Development of Nomographs for the Evaluation of Lighting Energy Performance in a Semi-infinite Office Space (중규모 사무공간에서 조명에너지 성능평가를 위한 노모그래프 개발에 관한 연구)

  • The purpose of this study was to analyze daylighting performance in a semi-infinite size office space for lighting energy conservation. DOE2.1E was used for simulations for the model space of $12\times12\times2.6m$. Nomographs were developed which could simulate work plane illuminance, glare index, energy consumption rate and energy reduction rate for daylighting design. Major results of simulations are as follows ; 1) When blinds facing south were installed, 43% of workplane illuminance diminished, but the flare index didn't exceed the recommended max-glare value. 2) In a semi-infinite office space facing south. energy consumption rate in the case space of 500 lux workplane illuminance is larger then case space of 300 lux workplane illuminance. Therefore, energy reduction rate is increased when the semi-infinite office faces south and naintains 300 lux workplane illuminance level.

Interaction Between the Quorum Sensing and Stringent Response Regulation Systems in the Enterohemorrhagic Escherichia coli O157:H7 EDL933 Strain

  • Quorum sensing and the stringent response are well-known regulation systems for the expression of virulence genes in enterohemorrhagic Escherichia coli (EHEC). However, how these two systems interact is not well known. E. coli strains with mutations in two regulation systems, ${\Delta}luxS$ (ECM101) and ${\Delta}luxS{\Delta}relA{\Delta}spoT$ (ECM201), and the ${\Delta}luxS$ complement strain to ECM201 (ECM202) were created from EHEC O157:H7 EDL933 to investigate how the regulatory systems interact. The phenotypic changes of the mutant strains were characterized and compared with the wild type. The mutant strains exhibited no obvious growth defects, although acid resistance and cellular cytotoxicity were decreased significantly in all the mutant strains. Phenotypic characterization revealed that mutations in the stringent response system (ECM201 and ECM202) influenced the metabolic (defective utilization of arabinose and L-sorbose) and enzymatic activities (decreased trypsin activity, and increased ${\alpha}$-glucosidase activity). In contrast, the quorum sensing system mutant (ECM101) did not display these phenotypes. The motility of the quorum sensing system mutant (ECM101) was unchanged, but mutation in the stringent response system influenced the motility. Our results suggest that quorum sensing interacts with the stringent response regulation system.

  • In this study, the amino-terminal half truncated lump and the whole lump genes from Photobacterium phosphoreum coding for the lumazine protein were cloned by polymerase chain reaction and expressed in Escherichia coli. To identifiy of the binding site of the ligand or substrate, the amino acid identities from the sequences of the lumazine protein, yellow fluorescent protein, and riboflavin synthase from different organisms were also compared and analyzed.

The Study on Environmental Sanitation for Book-lending Shops in Inchon Area (일부지역 대본업소의의 환경위생학적 조사 (조명과 CO를 중심으로))

