A study on wearing practice and improvement of personal protective equipment for participant handling livestock carcass infected with virulent avian infectious agents (가금류 악성 전염병 감염축 처리자의 보호 장비 착용실태 조사 및 개선에 관한 연구)

  • This study was aimed to investigate factors that affected the status of wearing personal protective equipment (PPE) for handling poultry carcasses with potential exposure to virulent avian infectious agents. A large outbreak of high pathogenic avian influenza (HPAI) occurred in South Korea in 2014. Many public officers participated for euthanizing and handling livestock carcass. However, several safety issues with using PPE were revealed. Therefore, a questionnaire survey was conducted for 340 people who participated euthanasia and carcass disposal in the place where HPAI mainly occurred in 2014. It was found that 31.8% of the respondents had ever taken off their protective equipment during operations because of its inconvenience. The most inconvenient PPE was goggles (54.6%), followed by mask (20.2%), latex gloves (11.6%), shoe covers (5.9%) and protective clothing (5.3%). The main complaints about this individual PPE was unclear sight, damp emitted toward eye, sweating, tearing easily and sweating, respectively. Considering such problems of PPE, new possible directions for improvement of gloves and goggles were suggested. With newly developed rubber coating fabric gloves and conventionally using latex and fabric gloves, H3N2 influenza virus transmission experiment was conducted. Rubber coating fabric gloves showed similar efficiency for blocking virus transmission with latex laboratory gloves and were not easily torn by sharp claws of chicken. In addition, air flow control safety eyewear was suggested to minimize moisture formation. The air flow control system efficiently suppressed moisture formation inside the goggles. Therefore our study will provide more specific directions about new PPE development for safety protection of actual wearers.

Occurrence of postweaning multisystemic wasting syndrome in pigs in Jeju (제주지역 양돈장 자돈에서 발생한 이유후 전신성 소모성 증후군의 증례)

  • Multiplex PCR and immunohistochemistry were used to detect and differentiate between porcine circovirus (PCV) type-I and the PCV associated with postweaning multisystemic wasting syndrome (PMWS). Unique DNA product to PCV type-II was confirmed the some organs including lymph nodes, tonsil and spleen from eight pigs in Jeju by multiplex PCR. In this study, the samples were tested by a multiplex PCR assay to determine the type of PCV in each case; all cases were PCV type-II positive. PCV type-II was identified not only in typical PMWS cases, but also in field cases submitted with various clinical histories, some of which were not suggestive of PMWS. Immunohistochemically PCV type-II antigen was detected in macrophage-like cells in the tonsil, liver, lymph nodes and spleen, while some hepatocytes and renal tubular epithelial cells were also positive to the virus. This study suggested that PCV type-II is one of the causative agents of PMWS as well as the major type of PCV in the affected pigs in Jeju.

The Impact of COVID 19 on the Meat Supply Chain in the USA: A Review

  • The COVID 19 pandemic resulted in a considerable influence on the world economy. Being a big sector of the economy, the food supply chain struggled. The meat supply chain was most notably affected as every part of the supply chain from farm to shelf was closely inter-related. With the closure of businesses and restaurants the demand for at home food from grocery stores increased. Meat production facilities were impacted when the virus spread to the workers causing facilities to close or line speeds to slow. The combination of these two issues, in turn, led to there being less meat on the shelves. With less meat animals being harvested, there was less demand for livestock leading to farmers having an excess in slaughter ready animals. The decreased demand for livestock led to economic issues as money was lost in multiple sections of the supply chain. Aside from the economy and supply chain issues, other issues include concerns over the safety of meat products due to decreased safety protocols to increase line speed. Additionally, concerns of animal welfare with the excess of animals being culled were raised due to decreased capacity in processing facilities. While this review paper mainly focuses on characterizing the impact of COVID 19 on the meat supply chain in the USA, the compiled information should be able to provide practical insights to the meat/food industry across the globe to develop potential mitigating strategies against the COVID 19 and/or any similar pandemic incidences in the future.

Analysis of Nucleotide Sequence Encoding VP2 Protein of Infectious Bursal Disease Virus Detected in Korea (국내 분리 닭 전염성 F낭병 바이러스의 VP2 단백질 생산 유전자의 염기서열 분석)

