  • Ion exchange performence to remove nitrate in water studied using commercially available strong vase anin exchange resin of $Cl^{-}$ type in the batch reactors. Anion exchange resin was more effective than activated carbon or zeolite. With large resin amount or high temperature or low initial concentration, nitrate removal characteristics for a typical gel-type resin was increased. The curves showed the generally accepted selectivity sequence as ${SO_4}^{2-}>{No_3}^->NO_{2-}>{HCO_3}^-$.

  • The sustained release of piracetam by complexation with ion exchange resin was prepared. The complex was coated with Eudragit RS100 and their releases in vitro were conducted with various different kinds of medium solution. Dissolution rate increased as ionic strength, acidity and drug concentrations increased and reached its maximal plateau concentration within 5 min. Based on these data, The controlled release of piracetam using ion-exchange piracetam complex coated with Eudragit RS100 could be available.

  • A fructosyltransferase from Aureobasidium pullulans was immobilized onto a polystyrene-type anionic ion-exchange resin and the production of fructo-oligosaccharides was Investigated by the immobilized enzyme. The optimum pH and the temperature of immobilized enzyme were found to be pH 5.0, $55^{\circ}C$ respectively. The thermal stability of the enzyme was greatly enhanced after immobilization. The reaction profiles of the immobilized enzyme was almost identical to those of the free cells and the soluble enzyme. The immobilized enzymes were stable up to 20 cycles without loss of initial activity in a repeated-batch operation $50^{\circ}C$.

Simultaneous Saccharification and Extractive Fermentation for Lactic Acid Production (동시당화 및 추출발효에 의한 Lactic Acid 생산)

  • lactic acid production from cellulose by simultaneous saccharification and fermentation(SSF) was studied. The SSF using cellulase enzyme Cytolase CL and Lactobacillus delbrueckii was strongly inhibited by the end product(lactic acid). An ion-exchange resin(RA-400) was used for in-situ product removal during SSF. The sorption capacity of the resin was 200mg/g-resin. The simple SSF and the extractive SSF resulted in lactic acid concentrations of 30.4g/L and 32.0g/L, respectively, at the initial substrate concentration of 50g/L. A model was developed to simulate the extractive SSF. The lactic acid conversion for the initial substrate of 100g/L was estimated to be improved from 60% to 09% by in-situ product removal. The experimentally determined kinectic parameters were pH dependent, and fitted as empirical expressions to establish their values at different pH's. Lactic acid productivity was predicted to be maximum at pH 4.5-5.0.

  • Various methods such as lel-mtration on Sephadex G-SO, 75, 100 Sephacry, and Ultro gel, and lon-exchanle chromatoaraphy on Amberilte and carboxymethyl (CM)-cellulose, and Fast Protein liquid Chromatolraphy (FPLC) were applied for the purification of enterotoxin B from Staphylococcus aureus ATCC 14458 and compared one another. lon-exchanle chromatography on Amberllte resin was good enough to remove non-entrotoxln materials in culture, convlnient to use and fast although tbe purity was less tban 70%. However, CM-cellulose showed to be better purity and yield tban those of Amberilte resin. The yields of these two resins for ion-exchange cbromatograpby were about 70% and 75%, respectively. When the gel-filtration methods on Sepbadex G-50, 75, 100, Sepbacryl, and Ultro lei were applied, the purities were about 90%. FPLC was found to be tbe most efficient metbod in terms of purity (96%) and speed. For the purification of sample with large volume, particularly, tbe combined metbod, gel-mtration after Amberlite can be also used efficiently. Tbe purified toxin was found to be identical to type B enterotoxin used for reference standard by Oucbterlony immunodiffusion test.

Development of New Omeprazole-lon Exchange Resin Complex (이온교환수지를 이용한 새로운 오메프라졸 복합체 개발)

  • Omeprazole(OMZ)-cholestyramine(CHL) and various OMZ-Dowex resin complexes were prepared by reaction between OMZ and activated resins in 0.1N NaOH solution. And their physical properties were tested by means of infrared(IR), differential scaning caloimeter(DSC), X-ray diffraction. Chemical stability of OMZ-CHL was increased markedly compared with OMZ and the decomposition of OMZ-CHL followed the pseudo first-order kinetics and the rate constants were $2.743{\times}10^{-4}/day$ at $20^{\circ}C$, $7.83{\times}10^{-3}day^{-1}$ under 80% RH and $1.68{\times}10^{-2}day^{-1}$ under UV radiation, respectively. On the other hand, the rate constants of OMZ were $2.996{\times}10^{-4}day^{-1}$ at $20^{\circ}C$, $1.17{\times}10^{-2}day^{-1}$ under 85% RH, and $4.07{\times}10^{-2}day^{-1}$ under UV radiation, respectively. The rates of dissolution of OMZ-CHL bulk and OMZ-CHL tablet were 100% and more than 85% in 15 minutes, respectively, which were increased than OMZ base and OMZ-tablet. In the acute toxicological test, the value of oral $LD_{50}$(mouse) was 4.608 g/kg. OMZ-CHL was pelletized using lactose, polyethyle neglycol(PEG), D-sorbitol, Avicel PH 101, sodium laurylsulfate and polyvinylpyrrolidone(PVP) K-30, and enteric coated with HPMCP, Myvacet, acetone, ethanol and cetanol, of which dissolution rate was found to be more than 85% in 10 minutes. From the above results, it was found that OMZ-CHL is a useful means for development of new oral dosage forms of OMZ.
