  • Objectives In this study, we evaluated the therapeutic effects of Gami-Bojungikgitang and Gami-Jwagwieum for bleomycin-induced lung fibrosis in mice. Methods Extracted lyophilization, Gami-Bojungikgitang (96g) and Gami-Jwagwieum (118g) boiled, filtered, depressed, concentrated, and are obtained. They were divided into five groups: normal, group IA; Animal group treated with bleomycin observed on the 21th day, group IB; Animal group treated with bleomycin observed on the 42th day, group IIA; Animal group treated with bleomycin and Gami-Bojungikgitang. Gami-Jwagwieum observed on the 21th day, group IIB; Animal group treated with bleomycin and Gami-Bojungikgitang/Gami-Jwagwieum observed on the 42th day. Mice are used on the 42th day and as a result, bronchoalveolar lavages fluid is obtained. Counting total number of cells, different ratio of macrophage, lymphocyte, and neutrophil are established. Results In animal group treated with bleomycin and Gami-Bojungikgitang, total cell count decreased by 50% in 3 weeks compared to animal group with non-administrated Gami-Bojungikgitang. However, total cell count in 6 weeks increased compared to 3 weeks although total cell count still decreased compared to animal group with non-administrated Gami-Bojungikgitang. In the view of differential cell counts in bronchoalveolar lavages fluid in treatment group on 3 and 6 weeks, neutrophile was a few and lymphocyte decreased. In animal group treated with bleomycin and Gami-Jwagwieum, total cell count decreased by 50% in 3 and 6 weeks compared to animal group with non-administrated Gami-Jwagwieum. In the view of differential cell counts in bronchoalveolar lavages fluid in treatment group on 3 and 6 weeks, lymphocyte also decreased. Conclusions Gami-Bojungikgitang and especially Gami-Jwagwieum for bleomycin-induced lung fibrosis in mice were effective in total cell count and differential cell count.

  • This experiment was carried out to elucidate the sugar composition of polysaccharides and the structural features of water-soluble polysaccharides(WSP) isolated from Korean oak mistletoe, Loranthus yadoriki Sieb. The 48-hours ball-milled meals of extractive-free dried mistletoe sawdusts were extracted with distilled water for $24hrs{\times}2$ at room temperature. The extracts poured into 95% ethyl alcohol to precipitate. The separated precipitate of WSP, in form of yellowish white powder by lyophilization, was fractionated into four subfractions of WSP-1, WSP-2, WSP-3 and WSP-4 by anion exchange chromatography on DEAE-cellulose column. The sugar composition of WSPs was analyzed by GLC in form of their glycitol acetates, and the structure of polysaccharides in Fractions WSP-1 and WSP-2 was determined by FT-IR and GC-MS after methylation through and acetylation. The sugars of WSPs from Korean oak mistletoe, Loranthus yadoriki, are majorly arabinose and galactose in stem, galactose in leaves very high in content and showed difference in composition and monomeric units between stems and leaves. D-galactose, D-glucose and L-arabinose are the simple sugars consisting of polysaccharides in WSP-1. ($1{\rightarrow}3$)-Linked galactan is the bakcbone with side chain of ($1{\rightarrow}5$)- -L-arabinofuranosyl residues and ($1{\rightarrow}6$)- -D-galactopyranosyl residues, and ($1{\rightarrow}4$)-linked glucan also presents. ($1{\rightarrow}4$)-Linked rhamnogalacturonan and ($1{\rightarrow}4$)- and ($1{\rightarrow}3$)-linked galactan present in WSP-2.

  • 1) 본실험조건(本實驗條件)에서 GLC에 의하여 AsA의 최저(最低)농도(濃度) 1mg%까지 정량가능(定量可能)하였다. 2) TMS 반응조건(反應條件)으로서 동결건조시료(凍結乾燥試料)에는 반드시 TMS 혼합시액중(混合試液中)에서 추출(抽出)과 TMS 화(化)를 동시(同時)에 시켜야만 98% 이상(以上)의 회수율(回收率)을 얻을 수 있다. 3) AsA 농도(濃度)가 1mg%이상(以上)인 시료(試料)에 대(對)하여 GLC에 의한 미량정량(微量定豊)을 확립(確立)하여 특(特)히 방해물질(妨害物質)이 많은 일반식품(一般食品)의 정량(定量)에 적용(適用)이 가능(可能)하도록 하였다. 4) 본실험(本實驗)에 사용(便用)한 일본전자(日本電子)의 750형(型)의 eletron meter의 성능(性能)보다 감도(感度)가 300배(倍)인 20K 형(型)을 사용(使用) 한다면 혈청중(血淸中) AsA의 정량(定量) 및 그 이하(以下)의 농도(濃度)인 미량정량(微量定量)도 충분(充分)히 가능(可能)하리라 생각된다.