  • The VP2 gene of infectious bursal disease virus (IBDV) Chinju which was previously detected in Chinju, Korea was cloned and sequenced to establish the information for the development of genetically engineered vaccines and diagnostic reagents against IBDV. The nucleotide sequence of the entire Chinju VP2 gene consisted of 1,356 bases long encoding 452 amino acids in a single open reading frame (ORF). It consisted of 368 adenine (27.1%), 363 cytosine (26.8%), 339 guanine (25.0%) and 286 thymine (21.1%) residues. The predicted $M_r$ of the Chinju VP2 protein was 48 kDa, and the protein contained 13 phosphorylation sites by protein kinase C, casein kinase II or tyrosine kinase, whereas 3 asparagine-linked glycosylation sites were recognized. The nucleotide sequence of Chinju VP2 ORF had a very close phylogenetic relationship with 98-99% homology to that of the very virulent IBDVs (vvIBDVs) HK46, OKYM, D6948, UK661, UPM97/61 and BD3/99. Also, the Chinju VP2 protein revealed a very close phylogenetic relationship with 99-100% homology to that of these vvIBDVs. The Chinju VP2 protein had 100% amino acid identity in the variable region of residues 206-360 with that of the D6948, HK46, OKYM and UK661, as well as 100% identity in two hypervariable regions of residues 212-224 and 314-324 with those of the D6948, HK46, OKYM, UK661, UPM97/61 and BD3/99. The amino acid sequence of the chinju VP2 protein contained a serine-rich heptapeptide of SWSASGS as in these vvIBDVs.

Analysis of Environmental Impacts using LCA for the Carcass Burial (전과정평가를 활용한 가축매몰지의 환경영향 분석)

  • The foot and mouth disease and AI were highly contagious. The virus can be transmitted in a number of ways, including close-contact animal to animal spread, long-distance aerosol spread and fomites, or inanimate objects, typically fodder and motor vehicles. A lot of burial sites were constructed in a short time for preventing the rapid spread of the virus. The carcass burial sites have a risk potential because the sites were constructed without any appropriate and systematic management. It resulted from lacking of time, equipments and man power. The carcass burial sites more than 4,700 constructed in 2011. Approximately 7 million poultry and 3.5 million livestock including head of cattle and swine were buried in farm land. It is time to be concerned if the secondary pollutions occur from the burial sites. The environmental impacts should be analyzed for managing the burial sites effectively and minimizing damages and risks to the environment and human health. This study was to analyze environmental impacts of the process of carcass burial construction using a life cycle assessment methodology. All input data of raw materials and energy usage were collected and the inventory was constructed. The results showed that 1 ton of carcass burial of the environmental impacts were $0.51yr^{-1}$ for ADP, 0.09 kg of 1,4DCB-eq for FAETP, 31.17 kg of $CO_2-eq$ for GWP, 0.04 kg of $C_2H_4-eq$ for POCP, 0.06 kg of $SO_2-eq$ for AP.

An Epidemiological Study on Biosecurity Practices on Commercial Pig Farms in Korea: Risk Factors for Porcine Reproductive Respiratory Syndrome Virus Infection (국내 양돈장의 차단방역 수준에 대한 역학적 연구: 돼지생식기호흡기증후군 위험요인 분석)

  • Although researches have highlighted the important role of enhanced farm biosecurity to reduce the severity and prevalence of diseases in livestock, to date there has been little study in Korea on farmers' adoption of biosecurity measures to control porcine reproductive and respiratory syndrome virus (PRRSV) infection. To mitigate the risk of PRRSV infection in pigs, the risk factors by which PRRSV is introduced in pig farms must be determined. The primary aim of this study was to investigate pig producers' perceptions about on-farm biosecurity practices. We also analyzed data obtained from a cross-sectional study on 196 farrow-to-finish farms conducted between March 2013 and February 2014 to identify risk factors for PRRSV infection at farm level. Standardized questionnaires with information about basic demographical data and management practices were collected in each farm by on-site visit of trained veterinarians. Farms were classified as negative or positive through the use of infection profiles that combined data on PCR positive pigs and serological testing including antibody titer, sero-conversion pattern at each age category, and vaccination status. Data on biosecurity practices, farm management and environmental characteristics were analyzed using multivariate ordinal logistic regression. Generally, the biosecurity level in the pig farms included in this study were insufficient to reduce/prevent the risk of PRRSV infection given the high pig density areas and the considerable extent of vehicle movement. Factors associated with PRRSV infection were those where owners used on-farm vaccination programs had a lower risk of infection (OR = 0.19, 95% CI 0.06-0.61). The results from the analysis may guide to tailor biosecurity measures in the reduction or prevention of PRRS to the specific circumstances of pig farms in different localities of the world. To the best knowledge of the authors, this is the first study to report information on the biosecurity practices currently implemented on Korean pig farms.

