ZNF552, a novel human KRAB/C2H2 zinc finger protein, inhibits AP-1- and SRE-mediated transcriptional activity

  • In this study, we report the identification and characterization of a novel C2H2 zinc finger protein, ZNF552, from a human embryonic heart cDNA library. ZNF552 is composed of three exons and two introns and maps to chromosome 19q13.43. The cDNA of ZNF552 is 2.3 kb, encoding 407 amino acids with an amino-terminal KRAB domain and seven carboxyl-terminal C2H2 zinc finger motifs in the nucleus and cytoplasm. Northern blotting analysis indicated that a 2.3 kb transcript specific for ZNF552 was expressed in liver, lung, spleen, testis and kidney, especially with a higher level in the lung and testis in human adult tissues. Reporter gene assays showed that ZNF552 was a transcriptional repressor, and overexpression of ZNF552 in the COS-7 cells inhibited the transcriptional activities of AP-1 and SRE, which could be relieved through RNAi analysis. Deletion studies showed that the KRAB domain of ZNF552 may be involved in this inhibition.

  • 동자개, Pseudobagrus fulvidraco (Richardson)의 명확한 생식년주기를 밝히기 위하여, 본 종을 1992년 5월부터 1993년 6월까지 전라남도 영산강 상류로부터 채집하여 암수의 GSI, HSI, 비만도, 난모세포 발달 단계의 분포빈도, 그리고 생식소와 간의 조직학적인 주년 변화를 조사하였다. 이들 결과를 토대로 하여 생식년주기를 다음과 같은 5단계로 구분 할 수 있었다. 1) 성장기 (4월$\~$5월초): 암컷의 GSI값이 증가하고 주변인기 난모세포들이 점진적으로 성장하였으며, 수컷의 세정관 상피에서는 정원세포들이 활발히 분화되기 시작하였다. 2) 성숙기 (5월$\~$6월초): 암수 모두 GSI가 빠르게 증가하고 여러 단계의 난모세포들이 관찰되었으며, 정소 내에는 정자 형성이 활발하게 일어났다. 3) 성숙 및 산란기 (6월$\~$8월): 암컷의 GSI는 높은 값을 유지하였고, 난모세포들은 많은 양의 난황을 축적하였으며, 수컷의 세정관에는 대부분 정자로 채워져 있었다. 4) 퇴화기 (9월$\~$11월): 암수 GSI는 감소되고, 난소 내에는 대부분 주변인기 세포들로 채워져 있고, 간세포에는 지방구들이 많이 축적되었다. 5) 휴지기 (12월$\~$다음해 3월): 암수 GSI는 낮은 값을 유지하며, 난소 내의 주변인기 세포의 증가와 정자 형성은 보이지 않았다.

넙치(Paralichthys olivaceus) HSP90$\beta$ 유전자의 분자생물학적 연구 (Molecular Biological Studies on the Stress Protein HSP90$\beta$ Gene from Flounder (Paralichthys olivaceus))

  • 열 충격단백질(Heat shock protein : HSP)은 온도 스트레스에 대하여 세포 내에서 발현되는 단백질이다. HSP의 중요 분류군의 하나가 HSP90 family 이다. 여러 종류의 포유동물과 조류에서 HSP 유전자 특성에 대한 연구가 많이 진행되었다. 본 연구에서는 넙치(Paralichthys olivaceus)로부터 제조한 넙치 뇌 cDNA 유전자 은행을 이용하여 넙치 HSP90 cDNA 유전자를 분리하여 구성 염기서열의 특성을 밝혀 내었다. 염기서열의 분석결과 넙치의 hsp90$\beta$ 유전자는 2,791 개의 뉴클레오타이드로 구성되어 있고, 726개의 아미노산 잔기가 암호화되어 있었다. 넙치 hsp90$\beta$ 유전자는 European sea bass와 96.6% zebrafish와 92.9%, Atlantic salmon와 92.0%, 그리고 사람과는 89.5%의 염기서열 상동성을 지니고 있었다. 또한 HSP90 아미노산 서열을 바탕으로 척추동물 종들과의 진화계통수를 구축하였다. 넙치 hsp90$\beta$ 유전자의 mRNA의 분포 정도를 RT-PCR를 이용하여 조사하였다. hsp90$\beta$ 유전자는 조사한 모든 조직(뇌, 간, 신장, 근육, 비장)에서 높은 수준으로 발현이 되고 있었다. 또한, 넙치 hsp90$\beta$ 단백질을 대량발현하기 위하여 대장균에서 발현을 유도하였다.

