The purpose of this study was carried out to investigate the effect of scoparone(6, 7-dimethoxyco-umarin) on liver function. Sprague-Dawley rats were treated with scoparone at a dose of 20mg/kg body weight for 5 days. Hepatic bile flow, liver weight, BSP(bromosulfophthalein) biliary excretion, alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activities, malondialdehyde production and lactate dehydrogenase(LDH) release were assayed. Among them, ALT and AST activities, malondialdehyde production and LDH release were assayed by using primary hepatocyte cultures at a concentration of 0.1mg/ml. Scoparone treatment had no effect on liver weight and hepatic bile flow. Scoparone treatment not only increased BSP biliary excretion, but also recovered the decreased BSP biliary excretion by CCl4, Also scoparone significantly decreased with the increases of ALT and AST activities, malondialdehyde production and LDH release induced by CCl4. These results suggested that scoparone could protect the liver damage by chemicals via promoting the liver excretory function.
The relationship of gonad weight and liver weight with body weight of six fish species viz; Catla catla, Labeo rohita, Cirrhina mrigala, Hypophthlamichthys molitrix, Ctenopharyngodon idella and Cyprinus carpio under the influence of artificial feed, broiler manure, buffalo manure, N:P:K (25:25:0) and a control pond were examined after a rearing period of one year. The positive relationship between gonad weight and body weight was significant which showed the dependence of gonadal development on body weight in all the six fish species. The currelation coefficients were higher in female fishes. However, the major carps had a much smaller proportional gonadal weight as compared with Chinese carps and a common carp due to their faster growth rate. The overall comparison of six fish species under different experimental treatments revealed that highest liver weight was observed for C. idella closely followed by C. carpio. The maximum correlation value was observed with H. molitrix under the broiler manure. The maximum Gonadosomatic Index (GSI) remained as 32.63 for C. carpio followed by C. idella. The maximum value for Hepatosomatic Index (HSI). remained 1.99 for C. idella followed by C. mrigala.

Ahnak, a large protein first identified as an inhibitor of TGF-β signaling in human neuroblastoma, was recently shown to promote TGF-β in some cancers. The TGF-β signaling pathway regulates cell growth, various biological functions, and cancer growth and metastasis. In this study, we used Ahnak knockout (KO) mice that underwent a 70% partial hepatectomy (PH) to investigate the function of Ahnak in TGF-β signaling during liver regeneration. At the indicated time points after PH, we analyzed the mRNA and protein expression of the TGF -β/Smad signaling pathway and cell cycle-related factors, evaluated the cell cycle through proliferating cell nuclear antigen (PCNA) immunostaining, analyzed the mitotic index by hematoxylin and eosin staining. We also measured the ratio of liver tissue weight to body weight. Activation of TGF-β signaling was confirmed by analyzing the levels of phospho-Smad 2 and 3 in the liver at the indicated time points after PH and was lower in Ahnak KO mice than in WT mice. The expression levels of cyclin B1, D1, and E1; proteins in the Rb/E2F transcriptional pathway, which regulates the cell cycle; and the numbers of PCNA-positive cells were increased in Ahnak KO mice and showed tendencies opposite that of TGF-β expression. During postoperative regeneration, the liver weight to body weight ratio tended to increase faster in Ahnak KO mice. However, 7 days after PH, both groups of mice showed similar rates of regeneration, following which their active regeneration stopped. Analysis of hepatocytes undergoing mitosis showed that there were more mitotic cells in Ahnak KO mice, consistent with the weight ratio. Our findings suggest that Ahnak enhances TGF-β signaling during postoperative liver regeneration, resulting in cell cycle disruption; this highlights a novel role of Ahnak in liver regeneration. These results provide new insight into liver regeneration and potential treatment targets for liver diseases that require surgical treatment.
The purpose of this experiment was to compare the effects of tryptophan administration on nutritional status of female rats which consumed reserpine and 6% casein diet with different carbohydrate contents(87%, 65%, 44% respective). Final body weight, body weight gain, FER, plasma amino acid concentration and microsomal cytochrome P 450 content in liver were measured and microscopic structure of hepatocytes was observed. In low-protein diet, the higher the carbohydrate content of diet was, the lower the damage was in the rat's liver. Tryptophan administration after dose of reserpine induced more effective recovery from liver damage of rats in high carbohydrate diet group than that in low carbohydrate diet group. In conclusion, the general nutritional assessments such as final body weight and body weight gain provided better estimate of the degree of structural changes in hepatocytes than functional assessment such as plasma amino acid concentration or liver microsomal cytochrome P450.
This study was performed to investigate the effect of chitosan and NOCC from different sources and of different molecular weights on cadmium toxicity. Sprague-Dawley rats were blocked into 26 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400 ppm in diet. Various forms of chitosan and NOCC were given at the level of 0 or 4%(w/w) of diet. Cd toxicity was alleviated by various chitosan and NOCC supplements. However, the alleviating effects were different with fiber source(crab and shrimp), type(chitosan and NOCC), and molecular weight (low, medium, and high). Molecular weight had no significant effect. Compared with shrimp-source-fiber-fed groups, crab-source-fiber-fed groups showed lower Cd concentrations in blood and kidney, lower kidney metallothionein concentration, and lower liver and kidney MT-Cd contents. Compared with NOCC -fed groups, chitosan-fed groups showed lower intestine and liver Cd concentrations, lower liver and kidney MT concentrations and MT-Cd content, and higher fecal Cd excretion. Among cadmium-exposed group, low molecular weight shrimp chitosan group showed low Cd concentrations of liver and kidney, high fecal Cd excretions, and the lowest intestine and liver MT concentrations and liver MT-Cd content. In summary, a crab source fiber was more effective than shrimp source fiber, chitosan was more effective than NOCC, and the most effective one in alleviating Cd toxicity was low molecular weight shrimp chitosan.
