위, 소장, 대장의 하합혈 침구자극이 Loperamide로 유발된 변비의 장관 운동성에 미치는 영향 (Effects of Acupuncture and Moxibustion at Lower Sea Points on the Intestinal Motility with Loperamide-Induced Constipation in Rats)

  • Objectives : The aim of this study was to observe effects of manual acupuncture(MA), electro-acupuncture(EA) and moxibustion (MO) on the rat with loperamide-induced constipation. Methods : This study measured small intestinal motility and number of fecal pellets in rats with loperamide induced constipation. MA and EA(intensity, 5 times muscle twitch threshold) was applied for 30 minutes and MO was treated on 5 times moxa cautery to the groups divided with age and sex. Results : The small intestinal motility was decreased by ST36 EA and ST37 EA in 5 weeks male group and ST37 EA in 7 weeks male and female groups, and ST36 MO in 7 weeks female group, but it was increased by ST39 EA in 7 weeks male group and ST37 MO and ST39 MO in 7 weeks male group. The number of fecal pellets was decreased by MA, EA, MO of ST36 in all group except 5, 7 weeks male groups only, and MA, EA of ST37 in 5, 7 weeks male group and ST37 MO in 7 weeks female group, and ST39 EA in 5 weeks male group and 7 weeks female group and ST39 MO in 7 weeks female group, but increased by ST37 EA in 5 weeks female group and ST39 EA in 7 weeks male group. Conclusions : Although these differences do not have a established tendency, it suggests that acupuncture and moxibustion are experimentally dependent upon the sex and age on intestinal motility in rats.

  • Background: Constipation is one of the most common functional gastrointestinal disorder. The present study examined the ability of water extract of fermented (FRC) and non-fermented (NFRC) roasted Cassia tora to improve intestinal function and reduce constipation in a rat constipation model. Methods and Results: Different concentration of FRC and NFRC were orally administered loperamide (5 mg/kg; LOP) reduced the number, weight, and water content of feces, as well as intestinal transit motility. However, 24 h-(24 hour fermented roasted-Cassia tora) 300 mg/kg FRC administration increased the number, weight, and water concent of feces, compared to that seen in the LOP group, and also improve intestinal transit mitility and, the thickness of distal colon and mucous fluid. Conclusions: The results of the present study indicated that LOP-induced constipation was improved by treatment with FRC. Therefore FRC could be used to develop functional foods or natural medicine for constipation. However, further study is needed to clarify how fermentation improves the medicinal properties of roasted C. tora.

  • Objectives : The purpose of this study was to observe the effects of acupuncture and electro-acupuncture(EA) at $LI_{11}$ on intestinal motility in rats. Methods : We made over-activated and suppressed state of intestinal motility with carbachol and loperamide in rat and carried out acupuncture with needle retained(NR), low frequency(2 Hz) EA and high frequency(100Hz) EA at $LI_{11}$ before or after the administration of carbachol or loperamide. The charcoal travel rate was measured to evaluate the intestinal motility. Results : 1. NR, 2Hz EA and 100Hz EA at $LI_{11}$ showed no significant influences on intestinal motility of rat in normal state. 2. All of the pre-treatment and post-treatment of NR, 2Hz EA and 100Hz EA at $LI_{11}$ showed no significant effects than control group on intestinal motility of rat which was over-activated with carbachol. 3. The pre-treatment of 2Hz EA and 100Hz EA at $LI_{11}$ on intestinal motility of rat which was suppressed with loperamide showed no significant influences but in the L-$LI_{11}$-EA(L) and L-$LI_{11}$-EA(H) groups, intestinal motility was significantly increased than L-control group. Conclusions : These results suggest that acupuncture and EA on $LI_{11}$ have preventive effect and therapeutic effect on suppressed intestinal motility, and EA is more effective than NR.

  • Objectives : The purpose of this study was to observe the effects of acupuncture and electro-acupuncture at $LI_4$ on intestinal motility in rat. Methods : We made over-activated state of intestinal motility with carbachol and suppressed state of intestinal motility with loperamide in rat and carried out needle retention acupuncture, low frequency electro-acupuncture and high frequency electro-acupuncture at $LI_4$ or sham point in rat devided into pre-treatment group and post-treatment group. and we resulted as follow for measuring charcoal travel rate to observe intestinal motility. Results : 1. Any of needle retention acupuncture, low frequency electro-acupuncture and high frequency electro-acupuncture at $LI_4$ have no significant influences on intestinal motility of rat in normal state. 2. All of the pre-treatment and post-treatment of needle retention acupuncture, low frequency electro-acupuncture and high frequency electro-acupuncture didn't significantly decrease intestinal motility in rat which over-activated with carbachol. 3. Pre-treatment of needle retention acupuncture and high frequency electro-acupuncture significantly increased intestinal motility in rat which suppressed with loperamide. 4. Post-treatment of needle retention acupuncture, low frequency electro-acupuncture and high frequency electro-acupuncture significantly increased intestinal motility in rat which suppressed with loperamide. Conclusions : These results suggest that acupuncture on $LI_4$ have preventive effect and therapeutic effect on suppressed intestinal motility, and especially high frequency electro-acupuncture is more effective.

