The role of calcium in the production of oxygen radical which causes reperfusion damage of ischemic heart has been examined. The reperfusion damage was indrced in isolated Langendorff perfused rat hearts by aortic clamping for 60 min followed by reperfusion with oxygenated Krebs-Henseleit solution with or without 1.25 mM $CaCl_2.$ On reperfusion of the ischemic hearts with the calcium containing solution, the release of cytosolic enzymes (LDH and CPK) increased abruptly. These increased release of enzymes were significantly inhibited by additions of oxygen radical scavengers (SOD, 5,000 U; catalase, 12,500 U) into the reperfusion solution. In the hearts isolated from rats pretreated with allopurinol(20 mg/kg orally, 24 hr and 2 hr prior to the experiments), the levels of enzymes being released during reperfusion were significantly lower than that of the control. However, in the hearts perfused with the calcium-free but oxygenated solution, the increase in the release of cytosolic enzymes during reperfusion was neither inhibited by oxygen radical scavengers nor by allopurinol pretreatment. For providing the evidence of oxygen radical generation during the reperfusion of ischemic hearts in situ, the SOD-inhibitable reduction of exogenously administered ferricytochrome C was measured. In the hearts perfused with the calcium containing solution, the SOD-inhibitable ferricytochrome C reduction increased within the first minute of reperfusion, and was almost completely inhibited by allopurinol pretreatment. When the heart was perfused with the calcium free solution, however, the reduction of ferricytochrome C was not only less than that in the calcium containing condition, but also was not so completely inhibited by allopurinol pretreatment. By ischemia, xanthine oxidase (XOD) in the ventricular tissue was changed qualitatively, but not quantitatively. In the heart made ischemic with the calcium containing condition, the oxygen radical producing O-form of XOD increased, while the D- and D/O-form decreased. However, in the ischemic heart reperfused with the calcium free condition, the D/O-form of XOD was elevated without significant increase in O-form of the enzyme. It is suggested from these results that the calclum may play a contributing role in the genesis of reperfusion damage by promoting the conversion of xanthine oxidase from the D/O-form to the oxygen radical producing O-form in the ischemic myocardium.
본 연구는 배암차즈기를 건조, 데침 및 발효에 따라 항염증 효능성분인 rosmarinic acid, homoplantaginin, luteolin의 함량 변화와 항산화 활성을 조사하였다. Rosmarinic acid는 열풍건조 시 16.42 mg/g, 데친 후 열풍건조 시 10.19 mg/g 순으로 검출되었으며, 동결건조 한 잎과 뿌리는 유의적인 차이가 없었다. 음건 및 동결건조 시에는 각각 8.69와 8.15 mg/g으로 유의성이 인정되었으며, 발효 후에는 0.05 mg/g으로 감소하거나 검출되지 않았다. 동결건조, 음건 및 열풍건조 한 배암차즈기는 $500{\mu}g/mL$의 농도에서 98.5% 이상의 ABTS radical 소거능을 나타내었으며, 데친 후 열풍건조 시에도 87.3%의 우수한 radical 소거능을 나타내었다. 뿌리는 잎에 비하여 ABTS radical 소거능이 낮았다. SOD 유사활성은 $1,000{\mu}g/mL$ 농도에서 6.1~27.8%로 측정되어 ABTS radical 소거능과 차이가 있었다. 배암차즈기에 함유되어 있는 주요 물질이며 항염증 효능 물질로 알려진 rosmarinic acid와 homoplantaginin은 차발효에 의해 거의 소실되기 때문에 배암차즈기를 기능성 차 또는 가공식품 원료로 개발하기 위해서는 차발효(산화발효)보다 동결, 냉풍 및 열풍건조가 적당할 것으로 판단된다.

