약용식물의 Peroxynitrite와 Hydroxyl radical 소거 활성 (Peroxynitrite and Hydroxyl Radical Scavenging Activity of Medicinal Plants)

  • 약용식물 9종의 물 추출물에 대한 항산화 효과를 탐색하기 위해 in vitro 조건에서 강력한 산화력과 독성을 갖는 $ONOO^-$${\cdot}$OH radical의 소거효과를 측정하였다. 그 결과, 감잎, 구기자, 꾸지뽕나무, 담쟁이덩굴, 마디풀, 비파나무, 으름덩굴, 참취 추출물은 10${\mu}g/ml$ 농도에서 50% 이상의$ONOO^$ 소거효과를 나타냈다. 이러한 $ONOO^-$ 소거효과는 양성 대조물인 penicillamine(94.08${\pm}$3.04)에 비해 낮았지만 비파나무 물 추출물(89.87${\pm}$4.57)이 다른 시료에 비해 가장 강력한 $ONOO^-$ 소거효과를 나타냈다. 또한, 가는잎쐐기풀과 감잎 물 추출물은 1mg/ml 농도에서 양성대조물인 thiourea 보다 효과적인${\cdot}$OH radical 억제효 과를 나타냈다. 결론적으로 9종의 약용식물은 $ONOO^-$${\cdot}$OH radical과 연관된 산화적 스트레스에 의한 세포와 조직의 손상을 억제시킬 수 있는 천연 항산화제로 밝혀졌다.

해수의 염 농도와 탁도가 전기, UV 및 전기+UV 공정의 Artemia sp. 불활성화에 미치는 영향 (Effect of Salt Concentration and Turbidity on the Inactivation of Artemia sp. in Electrolysis UV, Electrolysis+UV Processes)

  • This study was conducted to investigate the effect of salt concentration and turbidity on the inactivation of Artemia sp. by electrolysis, UV photolysis, electrolysis+UV process to treat ballast water in the presence of brackish water or muddy water caused by rainfall. The inactivation at different salt concentrations (30 g/L and 3 g/L) and turbidity levels (0, 156, 779 NTU) was compared. A decrease in salt concentration reduced RNO (OH radical generation index) degradation and TRO (Total Residual Oxidant) production, indicating that a longer electrolysis time is required to achieve a 100% inactivation rate in electrolysis process. In the UV process, the higher turbidity results in lower UV transmittance and lower inactivation efficiency of Artemia sp. Higher the turbidity resulted in lower ultraviolet transmittance in the UV process and lower inactivation efficiency of Artemia sp. A UV exposure time of over 30 seconds was required for 100% inactivation. Factors affecting inactivation efficiency of Artemia sp. in low salt concentration are in the order: electrolysis+UV > electrolysis > UV process. In the case of electrolysis+UV process, TRO is lower than the electrolysis process, but RNO is more decomposed, indicating that the OH radical has a greater effect on the inactivation effect. In low salt concentrations and high turbidity conditions, factors affecting Artemia sp. inactivation were in the order electrolysis > electrolysis+UV > UV process. When the salt concentration is low and the turbidity is high, the electrolysis process is affected by the salt concentration and the UV process is affected by turbidity. Therefore, the synergy due to the combination of the electrolysis process and the UV process was small, and the inactivation was lower than that of the single electrolysis process only affected by the salt concentration.

Quality Characteristics of Jelly Made from Fermented Red Ginseng Concentrate with Increased Ginsenoside Content by Enzyme Treatment

  • The purpose of this study is to investigate the physicochemical properties of jelly made from fermented red ginseng concentrate (FRGC) that can be easily absorbed and digested for the health promotion of the elderly. The pH of the jellies tended to decrease with increasing concentration of FRGC. Soluble solid content has significantly higher value when added more than 2%, and the water content of the sample was significantly lower when the FRGC was added 4%. As the amount of FRGC was increased, the total color difference increased, and the hardness of samples decreased significantly. On the other hand, the total ginsenoside contents of the FRGC was 45.50 mg/g. As the concentration of FRGC increased, the content of polyphenol and flavonoids increased. The increasing pattern of polyphenols and flavonoids showed a similar trend. As the content of FRGC increased, ABTS free radical scavenging activity significantly increased (p<0.05), and in the control, the minimum value (62.6 AEAC) and the 4% sample were highest (116.2 AEAC). DPPH radical scavenging activity was like that of ABTS radical scavenging activity. However, there was no significant difference in DPPH radical scavenging activity of 3% and 4% red ginseng jelly.