돼지장에서 분리한 Lactobacillus salivarius의 생균제로서 특성 (Probiotic Properties of Lactobacillus salivarius Isolated from Piglet Intestines)

  • 돼지장에 존재하는 많은 균총들로부터 생균제를 분리하기 위하여 생균제로서 필수 조건인 내산성, 내담즙산, 유해균 억제능력들을 조사하였는데 그 결과 다른 균들에 비해 생균제로서 작용 능력이 뛰어난 한 균주인 PA10을 얻을 수 있었다. 생균제로서 중요한 기능인 설사 유발 대장균과 Salmonella를 억제하기 위하여 돼지의 장으로부터 내산, 내담즙의 특성을 가진 미생물틀을 분리 검색하였다. 이들 중, 위산과 담즙에 잘 견디며, 대장균 및 Salmonella에 대한 억제 능력이 뛰어난 한 미생물을 분리하였는데 Lactobacillus salivarious로서 동정되었다. 이미 생물은 pH 3의 위산과 0.3% 담즙산에서 각각 2시간, 24시간 동안 약 50~70%의 생존률을 나타냈으며, 대장균 또는 Salmonella를 MRS배지에서 혼합 배양시 24시간안에 이들 유해균을 완전히 제거할 수 있었다. 이 실험에서 L salivarius로 명명된 이 균은 돼지장에 서식하는 수 많은 미생물중 내산, 내담즙산, 유해균 억제능을 동시에 만족 시킬 수 있는 특성을 가지는 균주임을 알 수 있었다. 앞으로 균주 보존 조건을 확립하고, 동물 실험에서도 유사한 결과를 얻을 수 있다면 이 분리균은 생균제로서 유용하게 이용될 수 있을 것으로 생각된다.

  • 토양(土壤) 및 하수(下水)에서 단백분해활성(蛋白分解活性)이 약하고 응유활성(凝乳活性)이 강한 SA-101 균주(菌株)를 분리(分離), 선정(選定)하여 Mucor sp. 동정(同定)하였다. 사용균주(使用菌株)에 의한 응유효소생산(凝乳酵素生産)은 밀기울 10 g에 0.1M HCl 을 함유한 수도수 8 ml를 가하여 $30^{\circ}C$에서 60시간의 배양(培養)이 가장 좋았다. Lactose 6%, NaCl 0.05%를 첨가(添加)하였을때 효소생산(酵素生産)이 촉진(促進)되었고 밀기울 g당(當) 9,810 단위(單位)의 효소생산(酵素生産)을 보였다. 유안염석(硫安鹽析)과 동결건조(凍結乾燥)등으로 조제한 조효소제품(粗酵素製品)의 MCA/PA비(比)는 Hansen rennet 보다 낮았으나 Meito rennet과 유사하였고 응유활성(凝乳活性)의 최적온도(最適溫度)는 1/100M $CaC1_2$를 함유한 10% 환원탈지유(還元脫脂乳)를 기질로 하였을 때 $60^{\circ}C$ 부근이었다.

종이의 산성화가 미생물의 분해능에 미치는 영향 (Effect of the paper acidity on the cellulolytic activity of fungi)

  • The effect of pH on degradation of paper by some fungi, which able to degrade cellulose, was investigated. Trichoderma koningii, Aspergillus nigerand Penicillium nigulosum were cultured at $28^{\circ}C$ for 16 days in the selective medium (PH3, PH4, PH5, PH6, PH7, PH8, PH9, PH10, PHC) containing paper as substrate. Each paper was pretreated with each pH buffer (pH 3∼pH 10, D.W.)prior to addition to the selective medium. Enzyme activities in the each culture medium were measured spectroph to metrically using C.M.C., Avicel, PNPG as the substrates for endoglucanase, exoglucanase and $\beta$-glucosidase, respectively. In all experimental fungi, the enzyme activities of PH3 and PH9 medium were usually much higher than those of other experimental groups. However in the PH6medium, enzyme activity was lower than other groups. To analyze the concentration and pattern of protein in the each culture medium, the medium was concentrated by lyophilization. The protein concentration of PH3 and PH9 medium were relatively high (T.koningii; 6.31mg, 6,19mg, A.niger; 1.62mg, 1.96mg, P.nigulosum;2.50mg, 2.73mg, respectively), but that of PH6 was relatively low. The protein pattern of each medium was analyzed by using SDS-PAGE and VDS Image Master Analysis Program. The concentrations of bands in the each lane were usually high at lane2 (PH3) and lane8 (PH9) and low at lane5 (PH6). Therefore, the incresed cellulolytic activity of fungus against acidified paper could be result of structural change and deterioration of paper caused by being acidified.