  • This survey was carried out for a month from Jan. 15th, 1979 to Feb, 15th, 1979 to study the sanitary environments of the "book-lending" shops (comic-book shops) in Inchon area, and the special emphasis was placed on their illumination and carbon monoxide gas, which plays an important part in the whole sanitary environment. 120 shops surveyed were chosen at random from 4 administrative districts, 30 shops from one district respectively. The results of the survey were as follows:1) The average space of the book-lending shops (63.3%) was 2.0~3.9 pyung, at 21 number of shops (17.5%) was 1.0~1.9 pyung, at 53 number of shops (10.8%) was 4.0~5.9 pyung, at 9 number of shops (7.5%) was 6.0~7.9 pyung. 2) The ages of 42.8 percent of the whole 888 customers range from 10 to 14 and those of 36.5 percent from 15 to 19, and those of 11.9 percent from 20 to 24, and 89.2 percent were male and 10.8 percent were females and the percent of spending time in shops was shown 10.2% in under 1 hr, 25.4% in 1 hr to 2 hrs, 46.5% in 2 hrs to 3 hrs and 17.8% in over 3 hrs. 3)The CO pollution was quite different by shops where the ventilator was equipped and where not equipped. In the shops equipped shown 36 shops (80.8%) and non-equipped shown 65 shops (86.7%), the pollution ratio of all the shops (120 shops) was 84.2 percent. 4) The CO Concentration of the ventilator equipped shops was shown 44.5 percent in 100~149 ppm, 150~199 ppm shown 33.3 percent and 22.2 percent was 200~250 ppm, In the non-equipped shops the CO concentration was shown 36.9 percent was 150~199 ppm and 200~250 ppm was 25.6 percent. The totally was 39.6 percent in 100~149 ppm. 5) 37.5 percent of the book-lending shops had the ventilation facilities but 62.6 percent had no ventilators. 6) The maxmum average illumination of the shops was 38.5 Lux and the minimum average illumination was 14.1 Lux, and the mean average illumination S.D. was 24.6\pm38.4. 7) 35.8 percent of the shops had the illumination of 20~29 Lux and 8.3 percent had that of more than 40 Lux. In other words 91.7 percent had the illumination of less than 40 Lux.

Sawdust Cultivation of Lentinula edodes Using a Detachable Plastic Bottle (분리형 용기를 이용한 표고버섯 톱밥재배 연구)

  • Currently, plastic bags are being used for sawdust cultivation of shiitake mushroom. However, due to serious environmental problems caused by the use of plastic bags, we studied the efficacy of bottle cultivation method to replace the sawdust bag method. Small detachable plastic bottles (400 g capacity) filled with Quercus spp. sawdust and wheat bran (4:1 w/w) media were incubated for 80 and 120 days. The weight loss (%) of the media was higher for the NIFoS 2464 strain at an approximate light intensity of 300 Lux than light intensity of 500 Lux; the light intensity was associated with the loss of sawdust medium-weight during the cultivation period. The highest yield was observed when the strain was cultivated for 80 days under dark conditions, 40 days under 500 Lux light, and air circulation fan speed of 30 rpm. When incubated for 120 days, mushroom yield in the bottle media was higher at 40 days of light exposure than 20 days of light exposure. In the bottle media incubated for 80 days under dark conditions, the mushrooms fruited due to repetitive water spraying on the top of the media and light stimulation during the fruiting period. The media could be separated from the bottles because the media shrank after the first harvest. These separated plastic bottles could be re-used for mushroom cultivation, thereby reducing the amount of plastic waste.

Red Color Light at Different Intensities Affects the Performance, Behavioral Activities and Welfare of Broilers

  • Red light (RL) marked higher weight gain (WG) and preference of broilers compared to other light colors. This study aimed to investigate how different intensities of RL affect the performance, behavior and welfare of broilers. RL treatments were T1 = high intensity (320 lux), T2 = medium intensity (20 lux); T3 = dim intensity (5 lux), T4 = control/white light at (20 lux) provided on 20L:4D schedule and T5 = negative control; 12 hours dark: 12 hours day light. Cobb strain broilers were used in a Complete Randomize Design with 6 replicates. WG, water/feed intake, feed conversion ratio (FCR), mortality, behavior and welfare were assessed. At 35 d, significantly (p<0.05) highest body weight ($2,147.06g{\pm}99$) was recorded by T3. Lowest body weight ($1,640.55g{\pm}56$) and FCR (1.34) were recorded by T5. Skin weight was the only carcass parameter showed a significant (p<0.05) influence giving the highest (56.2 g) and the lowest (12.6 g) values for T5 and T1 respectively. Reduced welfare status indicated by significantly (p<0.05) higher foot pad lesions, hock burns and breast blisters was found under T3, due to reduced expression of behavior. Highest walking ($2.08%{\pm}1%$) was performed under T1 in the evening during 29 to 35 days. Highest dust bathing ($3.01%{\pm}2%$) was performed in the morning during 22 to 28 days and highest bird interaction (BI) ($4.87%{\pm}4%$) was observed in the evening by T5 during 14 to 21 days. Light $intensity{\times}day$ $session{\times}age$ interaction was significantly (p<0.05) affected walking, dust bathing and BI. Light intensity significantly (p<0.05) affected certain behaviors such as lying, eating, drinking, standing, walking, preening while lying, wing/leg stretching, sleeping, dozing, BI, vocalization, idling. In conclusion, birds essentially required provision of light in the night for better performance. Exposed to 5 lux contributed to higher WG, potentially indicating compromised welfare status. Further researches are suggested to investigate RL intensity based lighting regimen that favors for both production and welfare of Broilers.

