Effects of Portulaca oleracea Powder on the Lipid Levels of Rats Fed a Hypercholesterolemia Inducing Diet

  • This study was initiated to investigate the effects of lyophilized purslane (Portulaca oleracea) powder (5% and 10%) on lipid levels in rats fed a hypercholesterolemia-inducing diet (1% cholesterol). During the four week study, there were no significant differences in either weight change or the food efficiency ratio between the group fed the hypercholesterol diet alone (HC) and the groups fed the purslane powder (HCPO-I and HCPO-II, 5% and 10% purslane, respectively). In serum, the levels of total lipid, total cholesterol and LDL-C decreased significantly for the group fed the 10% purslane powder (HCPO-II) in comparison with the group fed the 5% purslane powder (HCPO-I). The atherogenic index (AI) was reduced by about 51% for the group fed the 10% purslane powder (1.47) in comparison with the HC group (3.03). The activities of GOT, GPT, ALP and LDH decreased significantly for the groups fed the purslane powder in comparison with the HC group. Regarding liver tissue, the levels of total lipid, total cholesterol and triglyceride decreased significantly for the purslane powder-fed rats compared to the HC group. The fecal lipid profiles increased significantly as the amount of purslane powder was increased. Compared to the HC group, the fecal total cholesterol and triglyceride levels were higher in the group fed the 10% purslane powder by about 2.8 times and 2.3 times, respectively. For the serum and liver tissue, the content of lipid peroxide decreased significantly in the groups fed purslane powder compared to the HC group. The data from this experiment show an increase in the lipid levels discharged in feces, suggesting that the supplementation of purslane powder to a hypercholesterolemia-inducing diet reduces lipid levels.

  • This study was to determine whether blue honeysuckle lyophilized concentrated powder (BH) has positive effects on hypothyroidism and related liver damage in propylthiouracil-induced Hypothyroidism. Levothyroxine (LT4)-treated group was administrated by intraperitoneal injection with LT4, while BH or Flos Lonicerae lyophilized aqueous extract (LF)-treated groups were orally administrated for 42 days which two weeks pri or to PTU injection. The changes in organ weight, serum AST & ALT, serum lipid level, liver defense system were measured and the histopathology of the liver was observed. The oral administrations of 125, 250, and 500 mg/kg of BH showed favorable effects compared to LF on hypothyroidism-related liver damages through the enhance antioxidant defense system.

  • The objective of the present study is to determine whether blue honeysuckle lyophilized concentrated powder (BH) has favorable effects on hypothyroidism and related reproductive organ damage. Hypothyroidism was induced by 9 subcutaneous administration of propylthiouracil (PTU) for 28 days. Levothyroxine (LT4)-treated group was intraperitoneally injected with LT4 for the same period, while for BH (125, 250, and 500 mg/kg) or Flos Lonicerae lyophilized aqueous extract (LF, 250 mg/kg)-treated groups, the test materials were orally administrated for 42 days: two weeks before PTU injection and during PTU administration. The changes in serum thyroid hormone levels, serum male sex hormone levels, and testis antioxidant defense system were observed by histopathology of the thyroid gland, epididymis, prostate, and testis. The oral administrations of 125, 250, and 500 mg/kg of BH showed favorable effects compared to LF on hypothyroidism and related damages of reproductive organs through augmentation of the antioxidant defense system in the testis. In conclusion, BH is a promising new potent thyroid gland protecting agent.

Investigation of the Microbiological and Biochemical Properties of Kimchi in the Submerged Model System Designed for Fermented Sausages

  • The objective of this study was to investigate the potential of the application of lactic acid bacteria (LAB) from kimchi as a starter culture in the production of fermented sausages. To achieve this, a submerged model medium that contained LAB as part of a complex system of kimchi (0.5, 1.0, 1.0, 3.0, and 5.0%) and lyophilized kimchi powder (0.2 and 0.5%) was fermented for 120 h. During the fermentation period, the growth of total viable organisms and LAB, and the changes in the pH and the titratable acidity, were investigated. The initial LAB counts ranged from 6.4 to 7.7 Log CFU/mL for the kimchi media, and from 6.9 to 6.9 Log CFU/mL for the kimchi powder media. In all the kimchi batches, the LAB increased logarithmically, and the highest LAB counts (around 9 Log CFU/mL) were reached in 24 h. An evident lag phase of the LAB was observed in the kimchi powder samples and reached 8.8 Log CFU/mL in 8 h. The decrease in the pH and the formation of lactic acid were rapid in the kimchi batches, and reached pH values of 3.4-3.5 in 12 h. With these results, the LAB that was integrated with the addition of kimchi or kimchi powder demonstrated its potential utility as a substitute for starter culture.