Effect of Lactobacillus casei fermented Senna tora L. seeds and its active compound via muscarinic M3 signaling on the improvement of intestinal function in rats

  • We previously reported the potential of Senna tora L. seeds fermented by Lactobacillus casei (FSL) as a laxative agent in a loperamide-induced constipation rat model. Here, we examine the mechanism of action of FSL and its bioactive compound, revealed herein, on loperamide-induced constipation Sprague Dawley rat model. We identified the compound aurantio-obtusin (AO) using HPLC quantitative analysis. Rats were randomly assigned to six experimental groups (eight rats each)-normal and constipated groups (loperamide, FSL [100, 300, 500 mg/kg], and AO [1 mg/kg]). The FSL and AO-treated group showed an increase in the frequency, amount, and water content of feces in the constipated rat. Moreover, FSL and AO increased the intestinal transit speed in the constipated rat. Histological analysis revealed that FSL and AO recovered the intestinal mucus, the number of goblet cells, as well as thickness of the mucosa layer and muscle. Furthermore, the protein levels of the muscarinic acetylcholine receptor M3, which is involved in intestine contraction, were recovered in the FSL and AO-treated group. Its downstream signaling pathway (p-protein kinase C) was recovered by FSL and AO treatment. In conclusion, fermentation of S. tora L. seeds increases AO, which improves intestinal function, indicating that FSL is effective for treating constipation.

구멍쇠 미역의 변비 완화 효과 및 혈액지방에 미치는 영향 (Effects of Agarum cribrosum on the Improvements in Treating Constipation and Plasma lipid Profiles)

  • 구멍쇠 미역의 변비 개선 효능을 검정한 결과, 흰쥐의 사료섭취량과 체중 변화는 정상군에 비해 구멍쇠 미역 투여군이 유의적으로 식욕억제와 비만예방에 효과가 있을 것으로 사료된다. 장통과시간은 대조구 853.1분이었고, 구멍쇠 미역 5% 급이군에서 789.7분으로 감소하였다. 변비를 유발시키면서 식이섬유를 급이한 흰쥐의 체중 대비 간의 무게는 모든 실험군에서 정상군과 차이가 없었으며, 신장의 무게도 각 실험군에서 차이가 나타나지 않았다. 또한, 소장 및 대장의 길이는 변비의 유무와 식이섬유 급원에 따른 군간 유의적인 차이가 없었다. 분변량을 측정한 결과 정상군의 1일 평균 분변량은 2.84 g, 대조군은 1.34 g, 구멍쇠 미역 5% 급이군은 1.57 g, 구멍쇠 미역 10% 급이군은 2.02 g으로 구멍쇠 미역의 섭취가 변비 증상 호전 효과가 있을 것으로 판단된다. 분변의 수분함량 측정 결과, loperamide를 단독 투여한 대조군 분변의 수분함량이 정상군에 비해 유의적으로 감소하여 loperamide 투여로 인해 변비가 유발된 것을 확인하였으며, 구멍쇠미역 5% 및 10%가 함유된 사료를 급이한 군은 정상군에 비해 분변의 수분함량이 각각 1.6배 및 2.1배 증가하였다. 구멍쇠 미역을 급이한 흰쥐의 혈청 중 cholesterol 및 triglyceride 함량 변화측정 결과, 혈청 중 total cholesterol 함량은 구멍쇠 미역 5% 급이군에서 loperamide로 변비를 유발한 대조군에 비해 3.5% 감소하였으나 유의적인 차이는 없었으며, 구멍쇠 미역 10% 급이군은 40.21 mg/dL로 33.5% 감소하여 구멍쇠 미역의 섭취량이 높아질수록 흰쥐의 혈중 cholesterol 함량이 감소하였다. GOT 및 GPT는 각 실험군에서 차이가 나타나지 않았으나, BUN 농도는 대조군에 비해 구멍쇠 미역 10% 급이군에서 낮게 나타났으며, 혈중 포도당 농도는 각 실험군 간에 유의적인 차이가 없었다. 간 조직 내의 총콜레스테롤 분석 결과, 구멍 쇠 미역 급이군 모두 정상군 및 대조군에 비해 총콜레스테롤 함량이 감소하였으며, loperamide 투여에 의해 감소되었던 HDL-콜레스테롤은 구멍쇠 미역 급이 시 대조군에 비해 증가하였으나 유의적인 차이는 없었다. 또한, 중성지방 함량은 총 콜레스테롤과 마찬가지로 구멍쇠 미역 급이군이 정상군과 대조군에 비해 감소하였다. 이와 같은 결과로 보아 구멍쇠 미역은 장 이동률을 증가시켜 장운동을 촉진하리라 생각되며, 변비 해소 작용도 나타내어 장 기능 개선에 효과가 있을 것으로 판단된다.