천연약용식물의 활성산조종에 의한 산화적 손상의 보호 효과 (Protective Effect of Natural Medicinal Plants against Oxidative Damage Induced by Reactive Oxygen Species)

  • In order to evaluate anti -oxidant activities and protective effect against oxidatve damage, DPPH radical scavenging activity and lipid peroxidation inhibitory activity were measured among methanol extracts prepared from natural medicinal plants. Fourteen natural medicinal plants which were reported to have anti -oxidative or anti-inflammatory effects were selected based on our previous report. In addition to the total methanol extracts, n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions were prepared from each total extract. DPPH radical scavenging assay was performed against 14 total extracts and all samples showed dose-dependent activities in various extent. Among those, 6 samples, methanol extracts of Euryale ferox, paeonia suffruticosa, Areca catechu var. dulcissima, Cinnamomun cassia, Alpinia katsumadai and Betula platyphlla var. japonica showed IC$\sub$50/ value lower than 6.0 $\mu\textrm{g}$/ml. The highest DPPH radical scavenging activity was found in ethylacetate fraction of paeonia suffruticosa with IC$\sub$50/ value of 1.1 $\mu\textrm{g}$/ml. Analysis of lipid peroxidation inhibitory activity on hydrogen peroxide-induced oxidative damage in Chinese hamster lung fibroblast (V79-4) cells revealed that the highest inhibitory effect was observed in methanol extract of Betula platyhpylla var. japonica. Lipid peroxidation inhibitory activity was observed as a dose-dependent manner in all samples used in this study. Among fraction samples, ethylacetate fraction of Alpinia katsumadai had the strongest inhibitory activity with IC$\sub$50/ value of 0.9 $\mu\textrm{g}$/ml.

Nutritional Compositions of Ray and Effects of Steaming on Antioxidative Activities of Ray Hydrolysates

  • This study investigated the effects of steaming on the antioxidative activities of ray hydrolysates in vitro. Based on the results of 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, steamed ray hydrolysate possessed significantly higher antioxidative activities than raw ray hydrolysates (p<0.05). The 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity of steamed ray hydrolysate by pancreatin was slightly lower than raw ray hydrolysate at all concentrations. Raw ray hydrolysate displayed moderate hydroxyl radical scavenging activity, while steamed ray hydrolysate showed a greater increase in hydroxyl radical scavenging activity than raw ray hydrolysate at concentrations above 21.33 mg/mL, and it reached 58.21% at 42.67 mg/mL. The results of this study show that ray hydrolysates have potent free radical scavenging activities and reducing power, and the steaming has a partial impact on the antioxidative activity of ray hydrolysates.

Antioxidant Activity and Irritation Test of Extracts Obtained from Angelica dahurica

  • In this study, we assessed the free radical scavenging and xanthine oxidase inhibitory activities of extracts isolated from the dried roots and stems (including leaves) of Angelica dahurica. The irritation response from these extracts was also assessed to determine potential cosmetic use. Both sources of A. dahurica extracts exhibited radical scavenging properties to different extents. The free radical scavenging potency ($EC_{50}$) of the stems (including leaves) of A. dahurica was 243.33 ${\mu}g/mL$, which is significantly lower (p<0.01) than that observed of the roots (1,161.79 ${\mu}g/mL$). The inhibition values ($EC_{50}$) of xanthine oxidase were as follows: 435.19 ${\mu}g/mL$ (roots) and 434.66 ${\mu}g/mL$ (stems). We noted no significant differences between the two plant parts with regard to ability to inhibit xanthine oxidase activity. After the application of A. dahurica extracts to rabbits for skin and eye irritation tests, no negative effects were observed; therefore, the extracts are considered to be non-irritating to the skin and eye.

흑삼의 항산화 활성 (Antioxidant Activity of Black Panax ginseng)

  • This study was performed to investigate the antioxidant activity of extracts of black Panax ginseng (BGE) and its crude saponin (BGECS). The antioxidant activities of BGE and BGECS were evaluated for free radical scavenging activity against stable free radical (1,1-diphenyl-2-picrylhydrazyl) DPPH, nitrite, hydrogen peroxide and superoxide. In addition, the antioxidant activity of BGE and BGECS against peroxyl radicals, hydroxyl radicals and peroxynitrites were determined by the total oxy-radical scavenging capacity (TOSC) assay. As a result, BGE and BGECS were found to have a strong inhibitory activity with >90% against the DPPH radical at $1000{\mu}g/m{\ell}$ concentrations. Also, BGE and BGECS exhibited strong inhibitory activity with >80% against hydrogen peroxide at lower concentration ($125{\mu}g/m{\ell}$). Moreover, specific TOSC values (405 and 473 TOSC/mM) of BGE and BGECS against peroxynitrites were higher than GSH (347 TOSC/mM) used a positive control. These results suggest that BGE and BGECS could be useful to develop functional foods against disease related oxidative stress.