It has been well known that extended exposure to reactive oxygens causes severe damage to susceptible biomolecules. In this study, the effects of flavonoids including trifling and kaempferol from Ginseng leaves on single oxygen induced photohemolysis of erythrocytes and free radical scavenging activities were investigated . Each flavonoid aglycone (5-50UM) such as kaempferol, quercetin or baicalein exhibited a high protective effect against the photohemolysis. They protected the cells by scavenging 102 and free radicals. Although the free radical scavenging activities of the flavonoid glycosides were not much lower than those of their corresponding aglycones, their insolubility into lipid bilayers of membrane made them less effective in preventing the photohemolysis induced by 1O2. The 102 and free radical scavenging activities of flavonoids were estimated by the decomposition of the flavonoid by 1O2 and the bleaching of free radicals by the flavonoid, respectively. The solubilization of the flavonoid into micelle or erythrocytes was deduced from spectrophotometric and microscopic observations. The cooperation of L-ascorbic acid and a flavonoid, and a possible involvement of lipoxygenase or cyclooxygenase in the photohemolysis mechanism were discussed. This study was carried out to investigate the effect of Pine needle extract on lipid oxidation and free radical reaction in iron sources reacted with active oxygen species. The results were summarized as follow; The pine needle extracts didn`t show a distinct effect on reduction of lipid oxidation if the iron ion didn`t exist in oil emulsion. The pine needle extracts played role as a strong chelating agents to bind iron ion if Ferrous iron(Fe\ulcorner) exist in oil emulsion. Ferric iron(Fe) was lower effect than Ferrous iron(Fe) on free radical reaction in oil emulsion. And also, the Fe\ulcorner reacted with pine needle extract did not show distinct effect on free radical reaction, compared to Fe\ulcorner reacted with pine needle extract. And also, Pine needle extracts reacted with H\ulcornerO\ulcorner were tended to show a low oxygen scavenging ability in case of H\ulcornerO\ulcorner only was existed, compared to those of H\ulcornerO\ulcorner + Fe\ulcorner complex. Pine needle extracts were the most powerful Fe\ulcorner binding agents, compared to other strong synthetic antioxidants such as EDTA and DTPA.
In order to examine oxidative stress of reactive oxygen species and the antioxidant effect of Citri Reticulatae Pericarpium (CRP) extract, human skin melanoma cells were treated with various concentrations of hydrogen peroxide ($H_2O_2$). Antioxidant effect of CRP extract on $H_2O_2$-induced cytotoxicity, cell viability, DPPH-radical scavenging activity and superoxide dismutase (SOD)-like activity. In this study, $H_2O_2$ decreased cell viability of cultured human skin melanoma cells in dose- and time-dependent manners, and then, midcytotoxicity value (MCV) was determined at $60\;{\mu}M$ after human skin melanoma cells were cultured for 5 hours in the media containing $20{\sim}60\;{\mu}M$ of $H_2O_2$, respectively. The $H_2O_2$ was on cultured human skin melanoma cells because MCV of $H_2O_2$ was lower than $100\;{\mu}M$. In the antioxidant effect of CRP extract, CRP extract increased cell viability DPPH-radical scavenging activity and SOD-like activity. From these results, it is suggested that $H_2O_2$ was very toxic on cultured human skin melanoma cells. And also, CRP extract has the antioxidant effect on $H_2O_2$-induced cytotoxicity.

To evaluate the effect of occlusive skin on the activity of cutaneous oxygen free radical metabolizing enzymes in rats, the dorsal skin was covered with closed glass chamber shaped petri dish, 46 mm in diameter and 10 mm in height and sealed by an adhesive. Five day-occluded group showed more increased activity of xanthine oxidase (XO) than that of control, and the activity of five day-occluded group was higher than that of ten day-occluded group. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx) were significantly higher in ten day-occluded group than in control or five day-occluded group. All the more, five day-occluded group showed the decreasing tendency of SOD and GPx activities compared to those of control. On the other hand, the cerrous perhydroxide deposits were observed in the intercellular space of the stratum basale in five day-occluded group under the electronic microscope using a cytochemistry method. Futhermore, the degree of cerrous perhydroxide reaction was lower in ten day-occluded group than in five day-occluded group. In conclusion, the increased XO activity and the decreased SOD and GPx activities are likely to responsible far the accumulation of $H_2O_2$ in five day-occluded group.
