Investigation on the Microbiological and Biochemical Properties of Kimchi in the Solid-state Model System Designed for Fermented Sausages

  • The objective of this study was to investigate the potential of the application of kimchi LAB as starter culture in the production of fermented sausages. For this, the solid-state model media composed to simulate the substantial conditions of meat mixtures were fermented for 120 h after the treatment with different concentrations of kimchi (0.5, 1.0, 1.5, 3.0, and 5.0%) and lyophilized kimchi-powder (0.2 % and 0.5%). During the fermentation period, the growth of total viable cells and LAB, and the changes of pH and titratable acidity were investigated. The initial LAB counts ranged from 7.18 to 8.34 Log CFU/ mL for kimchi media and from 6.93 to 6.94 Log CFU/mL for kimchi-powder media depending on the added concentrations. The kimchi LAB in this study were not influenced by the immobilized condition for their adaptation and growth by showing no lag phase and thus acted similar as in the submerged medium. The initially increased counts reached around 9 Log CFU/ mL in 12 h independent of the concentrations of a ded kimchi. However, the growth and metabolic activity of kimchi-powder LAB were influenced by the immobilized condition. Supposedly, as the nutrient supply in solid-state depended solely on diffusion, these differences in the souring properties were caused by the LAB topography in the medium matrix. Nevertheless, the differences in the numbers of LAB between two media were less than 0.5 Log units and the pH drop in the solidstate batches was quite rapid and reached low values. Therefore, it can be assumed that kimchi and kimchi-powder LAB showed the utility as the substitute of commercial starter culture even without a rehydrating pretreatment.

  • For a probiotic yoghurt it is desirable to utilize lactic acid bacteria with a high survival rate and beneficial function to human beings. We have examined a variety of lactic acid bacteria to assess the acid and bile tolerance and antibiotic resistance. In addition, an in vitro culture experiment was performed to evaluate their ability to reduce cholesterol levels in the growth medium. Thirteen strains were selected from in vitro cholesterol assays and fed to Sprague-Dawley rats with a high-cholesterol diet. Among the 13 strains tested, 8 strains were shown to reduce serum cholesterol levels significantly after 24 days of administration in vivo. Rats were fed lyophilized yoghurt powder fermented with a combination of 3 selected strains: Bifidobacterium infantis AM-220, Lactobacillus AM-245, and Styeptococcus MA-1. The levels of total cholesterol and low density lipoprotein were significantly lower (p$\leq$0.05) in rats fed the yoghurt powder compared with control group. These studies suggest that yoghurt fermented with appropriately selected lactic acid bacteria may have a anticholesterolemic effect.

Effects of Red Snow Crab Chionoecetes japonicus Leg Meat Powder on the Quality Characteristics of Noodles (홍게(Chionoecetes japonicus) 다릿살 분말 첨가가 국수의 품질 특성에 미치는 영향)

  • In this study, we investigated the quality characteristics of noodles supplemented with lyophilized leg-meat powder from the red snow crab, Chionoecetes japonicus (CP). The noodles were prepared by adding 0, 2, 4, 6, 8, 10% of CP on a wheat-flour basis. The ash, crude protein and crude lipid contents of noodles increased with increasing CP content. The cooked weight, volume and water absorption values of noodles decreased, whereas the turbidity increased, with increasing CP content. The L values of the noodles decreased with increasing CP content, whereas the a and b color values increased. The hardness, gumminess and chewiness values increased with increasing CP content increased, while springiness and cohesiveness showed no significant differences among the groups. In the sensory evaluation, cooked noodles with 4-6% CP had greater overall acceptability than the noodles with other CP levels.

The Quality Characteristics and Processing of Madeleine Containing Red Snow Crab Chionoecetes japonicaus Leg-Meat Powder (홍게(Chionoecetes japonicus) 다릿살 분말을 첨가한 마들렌의 제조 및 품질특성)

  • In this study, we investigated the quality characteristics of madeleine combined with lyophilized red snow crab Chionoecetes japonicus leg-meat powder (CM). The madeleine was prepared by the addition of wheat flour containing 0, 1, 5, 10, 15 and 20% concentrations of CM. Moisture content, pH, and baking loss rate did not differ significantly among groups. Light coloration significantly decreased in a dose-dependent manner, while red and yellow coloration significantly increased. Hardness, chewiness and gumminess significantly increased in a dose-dependent manner, while cohesiveness, springiness and adhesiveness did not differ from CON (madeleine containing 0% CM). In terms of sensory characteristics, appearance and color did not differ compared to CON, while taste, flavor and overall preference increased in a dose-dependent manner, with CM10 having the highest score among the groups. The present study assesses the potential of this improved formula as a convenience food.