감초에서 분리된 데하드로글라이아스페린 C에 의한 마우스 모델계에서 quinone reductase 활성의 조절 (Regulation of Quinone Reductase Activity in Mice by Dehydroglyasperin C Isolated from Licorice)

  • 예로부터 다양한 방면에서 약용식물로 널리 사용되어 온 감초에서 분리한 dehydroglyasperin C (DGC)는 이전의 연구에서 세포 모델계에서 비교적 높은 항산화 능력과 2상 해독효소계 활성을 유도하는 것이 확인되었으나, 동물 모델계에서 DGC의 항산화능과 2상 해독효소계를 평가하였을 때, 유의적 결과가 관찰되지 않았다. 따라서 DGC 처리시간에 따라 마우스의 장기 및 혈장에 어떠한 영향을 미치는지를 알아보고자 0, 2, 4, 6, 8, 12, 24 시간 동안 DGC를 처리한 후, 장기별 QR 유도활성과 간과 신장에서의 단백질 발현 패턴, 혈장의 항산화능력을 측정하였다. 그 결과 DGC 처리에 의한 QR 유도활성은 위, 대장에서 시간에 따라 변화하는 경향을 보였고, 간, 신장, 소장, 폐에서는 큰 경향성이 나타나지 않았으며, 2상 해독효소의 단백질 발현 패턴은 간에서는 역시 큰 경향성이 나타나지 않았고 위에서는 QR 유도활성과 유사한 경향이 나타남을 확인하였다. 혈장의 DGC 처리 시간에 따른 항산화활성은 시간에 따라 유의적으로 값이 증가한 것을 확인할 수 있었다. 결론적으로 DGC는 처리 시간에 따라 각 장기 및 혈장에 각기 다르게 영향을 미치는 것으로 사료되며, 앞으로도 추가적인 연구를 통하여 DGC의 효능을 보다 구체적으로 검증하는 것이 필요한 것으로 생각된다.

늑막강내 저류액의 임상적 고찰 (Clinical Evaluation of Pleural Fluid)