  • The antioxidative properties of lactobacilli originating from humans (Lactobacillus acidophilus KCTC 3111, Lactobacillus jonsonii KCTC 3141, Lactobacillus acidophilus KCTC 3151, and Lactobacillus brevis KCTC 3498) were investigated using in vitro methods, including inhibition of lipid peroxidation, resistance to hydrogen peroxide and hydroxyl radical, hydroxyl radical scavenging activity, and glutathione peroxidase (GPX) activity. L. acidophilus KCTC 3111 showed the highest inhibition of lipid peroxidation in both intact cells (49.7%) and cell lysate (65.2%). This strain exhibited resistance to hydrogen peroxide and hydroxyl radical, which was viable for 7 h in the concentration of 1.0 mM hydrogen peroxide. In addition, this strain showed high hydroxyl radical scavenging activity. In the GPX activity assay, the highest activity was measured in L. brevis 3498. GPX activity of L acidophilus 3111 was lower than that of L. brevis 3498.

흰쥐에 있어서 구기자 알콜 추출물이 Oxygen Free Radical 및 Alcohol 대사효소 활성에 미치는 영향 (Effect of the Ethanol Extract of Lycium chinense on the Oxygen Free Radical and Alcohol Metabolizing Enzyme Activities in Rats)

  • To investigate an effect of the ethanol extract of Lycium chinense(EELC) on the activities of enzymes scavenging oxygen free radicals or detoxicating alcohol. The ground Lycium chinense was extracted with 30% edible ethanol and then diluted with 6% ethanol to contain 2% EELC(w/v). Three different groups of male Sprague-Dawley rats had taken a drink EELC, ethanol(ETH) or water(control), respectively for 2 months. At the end of experimental period, the animals were sacrificed and obtained the following findings. The EELC-treated animals showed the highest activity of hepatic glucose-6-phosphatase among three groups. The activities of xanthine oxidase and cytochrome p-450 from EELC treatment group were lower than those from ETH-treated group. However, the activity of superoxide dismutase was higher in the EELC-treated group than the ETH-treated(p<0.005). Furthermore, hepatic alcohol or aldehyde dehydrogenase activity, and glutathione content and glutathione peroxidase were significantly higher in EELC-treated animals than in ETH-treated those. The activity of glutathione S-transferase in liver was appeared the orderly higher value in EELC, ETH and control-treated group. As the result, EELC may affect the reduction of oxygen free radical production and help the detoxication of ethanol.

해양오염의 진단을 위한 생화학적 오염지표에 관한 연구 X. 남해산 홍합 (Mytilus coruscus)의 산소 라디칼 및 제거효소의 변화 (Study on Biochemical Pollutant Markers for Diagnosis of Marine Pollution X. Changes in Oxygen Radicals and Their Scavenger Enzymes of the Mussel (Mytilus coruscus) in the South Sea)

  • 해양오염의 진단을 위한 생화학적 오염지표 설정의 기초연구의 일환으로, 오염이 심각한 남해산 홍합(Mytilus coruscus)의 근육중의 활성산소종 및 그들의 제거효소의 활성을 $분석{\times}평가하였다.$ 이들 홍합의 근육중의 단백질의 향량은 남해안 양식산 홍합의 근육중의 단백질 함량이 동해안의 포항의 자연산 홍합(대조군)의 단백질 함량에 비해 4~14%나 유의적으로 감소하였다.(p<0.05-0.01).남해안의 양식산 홍합의 근육중의 수퍼옥시드 라디칼의 생성량은 대조군으로 사용한 동해안 포항의 자연산 홍합의 근육중의 수퍼옥시드 라디칼의 생성량 대비 82~138%정도나 높앗다. 또한 남해안의 양식산 홍합의 근육중의 히드록시 라디칼의 생성량은 대조군으로 사용한 동해안 포항의 자연산 홍합의 근육중의 히드록시 라디칼의 생서량 대비 9~25%정도나 높았다.남해안의 양식산 홍합의 근육중의 SOD의 활성은 동해안 포항의 자연산 홍합의 근육중의 SOD의 활성대비 16~28%정도나 낮았다. 이상의 영구결과에서 볼 때 남해안 홍합의오염 지표로서는 단백질 함량의 감소, 활성산소종의 증가 및 활성산소의 제거효소로서 SOD의 활성감소로 평가각 가능하다라는 사실을 확인할 수 있었다.