Bovine papillomavirus detection from bovine teats using immunohistochemistry and electronmicroscopy (면역조직화학염색 및 전자현미경검사를 이용한 소 유두로부터 소유두종바이러스 검출)

  • We examined the teats of slaughtered cattles to investigate the prevalence of papilloma and to detect bovine papilloma virus (BPV) from those samples. Among 880 heads, 432 (49.0%) was Holstein and 448 (51.0%) was Korean native cattle. Grossly, out of 432 heads (Holstein), 263 (60.8%) had papilloma lesions on teats. However, out of 448 heads (Korean native cattle), only 33 (7.4%) had papilloma lesions on teats. Immunohistochemically, BPV antigen was detected in 35 heads (22.9%) of 153 (Holstein) which had papilloma lesions. But, in Korean native cattle, BPV antigen was detected in 1 head (3.8%) of 26. Electronmicroscopically, in Holstein papilloma cases, BPV particles was detected in 31 heads (39.2%) of 79. This study revealed that papilloma was very prevalent in holstein teats, Korea and the prevalence of papilloma in Holstein was 8 times higher than that of Korean native cattle. Moreover, it was needed to consider gross and histopathological lesions to confirm BPV infection on teats because the sensitivity of immunohistochemistry and electronmicroscopy was not high.

Effect of a trivalent (FPV, FHV, FCV) inactivated vaccine in kittens (고양이 3종(FPV, FHV, FCV) 불활화 백신의 효과)

  • This study tested the effect of a trivalent (feline panleukopenia; FPV, feline viral rhinotracheitis; FHV, feline calicivirus infection; FCV) inactivated vaccine in cats. The vaccine was tested for the safety in guinea pigs, mice and cats. Also, it was tested for the efficacy in cats. The vaccine was inoculated to cats at 7~9 and 10~12 weeks of age (conventional schedule) and the serological response to vaccination was assessed and was compared to the unvaccinated group. All cats were bled by jugular venipuncture for FPV, FHV and FCV specific serological test (virus neutralizing antibody, VN) at 7~9, 10~12 and 13~15 weeks. After last bleeding, all cats were inoculated with each virus (FPV : orally $2ml\;10^{7.5}\;TCID_{50}/ml$, FHV : nasally $1ml\;10^{7.0}\;TCID_{50}/ml$ and FCV : nasally $1ml\;10^{7.0}\;TCID_{50}/ml$). The Vaccine verified excellent protective effect in guinea pigs, mice and cats. The VN antibody titers of the unvaccinated group cats against FPV, FHV and FCV were <2~16, on the other hand the vaccinated group cats were $512{\sim}{\geq}4096$, 64~1024 and 64~1024, respectively. When all cats were challenged with virulent viruses, the survival rates of the vaccinated group cats were over 80%, while the survival rates of the unvaccinated group cats were less 20%. The typical clinical signs were not observed in the vaccinated group cats, but the typical clinical signs and histopathological lesions were observed in the unvaccinated group cats. As the result of tests, the VN values obtained in this study appeared to be high enough to protect cats from viral challenges. The trivalent (FPV, FHV, and FCV) inactivated vaccine seemed to be very effective, for prevention of feline viral diseases (FPV, FHV, and FCV).

Aerodynamic Approaches for the Predition of Spread the HPAI (High Pathogenic Avian Influenza) on Aerosol (고병원성 조류인플루엔자 (HPAI)의 에어로졸을 통한 공기 전파 예측을 위한 공기유동학적 확산 모델 연구)

  • HPAI (High pathogenic avian influenza) which is a disease legally designated as an epidemic generally shows rapid spread of disease resulting in high mortality rate as well as severe economic damages. Because Korea is contiguous with China and southeast Asia where HPAI have occurred frequently, there is a high risk for HPAI outbreak. A prompt treatment against epidemics is most important for prevention of disease spread. The spread of HPAI should be considered by both direct and indirect contact as well as various spread factors including airborne spread. There are high risk of rapid propagation of HPAI flowing through the air because of collective farms mostly in Korea. Field experiments for the mechanism of disease spread have limitations such as unstable weather condition and difficulties in maintaining experimental conditions. In this study, therefore, computational fluid dynamics which has been actively used for mass transfer modeling were adapted. Korea has complex terrains and many livestock farms are located in the mountain regions. GIS numerical map was used to estimate spreads of virus attached aerosol by means of designing three dimensional complicated geometry including farm location, road network, related facilities. This can be used as back data in order to take preventive measures against HPAI occurrence and spread.

  • Simulation using the virtual reality has been applied in various fields such as exercise, education, disaster simulation training, but there is a little research on disease caused by virus transmission. In this study, we conducted simulation studies and analysis of avian influenza disaster response training. The annual avian influenza virus is recurring every year in Korea, but there are still few solutions and preventive measures for the preventing the avian influenza. The avian influenza can cause a great deal of societal harm and enormous economic damage. Prophylaxis is important because livestock epidemics, such as avian influenza and foot-and-mouth disease, have a large impact on farm households. Therefore, we proposed and analyzed contents that can be avoided through simulation of avian influenza disaster response presented in this study.
