PSA and BMPR-IB was observed differently within level of at least 0.01 pM testosterone compared with non-treated control. These observations suggest that the detection of PSA and BMPR-IB mRNA by the quantitative RT-PCR in LNCaP cells is very sensitive method to identify the endocrine disruptors to have the androgenic effects.

The Kinetics Study of Ozone with Sulfur Dioxide in the Gas Phase (기체 상태에서의 오존과 아황산가스의 반응연구)

  • The kinetic of the gas phase reactions of ozone(0.5 torr) with sulfur dioxide was studied. The SO2 reaction was conducted in the 7∼22 torr range at 90∼155$^{\circ}$C. The reaction rate was faster than the reaction rate of O$_3$ in the presence of CO$_2$ alone. The reaction of O$_3$ with SO$_2$ follows the rate law: -d(O$_3)/dt=k_0(SO_2)(M)(O_3)+2k _1(SO_2)(O_3$). The first term of this rate law arises from a third order molecular reaction predominating in the lower temperature range and gave a rate constant k$_0$ = (9.35 $\pm$ 8.6) ${\times}$ 10$^9$e$^{-(11.05{\pm}2.04)kcal/RT}(M^{-2}s^{-1}$). The second term of the above rate law derived from a second order thermal decomposition reaction which was the major part of the reaction and gave a rate constant k$_0 =(9.35{\pm}8.6){\times}10^9e^{-(11.05{\pm}2.04)kcal/RT}(M^{-2}s^{-1}$). The overall reaction proceeds with kinetics of complex order composed mainly of second order and third order components.

Experession of Vasoactive Intestinal Peptide in the Hypothalamus of Fasting and Anorexia Mutant Mice (anx/anx) (절식시킨 생쥐와 식욕부진 돌연변이 생쥐의 시상하부에서 Vasoactive Intestinal Peptide의 발현)

  • The present study was conducted to identify the mechanism about the regulation of appetite by examining the expression patterns of vasoactive intestinal peptide in the hypothalamus of either fasted for 24 hours or anorexia mutant mouse. In order to investigate expression pattern of the vasoactive intestinal peptide, immunohisto-chemistry was employed along with reverse transcription polymerase chain reaction (RT-PCR) and dot blotting. Immunohistochemistry has shown that level of expression of vasoactive intestinal peptide and appetite-suppessing neuropeptide, was lower in the suprachiasmatic nucleus (SCN) and higher in the paraventricular nucleus (PVN) of the anorexia mutant group than in the comparable regions in the control group. This pattern was repeated in the fasting group, which also showed lower and higher levels of vasoactive intestinal peptide expression in the SCN and PVN respectively, In contrast, the vasoactive intestinal peptide mRNA level in the entire hypothalamus via RT-PCR and dot blotting was similar in the fasting and control groups, while it was significantly increased in the anorexia mutant group.