Habitat Characteristics and Distribution of Cymbidium kanran Native to Jejudo, Korea (제주한란의 자생환경 특성 및 분포에 관한 연구)

  • This work carried out to define the characteristic of Cymbidium kanran habitat at Mt. Halla in Jejudo, Korea from the ecological point of view including geological and topographical features, air and soil temperature, relative humidity, fluctuations of light intensity, habitat vegetation, distribution altitude, area limit, and etc. And another goal of this study consider the conservation counterplan of the cymbidium habitat. Natural distribution areas of the cymbidium were observed more abundantly on the well drained south and east-facing slopes. Soil acidity was ranged from pH 4.1 to 5.3, and electric conductivity was ranged from 176.4 to 299.9 us/cm (average 215.3 us/cm). Base-saturation percentage of the habitat soil was below 50%, bulk density 0.42g/$cm^2$, particle density 2.05g/$cm^2$, humus content 26%, total nitrogen 0.82%, available phosphate 4.2 mg/kg, exchangeable potassium 0.63 Cmol/kg, calcium 0.44 Cmol/kg and magnesium 0.67 Cmol/kg. Annual mean air temperature was $15.4^{\circ}C$, however, air temperature was ranged $11.7{\sim}18.2^{\circ}C$ in spring, $21.2{\sim}23.8^{\circ}C$ in summer, $12.8{\sim}22.0^{\circ}C$ in fall and $5.5{\sim}7.8^{\circ}C$ in winter season. Annual mean soil temperature at depth of 10cm was $13.2^{\circ}C$ And minimum value was recorded $4.7^{\circ}C$ on January, and maximum value $22.5^{\circ}C$ on August. Relative humidity was ranged 90.8~94.7% in summer, 80.8~91.5% in fall and 77.6~84.2% in winter season. Minimum value was 56.5% on December, and maximum value was 100% on July and August. Light intensities were ranged from 400 to 1,800 lux at the greater part of Cymbidium kanran sites in Jejudo. Summer regarded as an lower light intensities was recorded to be range of 500~600 lux; however, autumn and winter were shown higher light regimes ranged from 3,500 to 3,800 lux. Therefore, one must be suprised that the cymbidium grow at the light condition of 6 lux (minimum) or 10,000 lux (maximum). Tree species keeping higher frequency rate and density were Eurya japonica, Camellia japonica, Castanopsis cuspidata, Carpinus laxiflora and Pinus densiflora. Number of trees growing in a 5${\times}$5m quardrat was 35 as an average, and proportion of evergreen versus deciduous was 5:1. Distribution altitude of the orchid habitat was ranged from 120m (low) to 840m (high) from sea level on the south facing slope of Mt. Halla, and was ranged eastern borderline of Gujwaup, Bukjejugun to western boundaries of Jungmundong, Seogwipo city. For the stable conservation of Cymbidium kanran habitat, sunlight regimes must be increased more by means of cutting trees or twigs in the site.