Evaluation of Gelation Properties of Salt-Soluble Proteins Extracted from Protaetia brevitarsis Larvae and Tenebrio molitor Larvae and Application to Pork Myofibrillar Protein Gel System

  • The purpose of this study was to investigate the functional properties of salt-soluble proteins obtained from Protaetia brevitarsis (PB) and Tenebrio molitor (TM) larvae, the interaction between these proteins and pork myofibrillar protein (MP) in a gel system. The gel properties of salt-soluble protein extracts showed that the PB had a higher viscosity than the TM protein. However, the TM protein had higher gel strength compared with the PB protein. The gelation characteristics of the pork MP gel systems added with lyophilized insect salt-soluble protein powder showed to decrease slightly viscosity compared with MP alone. Adding the TM or PB protein powder did not affect the pork MP's hydrophobicity and sulfhydryl group levels. Furthermore, the protein bands of the MP did not change with the type or amount of insect salt-soluble protein. The cooking yields of the pork MP gels containing PB or TM protein powder were higher than those without insect protein. Regardless of the type of insect salt-soluble protein added, the pork MP's gel strength decreased. Furthermore, as the level of insect powder increased, the surface protein structure became rough and porous. The results demonstrated that proteins extracted from PB and TM larvae interfered with the gelation of pork MP in a gel system.

동결 건조법을 이용한 옥천산 캡슐 제형 개발에 관한 연구 (A Study of Okchun-San Formulation for Freeze Drying Capsulation)

  • Objective : Okchun-San(OCS) is known as an effective herbal medicine on Type 2 diabetes. We performed to change OCS formulation for freeze drying capsulation. Methods The mixtures of OCS were extracted with water. finally, the filtered solution were evaporated and lyophilized to dry granules. The various ratio of excipients were studied to determine the formation for capsulizing. The samples were inspected for any difference in color, taste and appearance. Results: The prepared form of OCS were dried and weighted 260kg. The lyophilized dry powder yielding 40kg. The suitable ratio of OCS-dry powder and excipient was 10:1. The average weights of On and placebo capsules were $440{\pm}5.28mg$, $465{\pm}7.95mg$, respectively. There was no notable change in color and appearance for both capsuled samples throughout the study period. Conclusions: Therefore in can be concluded that freeze drying capsulation is appropriate form of OCS.

전자선 조사 동결건조 김치분말의 품질 특성 (Effect of Irradiation on the Quality Characteristics of Lyophilized Kimchi Powder)

  • 미숙성김치와 숙성김치를 동결건조한 후 방사선량 별로 나누어 이에 대한 이화학적 분석, 잔존 미생물수 분석 및 관능평가를 통하여 김치의 양념소재화 가능성을 평가하였다. 미숙성김치와 숙성김치는 pH 및 적정산도는 차이가 있었으나 이의 동결건조물의 방사선 조사 처리구들 간에는 큰 차이가 없는 것으로 나타났다. 방사선을 조사하기 전 숙성김치의 색도는 미숙성김치에 비해 L, a 및 b값에서 다소 높은 값을 보였으며, 조사처리 후에는 조사선량의 강도에 의한 영향은 거의 받지 않았고 미숙성김치는 숙성김치보다 색상에서 다소 변화를 보였다. 미숙성김치분말보다 숙성김치분말은 미생물에 대한 방사선 조사에 의한 살균 효과가 전반적으로 높게 나타났으며, 30 kGy 조사 시에는 특히 총균수와 젖산균이 검출되지 않았다. 숙성김치분말에는 ethanol, 1-propene-3-methyl, disulfide dimethyl 및 disulfide methyl propyl 등의 휘발성 성분이 다소 많았고 특히 disulfide dipropyl 성분은 숙성김치분말에만 존재하는 특성을 보인 반면 그 외의 성분에서는 오히려 미숙성김치분말에 많은 것으로 나타났다. 방사선을 조사함에 따라 숙성김치분말에서는 2-butanone과 acetic acid, 핵산 및 butanal 2-methyl 성분 등이, 미숙성김치분말에서는 hexane 및 butanal 2-methyl 성분이 새롭게 생성되거나 증가되는 경향을 보였으며 전반적으로 황화물의 함량이 감소하는 경향을 보였다. 조사한 김치분말에 대하여 이미, 이취 및 매운맛에 대한 관능평가결과 모두 조사량이 증가함에 따라 이미와 이취는 증가하였으나 조사랑 30 kGy 까지는 수용가능한 것으로 평가되었다.