  • Objectives : The aim of this study was to observe the effect of acupuncture and high frequency electroacupuncture at $ST_{39}$ on intestinal motility in rats. Methods: We made over-activated state of intestinal motility with carbachol and suppressed state of intestinal motility with loperamide in rat and carried out needle retention acupuncture and high frequency electroacupuncture at $ST_{39}$ or sham point in rat divided into pre-treatment group and post-treatment group. We fed charcoal to them after the treatment and measured the travel rate of charcoal in the gastrointestinal tract to observe intestinal motility. Results : Examining intestinal motility of normal rat treated with needle retention acupuncture at $ST_{39}$ significantly suppressed intestinal motility. Pre-treatment of needle retention acupuncture significantly suppressed intestinal motility in rat which over-activated with carbachol. Pre-treatment and post-treatment of high frequency electroacupuncture significantly suppressed intestinal motility in rat which over-activated with carbachol. All of the pre-treatment and post-treatment of needle retention acupuncture, high frequency electroacupuncture showed no significant effect than control group on intestinal motility of rat which was suppressed with loperamide. Conclusions : These results suggest that acupuncture and high frequency electroacupuncture at $ST_{39}$ have preventive and therapeutic effects on over-activated intestinal motility, and high frequency electroacupuncture is more effective.

장기능개선제-신소재(KTG075)의 대장관 내 mucin 2 분비에 미치는 영향 (Effect of Intestinal Function Enhancer (KTG075) on Mucin 2 Secretion)

  • 본 연구에서는 장 기능 개선 및 변비질환의 예방 및 치료에 효과적인 식물성 복합추출물인 장기능개선제-신소재(KTG075)의 대장관 내 점액질의 분비에 미치는 영향, 특히 대장에 가장 많이 분비되는 mucin 2 분비에 미치는 영향을 알아보고자 하였다. Mucin(MUC)은 그 구조에 따라서 여러 아형이 있는데, 아형에 따라서 조직 분포가 다르며 대장에서 가장 많이 분비되는 mucin의 아형은 mucin 2로서 mucin 2에 대한 항체(Biogenex AM358)를 사용하여 면역조직화학법으로 mucin 2를 관찰 시, 변비유발군에서는 mucin 2(연갈색)로 염색된 세포가 현저히 감소되나 KTG075 투여 시 뚜렷하게 mucin 2의 염색이 증가되었다. 또한 alcian blue 염색으로 점액질층을 관찰 시 점액질 두께도 변비유발군에서는 현저히 감소되었고 KTG075투여군에서는 점액질층이 거의 정상 수준으로 증가되었다. 이러한 결과는 변비유발군에서의 mucin 2의 생성과 분비가 감소되나 KTG075 투여군에서는 장 기능을 활성화시킴으로써 mucin 2의 생성과 분비를 증가시켜 장관 내 윤활도가 유지되고 장관 운동을 증가시켜 배변을 용이하게 하여 변비 또는 스트레스 등에 의해 저하된 장 기능을 개선시킴을 확인할 수 있었다.

함초 추출물의 장 기능 개선과 변비 해소 효과 (Effects of Slander Glasswort (Salicornia herbacea L.) Extract on Improvements in Bowel Function and Constipation Relief)

  • 함초 추출물의 장내 이동 효과를 살펴보기 위하여 함초 추출물을 식수에 첨가하여 장 이동률을 살펴본 결과 대조군에 비해 함초 추출물을 처리한 군에서 활성탄 식이의 장 이동률은 8.5-16.4% 증가되었다. 함초 추출물의 변비해소효과는 함초 추출물을 식수에 녹여 공급한 결과 대조군에 비해 함초 추출물을 3 mg/mL 처리한 군에서의 변량이 식수 섭취량과 관련하여 최대 156% 증가하였다. 또한 함초 요구르트의 변비해소효과는 0.25% 함초 요구르트의 공급으로 변비 유발기간 내 변량이 최대 150% 증가되었으며 처리군에 비해 loperamide를 계속 섭취함에도 불구하고 변비가 해소되었음을 알 수 있었다. 이상의 결과로 볼 때 함초 추출물은 장 이동률을 증가시켜 장운동을 촉진할 수 있으리라 생각되며 또한 변비 해소작용까지 나타내어 장 기능 개선에 효과를 나타내는 물질이라 생각된다.