Effect of Multi-walled Carbon Nanotube Dispersion on the Electrical, Morphological and Rheological Properties of Polycarbonate/Multi-walled Carbon Nanotube Composites

  • The effect of a multiwalled carbon nanotube (MWCNT) dispersion on the electrical, morphological and rheological properties of polycarbonate (PC)/MWCNT composites was investigated, with and without pretreating the MWCNTs with hydrogen peroxide oxidation and lyophilization. The resulting PC/treated MWCNT composites showed higher electrical conductivity than the PC/untreated MWCNT composites. The morphological behavior indicated the treated composites to have greater dispersion of MWCNTs in the PC matrix. In addition, the electromagnetic interference shielding efficiency (EMI SE) of the treated composites was higher than that of the untreated ones. Rheological studies of the composites showed that the complex viscosity of the treated composites was higher than the untreated ones due to increased dispersion of the MWCNTs in the PC matrix, which is consistent with the electrical conductivity, EMI SE and morphological studies of the treated composites. The latter results suggested that the increased electrical conductivity and EMI SE of the treated composites were mainly due to the increased dispersion of MWCNTs in the PC matrix.

Improving the Viability of Freeze-dried Probiotics Using a Lysine-based Rehydration Mixture

  • The probiotic market is constantly continuing to grow, concomitantly with a widening in the range and diversity of probiotic products. Probiotics are defined as live microorganisms that provide a benefit to the host when consumed at a proper dose; the viability of a probiotic is therefore of crucial importance for its efficacy. Many products undergo lyophilization for maintaining their shelf-life. Unfortunately, this procedure may damage the integrity of the cells due to stress conditions during both the freezing and (vacuum-) drying process, thereby impacting their functionality. We propose a lysine-based mixture for rehydration of freeze-dried probiotics for improving their viability during in vitro simulated gastric and duodenum stress conditions. Measurement of the zeta potential served as an indicator of cell integrity and efficacy of this mixture, while functionality was estimated by adhesion to a human enterocyte-like Caco-2 cell-line. The freeze-dried bacteria exhibited a significantly different zeta potential compared to fresh cultures; however, this condition could be restored by rehydration with the lysine mixture. Recovery of the surface charge was found to influence adhesion ability to the Caco-2 cell-line. The optimum lysine concentration of the formulation, designated "Zeta-bio", was found to be 0.03 M for improving the viability of Lactiplantibacillus plantarum Lp-115 by up to 13.86% and a 7-strain mixture (400B) to 41.99% compared to the control rehydrated with distilled water. In addition, the lysine Zeta-bio formulation notably increased the adherence ability of lyophilized Lp-115 to the Caco-2 cell-line after subjected to the in vitro stress conditions of the simulated gastrointestinal tract passage.

베타카로틴 함유 키토산/하이알루론산 나노캡슐의 용해도 및 재분산성 특성 (Properties of β-carotene-loaded chitosan/hyaluronic acid nanocapsules: solubility and redispersibility)

  • β-Carotene의 용해도를 증진시키기 키토산을 기본 피복물질로 사용하면서 TPP와 HA를 단독 또는 복합적으로 사용하여 세 종류(CS/TPP, CS/TPP/HA와 CS/HA)의 나노캡슐을 제조하였다. 모든 종류의 나노캡슐에서 95%가 넘는 높은 포집효율이 관측되었다. β-Carotene 에멀젼의 저장안정성이 취약한 pH 2-3 조건에서 8일동안 저장하였을 때 CS/TPP/HA와 CS/HA 나노캡슐의 물리적 특성뿐만 아니라 β-carotene의 용해도 또한 안정적으로 유지되는 경향을 나타냈다. β-Carotene의 용해도를 나노캡슐의 건조 전과 후 관측한 결과, 대조군에 비해서 나노캡슐화에 의해서 유의적으로 향상된 것을 확인할 수 있었다. 특히 CS/HA 나노캡슐은 용해도와 재분산성의 측면에서 모두 우수한 특성을 나타냈다. 본 연구를 통해서 CS/HA 나노캡슐은 β-carotene과 같은 난용성 활성성분의 용해도를 증진시킬 수 있는 식품산업에 활용가능한 전달체로서 이용 가능성이 높을 것으로 판단된다.