Optimization of Ultrasound-assisted Extraction of Phenolic Compounds from Salicornia herbacea Powder

  • Salicornia herbacea is rich in natural minerals, dietary fibers, and potentially health-promoting phenolic compounds. In this paper, an experimental design was applied for the optimization of the ultrasound-assisted extraction of phenolic compounds from lyophilized Salicornia herbacea powder. The experiments were conducted in accordance with a five-level, three-variable central composite rotatable design (CCRD), and the effects of solvent concentration, extraction time, and extraction temperature were evaluated via response surface methodology (RSM). The optimal extraction conditions were as follows: ethanol concentration, 76.80%; extraction time, 20 min; and extraction temperature, $33.21^{\circ}C$; and the solvent concentration was the most significant parameter in this process, under which the predicted total phenolic content was 49.91 mg GAE/g sample.

Enzyme Inhibitory and Antioxidant Activities of Fruit and Stem of Opuntia ficus india var. saboten

  • The objective was to verify biological activities of fruit and stem of prickly pear(Opuntia ficus indica L. var, saboten Makino). We have determined inhibitory activities on enzymes, such as dopamine ${\beta}$-hydroxylase(DBH), monoamme oxidase A and B(MAO-A, B), and antioxidant activity, in vitro. We purchased dried stem powder and lyophilized fruit powder of prickly pear from CheJu Island, and prepared the extracts with 80% of methanol. The fruit extract showed stronger inhibitory effects on MAO-A and -B and antioxidant activity compared. to the stem extract, on fractionation with hexane, ethyl acetate, butanol and water. Both the stem and the fruit extracts with ethyl acetate showed stronger enzyme inhibitory and antioxidant activities than other extracts. Now we are isolating active principles from both ethyl acetate extracts.

Development of powder with increased rutin content from mulberry leaves for the application of food materials

  • We studied on improvement method of rutin content using mulberry leaf powder. Mulberry leaves were collected and then hot-air dried and powdered for experiment. As a result, we have developed a pre-treatment method that extracts mulberry leaf powder with water or fermented alcohol with reflux extractor and then increases the rutin content by improving the process. Citric acid (0.1 ~ 1%) and 1000 ml fermented alcohol (50 ~ 95%) or water (10 ~ 50 times) was extracted with 100 g of mulberry leaf powder using a reflux extraction device ($80{\sim}90^{\circ}C$, 1 hour, twice). The extracts were collected, filtered and concentrated. For the recrystallization, the concentrate was dissolved by adding distilled water and allowed to stand at a low temperature. Then, the supernatant was discarded by centrifugation, and only the residue was lyophilized to prepare a final powder. As a result, regardless of the concentration of citric acid added, the content of rutin was higher in 90% fermented alcohol extract. Whereas, in the case of extracting with water, citric acid 0.5% was added to water 25 times as much as the weight of mulberry leaf powder, and 2274.4 (mg / 100g) of rutin content was highest in the case of refluxing twice at $80^{\circ}C$ for 1 hour. The powder with increased rutin content is expected to be applicable to various foods as a food additive. In addition, it can contribute to the improvement of the farm income by promoting consumption of mulberry leaf while satisfying the consumers' desire for functional food intake.