Effect of Substrates and Lyoprotectant on the Survival Ratio of Lyophilized Bacillus sp. SH1RP8 (동결건조 보호제와 기질이 동결건조된 Bacillus sp. SH1RP8의 생존율에 미치는 영향)

  • In order to develop an eco-friendly biofertilizer, a plant growth promoting rhizobacterium (PGPR), Bacillus sp., SH1RP8 was investigated. SH1RP8 was lyophilized via freeze-drying along with other protective agents that protect cells from lysis. The freezedried powder of Bacillus sp. SH1RP8, containing 5% skim milk (w/v), exhibited the highest survival rate of 30.6% among all the protective agents (skim milk, glucose, and peptone). The lyoprotective effect of the skim milk, mixture including 5% skim milk, and substrates on the survival of the test strain was examined. Control group was added only skim milk and test groups were added skim milk and other substrates. As a result, the group supplemented with both glycerol and 5% skim milk showed the protective effect much higher by 214.29% than the control group. Freeze-dried Bacillus sp. SH1RP8 could be a good candidate as a potential biofertilizer due to its effective PGPR activity.

Tumor Cell Cytotoxicity of Gamma Irradiated Chaga Mushroom Extract (감마선 조사된 차가버섯 추출물의 암세포 활성 억제)

  • This study was investigated the effect of tumor cell cytotoxicity of gamma irradiated Chaga mushroom extract (CME). CME was prepared by hot water extraction at $70^{\circ}C$ for 4 hours and lyophilized. $Ten\;mg\;ml^{-1}$ of lyophilized CME powder was dissolved with deionized water and then irradiated at the doses of 10, 50, 100, and 150kGy by cobalt 60 gamma irradiator. The gamma-irradiated and non-irradiated CME were treated into the cancer cell, including human stomach cancer and human colon cancer. Cytotoxicity against the cancer cell was increased in gamma-irradiated CME and antioxidant activity was also increased in gamma-irradiated CME, as irradiation dose increased. Therefore, it was considered that gamma irradiation was effective method for improvement of the cancer cell cytotoxicity and antioxidant activity of Chaga mushroom extract.

  • This study investigated the effect of gamma irradiation on immune enhancing activity of Chaga mushroom extract (CME). CME was prepared by hot water extraction at $70^{\circ}C$ for 4 hours and lyophilized. Lyophilized CME powder was dissolved with deionized water at $10mg\;ml^{-1}$ and then irradiated at the doses of 10, 30 and 50 kGy by cobalt 60 gamma irradiator. The gamma-irradiated and non-irradiated CME were treated into the splenocyte separated from mouse. Cell proliferation and cytokine production of the immune cells were increased by gamma-irradiated CME and these increases were more prominent when CME was irradiated at higher doses. Therefore, it is considered that gamma irradiation can be an effective method for improvement of the immunomodulating activity Chaga mushroom extract.

Variations in total phenols, total anthocyanins, and antioxidant activity levels in black chokeberry (Aronia melanocarpa) fruits subjected to dry and moist heat treatments

  • The present study investigated the effects of dry and moist heat treatments on total phenols, total anthocyanins, and antioxidant activity levels in black chokeberry (Aronia melanocarpa) fruits. Lyophilized chokeberry powder samples were heated in a drying oven at 60, 100, 160, 180, and 200℃ for 20, 40, or 60 min. Finely ground fresh chokeberry fruits were heated in water at 60, 80, and 100℃ for 20 min, and bioactive compound and antioxidant activity levels were measured. The bioactive compounds and antioxidant activity decreased with increasing temperature and treatment duration. Antioxidant activity was preserved at 160℃ or lower without significant loss for dry heating, whereas moist heat treatment increased both bioactive compounds and antioxidant activity with increasing temperature.

  • Sterile Products may be broadly classified into two main categories, according to the manner in which they are produced: those which are sterilized after the product has been filled and sealed in the final container(s) ("terminally sterilized" products) and those where the sterilization stage (or stages) takes place is it before or after the bulk product filled in to final container. In this latter instance, all subsequent processing (typically, the filling and sealing operations) must be conducted aseptically in order to prevent recontamination of the sterilized product. The two most common pharmaceutical applications of aseptic processing methods are (a) the filling of liquid products following sterilization by filtration and (b) the filling of previously sterilized bulk powder products. An aseptic processing operation should be tested using a microbiological growth medium (media fill) during lyophilized injection formulation, filling, loading, lyophilisation, stoppering, and unloading activities.

Pharmacognostical Studies on Alisma Plants (택사의 생약학적 연구)

  • In order to clarify the originality of several Alisma Rhizomes, we investigated the differences of their morphological, anatomical and physiochemical characteristics. Morphological observation shows the variability of Alisma shape and quality. The diversities in morphological shapes and qualities of tuber were observed among different origins. Chinese product looked like corm shape, but korean's irregular shape. The microscopic observation of korean Alisma revealed the presence of irregular vascular bundle which was scattered transversely and longitudinaly, which was different from that of the rhizome. This facts suggest that the portion of crude drug in Alisma should be the corm rather than the rhizome. In the physicochemical differences, chinese Alisma contains larger amount of mineral elements such as K, Na and Ca than korean Alisma. On the contrary, protein contents in lyophilized powder of aqueous extracts in korean Alisma rhizome was significantly higher than chinese Alisma rhizome. The leaves and stems contained larger amount of Ca and Na than tuber, and may be used as the osmotic diuretics for chinese traditional medicine.