Photosensitive resin of azide type is good for resolution and inner solvent, but it is really problem to development of practical use because fanctional groups of polymer has many hydrophilic radicals. By careful attention to this point, this study was investigated synthesis term, photo property and development property of composed photosensitive resin of azida type, it is to this effect. 1) H-NMR spectrum of compared DABCI showed amion redical by $\delta$6.0~6.1ppm to substitude for azide radical by amino radical by $\delta$8.9~9.45ppm, and FT-IR absorption spectra showed the absorption bends at 2100cm. 2)FT-IR absorption spectra of PHS1-DAB, PHS2-DAB, CMM-DAB and CHM-DAB showed azida radical pick to be lost at after irradiation by UV light. 3) According to exposuer change of PHS1-DAB, PHS2-DAB, CMM-DAB and CHM-DAB, absorption maximum value of UV spectrum change was 280nm. 4) to compared relative sensitivity of compared photosensitive resin, PHS2-DAB was the best and to compared insolubility rate of compared photosensitive resin, CMM-DAB was the lower. 5)Solubility if NaOH was the best by 1.0mol/$\ell$ and solubility of developing solution of ethanol to water was it in the ratio of 4 to 1.
Water extract from Pinus densiflora (WPD) was investigated for its antioxidant activity and its ability to provide protection from DNA damage. A series of antioxidant assays, including a 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging assay, a reducing power assay, a metal-chelating assay, a superoxide radical scavenging assay, and a nitrite scavenging ability, as well as a DNA damage protection assay were performed. Total phenolic content was found to be 211.32 mg Tan/g WPD. The extract scavenged 50% DPPH free radical at a concentration of 21.35 ${\mu}g/mL$. At that same concentration, the reducing power ability of WPD was higher than that of ${\alpha}$-tocopherol. The extract chelated 68.9% ferrous ion at the concentration of 4 mg/mL. WPD showed better nitrite scavenging effect at the lower pH. Meanwhile, WPD exhibited a strong capability for DNA damage protection at 1 mg/mL concentration. Taken together, these data suggest water extract from Pinus densiflora could be used as a suitable natural antioxidant.
The results of reconstruction of the lower esophagus with jejunum in a total of 24 cases of primary carcinoma of the lower third esophagus and gastroesophageal carcinoma were presented, and clinical values of substitution for the esophagus with jejunum were also discussed. They were operated in the department of thoracic and cardiovascular surgery, Hanyang University Hospital during the period of 9 years from 1972 to 1981. Surgical managements to lower esophageal reconstruction with jejunum were carded out with not the same procedure in all cases studied, but with three different procedure mentioned below/ In 13 cases of lower third esophagectomy with or without partial `8astrectomy of a total of 24 cases, interposition of jejunum between the esophagus and the stomach were performed after the fashion to esophagojejunostomy with mobilized jejunal loops and 8astro-JeJunostomy with side to side anastomosis. In 7 cases of lower third esophagectomy and total gastrectomy, the continuity of the esophagus were performed the fashion to esophagojejunostomy with mobilized jejunum. In 4 cases of unresectable gastro-esophageal carcinoma, bypass operation of the lower esophagus and the stomach were performed after the fashion to esophagojejunostomy with side to and anastomosis. After the bypass operation, it was observed that oral feeding to the patients was excellent. Following these consecutive series of 20 cases of radical operation for lower esophageal carcinomas and 4 cases of bypass operation for unresectable gastroesophageal carcinomas, no complication such as postoperative leakage and stenosis from anastomotic site or Infection In operating area and operative death were observed.