Comparison of Photosynthetic Rates among Panax Species and Cultivars (인삼의 종 및 품종간 광합성 특성비교)

  • The study was carried out to investigate the effects of light intensity, temperature and seasonal trends on the photosynthesis as well as the physiological characteristics of Panax species and cultivars. Four-year-old plant of Violet-stem variant, Yellow-berry variant, Mimaki and Russian ginseng of Panax ginseng C. A. Meyer and American ginseng of Panax quiquefolium L. were used for study. These Panax species and cultivars were cultivated under the straw shading. The light saturation point of leaves in Violet-stem variant, Yellow-berry variant, Mimaki and American ginseng were 15,000 lux, but that of Russian ginseng was 10,000 lux. The optimum air temperature on the phtosynthesis of Violet-stem variant, Yellow-berry variant, Mimaki and American ginseng were 20$^{\circ}C$ but that of Russian ginseng was 15$^{\circ}C$ under 15,000 lux. The photosynthetic rates were increased in order of Russian ginseng, Mimaki, Yellow-berry variant, Violet-stem variant and American ginseng. The respiration rates of ginseng leaves were increased according to the increasing of temperature, but it was not different among Panax species and cultivars. Stomata frequency of American ginseng was highest, that of Russian ginseng lowest, while the length of stomata was reverse. The total chlorophyll content of American ginseng and specific leaf weight of Mimaki were higher than other ginseng cultivars. The root weight of American ginseng was heavier than others, but that of Russian ginseng was lighter. The num ber of leaflets per plant of 2-year-old plant and the root weight of 6-year-old plant were increased in order of Russian ginseng, American ginseng, Mimaki, Yellow-berry variant and Violet-stem variant.

Conditions for Stable light Production of Recombinant Escherichia coli Containing Lux Operon and Sensitivity to Toxic Chemicals (Lux operon을 함유한 유전자 재조합 Escherichia coli의 발광 안정화 조건 및 독성물질에 대한 민감성)

  • Recombinant E. coli DH5 ${\alpha}$/pSB311 was made by cloning the genes encoding bacterial luciferase and aldehyde substrate proteins from Photohabdus luminescense, to complement defects of Lumistox, which is normally used in bioassays to monitor toxic substances in water environmental systems. The conditions for stable light production by the recombinant strains were investigated with respect to cell growth stage, cell number, and buffer conditions. The optimum growth stage was a middle-exponential stage with an OD$_{660nm}$ value of 0.6-0.7. ADout 10$^{6}$-10$^{7}$ cells per test tube was optimum for stable light emission. The effect of buffer was not significant if an optimum viable cell number was maintained. The bioluminescence of the recombinant E. coli harboring the lux operon of Photohabdus luminescense was not affected by temperature, while the bioluminescence of Lumistox was temperature sensitive. The recombinant E. coli was more sensitive to heavy metals (Cd, Cu, Hg, Zn) than Lumistox, because it does not require high concentrations of NaCl in the buffer.

Rooting Performance Using Cuttings and Analysis of Light and Soil Environmental Characteristics for Indoor Plants of Winter Daphne (Daphne odora Thunb) (서향의 삽목번식 방법과 실내도입을 위한 광, 토양에 관한 연구)

  • This research was done to identify the mass propagation method in winter daphne (Daphne odora Thunb) using its softwood cuttings and to investigate its plant characters established at different light and soil conditions as an indoor plant. Cuttings from winter daphne were taken and grown in different treatment consisted of rooting media (perlite, vermiculite, perlite + vermiculite (1 : 1) and commercial horticulture media soil), indole butyric acid (IBA) hormone concentrations (0, 100, 500, 1000 ppm and Rootone) and date of cutting. Transplants were grown at different light intensities (100, 1000, 2500 lux and control) and growing media. Results showed that cuttings grown in perlite + vermiculite (1 : 1) gave higher percentage (100%) rooting. Cuttings treated with Rootone and IBA 100 ppm showed good rooting growth and cutting taken in June, 25 gave the highest rooting (96.7%). The best plant growth obtained at 1000 lux (56~60 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) followed by 2500 lux (125~130 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) and scoria mixed with commercial horticulture media soil showed better growth of transplants.