Objectives : This study was performed to prove the preventive effect of water extracts from Artemisiae Iwayomogi Herba on hyperlipiderma and liver damage induced alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And Artemisiae Iwayomogi Herba extract was administrated for the same period. We measured the serum component in rat's blood. body weight and weight of liver. Result : At first, we observed preventive effects of Artemisiae Iwayomogi Herba on hyperlipiderma induced by alcohol. Artemisiae Iwayomogi Herba group showed significant decrease of total cholesterol and triglyceride in comparison with those of the control group. Artemisiae Iwayomogi Herba group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. At second, we observed preventive effects of Artemisiae Iwayomogi Herba on liver damage induced by alcohol. Artemisiae Iwayomogi Herba group showed significant decrease of GOT, ALP and LDH in comparison with those of the control group. Artemisiae Iwayomogi Herba group showed significant increase of liver weight in comparison with those of the control group. Conclusion : Reviewing these experimental results, it appears that water extracts from Artemisiae Iwayomogi Herba have pharmaceutical preventive efficacy on hyperlipiderma and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of Artemisiae Iwayomogi Herba.
Objectives: The aim of this report is to suggest that severe weight loss diagnosed with a pattern of liver-spleen disharmony can be effectively treated with Korean Medicine (acupuncture and herbal medication). Methods: This is a clinical case report of a patient with severe weight loss diagnosed with a pattern of liver-spleen disharmony. The patient was treated with Samryungbeakchul-san (Canlingbaizhu-san), Hyangsayangyi-tang (Xiangshayangwei-tang), and Haewoolhwajoong-tang (Jieyuhezhong-tang) two to three times a day and with acupuncture and moxibustion two to three times a week. Results: After the short-term treatments described above, the patient was gradually recovering from severe weight loss and many accompanying symptoms, such as diarrhea, insomnia, insufficient food intake, and others. Conclusions: The results show the possibility that severe weight loss diagnosed with a pattern of liver-spleen disharmony could be effectively treated with Korean Medicine (acupuncture and herbal medication).
Goose fatty liver is one of the most delicious and popular foods in the world, but there is no reliable genetic marker for the early selection and breeding of geese with good liver-producing potential. In our study, one hundred and twenty-four 78-day-old Landes geese bred in Shunda Landes goose breeding farm, Jiutai, Jilin, China were selected randomly. The fatty livers were sampled each week after overfeeding during a three week period. Polymerase chain reaction-single strand conformation polymorphism and DNA sequencing were used to identify single nucleotide polymorphisms (SNPs) of fatty acid synthase (FAS), which is an important enzyme involved in the synthesis of fat under both physiological and pathological conditions. Least-squares correlation was established between these SNPs and fatty liver weight, abdominal fat weight, and intestinal fat weight of the overfed Landes geese, respectively. The results showed that fatty liver weight of geese with EF and FF genotypes (amplified by primer P1) was significantly higher than that of the EE genotype (p<0.05), and liver weight of CD and DD genotypes (amplified by primer P2) was significantly higher than that of the CC genotype (p<0.05). Different genotype combinations showed different liver weights, and from highest to lowest were ABDD, DDEF, DDFF, DDEE, ABEF, ABFF, AADD, and CDEF. Further analysis of DNA sequencing showed that there were two SNPs within the 5' promoter region the FAS gene. The geese of EF and FF genotypes carried a change of T to C, and the geese of CD and DD genotypes carried a change of A to G. The changes of the bases could potentially influence the binding of some transcription factors to this region as to regulate FAS gene. To our knowledge, this is the first report of SNPs found within the 5' promoter region of the Landes goose FAS gene, and our data will provide an insight for early selection of geese for liver production.
Experiment was performed to investigate the immunotoxicity of cadmium administered orally and the effect of ginseng petroleum ether fraction on it. Mice were given 3, 30, or 300 ppm cadmium as cadmium chloride orally in the drinking water and injection of ginseng petroleum ether fraction intraperitoneally for 4 weeks. Mice were sensitized and challenged with sheep red blood cells (S-RBC). Humoral immune response was evaluated by antibody production and Arthus reaction. Body weight gain, spleen weight, thymus weight, and liver weight were also measured. In the present study, cadmium generally suppressed the humoral immunity. There was decrease in the rate of body weight gain and liver weight to body weight by cadmium-administration. Ginseng petroleum ether fraction showed restoring effect, to some extent, on the decrease in PFC and in the rate of liver weight to body weight by cadmium-administration. But it more suppressed Arthus reaction.
The main experiments was investigated the skin tissue damage changing for the skin bum having influence on the skin and the liver and also observed the radical liver weight, ALT in the serum, the fluctuating of AST for the skin bum causing to the liver damage. Anatomically the edema formation of skin after thermal injury was showed, and skin bum increased liver weight (% of body weight, p<0.05) and the activity of serum aniline aminotrasferase (p<0.05), and also histologically induced wes of epidermal layer, protein degeneration of connective tissue, local hemorrhage and degeneration of glandular epithelium in the skin tissue. Liver tissue showed the evidences of postbum damage, they were sinusoidal dilatation, cell swelling, infiltration of inflammatory cells.