  • Author made a clinical study of 248 cases of pleural effusion patients who were diagnosed and treated at departments of chest surgery and internal medicine, Pusan National University Hospital, during the period from Jan. 1983 to Dec. 1985. The age distribution ranged from 1 to 76 years old and the ratio of male to female was 1.38:1. The cardinal symptoms were chest pain[69.4%], dyspnea[66.1%], cough[57.7%], fever[37.1%], sputum[26.2%], general malais[13.7%] and cyanosis[1.6%] in this order. The causes of pleural effusion were pulmonary tuberculosis[42.4%], pneumonia[23.0%], malignancy[16.5%], congestive heart failure[9.3%], liver cirrhosis[2.8%] and nephrosis[2.0%] in this order. The protein in the pleural effusions was 1.61*0.90[mean*SD] gm% in transudate and 5.05*1.10[Mean*SD] gm% in exudate. In 34 cases[89.5%]out of 38 transudates, the protein was under 3 gm% and in 201 cases [95.7%] out of 210 exudates, the protein was over 3 gm%. The protein ratio of pleural effusion to serum was 0.2650.11[Mean LSD] in transudates and 0.73*0.12[Mean LSD] in exudate. The ratio under 0.5 was in 36 cases[94.8%] out of 38 transudates and over 0.5 was in 206 cases[98.1%] out of 210 exudates. The LDH in the pleural effusion was 114.7550.3[mean*SD] units / ml in transudate and 627.05325.9[mean*SD] units / ml in exudate. The LDH less than 200 units / ml was in 36 cases[94.6%] out of 38 transudates and more than 200 units / ml was in 199 cases[94.7%] out of 210 exudates. The LDH ratio of pleural effusion to serum was 0.34k 0.11[mean*SD] in transudate and 1.15*1.12[mean*SD] in exudate. The LDH ratio of pleural effusion to serum was less than 0.6 in 36 cases[94.8%]out of 38 transudates and more than 0.6 in 200 cases[95.2%] out of 210 exudates. Etiologic organisms were confirmed in 78 cases[48.1%] among the requested 162 cases. In the 78 cases of etiologic organisms, staphylococcus was 33 cases[20.3%], streptococcus 24 cases[14.8%], Klebsiella pneumonia 7 cases[4.3%], pseudomonas 6 cases[3.7%], E. coli[3.1%], enterobacter 3 cases[1.9%]. 43 patient of pleural effusion from malignancy were undergone three or more thoracenteses. In 13 cases[31.7%], three specimen were negative and in 7 cases[17.1%], three specimens were positive for malignancy. In the remaining of 21 cases[51.2%], malignant cells were found in one or more of the specimens but not in all. Methods of treatment of pleural effusion by closed thoracotomy was 188 cases[75.8%], thoracentesis 27 cases[10.9%], decortication 16 cases[6.5%], thoracoplasty 6 cases[2.4%] and decortication with thoracoplasty 3 cases[1.2%].

$^{99m}Tc-HMPAO$를 이용한 자가백혈구표지 및 그를 이용한 염증병소의 스캔 (Inflammation Scan Using $^{99m}Tc-HMPAO$ Labelled Leukocytes)

  • Inflammation scan using radiolabelled leukocytes has high sensitivity and specificity. Several methods for labelling leukocytes have been evaluated using P-32 diisopropyl fluorophosphate (DFP-32), H-3 thymidine, Cr-51 chromate, Ga-67 citrate and Tc-99m-sulfur colloid. In-111-oxine has proved so far to be the most reliable agent for labelling leukocytes. In-111-oxine is, however, expensive, not easily available when needed, and its radiation dose to leukocytes is relatively high. Moreover, resolution of the resultant image is relatively poor. Tc-99m is still the agent of choice because of, as compared with the indium, its favorable physical characteristics, lower cost and availability. Now the technique for labelling the leukocytes with technetium is successfully obtained using the lipophilic HAPAO with higher efficiency for granulocytes than for other cells. With this technique it is possible to label leukocytes in plasma to improve the viability of the leukocytes. Inflammation scan using Tc-99m-HMPAO has been evaluated in several laboratories, and difference in methods for separation and labelling accounts for difference in efficiency, viability and biodistribution of the labelled leukocytes. We performed inflammation scan using leukocytes labelled with Tc-99m-HMPAO in three dogs 24 hours after inoculation of live E. Coli and A. Aureus in their right abdominal wall. We separated mixed leukocytes by simple sedimentation using 6% hetastarch (HES) and labelled the leukocytes with Tc-99m-HMPAO in 20% cell free plama diluted with phosphate buffer solution(Fig. 1). Uptake was high in the liver and spleen but is was minimal in the lungs on whole body scan. Kidneys and intestine showed minimal activity although it was high in the urinary bladder(Fig. 2). Uptake of labelled leukocytes in the inflammation site was do(mite on 2 hour-postinjection scan and abscess was clearly delineated on 24 hour-delayed scan with high target-to-nontarget ratio(Fig. 3, 4). Inflammation scan using mixed leukocytes labelled with Tc-99m-HMPAO is very sensitive and specific in early detection of inflammation.

The Beneficial Effects of Ferulic Acid supplementation during In Vitro Maturation of Porcine Oocytes on Their Parthenogenetic Development

  • Ferulic Acid (FA) is a metabolite of phenylalanine and tyrosine, a phenolic compound commonly found in fruits and vegetables. Several studies have shown that FA has various functions such as antioxidant effect, prevention of cell damage from irradiation, protection from cell damage caused by oxygen deficiency, anti-inflammatory action, anti-aging action, liver protective effect and anti-cancer action. In this study, we investigated the maturation rate, intracellular glutathione (GSH) and reactive oxygen species (ROS) of porcine oocytes by adding FA to the in vitro maturation (IVM) medium and examined subsequent embryonic developmental competence at 5% oxygen through parthenogenesis. There is no significant difference between the control group ($0{\mu}M$) and treatment groups ($5{\mu}M$, $10{\mu}M$, $20{\mu}M$) on maturation rates. Intracellular GSH levels in oocyte treated with $5{\mu}M$ of FA significantly increased (P < 0.05), and $20{\mu}M$ of FA revealed significant decrease (P < 0.05) in intracellular ROS levels compared with the control group. Oocytes treated with FA exhibited significantly higher cleavage rates (79.01% vs 89.19%, 92.20%, 90.89%, respectively) than the control group. Oocytes treated with $10{\mu}M$ showed significantly higher blastocyst formation rates (28.3% vs 40.3%, respectively) after PA than the control group. Total cell numbers in blastocyst of $10{\mu}M$ FA displayed significantly higher (39.4 vs 51.9, respectively) than the control group. In conclusion, these results suggested that treatment with FA during IVM improved the developmental potential of porcine embryos by increasing intracellular GSH synthesis and reducing ROS levels. Also, there was an improvement of cleavage rate, blastocyst formation and total cell numbers in blastocysts. It might be associated with Keap1-Nrf2 pathway as an antioxidant regulate pathway that plays a crucial role in determining the sensitivity of cells to oxidative damages by regulating the basal and inducible expression of enzymes which is related to detoxification and anti-oxidative effects, stress response enzymes and/or proteins and ABC transporters.

실험적으로 기니픽에 유발시킨 렙토스피라병에 대한 미생물학적 및 병리학적 연구 (Microbiological and Pathological Features of Experimentally Induced Leptospirosis in Guinea Pigs)

  • To understand the pathogenesis of anicteric leptospirosis with severe pulmonary hemorrhage occured in Korea, the microbiological and pathological features were observed in the experimentally induced leptospirosis in guinea pigs infected with a virulent strain of Leptospira interrogans isolated from the patient at Wonju, Korea, and the results are summarized as follows. 1. The main pathological features were widespread hemorrhages, especially affecting lung, skeletal muscles, retroperitoneal and perirenal adipose tissues. The hemorrhages accompanied inflammatory process especially of vasculitic pattern as well as occasional coagulation necrosis in the liver, skeletal muscle, and myocardium. The main inflammatory cells were of plasma cell even in the fairly early stage of the infection. 2. Those pathologic changes were more exaggerated in the inoculation site. 3. Within 144 hours of infection, the longer the infection time, the more antigens were observed in the tissues, and the severer the pathologic changes. 4. Leptospiral antigens were detected at first by indirect immunofluorescent and immunoperoxidase technics. As the infection time extended, the antigens were observed in all of the tissues examined except in the skeletal muscle. The shape of the antigens was spiral or thread-like within 72 hours of infection. As the infection progressed, they became fragmented and granular. 5. Leptospires were detected in the blood within 144 hours of infection by darkfield microscopic examination. Thereafter, none was observed. 6. Antibody to leptospires were detected as early as 72 hours of infection. In summary, the virulent strain of L. interrogans used in this experiment induced widespread hemorrhages with inflammatory reaction especially in lung, skeletal muscles, and retroperitoneal adipose tissue. With these findings, it is suggested that the direct toxic effect of leptospires might playa great role in the pathogenesis of this infection.

복부 고형장기 손상을 동반한 안정 골반골 골절의 특성 (Characteristics of Stable Pelvic Bone Fractures with Intra-abdominal Solid Organ Injury)

  • Purpose: This study analyzed the characteristics of stable pelvic bone fractures with intra-abdominal solid organ injury. Methods: Medical records were retrospectively reviewed from January 2000 to December 2009 of patients with stable pelvic bone fractures. A stable pelvic bone fracture according to Young's classification is defined as a lateral compression type I and antero-posterior compression type I. Subjects were divided into two groups, one with (injured group) and one without (non-injured group) intra-abdominal solid organ injury, to evaluate the dependences of the characteristics on the presence of an intra-abdominal solid organ injury. Data including demographics, mechanism of injury, initial hemodynamic status, laboratory results, Revised Trauma Score (RTS), Abbreviated Injury Scale (AIS), Injury Severity Score (ISS), amount of transfusion, admission to intensive care unit (ICU), and mortality were analyzed. Results: The subjects were 128 patients with a mean age of 42 years old, of whom were 67 male patients (52.3%). The injured group had 21 patients(16.4%), and the most frequent injured solid organ was the liver. Traffic accident was the most common mechanism of injury and lateral compression was the most common type of fracture in all groups. Initial systolic blood pressure was lower in the injured group, and the ISS was greater in the injured group. Arterial pH was lower in the injured group, and shock within 24 hours after arrival at the emergency department was more frequent in the injured group. Transfused packed red blood cells within 24 hours were 8 patients(38.1%) in the injured group and 11 patients(10.3%) in the non-injured group. Conservative treatment was the most common therapeutic modality in all groups. Stay in the ICU was longer in the injured group, and three mortalities occurred. Conclusion: There is a need to decide on a diagnostic and therapeutic plan regarding the possibility of intra-abdominal solid organ injury for hemodynamically unstable patients with stable pelvic bone fractures and for patients with stable pelvic bone fractures along with multiple associated injuries.

에볼라 바이러스 진단법과 개발 동향에 관한 고찰 연구 (Study on Laboratory Diagnosis of the Ebola Virus and Its Current Trends)

  • 2013년 12월 말, 에볼라 바이러스는 서아프리카에서 발발했다. 기니를 시작으로 라이베리아, 시에라리온으로 빠르게 퍼지게 되었다. 에볼라 바이러스(자이르형 에볼라 바이러스)는 외피, 비분절, 음성단일가닥 RNA바이러스이다. 에볼라 바이러스는 인수공통 전염병이다. 바이러스는 처음 감염된 야생동물과 접촉 한 후, 혈액, 땀, 소변, 정액, 모유 등의 체액과의 직접적인 접촉을 통해 사람 대 사람으로 전염된다. 그러나 공기로 전염되지는 않는다. 잠복기는 2~21일이다. 에볼라 바이러스는 내피세포, 단핵 식세포, 간세포를 감염시킨다. 감염 후 바이러스가 숙주의 면역 시스템을 회피하기 위해 여러 메커니즘을 사용한다. 이것은 혈관, 간 등의 내부 조직 및 기관에 막대한 피해를 주어 죽음에 이르게 한다. RNA 바이러스에 대한 대부분의 실험은 역전사 중합효소 연쇄반응(RT-PCR)라 기술에 의존한다. 이 방법은 매우 민감하지만 숙련된 과학자, 전원 공급 장치 요구하며 비싸다. 스트립 분석기법(효소면역분석법, ELISA)은 에볼라 바이러스 항원 또는 항체를 검출한다. 이 기법은 저렴하며, 전기, 냉장 장치가 필요하지 않다. 실험적인 치료 및 백신 개발에 관한 지속적인 노력에도 불구하고, 에볼라 바이러스 질환은 현재 치료법에 제한이 있다. 그러므로 신속하고 정확한 진단이 환자관리, 감염예방, 관리대책에 있어서 매우 중요하다.
